Modification of diurnal changes in the ultrastructure of synaptic ribbons of the turtle.

Synaptic ribbons of rod cells of the red-eared turtle under light-dark (LD) cycle, continuous dark (DD) and continuous light (LL) were examined by electron microscopy. In the LD cycle, several stick-shaped (plate-shaped, three-dimensionally) ribbons formed multilayered ribbon complexes at 1000 hr, and they turned into aggregates of rounded ribbons at 1900 hr (1 hr after light-off). In DD, stick-shaped ribbons were situated singly near presynaptic membranes. Large ribbon complexes consisting of 7-8 parallel layers, as shown in the LD cycle, were not observed. Aggregates of rounded ribbons, or club-shaped ones could not be observed. In LL, aggregates of irregular ribbon fragments were noted. None of the ribbons in this regimen showed sharp contours. Cyclic LD illumination is necessary to form multilayered ribbon complexes and rounded ribbons. Internal circadian rhythm contributes little part to the diurnal changes in the structure of the synaptic ribbons.

Ciliocytophthoria (ccp) in nasopharyngeal smear from patients with acute tonsillitis.

Ciliocytophthoria (ccp) in nasal or nasopharyngeal secretion from the patients with acute tonsillitis, acute rhinosinusitis and allergic rhinitis was studied. The ratio of the presence of ccp among acute tonsillitis, acute rhinosinusitis and allergic rhinitis was compared. Acute tonsillitis had a much more positive ccp test than other diseases. This might be related to the anatomical feature of palatine tonsil having a complicated hollow-crypt. The presence of Haemophilus influenzae was the most frequent in patients with positive ccp test. Further study will be necessary to determine possible relationship between viral infection and bacteria or endotoxin from bacteria.

Atrial natriuretic peptide, angiotensin, norepinephrine and electrolyte in cerebrospinal fluid of essential hypertension.

We determined concentrations of atrial natriuretic peptide (ANP), angiotensin (Ang), norepinephrine (NE) and electrolyte in plasma and cerebrospinal fluid (CSF) to study possible roles of these substances within the brain in human hypertension. Blood and CSF samples were obtained from 10 patients with mild to moderate essential hypertension (EHT) aged 40-65 y and 10 age-matched normotensive subjects (NT) on a regular salt diet (8 g/day). Levels of ANP, NE, Na, K, Ca and Cl in CSF and plasma were comparable between EHT and NT. Plasma renin activity, plasma and CSF Ang II were lower in EHT than NT. CSF Ang III tended to be lower in EHT. There was no correlation between CSF and plasma ANP, or between CSF and plasma Ang II. Our results indicate that CSF levels of ANP may not be altered in middle aged patients with mild to moderate hypertension. It is also suggested that Ang II, NE and sodium in the central nervous system may not have important roles in hypertension of those patients.

Isolation and in vitro growth of glioma-infiltrating lymphocytes, and an analysis of their surface phenotypes.

The present investigation was conducted in order to examine the feasibility of isolating and growing glioma-infiltrating lymphocytes in vitro as possible effector cells for use in new adoptive immunotherapy. Eight surgical specimens obtained from patients with malignant astrocytomas were treated by enzyme dispersion; the cells were separated on a density gradient and grown in the presence of human recombinant interleukin-2. The cultured lymphocytes were tested for cell-surface markers by using monoclonal antibodies in a flow cytometric analysis. In all cases the glioma-derived lymphocytes were grown in culture for several weeks with substantial increases in cell numbers (at least 5 X 10(8) cells). The mature T cell population (CD3, 89%) was found to have an increased proportion of the cytotoxic/suppressor phenotype CD8 (55%) as compared to peripheral blood lymphocytes (PBL's). Eighty-six percent of the cultivated lymphocytes expressed HLA-DR. The IL-2 receptor was predominantly expressed on the helper subset (CD4-positive). Otherwise, anti-CD16, which specifically reacts with natural killer (NK) cells, did not stain significantly more of the cultured gliomaderived lymphocytes compared with lymphocyte-activated PBL's. These results corroborate the observations made with conventional immunohistochemical examination. It has been demonstrated that T lymphocytes isolated from human cancers are enriched for specific reactivity to their autochthonous tumor cells. These experiments support the possible use of glioma-infiltrating lymphocytes as a new treatment for patients with malignant glioma.

[Five cases of primary intracranial germ cell tumor treated by combination chemotherapy with cisplatin].

Five cases of non-germinoma germ cell tumor which received combination chemotherapy with cisplatin were studied to elucidate the efficacy of chemotherapy. The patients' ages ranged from 7 to 24 years old. Three of the patients were male and two were female. Locations of the tumors were suprasellar in three cases, pineal in one case and basal ganglia in the other. Three of the 5 tumors were histologically verified as yolk sac tumors, embryonal carcinoma and HCG_producing germinoma. Two were histologically unverified HCG-producing tumors. Following the reception of PVB therapy (cisplatin, vincristine and bleomycin) by all the patients, three of the HCG-producing tumors showed complete remission and two of AFP producing tumors demonstrated partial remission, and progressive healing process, respectively. Renal dysfunction in two case, leukopenia in two cases and lung fibrosis in one cae were noted as side effects. Two cases were treated only with cisplatin because ling fibrosis due to administration of bleomycin was intractable and harmful. Also the time when irradiation. It seemed better to administer chemotherapeutic agents prior to or during irradiation on account of the fact that, after radiotherapy, there is reduced blood flow in the tumor region. Also, CDDP acts as a radiosensitizer. Therefore, individual administration of cisplatin during irradiation was recommended for the initial chemotherapy against the case of non-germinoma germ cell tumors, in which an intractable side effect might be expected.
