Nasal absorption of secretin in rats was enhanced in an acid solution, and the maximum absorption was observed at a sodium chloride solution molarity of 0.462. In order to examine reasons for the variation of absorbability caused by the change of pH and osmolarity in secretin preparations, both a pretreatment study, in which the nasal mucosa was treated with placebo prior to the administration of a secretin preparation, and a histological study were conducted in rats. The nasal absorption of secretin was determined by measuring the increased secretin of pancreatic juice. Similar profiles of nasal absorption, both after intranasal administration of secretin preparations and as a result of pretreatment effects, were obtained in studies of the effects of pH and osmolarity. However, in the pH-effect study, the absorption with the use of active preparations was observed to be significantly larger than that with the pretreatment effect below a pH of 4.79, and significantly smaller than that with the pretreatment effect at a pH of 7 to 8. The results of histological studies revealed structural changes of the epithelial cells of the nasal mucosa at pH 2.94, and shrinkage of epithelial cells was observed at a sodium chloride solution molarity of 0.462.

Whisker-like crystals appeared on the surface of tablets that contained lactose or mannitol, a hygroscopic material such as docusate sodium, magnesium chloride, or potassium acetate, and other ingredients stored in an atmosphere of high relative humidity. The crystals were observed under a scanning electron microscope and were measured using differential scanning calorimetry and TLC. The crystals contained lactose or mannitol.
The effects of dose, pH, and osmolarity on the nasal absorption of a secretin solution were studied in rats. The nasal absorption of secretin was determined by measuring the increased secretion of pancreatic juice, and this was compared with the same response on intravenous administration. The relative bioavailability of secretin on intranasal administration compared with intravenous administration, as estimated from the pharmacological response, was approximately one-tenth. It was enhanced in acid solution and reached a maximum in a 0.462 M sodium chloride solution.
A prolonged weak illumination has been reported to induce an off-response in transretinal potential of bullfrog retina immersed in a low Ca2+, Ba2+ and Na-aspartate-containing solution. Changes in the interstitial current accompanying the generation of the off-response were studied by inserting a pair of micropipettes into the bullfrog retina. A transient increase in proximal-to-distal current above the dark level was observed when the off-response was developing (off-current). The current divergence of the off-current indicated that the source of the off-current was located about 75 micron below the receptor surface. In the absence of Ba2+, similar changes in currents were also observed, although no detectable off-response was observed in the transretinal potential. Findings suggest that regardless of the presence of Ba2+, current flowing outward through the plasma membrane of the rod inner segments increases transiently when the off-response is developing.
Stiff skin syndrome was first documented by Esterly and McKusick. We saw a patient with this disease, as well as another who had a similar condition but a somewhat different histology. These two patients shared heritable stiff skin, restricted joint mobility, and absent mucopolysacchariduria. In stiff skin syndrome, which may have two variants, skin biopsy demonstrates large cells stained metachromatically by toluidine blue. These cells are shaped like dermal melanocytes or rounded cells and their presence may be directly related to the deposition of mucopolysaccharide in the dermis. In some cases they may invade the deeper tissues. Two cases of stiff skin syndrome that were in the Japanese literature are also discussed.
Relationships between the change in threshold of the fast PIII response and the rhodopsin photoproduct content were studied in the isolated bullfrog retina treated with Ba2+ and aspartate. A reduction in the extracellular Ca2+ concentration from 1.0 to 0.01 mM caused an increase in the decay rate of 380 nm absorbing photoproducts (metarhodopsin II and retinal) and metarhodopsin III. In both normal and low Ca2+ solutions, the threshold change after bleaching of about 50% of rhodopsin was closely related to the 380 nm absorbing photoproducts. The results suggest that the intermediate adaptation process of rods may be regulated by the 380 nm absorbing photoproducts.
