[Production of polyclonal antibody against HBD-2 by immunization with recombinant GST-HBD-2 fusion protein].

For the purpose of detecting the HBD-2 expression at protein level, the recombinant prokaryotic expression vector pGEX-1lambdaT-HBD-2 was constructed and the E. coli-based product of GST-HBD-2 fusion protein was prepared. When rabbit was immunized with the fusion protein, the anti-serum against HBD-2 was produced. After caprylic acid and ammonium sulfate precipitation, high titer of specific polyclonal antibody against HBD-2, which was detected by ELISA and Western blot, was obtained. This result suggests that recombinant peptide fusion protein could be used instead of the conjugate of peptide-albumin or peptide-thyroid globulin to produce antibody. The obtained antibodies could be used for revealing the tissue distribution of HBD-2 and the regulation of its gene expression.

Electronic visual prosthesis.

Retinitis pigmentosa (RP) and age-related macular degeneration (AMD) are incurable diseases that result in profound vision loss due to degeneration of the light sensing photoreceptors. However, the discovery that direct electrical stimulation of the retinal neurons creates visual sensation has inspired prosthetic devices aimed to restore useful vision in RP/AMD patients. The approach to one such electronic visual prosthesis is described in this article. The prosthesis consists of an external unit and an internal unit. The communication link has three components--power and data transfer from the external to the internal unit, and data transfer from the internal to the external unit. A novel method of integrating power transfer and back telemetry is described here. The goal is to design a stimulator chip with a small area with low power consumption. This chip, capable of stimulating 60 dedicated electrodes, is fabricated using AMI 1.2 microm process technology and the results are presented. Improvements in the design to increase the number of outputs to 1,000 have been discussed. The new circuit is aimed at increasing the circuit density, reducing power per stimulus, and meeting the requirements more closely than the previous designs. The results of the designed chip are presented.

[Bifidobacterium cell wall proteins induced beta-defensin 2 mRNA expression in human intestinal epithelial cells].

OBJECTIVE: To examine Bifidobacterium mediated induction of human defensin 2 mRNA expression in human intestinal epithelial cells and define the bioactive components of Bifidobacterium. METHODS: Bifidobacterium was cultured in MRS medium in anaerobic incubator and its cell wall was isolated by sonication and centrifugation. The cell wall proteins of Bifidobacteria were extracted with 2% SDS. The level of hBD-2 mRNA in HT-29 cells was detected by reverse-transcription PCR and Northern blot analysis. RESULTS: There was no detectable signal of hBD-2 mRNA in the unstimulated HT-29 cells. However, the heat-killed Bifidobacteria, cell wall of Bifidobacteria, and cell wall proteins remarkably induced hBD-2 mRNA expression in HT-29. CONCLUSION: Bifidobacterium or its cell wall proteins can induce hBD-2 gene expression in human intestinal epithelial cells, which may play a role in the mechanisms of innate defense against pathogens in the intestine.
