Effect of denervation on acetylcholine synthesizing activity and nicotine-like binding sites in rat hind limb muscles.

The acetylcholine synthesizing (ACh-s) activity and the binding of the nicotine ligands 3H-alpha-bungarotoxin (alpha Btx) and 3H-d-tubocurarine (d-TC) were analysed in rat hind limb muscles 1-32 days after denervation. Muscles of the contralateral leg served as controls. ACh-s activity was considerably lower in tibialis anterior as compared to the extensor digitorum longus and the peroneus longus muscles. After denervation there was a rapid decrease in ACh-s activity in parallel to a considerable increase in alpha-Btx binding. The binding of d-TC was essentially unchanged following denervation indicating the special characteristics of the newly formed receptors. ACh-s activity can be differentiated into one specific (choline acetyltransferase) and one unspecific component.

Acetylcholine synthesizing activity and nicotinic binding sites in rat hind limb muscles during reinnervation.

Acetylcholine synthesizing (ACh-s) activity and binding of the nicotinic ligands 3H-alpha-bungarotoxin (alpha-Btx) and 3H-d-tubocurarine (d-TC) were analysed in rat hind limb muscles for up to 6 months after a cryolesion of the sciatic nerve. Muscles of the contralateral leg served as controls. After cryolesion there was a rapid decrease in ACh-s activity parallel to a considerable increase in alpha-Btx binding. A clearcut increase in ACh-s activity was first seen at 32 days and complete restoration in ACh-s activity was seen from 2-3 months postoperatively. From the 16th day there was a rapid decrease in alpha-Btx bindings sites and initial values were seen at the 32nd day postoperatively. D-TC binding sites were relatively unchanged during the experimental period. The first neurophysiological signs of reinnervation were seen at 16 days. The rats were observed to move normally about 2 months postoperatively. Normalization of alpha-Btx binding sites seems to be an earlier sign of reinnervation than normalization of ACh-s activity.

Radioimmunoassay of serum myoglobin in neuromuscular diseases.

Radioimmunoassay of serum myoglobin was performed in 85 patients with muscular symptoms. Elevated levels were found in 93% of patients with myogenic myopathy, in 54% with myasthenia gravis and in 50% with neurogenic myopathy. All 11 patients with polymyositis had elevated myoglobin concentrations. In six of seven patients with polymyositis, who were followed up with repeated determinations, a clear relationship between myoglobin levels and clinical course was found. In general serum myoglobin seemed to be a more sensitive indicator of muscle disease than creatine kinase.
