Quantitative structure-activity relationship models for prediction of the toxicity of polybrominated diphenyl ether congeners.

Levels of polybrominated diphenyl ethers (PBDEs) are increasing in the environment and may cause long-term health problems in humans. The similarity in the chemical structures of PBDEs and other halogenated aromatic pollutants hints on the possibility that they might share similar toxicological effects. In this work, three-dimensional quantitative structure activity relationships (3-D-QSAR) models, using comparative molecular field analysis (CoMFA) and comparative similarity indices analysis (CoMSIA), were built based on calculated structural indices and a reported experimental toxicology index (aryl hydrocarbon receptor relative binding affinities, RBA) of 18 PBDEs congeners, to determine the factors required for the RBA of these PBDEs. After performing leave-one-out cross-validation, satisfactory results were obtained with cross-validation O2 and R2 values of 0.580 and 0.995 by the CoMFA model and 0.680 and 0.982 by the CoMSIA model, respectively. The results showed clearly that the nonplanar conformations of PBDEs result in the lowest energy level and that the electrostatic index was the main factor reflecting the RBA of PBDEs. The two QSAR models were then used to predict the RBA value of 46 PBDEs for which experimental values are unavailable at present.

Electrochemical preparation of composite polyaniline coating and its application in the determination of bisphenol A, 4-n-nonylphenol, 4-tert-octylphenol using direct solid phase microextraction coupled with high performance liquid chromatography.

For SPME-HPLC, metal wires with better mechanical strength are preferred over the fused silica fibers. In this article, a novel composite polyaniline (CPANI) doped with PEG and polydimethylsiloxane coating (CPANI fiber) was prepared on a stainless steel wire by a three-electrode system: the fiber was used as the work electrode, a calomel electrode and a platinum electrode were used as the reference and the counter electrodes, respectively. To evaluate the new CPANI coating, the coating was used to extract three kinds of phenols (bisphenol A, 4-n-nonylphenol, and 4-tert-octylphenol) in water samples by direct-SPME mode and then desorbed in commercial SPME-HPLC interface to separation. The extraction procedure was also optimized. Five real water samples were investigated. Good recoveries were gained when environmental samples were analyzed.

Gas chromatography/ion trap mass spectrometry applied for the determination of polybrominated diphenyl ethers in soil.

A gas chromatography/ion trap mass spectrometry (GC/ITMS) method was developed for the determination of polybrominated diphenyl ethers (PBDEs). ITMS parameters were optimized in order to achieve the best sensitivity for the PBDE analysis. Tandem mass spectrometry, along with an isotope dilution internal standard method, was used for the quantitation. Chromatographic windows were developed for mono- to hepta-BDEs, depending on the retention times when a 30-m GC column was used. A different 15-m column was used to analyze deca-BDE. Environmental soil samples collected from an electronic waste recycling site were prepared by using Soxhlet extraction and column chromatographic cleanup. Average recoveries of 61-118% were obtained for the 13C-labeled PBDE internal standards spiked in the samples prior to sample preparation. The accuracy represented by relative analytical errors was -24% to 18%, and the precision (relative standard deviation) was 11-26% (n=8). The method detection limits ranged from 0.013-0.25 ng/g for the PBDEs in soil.

Determination of polybrominated diphenyl ethers in soil and sediment from an electronic waste recycling facility.

Soil and sediment collected in the vicinity of an open electronic waste disposal and recycling facility located in Guiyu, Guangdong, China, were analyzed for the levels of common polybrominated diphenyl ethers (PBDEs) by using soxhlet extraction and gas chromatography/mass spectrometry. The PBDEs were detected in the soil and sediment samples at levels of 0.26-824 ng/g (dry weight). The obtained isomer profiles of PBDEs from the two soil samples collected from different contamination sites were found to be similar to various technical formulations of fire retardant products.

Study on the contamination of heavy metals and their correlations in mollusks collected from coastal sites along the Chinese Bohai Sea.

Mollusks living in seas can accumulate heavy metals, and may serve as excellent passive biomonitors. During a period of 1 year, bioaccumulation of As, Cd, Co, Cr, Cu, Fe, Hg, Mn, Ni, Pb, Se, and Zn was examined in nine kinds of mollusks (Rapana venosa, Neverita didyma, Scapharca subcrenata, Mytilus edulis, Amusium, Crassostrea talienwhanensis, Meretix meretrix, Ruditapes philippinarum, and Mactra veneriformis), which were collected at eight coastal sites along the Chinese Bohai Sea. Metal concentrations were directly determined by inductively coupled plasma mass spectrometer (ICP-MS). Two certified reference materials, dogfish muscle (DORM-2) and mussel (GBW 08571), were used to validate the methods, and the recoveries were within 83.72-112.30% of the certified values. Bioaccumulation of metals varied strongly among sampling sites and species. Statistical analysis (one-way ANOVA) indicated that different species examined showed different bioaccumulation of metals, and perhaps they could be used as potential biomonitors to investigate the contamination levels of heavy metals. Principal component analysis (PCA) and correlation analysis were used to study the relationships between these heavy metals. The results showed that, in nine mollusks' tissues, there are significant correlations between these elements in adjacent main group or subgroup in periodic table of elements.

Development and validation of an enzyme-linked immunosorbent assay for vitellogenin in Chinese loach (Misgurnus angaillicaudatus).

A competitive enzyme-linked immunosorbent assay (ELISA) was developed for the quantification of vitellogenin (Vtg) in the plasma of Chinese loach (Misgurnus angaillicaudatus). Vtg was isolated by anion exchange membrane chromatography from the plasma of 17beta-estradiol (E(2))-treated loach. Polyclonal antibody against Vtg was raised in rabbits, and the specificity of the antibody was assessed by Western blotting analysis. No cross-reactivity was observed with plasma from male loach. Plasma samples from vitellogenic females and E(2)-treated males were diluted parallel with the purified Vtg standard curve in the ELISA. The detection limit of the assay was 5.7 ng ml(-) (1) and a working range of the standard curve was between 15.6 and 1000 ng ml(-) (1). The intra- and inter-assay coefficients of variations were 6.9% and 10.4%, respectively. The recovery was 104.4%. The assay was applied for measuring vitellogenin concentration in the plasma from 24 wild Chinese loach from Lianyungang, Jiangsu Province, in October 2003. It was found that, in addition to females (plasma Vtg concentration ranging from 518.4 microg ml(-) (1) to 1922.3 microg ml(-) (1)), male loach showed low plasma Vtg--from undetectable (two fish) to 2.9+/-0.7 microg ml(-) (1) (mean+/-S.D.). The results indicate that this method provides a valuable tool for the study of estrogenic effect in Chinese loach.

Determination of 3-chloropropane-1,2-diol in liquid hydrolyzed vegetable proteins and soy sauce by solid-phase microextraction and gas chromatography/mass spectrometry.

Headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography/mass spectrometry (GC/MS) method was developed to determine 3-chloropropane-1,2-diol (3-MCPD) in hydrolyzed vegetable protein and Chinese soy sauce. The 3-MCPD was firstly derivativized with phenylboronic acid in aqueous solution at 90 degrees C for 10 min, then extracted by HS-SPME and finally detected with GC/MS, parameters related to both the derivative reaction and the HS-SPME process were optimized. The proposed method has a linear range of 0.0194-394 microg g(-1), a detection limit of 3.87 ng g(-1) (S/N = 3), and a precision of RSD = 7.5% (n = 5). Seventeen real samples, including four HVPs and thirteen soy sauce samples, were analyzed to examine the feasibility of the proposed procedure; with a concentration of 3-MCPD and acceptable recoveries at 0.71 microg g(-1) spiked levels were obtained. Being simpler, faster and more environmentally benign than the existing methods, this method is accurate and suitable for routine analysis.

[Determination of polybrominated diphenyl ethers in sewage sludge by high resolution gas chromatography coupled with high resolution mass spectrometry].

Polybrominated diphenyl ethers (PBDEs) are a group of brominated flame retardants, which are manufactured in large quantities and widely used in a variety of consumer goods. Recently they spread ubiquitously as environmental contaminants. In order to investigate the pollution level of PBDEs in different environmental samples, a method has been established by using high resolution gas chromatography coupled with high resolution mass spectrometry (HRGC-HRMS). The method was used to analyze biosamples obtained from the international inter-calibration. The results were satisfactory by analyzing z-score figures. Then, PBDE contents in three samples of sewage sludge were investigated by the method after Soxhlet extraction and multi-layer silica gel chromatographic separation. The number of PBDEs determined was 19. From the results, we can see that the contamination level of PBDEs in the sample from Beijing was higher than those from two other areas. The method was validated and applied for the analysis of environmental samples.

Dechlorination and destruction of 2,4,6-trichlorophenol and pentachlorophenol using hydrogen peroxide as the oxidant catalyzed by molybdate ions under basic condition.

The dechlorination and destruction of 2,4,6-trichlorophenol (TCP) and pentachlorophenol (PCP) under basic condition using hydrogen peroxide as the oxidant catalyzed by molybdate ions have been studied. Under ambient conditions of temperature and pressure, more than 95% of millimolar solutions of TCP and PCP can be converted to CO2 and CO, chlorinated and nonchlorinated carboxylic acid in 40 min. Up to 2.4 chloride ions per TCP and 3.5 chloride ions per PCP were released. TOC measurements indicated that 18% and 11% of the carbon was mineralized for TCP and PCP respectively after an hour of reaction. The results of ESR measurements suggested that the reaction possibly proceeded via the pathway with singlet oxygen.

Sensitivity enhancement of chlorinated phenols by continuous flow liquid membrane extraction followed by capillary electrophoresis.

This paper describes a new analytical system, based on the combination of continuous flow liquid membrane extraction (CFLME) enrichment and capillary electrophoresis (CE) separation, for analysis of chlorinated phenols in water samples. Five chlorinated phenols including 3-chlorophenol (3CP), 4-chlorophenol (4CP) 2,4-dichlorophenol (DCP), 2,4,6-trichlorophenol (TCP), and pentachlorophenol (PCP) were separated by CE with Tris/sodium dihydrogen phosphate solution containing methanol 1% (v/v) as the run buffer. CFLME related parameters were investigated and optimal enrichment was obtained by using 0.3 mol L(-1) Tris as acceptor and with a sample pH 5.0, a sample flow rate of 4.0 mL min(-1), and an enrichment sample volume of 150 mL. The detection limit (S/N= 3) was 6.9, 1.0, and 1.7 ng mL(-1) for DCP, PCP, and TCP, respectively. The reproducibility (RSD%, n = 6) was 5.7 for DCP, 2.5 for PCP, and 2.8% for TCP (n = 6). The proposed method was applied to the determination of chlorinated phenols in spiked water samples with relatively satisfactory recoveries.

Determination of 4-tert-octylphenol, 4-nonylphenol and bisphenol A in surface waters from the Haihe River in Tianjin by gas chromatography-mass spectrometry with selected ion monitoring.

The estrogenic pollutants 4-tert-octylphenol (OP), 4-nonylphenol (NP) and bisphenol A (BPA) were determined in surface water samples from the Haihe River, Tianjin, China. The analytes were extracted and concentrated from 300 ml acidified water samples by liquid-liquid extractions using dichloromethane, derivatized with trifluoroacetic anhydride, and quantified by gas chromatography-mass spectrometry (GC-MS) with selected ion monitoring (SIM). Among the samples collected from 14 sampling sites, only one sample was found to have a relatively high concentration of BPA (8.30 microgl(-1)) and NP (0.55 microgl(-1)). The concentrations of OP, NP and BPA in the other samples were in the range of 18.0-20.2, 106-296 and 19.1-106 ngl(-1), respectively. Recoveries for OP, NP and BPA in the spiked water samples were all over 80%.

A rapid two-step chromatographic method for the quantitative determination of vitellogenin in fish plasma.

By combining anion-exchange membrane purification with high-performance size-exclusion chromatography (HPSEC) analysis, a two-step chromatographic method was developed for the determination of vitellogenin (Vtg) in fish plasma. Most plasma protein interferences can be removed during anion-exchange membrane purification process. Vtg is eluted from the size-exclusion chromatography column with a retention time of about 9 min and is characterized based on the native molecular weight, with a limit of quantification of 20 microg Vtg mL(-1)plasma. The spiked recovery and interassay variability were better than 80% and 4.8%. This method was successfully applied to analyze the plasma Vtg levels of loach (Misgurnus angaillicaudatus) and sea catfish (Enchelyopus elongatus). In addition to all the female fish, Vtg is detected in 75% of male loaches and 100% of male sea catfish. The result indicates that some chemicals or unknown factors with estrogenic activity have induced male fish to produce Vtg.

Multiwalled carbon nanotubes as a solid-phase extraction adsorbent for the determination of bisphenol A, 4-n-nonylphenol, and 4-tert-octylphenol.

The adsorptive potential of multiwalled carbon nanotubes (MWNTs) for solid-phase extraction of bisphenol A, 4-n-nonylphenol, and 4-tert-octylphenol was investigated for the first time. The three analytes are quantitatively adsorbed on a MWNTs-packed cartridge, then the analytes retained on the cartridge are quantitatively desorbed with suitable amounts of methanol. Finally, the analytes in the methanol eluate are determined by high performance liquid chromatography-fluorometric detection. Parameters influencing the extraction efficiency, such as volume of the sample solutions, pH of the sample, and the eluent volume, were examined. Comparative studies showed that MWNTs were superior to C18 for the extraction of the more polar analyte bisphenol A and at least as effective as C18 for the extraction of 4-n-nonylphenol and 4-tert-octylphenol. Compared to XAD-2 copolymer, MWNTs exhibited a better property for the extraction of all three analytes. The developed method has been applied to determine bisphenol A, 4-n-nonylphenol, and 4-tert-octylphenol in several environmental water samples. The accuracy of the proposed method was tested by recovery measurements on spiked samples, and good recovery results (89.8-104.2%) were obtained. Detection limits of 0.083, 0.024, and 0.018 ng mL(-1) for bisphenol A, 4-n-nonylphenol, and 4-tert-octylphenol, respectively, were achieved under the optimized conditions.

Membrane chromatographic method for the rapid purification of vitellogenin from fish plasma.

A membrane chromatographic method was developed for the rapid purification of vitellogenin (Vtg) from the plasma of 17beta-estradiol induced loach (Misgurnus angaillicaud atus) and carp (Cyprinus carpio). The time required for the proposed procedure is less then 10 min at a flow-rate of 5 ml/min of the mobile phase, and 0.5 ml of fish plasma could be separated in one cycle. Multistep gradient elution was more suitable for the separation than linear gradient elution. Under optimized conditions, a single Vtg peak can be obtained and its identity was confirmed by SDS-PAGE and gel-permeation chromatography assessment. This method is rapid and easy to operate compared to conventional HPLC and FPLC columns for Vtg separation.

Study on the interaction between nucleic acids and acetamiprid.

The interaction between deoxyribonucleic acid (DNA) and acetamiprid was studied. It was found that the fluorescence of acetamiprid could be enhanced in the presence of DNA in sulfuric acid solution. The excitation and emission wavelength of acetamiprid was 291 nm and 587 nm, respectively. Under optimal conditions, the calibration graph is over the range of 0.1-10 micromL(-1). The calibration limit is 0.06 microg mL(-1) (S/N = 3). The determination results of DNA in yeast cell and golden staphylococcus samples by this method were satisfactory. The mechanism of the reaction is discussed.

Separation and determination of methyl tert-butyl ether and its degradation products by a laboratory-constructed micro-cryogenic chromatographic oven.

A laboratory-made micro-cryogenic chromatographic oven was mainly improved in size, which was controlled at 6 x 6 x 2.5 cm. A thermoelectric system was used to cool the capillary column instead of the traditional liquid cryogen. A cold block connected to the cryogenic module was directly solidified at room temperature with thermally conductive adhesive so that the uniformity of transferring heat was greatly improved, and the size of the system was reduced. Moreover, this system was inexpensive and convenient for both operation and control. The newly developed device coupled with headspace solid-phase microextraction (SPME) was successfully applied to the determination of methyl tert-butyl ether (MTBE) and its degradation products. During the analysis procedure, a 65 microm polydimethylsiloxane/divinylbenzene (PDMS/DVB) fiber was used to extract MTBE and its degradation products. The extraction was controlled at 50 degrees C for 30 min and the NaCl content in the sample was maintained at 35%. The limits of detection (LODs) ranged from 0.006 microg mL(-1) (for MTBE) to 0.206 microg mL(-1) (for methyl acetate) and the relative standard deviations (RSD%) were below 4%. The spiked recoveries for the developed method were evaluated using various water samples as a matrix.

Solid-phase extraction of several phthalate esters from environmental water samples on a column packed with polytetrafluoroethylene turnings.

A method for the determination of several phthalate esters in water samples using solid-phase extraction coupled to high-performance liquid chromatography was developed. A polytetrafluoroethylene turnings packed column was used as a SPE adsorbent material. The analytes were sorbed on polytetrafluoroethylene turnings, from which they were eluted by a small amount of acetonitrile, followed by a RP-HPLC-UV analysis. Detection limits (3sigma) of 4.7, 3.1, 3.6, 4.3, and 5.8 ng mL(-1) for di-n-butyl-phthalate, di-cyclohexyl-phthalate, di-n-octyl-phthalate, di-n-nonyl-phthalate and di-iso-decyl-phthalate, respectively, were achieved. Acceptable recovery results (92.1-127.5%) were obtained on five phthalate esters spiked samples.
