Investigation of the pathogenesis of glaucoma capsulare with special discussion of alpha 1 Lp and Cp in aqueous humor.

The aqueous humor proteins of exfoliation syndrome were studied qualitatively by crossed immunoelectrophoresis and compared with normal controls. Two kinds of macromolecules (alpha 1-lipoprotein and ceruloplasmin) were detected more frequently in exfoliation syndrome than in the normal controls. Alpha 1 Lp was detected in eight of ten patients with exfoliation syndrome, but in none of the eight controls. Cp was detected in seven of ten patients with exfoliation syndrome, but in only one of the eight controls. The presence of these macromolecules in the anterior chamber suggests high permeability of vessels in the exfoliation syndrome. This interpretation is also supported by the fact that neovascularization is sometimes present in the iris of the exfoliation syndrome. In the aqueous humor from a patient with diabetic retinopathy, ten kinds of aqueous humor proteins were identified.

Facilitatory effects of intermittent photic stimulation on visual cortical kindling.

There are few reports on kindling preparation by daily electrical stimulation to the primary visual cortex. In our previous studies on cats, it was demonstrated that repeated stimulation to the primary visual cortex brought a similar progress in seizure development as in amygdaloid kindling, although it took more trials. In this study, we examined the effects of intermittent photic stimulation (IPS) (2.5 Hz) on the visual cortical kindling preparation to clarify more underlying mechanisms of photosensitivity. Under IPS, afterdischarges synchronizing to IPS were seen and a duration of afterdischarge was extremely prolonged, even in the early stage of the kindling process. The threshold of stimulus intensity inducing afterdischarge was significantly reduced and the generalized convulsive seizure was obtained with fewer times of stimulation, as compared to visual cortical kindling without IPS. In visual cortical kindling myoclonic responses were sometimes caused by IPS for a few minutes immediately after cessation of the electrically induced generalized seizure. Such phenomena were never seen in amygdaloid kindling. These results indicate that IPS facilitates a seizure development in all stages of visual cortical kindling.

Studies of human tear proteins--1. Analysis of tears from normal subjects by crossed immunoelectrophoresis.

Tears were collected from 10 normal subjects by two methods, 1) micropipetting after stimulation with an onion slice, and 2) Schirmer 1 test using a filter paper strip. The tears in the 5 x 5 mm part of the filter paper placed within the conjunctival sac were extracted with a 0.9% NaCl solution. The tear samples so obtained were analyzed by crossed immunoelectrophoresis. Nineteen proteins were identified by the use of monospecific antisera. Ten tear-specific proteins including the specific tear prealbumin of Bonavida et al., lactoferrin and lysozyme were identified by the intermediate gel method. Some differences were found in the pattern of the tear-specific proteins between the tears collected by the two methods, but the pattern was basically similar. Some individual differences were also found. The levels of serum proteins including albumin and IgG varied considerably, but it was thought to be due to dilution by reflex lacrimation during tear sampling. On the other hand, the pattern of the tear-specific proteins was fairly constant. The filter paper method allowed tear protein analysis, even in patients with 0 mm wetting by the Schirmer 1 test. A study of the length-volume relationship of the filter paper wetting showed that the tears contained in the 5 x 5 mm portion of the filter paper were about 1.8 microliters. The filter paper method was thought to offer a reliable clinical tool for the study of the tear proteins in various pathological conditions of the lacrimal secretion.

A new approach to the reactive afterdischarge in the development of kindling.

Simultaneously recorded multielectrode EEG data obtained during the course of amygdaloid kindling in cats were quantitatively analyzed by means of a multidimensional autoregressive model. By the use of this method, the afterdischarge was objectively separated into propagated and reactive discharge respectively in the frequency domain and it permitted the study of the progressive development of reactive afterdischarge which has been suggested to have a functional relationship with seizure development. As a result, marked augmentation and a tendency for synchronization of reactive afterdischarge were noted in the midbrain reticular formation and anteroventral part of the thalamus along with seizure development. Furthermore, a remarkable contribution to producing afterdischarges at other brain sites was revealed. Such contribution of the reticular system as a pacemaker was also shown by analysis of the background EEG. From the above findings obtained in kindling preparation of cats through multidimensional autoregressive analysis, a new approach to the mechanism of kindling phenomenon was demonstrated and a possible mechanism was also discussed.
