Protein targeting towards the thylakoid lumen of chloroplasts: proper localization of fusion proteins is only observed in vivo.

Routeing of fusion proteins to the thylakoid lumen of the chloroplast was compared in vitro and in vivo. The Escherichia coli protein beta-lactamase was used as a passenger to study this intraorganellar sorting process. The first step, translocation of beta-lactamase into the chloroplast stroma, occurs properly both in vitro and in vivo and is dependent on the presence of a transit peptide in the protein construct. The second step, targeting towards the thylakoid lumen, is more complicated as was also observed previously when other passenger proteins were used. In vitro, the presence of a thylakoid transfer domain is not enough for routeing and proper processing. Only when the complete thylakoid lumen precursor plastocyanin was fused to beta-lactamase was the fusion protein processed adequately, but routeing was still incomplete. However, in vivo, the information present in the thylakoid transfer domain was the only requirement for proper transport towards the thylakoid lumen. These data show that in vivo, the only requirement for targeting of passenger proteins towards the thylakoid lumen is the presence of a transit peptide and a thylakoid transfer domain. Furthermore, we demonstrate that the in vitro import system does not necessarily reflect the in vivo situation with respect to intraorganellar sorting.

Factors affecting dystocia and birth weight in Grati cattle in Pujon, East Java.

This paper discusses factors affecting dystocia and birth weight in Grati cattle in three villages in the Pujon district, East Java. Records of birth weight, dystocia score, village, season, parity, sex, artificial versus natural service, and age and weight of dam, were obtained from 268 parturitions. Village and weight of the dam had a significant effect on birth weight but not on dystocia. Parity had a significant effect on dystocia score. More abnormal births occurred with AI using exotic bulls. Calves from an abnormal birth were on average more than three kg heavier. First parity births combined with Al with exotic bulls had a particularly high incidence of dystocia. It is concluded that farmers in Pujon can reduce the incidence of dystocia in heifers by using local bulls only.

The effect of dantrolene sodium in relation to blood levels in spastic patients after prolonged administration.

In 25 patients with spasticity, pharmacokinetics and effects of dantrolene sodium were investigated after prolonged administration. A beneficial effect occurred in seven patients. The results were better on 100 mg daily than on a higher daily dose. An increase of the daily dose from 200 to 400 mg was not associated with higher blood levels. Many side effects were noted such as: anorexia, nausea, drowsiness, depression and muscle weakness. From this study we conclude that dantrolene sodium is a muscle relaxant with a weak to moderate effect in patients with spasticity; the effect at doses higher than 200 mg daily is probably poor.

Further analysis of the type differences of rat-liver mitochondrial DNA.

1. The sequence of the small Hind III fragment F of rat-liver mitochondrial DNA (mtDNA) type A and type B was determined in order to investigate the nature of the differences between the two types of mtDNA and to determine its position in the Hind III fragment map. 2. The three differences found were point mutations. No deletion or insertion and no modification was observed. Two of the three differences affect the sequences which are recognition sites for Eco RI, Alu I and Taq I in type A, but not in type B, mtDNA. 3. The presence of an Eco RI restriction site only within the Hind III fragment F of type A shows that the fragment is situated in between the Hind III fragments A and E. 4. In one of the six reading frames, the Hind III fragment F contains the code for a carboxyl-terminal end of a polypeptide in which the three mutations do not lead to alterations in the possible aminoacid sequence. 5. The restriction sites for Taq I and a number of the sites for Alu I and Hae III were mapped. 6. The positions of the Hap II fragment J, and of a Hind III fragment G on the mtDNA were determined.

Studies on the function of two adjacent N6,N6-dimethyladenosines near the 3' end of 16S ribosomal RNA of Escherichia coli. IV. The effect of the methylgroups on ribosomal subunit interaction.

The effect of the presence or absence of the methylgroups of the m2(6)Am2(6)A sequence near the 3' end of 16S rRNA of Escherichia coli on the interaction of the ribosomal subunits has been studied, using wild-type (methylated) and mutant (unmethylated) ribosomes. Subunit exchange experiments and competitive association experiments show a strong preference of the 50S subunit for association with methylated 30S subunits. The results indicate that the equilibrium constant of the reaction 70S in equilibrium with 30S + 50S is dependent on the methylgroups; mutant 30S.50S couples are less stable than wild-type 30S.50S couples. It is postulated that the methylgroups also stimulate the interaction between 30S subunits and initiation factor IF-3.

Effect of litter size at birth on lactation in mice.

The relative influence of litter size at birth and dam body weight at parturition on lactational performance was determined with five among-line crossfostering experiments involving 17 populations of mice. Included were lines selected for postweaning growth and maternal performance, and unselected controls. Twelve-day litter weight of a crossfostered litter standardized to six or eight pups was used as the criterion of lactational performance. Litter size at birth had a small positive correlation with 12-day litter weight when pooled within lines in each crossfostering experiment (.06 to .30, with mean of .17). The correlation between dam body weight at parturition and her crossfostered 12-day litter weight was consistently higher (.13 to .51, with mean of .35). Litter size was positively correlated with dam body weight at parturition (.33 to .56, with mean of .47). The partial correlation between litter size and 12-day litter weight, with dam body weight at parturition held constant, was reduced (-.07 to .18, with mean of .02) compared to the simple correlation. Multiple regression analyses indicated that dam body weight at parturition consistently accounted for more of the variation in 12-day litter weight than litter size. The data provide no evidence to support the hypothesis that increased fetal number or mass results in a greater lactational output by the dam.

To obtain more information about the arrangement of Hind III restriction fragments in the tRNA-rRNA region of the Neurospora crassa mitochondrial (mt) DNA we have cleaved the mtDNA with Hpa I and Hind II. We could construct additional cleavage maps for these enzymes. Hybridization of rRNAs to Hind II fragments confirmed the existence of an intervening region of about 2,300 basepairs in the 24S rRNA (Hahn et al., Cell, in press). About seven tRNA genes, among which the genes for tRNA1Ser and tRNAMetM, are located in a segment of about 5,000 bp separating the 24S and 17S rRNA genes. Another cluster of 14 tRNA genes is found adjacent to the other end of the 24S gene. The genes for tRNALeu1 and tRNAMetF are located in this cluster.

The heterogeneity of rat-liver mitochondrial DNA.

Two types of mitochondrial DNA (mtDNA) can be distinquished in an inbred strain of rats of the Wistar type. The population of DNA molecules of the liver of one single rat is homogeneous. This was shown for a number of 100 animals and confirms the data of other investigators. The two types of mitochondrial DNA, designated A and B, differ in their number of cleavage sites for the restriction endonucleases Eco RI (2sites), Hind II (1 site) and Hha I (1 site). No differences were found for the restriction enzymes Bam HI, Hap II, Hind III and Hpa I. The degree of sequence divergence of the two types of DNA is calculated to be roughly 5% on the basis of these observations. From 20 rats part of the liver was taken and the mtDNA was characterized. Heterologous and homologous crosses between type A and type B rats were made. Analysis of the offspring revealed strictly maternal inheritance of the A and B mtDNA traits. For purposes of base-sequence analysis and RNA.DNA hybridization the strain could easily be "purified" genetically.
