Ribulose-peptide in human semen: I. Isolation.

A substance apparently related to a compound consisting of phospho-peptide and ribulose-peptide in the yolk of chick embryos was isolated from acid soluble fractions of human semen by means of Dowex 1 (OH) form column chromatography. The treated precipitate of human semen was examined with an analytical ultracentrifuge by means of synthetic boundary cells and found to be a single component. The sedimentation coefficient was 0.80. The sugar component in the precipitate was identified as ribulose by paper chromatography, color reaction, absorption spectrum of the reaction product in the orcinol, and enzymatic analysis. Amino acids detected in the acid hydrolysate of the precipitate were lysine, histidine, arginine, aspartic acid, threonine, serine, glutamic acid, proline, glycine, alanine, valine, isoleucine, and leucine. From the above, this substance was determined to be a ribulose-peptide.

Ribulose-peptide in human semen: II. Synthesis.

In order to study the metabolism of a naturally occurring peptide containing D-ribulose in the semen, a pathway of D-ribulose synthesis from radioactively labeled glucose was analyzed. Semen obtained from ten healthy adults was used in this study. The seminal fluid of each person sampled was divided into two equal parts and each one was incubated with glucose-1-14C or glucose-6-14C at room temperature for 1 hr. The rate of incorporation into ribulose from glucose-6-14C was higher than that of glucose-1-14C. This suggests that the ribulose in semen might be converted from glucose through the pentose-phosphate cycle, and incorporated into the ribulose-peptide through an unknown pathway. Incorporation of the labelled 14C was also found in all of the amino acids which were detected in the acid hydrolysate of the ribulose-peptide.

Factors concerning the immunity to poxvirus infection.

Two kinds of early antigens were shown in vaccinia-infected cells, one heat (56 degrees, 30 min)-stable (ES antigen) and the other heat-labile (EL antigen). Both antigens do not induce circulating neutralizing antibody in rabbits, but resistance to intradermal inoculation of active virus was observed when animals were immunized with either antigen in combination with Freund's complete adjuvant. Vaccinia-specific haemagglutinin (VHA) associated with infected cell membrane was purified and its antigenicity was also studied. Rabbits showed a good antibody response to VHA and some animals also showed neutralizing antibody, though in a small amount, to either intracellular (ICV) or extracellular (EVC) virus. Rabbits immunized with membrane antigens obtained from either VHA-positive or negative virus in combination with Freund's adjuvant showed skin restance to challenge with active virus. It was suggested that antibody against ECV might be responsible for the resistance.

Metabolism of 1-(3-trifluoromethylphenyl)-3-(2-hydroxyethyl)quinazoline-2,4(1H,3H)-dione (H-88). II. Abosorption, distribution and excretion in rat, mouse, rabbit, monkey and man.

1. The maximum concentration radioactivity in blood occurred 2-4 h after oral administration of [14C]H-88 in mouse, rabbit and man. With rat, a maximum concentration was obtained 24 h after administration of the drug at a dose level of 60 mg/kg, but only 4 h at a dose of 6 mg/kg. Unchanged H-88 comprised about 50% of serum radioactivity in rat and mouse during the first few hours, but only a small proportion of the serum radioactivity in rabbit and man. 2. The distribution pattern of the radioactivity in rat given the drug at two dose levels was similar, and this differed slightly from mouse and considerably from rabbit. Autoradiograms in rat and rabbit confirmed the findings from the distribution studies, and the autoradiographic distribution pattern in monkey was similar to that in rabbit. 3. Of the administered radioactivity 25-30% was recovered from bile within 48 h in bile-duct-cannulated rats and rabbits. Excretion of radioactivity in respiratory CO2 was negligible in the rat.

[Influence of schizophyllan, streptomycin and rifampicin on histopathological changes in mice infected with tubercle bacilli (author's transl)].

Experimental tuberculosis in mice infected with streptomycin-resistant Mycobacterium tuberculosis SCHACHT strain was treated with streptomycin or rifampicin alone and in combination with schizophyllan. The histopathogical tests of various organs of the treated mice were carried out. 1) In the group of mice treated with streptomycin alone, the moderate focal proliferation of RE cells were seen at the beginning of infection. However, durable activation of RE cells and the prolongation of life-span could not be recognized as compared with control animals. 2) The treatment with streptomycin-schizophyllan combination appeared to be somewhat more effective than schizophyllan alone, the phagocytic capacity being more strongly stimulated. 3) In the group treated with rifampicin alone, the therapeutic effect could be exhibited by the direct antibacterial action of rifampicin, but the activation of RE cells was slight. When rifampicin was discontinued, the growth of tubercle bacilli was rapidly resumed, and the durable therapeutic effect seemed not to be expected. Degeneration of hepatic cells tended to develop. 4) In the group treated with rifampicin-schizophyllan combination, the antibacterial effect of rifampicin appeared to be potentiated by the strong activation of RE cells by schizophyllan, showing the durable therapeutic effects.
