Determination of three bioactive constituents in moutan cortex by capillary electrophoresis with electrochemical detection.

Capillary electrophoresis with electrochemical detection has been employed for the separation and determination of the three active constituents (paeonol, benzoyloxypaeoniflorin, and oxypaeoniflorin) in traditional Chinese medicine, Moutan Cortex (root cortex of Paeonia suffruticosa Andrews). The effects of several important factors, such as the concentration of running buffer, the separation voltage, the injection time, and the detection potential, were investigated to determine the optimum conditions. The detection electrode was a 300 microm diameter carbon-disc electrode at a working potential of +0.90 V (versus SCE). The three analytes could be well separated within 7 min in a 40 cm length fused-silica capillary at a separation voltage of 12 kV in a 50 mM borate buffer (pH 9.2). The relation between the peak current and the analyte concentration was linear over 3 orders of magnitude with detection limits (S/N = 3) ranging from 0.4 to 0.7 microM for all analytes. The proposed method has been successfully applied to the determination of paeonol, benzoyloxypaeoniflorin, and oxypaeoniflorin in real plant samples with satisfactory assay results.

Rapid determination of paeoniflorin and three sugars in Radix Paeoniae Alba by capillary electrophoresis.

A method based on capillary electrophoresis with electrochemical detection has been developed for the separation and determination of paeoniflorin, sucrose, glucose, and fructose in traditional Chinese medicine, Radix Paeoniae Alba. The effects of several important factors, such as the concentration of NaOH, the separation voltage, the injection time, and the detection potential, were investigated to determine the optimum conditions. The detection electrode was a 300-microm diameter copper disc electrode at a working potential of +0.60 V (versus SCE). The four analytes can be well separated within 8 min in a 40 cm length fused-silica capillary at a separation voltage of 12 kV in a 75 mM NaOH aqueous solution. The relation between the peak current and the analyte concentration was linear over about 3 orders of magnitude with detection limits (S/N = 3) ranging from 1 to 2 microM for all analytes. The proposed method has been successfully applied for the determination of the paeoniflorin and sugars in real plant samples with satisfactory assay results.

[Effects of chemokine receptor and its ligand on migration of ovarian cancer cells].

BACKGROUND & OBJECTIVE: Chemokine receptors express on many tumor cells, and closely correlate with migration and metastasis of tumor cells. This study was to investigate expressions of chemokine(C-X-C) receptor 4 (CXCR4) and chemokine (C-X-C motif) ligand 12 (CXCL12) in human ovarian epithelial tumor cells, and their effects on migration of tumor cells. METHODS: Expression of CXCR4 mRNA and protein in 15 specimens of epithelial ovarian cancer tissue, ovarian cancer cell line CAOV3, endothelial cell line HUVEC, and 10 specimens of normal ovary tissue were evaluated by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot. Expression of CXCL12 mRNA in retroperitoneal lymph nodes, and smooth muscle of fallopian tube from the same 15 epithelial ovarian cancer patients was tested by RT-PCR, quantity of CXCL12 in ascites of 15 patients was assayed using ELISA. Boyden Transwells was used to analyze effects of CXCL12, and cancerous ascites on chemotaxis of CAOV3, and HUVEC cells. RESULTS: (1) Expression levels of CXCR4 mRNA in ovary cancer tissues, CAOV3 cells, and HUVEC cells were 2.30+/-1.12, 1.89+/-1.20, and 1.68+/-1.11, respectively; those of CXCR4 protein were 1.35+/-0.14, 1.86+/-0.34, and 1.96+/-0.23, respectively; CXCR4 mRNA and protein can't be detected in normal ovarian tissues. (2) In 15 ovarian cancer patients, concentrations of CXCL12 in ascites were 632-9 326 pg/ml, and CXCL12 mRNA level in retroperitoneal lymph nodes was 1.14+/-0.87, CXCL12 mRNA can't be detected in smooth muscle of fallopian tube. (3) Recombinant human CXCL12 induced migration of CAOV3, and HUVEC cells, the chemotactic indices (CI) were 3.9+/-1.2, and 4.1+/-1.6, significantly higher than those of control (1.0+/-0.4, and 1.1+/-0.7) (P<0.05)u cancerous ascites induced migration of CAOV3 cells with CI of 1.9+/-0.8, significantly higher than that of control (P<0.05). CONCLUSION: CXCR4 and CXCL12 may play roles in metastasis of epithelial ovarian cancer by promoting migration of tumor cells and endothelial cells.

Induction of apoptosis in human ovarian epithelial cancer cells by antisurvivin oligonucleotides.

Survivin, an anti-apoptosis gene that is abnormally overexpressed in a variety of human tumors, may play an important role in the carcinogenesis and drug resistance of cancer. This study was designed to explore the effects of liposome-survivin antisense oligonucleotide (Lip-ASODN) on the growth and apoptosis of human ovarian cancer cell lines, A2780 and SKOV3. To investigate the use of survivin as a therapeutic target on ovarian cancer, we carried out transfections with Lip-ASODN to induce apoptosis in ovarian cancer cell lines, A2780 and SKOV3. The expression of survivin mRNA and relative protein were evaluated separately by quantitative real-time RT-PCR and Western blot analysis. Cell proliferation inhibition was determined by methyl thiazolyl tetrazolium (MTT) assay, and the induced cell apoptosis was examined using flow cytometry (FCM) after Lip-ASODN transfection. Our results showed that the overexpression of survivin led to infinite carcino-proliferation, and survivin expression in the survivin-positive ovarian cancer cell line A2780 and SKOV3 cells was significantly and gradually reduced when transfected with Lip-ASODN at concentrations of 200, 400 and 600 nM by degrees. Lip-ASODN transfection induced greater apoptosis rates in the human ovarian cancer cell lines A2780 and SKOV3 (p<0.05). The growth inhibition and apoptotic rates of tumor cells change when treated with different concentrations of Lip-ASODN. The cell growth inhibition peak rate was reached when increasing Lip-ASODN concentration to 600 nM. Furthermore, time course evaluation showed that survivin protein expression was inhibited by Lip-ASODN within 12 h after transfection. We concluded that down-regulation of survivin by a targeted antisense oligonucleotide appears to be an effective gene therapy approach in the treatment of ovarian cancer.

[Predicting efficacy of neoadjuvant cheomotherapy on resectable stage IIIA non-small cell lung cancer by multi-gene expressions].

BACKGROUND & OBJECTIVE: Neoadjuvant chemotherapy for stage IIIA non-small cell lung cancer (NSCLC) remains controversial. The role of the expressions of P53, K-ras, HER2, vascular endothelial growth factor (VEGF), epidermal growth factor receptor (EGFR), CD44, and matrix metalloproteinase-9 (MMP-9) in predicting efficacy of neoadjuvant chemotherapy on stage IIIA NSCLC is still unclear although they have been found to be related to prognosis. This study was to determine the predictive effect of multi-gene expression on treatment outcome of neoadjuvant chemotherapy for resectable stage IIIA NSCLC. METHODS: Expressions of p53, K-ras, HER2, VEGF, EGFR, CD44, MMP-9 in the patients enrolled in the prospective randomized controlled trial were detected by immunohistochemistry. The treatment efficacies of combination (neoadjuvant chemotherapy combined with surgery) group (36 patients) and surgery alone group (32 patients) were compared. RESULTS: The high gene expression rate was 58.3% in combination group, and 40.6% in surgery alone group(P=0.145). In combination group, no significant difference of disease-free survival rate (P=0.903) and survival time (P=0.238) was found between patients with histopathologic regression and patients without histopathologic regression; high gene expression had no correlation with pathologic regression (P= 0.862); the mean disease-free survival time was significantly lower in high gene expression subgroup than in low gene expression subgroup [(14.1+/-9.8) months vs. (27.2+/-13.6) months, P=0.032]; the 2-year disease-free survival rate was 38.1% in high gene expression subgroup, and 46.7% in low gene expression subgroup (P=0.607). CONCLUSIONS: Pathologic regression after neoadjuvant chemotherapy has no correlation with disease-free survival rate and survial time. The high gene expression maybe indicate high risk of postoperative metastasis; the necessity of postoperative chemotherapy needs further study.

[Reducing the influence non-linearity of detecting array on absorption spectrum with curve fitting].

In a micro spectral analytical system, the non-linearity response of the photoelectronic detecting array pixel will cause the deformation of the spectrum, especially in the measurement of absorption spectrum. In this paper, the factors that cause the deformation were studied and a corresponding solution was proposed to reduce such deformation. By using curve fitting, the non-linearity of the pixel can be corrected. This method was adopted to correct the absorption spectrum obtained by a micro spectral analytical system and the result showed that it is a efficient way to correct the non-linearity in the pixel of photoelectronic detecting array.

[Experimental testing of micro biochemical analytical system].

A micro biochemical analytical system based on a micro fiber spectrometer is introduced. Experiment was carried out to calibrate and test the analysis system. In the experiment, the absorption spectra of Fe2+ -ferroin solution bodies with different concentrations were obtained. The working curve shows a fine linearity of the analysis system. The authors also compared the experimental results obtained from 722-spectrometer and those from our analysis system. It was shown that their system can meet the requirement of practical use. This system also has many advantages, such as real-time whole spectrum analyzing and small volume, and is an ideal instrument for biochemical analysis.

[Effect of natural killer cell line NK-92 against human ovarian carcinoma cells in vitro and in vivo].

OBJECTIVE: To study the cytotoxic activity of NK-92 cells irradiated against human ovarian cancer. METHODS: NK-92 cells were exposed to different doses of radiation and assayed for proliferation by a standard (3)H-thymidine incorporation assay and cell count by using trypan blue exclusion. The cytotoxic activity of NK-92 cells against targets was measured in a standard (51)Cr-release assay in vitro. The effectiveness of irradiated NK-92 cells on ovarian cancer was compared with the control group of cancers (without injection of irradiated NK-92 cells). RESULTS: (1) In vitro:The proliferation of NK-92 cells was inhibited by radiation of 4, 8 and 16 Gy, respectively. From the (3)H-thymidine incorporation data, irradiation by 4 Gy reduced cell proliferation to 29% of control, while 8 Gy reduced proliferation to 6%. The cytotoxicity of NK-92 cells at 4 Gy 2 days following irradiation was approximately 42%-62% for ovarian cancer cell HO-8910, while it was 33%-58% at 8 Gy. (2) In vivo: Tumor size in treatment group was (0.047 +/- 0.019) cm(3) on day 30 after inoculation, and (0.167 +/- 0.021) cm(3) on day 40 and (0.343 +/- 0.022) cm(3) on day 50, while the sizes were smaller in treatment group (P < 0.01). In addition, the tumor group animals died between 74-82 days after injection of HO-8910 cells, while the treatment group animals were alive over 120 days (P < 0.01). CONCLUSION: Our study indicates that injection of irradiated NK-92 cells may be a potentially effective treatment for human ovarian carcinoma.

[Inhibitory effect of coxsackie adenovirus receptor on invasive and metastatic phenotype of ovarian cancer cell line SKOV3].

BACKGROUND & OBJECTIVE: Coxsackie and adenovirus receptor (CAR) was originally identified as the cellular receptor of 2,5-type adenovirus. Recent researches found that CAR is a subordinate cell adhesion molecule and plays an important role in the malignant phenotype changes of tumor cells. This study was to explore inhibitory effect of CAR on invasive and metastatic phenotype of ovarian cancer cell line SKOV3. METHODS: The expression of CAR in ovarian cancer cell lines SKOV3, CAOV3, SW626, and A2780 was detected by quantitative reverse transcription-polymerase chain reaction (RT-RCR) and Western blot. SKOV3 cells were transfected with the eukaryotic expression plasmid containing full-length CAR cDNA. The positive clones were screened by G418, and identified by RT-PCR, Western blot and ADV/GFP infection assay. The biological behavior changes of positively transfected cells were assessed by colony formation assay and cell adhesion assay. RESULTS: The expression of CAR was down-regulated in the 4 ovarian cancer cell lines, among which SKOV3 cells had no CAR expression. The mRNA and protein levels of CAR were obviously higher in CAR-transfected SKOV3 cells than in untransfected cells and mock-transfected cells; the adhesion ability of CAR-transfected SKOV3 cells was obviously enhanced. The colony formation count was significantly lower in CAR-transfected SKOV3 cells than in untransfected cells and mock-transfected cells (25.3+/-8.9 vs. 88.8+/-14.0, 82.5+/-19.4, P<0.05). CONCLUSION: Transfected CAR gene may enhance the adhesion ability of ovarian cancer SKOV3 cells, therefore, inhibit the malignant metastatic phenotype of SKOV3 cells.

[Enhancive effect of histone deacetylase inhibitor trichostatin a on transfection efficiency of adenovirus in ovarian carcinoma cell line A2780].

BACKGROUND & OBJECTIVE: The presence of Coxsackie and adenovirus receptor (CAR) on target cell surface is required for efficient adenovirus transfection; lack or down-regulated expression of CAR on cancer cells is the main cause of inefficiency of adenovirus-based gene therapy. This study was to evaluate enhancive effect of trichostatin A (TSA), a histone deacetylase (HDAC) inhibitor, on the transfection efficiency of adenovirus in ovarian carcinoma cell line A2780, and explore its possible application to adenovirus-based gene therapy. METHODS: mRNA and protein levels of CAR on A2780 cells were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot before and after treatment of TSA. Transfection efficiency of adenovirus was valued by flow cytometry (FCM). In vitro antitumor effect of adenovirus/thymidine kinase (ADV/TK) was detected by MTT assay. RESULTS: After treatment of TSA, mRNA and protein levels of CAR on A2780 cells were obviously increased. Transfection rates of adenovirus were (1.24+/-0.14)% in untreated group, (7.58+/-0.32)% in 5 nmol/L of TSA treated group, and (7.94+/-0.28)% in 100 nmol/L of TSA treated groups. In vitro antitumor effect of ADV/TK was 4-10 folds in TSA (5 or 100 nmol/L) treated groups compared with that in untreated group. CONCLUSION: TSA could enhance transfection efficiency of adenovirus in ovarian carcinoma cells, and may be useful in gene therapy for ovarian carcinoma.

[Diagnosis and treatment of cervical anastomotic fistula contaminating mediastinum or pleural cavity after esophagogastrostomy: a report of five cases with literature review].

BACKGROUND & OBJECTIVE: A special kind of fistula will be formed when cervical gastroesophageal anastomotic fistula occurs and its contents contaminate mediastinum or pleural cavity after esophagogastrostomy. The diagnosis and treatment are difficult to made. It is very dangerous if the treatment is incorrect. This study was to analyze the causes, preventive measures, diagnosis methods and standard, and appropriate treatments of this kind of fistula. METHODS: Clinical data of 5 patients who suffered cervical anastomotic fistula contaminating mediastinum or pleural cavity after esophagogastrostomy, treated in our hospital, were retrospectively analyzed; the related literature was reviewed. RESULTS: The morbidity of this kind fistula was 1.83%(4/219) in our hospital. All the 5 patients suffered fistula 1-13 days after esophagogastrostomy; bronchopleural fistula occurred in 2 patients. The main causes of this kind of anastomotic fistula were hypertensive anastomosis, low anastomotic location, incomplete suture of thorax-esophagus outlet, defection of surgical technique, and so on. The fistula could be diagnosed correctly when the patient suffered high fever, dyspnea and thoracalgia after operation, with cervical anastomotic external fistula which could not been healed when the wound was opened, X-ray-showed widened mediastinum and hydropneumothorax, and drainage, esophagoraphy and CT-confirmed anastomotic fistula. All patients were cured in a short term with sufficient drainage and irrigation, enough nutrition, appropriate use of antibiotics, and pyothorax dissection and muscle flaps transplantation for bronchopleural fistula. CONCLUSION: Most cervical anastomotic fistulas happen 1-13 days after esophagogastrostomy with critical conditions; timely diagnosis and proper treatment including operation could shorten the disease course and greatly decrease the mortality.

[Postoperative mortality after gastrectomy for gastric cancer: analysis of 1142 cases].

OBJECTIVE: To analyze postoperative morbidity and mortality after gastrectomy for gastric carcinoma and identify main risk factors influencing mortality. METHODS: A total of 1142 patients with gastric cancer received gastrectomy between January 1989 and April 2004. The patients were divided into three groups according to different period, the first group (n=405): from January 1989 to January 1994; the second group (n=377): from February 1994 to January 1999; the third group (n=360): from February 1999 to March 2004. Postoperative morbidity and mortality were compared among three groups, the risk factors influencing postoperative mortality were determined by multiple logistic regression analysis. RESULTS: The total postoperative morbidity and mortality for all patients were 11.2% (128/1142) and 3.6% (41/1142), respectively. The postoperative morbidity was 13.1%, 10.1%, and 10.3% in the first, second, and third group respectively, there was no significant difference in morbidity among the three groups (P > 0.05). The postoperative mortality was 4.7%, 3.4%, and 2.5% respectively (P > 0.05), there was no significant difference in mortality among the three groups (P > 0.05). The most common postoperative complication was anastomotic leakage (24.2%, 31/128). The following clinicopathologic factors were found to be correlated with postoperative mortality: stage IV; palliative excision; multivisceral resection; and preoperative complications (P< 0.05). Multivariate analysis revealed that the extent of lymph node dissection or surgical procedure were not main risk factors influencing mortality. CONCLUSION: Patients with advanced gastric cancer have a high risk of postoperative mortality. Unnecessary lymph node dissection or multivisceral resection should be avoided for patients with stage IV gastric cancer.

[Arthroscopic transtibial double-bundle posterior cruciate ligament reconstruction using calcaneal-tendon allograft].

OBJECTIVE: To evaluate the surgical approach and the clinical outcome of arthroscopic transtibial double-bundle and double-tunnel posterior cruciate ligament(PCL) reconstruction using calcaneal-tendon allograft. METHODS: Ten patients with knee instability caused by PCL rupture were treated by arthroscopic PCL reconstructions using calcaneal-tendon allograft only by anterior approach between March, 2002 and April, 2003. First, the square bone block 10 mm in diamter, which is connected with a band of calcaneal tendon 12 mm in diameter, was fixed into the tibial tunnel. Then, the tendon was divided into two parts with a proportion of 3 : 2 (7 mm : 5 mm), which were fixed into the anterolateral (AL) and posteromedial (PM) tunnels in medial femoral condyle respectively. The knees were fixed with plaster slabs or braces for 4 weeks after operation. The periods of follow-up were 12 to 48 months (average, 32 months),and the outcome was evaluated by Lachman's test, posterior drawer test (PDT), and Lysholm scorses. RESULTS: Knee instability was seen no more in the 10 cases after operation. Recovery of stability in coronary axis was apparent. The results of Lachman's test and PDT were both positive in every case before operation, but became negative after operation. Extension and flexion of knee joints became normal. Lysholm scores rose from 58 (pre) to 93 (post). Joint function of nine cases reached excellent level, one case reached good level, and the total rate of excellence and good was 100%. Clinical observations suggested that immunological rejection was mild (only one patient had a low fever); meanwhile, no surgical complication happened. During 48 months, no ligament looseness was observed. Satisfactory clinical outcome was achieved in the whole group. CONCLUSION: Double-bundle and double-tunnel PCL reconstruction using calcaneal-tendon allograft is a good surgical approach, which not only avoids injuring the donor area, but also accords with the anatomical structure of PCL. The short-term outcome has been proved, but long-term outcome needs more observations.

[The effect of hIL-4 on the expression of E-selectin and ICAM-1 on bovine aortic endothelial cells].

AIM: To investigate the effect of human interleukin-4 (hIL-4) on the expression of E-selectin and ICAM-1 on bovine aortic endothelial cells(BAEC) activated with TNF-alpha. METHODS: BAEC were cultured in different concentrations of hIL-4 for 2 h followed by treatment with 4 microg/L TNF-alpha for 6 h or 18 h. The expression of E-selectin and ICAM-1 on BAEC was detected by cell-ELISA. The effect of hIL-4 on viability of BAEC was detected by MTT colorimetry. RESULTS: Pretreatment of BAEC with hIL-4 inhibited the expression of E-selectin and ICAM-1 on BAEC activated by TNF-alpha in a dose-dependent manner (P<0.01). Both TNF-alpha and hIL-4 had no effect on the viability of BAEC. CONCLUSION: hIL-4 can suppress the expression of E-selectin and ICAM-1 on BAEC activated by TNF-alpha, which may contribute to the xenotransplant immune tolerance associated with hIL-4.

[Retrospective observation of curative effects on MDS refractory anemia with combination of all-trans retinoic acid, 1, 25-dihydroxyvitamin D3 and androgen].

This study was aimed to examine whether a combination of all-trans retinoic acid (ATRA), 1, 25-dihydroxyvitamin D(3) and androgen possesses the therapeutic value for the MDS-refractory anemia (MDS-RA), and to analyze the mechanisms in detail. 62 cases receiving a scheme of combination of ATRA, 1, 25-dihydroxyvitamin D(3) and androgen (group A) were monitored. The remaining 33 cases (group B) were provided with vitamin supplementation, chalybeate drugs, and one or two of the combination. Bone marrow aspiration and biopsy were performed for collecting the specimens at the baseline and afterwards. The conditions of the patients were monitored by means of weekly complete blood counts and the monthly examination, including toxicity test, physical examination, electrocardiography, and biochemistry panel. The results showed that after treating for 8 weeks in group A, 4 out of 62 patients showed complete remission and 12 patients showed partial remission according to the defined response criteria, and 43 patients (69.35%) showed hematological improvement (HI). The further treatment for 16 out of 62 patients (25.81%), 13 failures (10 deaths, 2 RAEB and 1 RAEB-T) and 3 transformations (M(2), M(3), M(5)) with a median survival interval of 26.25 months, were observed and interrupted for some reasons. However, partial remission was observed only in 3 patients in group B, and HI amounted to 51.51%. Furthermore, the disease progression was observed in 12 out of 33 patients (36.36%) with a median survival interval of 16 months, 9 failures (including 6 deaths, 2 RAEB and 1 RAEB-T) and 3 transformations (M(2), M(3), M(4)). The overall ratios of survival for 3 and 5 years in group A, which received the combination, reached to 69.24% and 53.72% respectively, in comparison with 52.23% and 31.34% in the patients of group B (log-rank, P = 0.016). The following requirements, if were met, would be significant for prognosis: the combination regiment, no transformation, children, no complication, female, 90-120 g/L of hemoglobin concentration, normal cellular bone marrow and uni-cytopenias (P < 0.05). Moreover, Cox regression showed that therapy, transformation and age are all the independent factors (P < 0.05). It is concluded that the combination of above mentioned 3 drugs may be effective and safe treatment for the patients with MDS-RA. Its relevant mechanisms can be involved in the combination, that elicits a wide range of pharmacological effects, such as differentiation, anti-tumor-promotion, anti-apoptosis, anti-angiogenesis, anti-cachexia and immunoregulation.
