[Association between hypertensive cerebrovascular stroke and renin-angiotensin system gene polymorphism from Chinese cohort in Shanghai].

OBJECTIVE: To evaluate the association between hypertensive cerebrovascular stroke and renin-angiotensin system gene polymorphism from Chinese cohort. METHODS: The polymorphisms of angiotensinogen(AGT) and angiotensin-converting enzyme(ACE) from 257 cases of simple essential hypertension(EH) and 218 cases of hypertensive stroke(131 hemorrhagic cases and 87 ischemic cases were detected by PCR-RFLP. RESULTS: The frequencies of DD genotype and D allele of ACE gene in ischemic stroke were significantly higher than those in hemorrhagic stroke and EH, and the odds ratios(OR) of DD/II genotype were 3.25(2.20-4.79) and 2.87(2.03-4.06), while the OR of D/I allele were 1.83(1.38-2.43) and 1.69(1.27-2.24) respectively. Otherwise, there was no difference in frequency distribution of Met235Thr mutation polymorphism and DD+TT/non DD non TT combined among the three groups. CONCLUSION: The I/D polymorphism of ACE gene may be one of the risk factors and susceptible genetic markers for ischemic stroke with essential hypertension in Chinese.

The hyperintense lesions of multiple sclerosis seen on proton density- and T2-weighted MR images have important clinical and research roles in the diagnosis, follow-up, prognosis, and treatment of the disease.

[Correction of blepharophimosis with a temporal periosteal flap].

OBJECTIVE: To Provide a dependable and effective method for the treatment of blepharophimpsos. METHODS: In Blaskovics canthoplasty, a periosteum flap raised from the lateral orbit margin was turned over and fixed to the end of superiou tarsur. The operation was performed on 11 patients (13 lids), with blepharophimosis. Of whom 8 patients (8 lids) were serious blepharophimosis associated with cryptophthalmus or hollow socket; 3 patients (5 lids) were moderate. The epicanthus in 3 cases (4 lids) was corrected by Spaeth's technique. RESULTS: The follow-up period was 10.5 months in average. 10 lids of blepharophimosis were cured; 2 were improved significantly and 1 improved. No severe complications were found. CONCLUSIONS: The periosteal flap acts not only as a palpebral supporting tissue but also forms lateral ligament. The corrective effects of this method for blepharophimosis are fairly good.

[Land use dynamic monitoring supported by remote sensing and GIS--a case study in Kenli County of Yellow River delta].

By remote sensing both visual interpretation and computer aided classification, the land use information of Kenli County of Yellow River delta in 1981, 1991 and 1995 was compared and analyzed. The unifying land use dynamic monitoring classification system was made, and the methods of land use dynamic monitoring both on land use type areas and on their spatial distribution were proposed. The changes of land use type areas of Kenli County were analyzed, and the spatial dynamics of land use types from 1981 to 1995 was analyzed under GIS support. In addition, the land use changes of outdoor monitoring plots were analyzed, and the basic patterns of land use dynamics of Kenli County were proposed.

[Determination of S-100 beta protein in cerebrospinal fluid of patients with metastatic carcinoma to central nervous system].

OBJECTIVE: To investigate the concentration of S-100 beta protein in cerebrospinal fluid (CSF) for 25 patients with metastatic carcinoma to central nervous system (CNS), and to evaluate its clinical value. METHODS: S-100 beta protein of CSF was measured by ELISA for 25 patients with metastatic carcinoma to CNS and 20 patients without any CNS disease (control group). The patients were divided into three categories according to severity of disease: severe group, moderate group and mild group. Meanwhile, CSF cytology was also detected. RESULTS: The level of S-100 beta protein of all patients was higher than that of control group (P < 0.01), and severe group > moderate group (P < 0.01) > mild group (P < 0.01). The percentage of CSF lymphocyte in severe group was lower than that of mild group and control group (P < 0.05 and P < 0.01). CONCLUSIONS: CSF S-100 beta protein level of patients with metastatic carcinoma may be related to the severity of their brain insult.

[Transposition of bilateral pedicled adrenal gland for the treatment of Cushing's disease].

OBJECTIVE: To study the therapeutic method of Cushing's disease. METHODS: 16 patients with Cushing's disease were treated by transposition of bilateral pedicled adrenal gland into the dorsal subcutaneous tissues. RESULTS: These patients were followed up for 4 to 48 months. No relapse was observed and 3 patients with hypoadrenocorticism were given small doses of steroid. CONCLUSIONS: This procedure may be a rational and satisfactory technique for the treatment of Cushing's disease.

[Relationship between estrogen receptor and Graves' disease].

OBJECTIVE: To study the relation of expression of estrogen receptor (ER) in thyroid tissues with Graves' disease (GD). METHODS: ER was detected in the tissues of 40 patients with GD and 15 patients with normal thyroid by immunohistochemical method. Gene expression of ER was studied in frozen slides of 26 GD thyroids and 10 normal thyroids by fluorescence in situ hybridization (FISH) together with image analysis system. RESULTS: In immunohistochemical detection, the positive rate of ER in GD group was 70% (28/40), but 13.3% (2/15) in the control group (P < 0.01). The positive degree of ER in female with GD was higher than that in male with GD (P < 0.05). The positive rate of ER in the female GD group below 35 years was 85.0% (17/20) and 50% (5/10) in the female GD group above 35 years (P < 0.05). In FISH, the optical density (A) value of the female GD group was 30.14 +/- 10.74, and in the male GD group 21.06 +/- 7.71, but in the control group it was 16.91 +/- 5.30 (P < 0.01). The A value of the female GD group below 35 years was 33.85 +/- 10.10, and 23.24 +/- 7.48 in the female GD group above 35 years (P < 0.05). CONCLUSIONS: The increasing of ER content is related to the development of GD. Estrogen may be a promoting factor for GD. The sex and age differences in GD development are associated with the effects of estrogen and ER.

[Study on the hepatitis virus infection among medical professionals].

OBJECTIVE: In order to find out the present situation of hepatitis virus infection among medical professionals. METHODS: 140 medical professionals were studied on their serum HBsAg, anti - HBs, anti - HCV using EIA, and HEV - IgG, HGV - IgG with ELISA. RESULTS: The infection rates of HBsAg, HCV, HEV and HGV of medical professionals were 7.8%, 1.4%, 7.1% and 7.1% respectively with an overall infection rate of 23.5%. The infection rates for the clinical group and the non - clinical group were 33.3% and 12.3% respectively [with significant differences (P < 0.01)]. The infection rates of HEV and HGV for the clinical group were 8.0% and 12.0% respectively. It was found no superimposing infection in non - clinical group but was in clinical group. CONCLUSION: The result suggested that the infection rate of hepatitis virus in the medical professionals in the clinical group was higher than in the non - clinical group.

[The lymphocyte subset and blood interleukin-2 level in children with Henoch-Schönlein purpura before and after cimetidine treatment].

The objective of this study was to evaluate the effect of cimetidine treatment on cellular immunity in children with Henoch-Schönlein purpura. T cell subset was detected by double marker flow cytometry and the level of IL-2 in peripheral blood was measured by MTT method, before and after cimetidine treatment. The results showed that CD3+, CD4+ T cell subset, CD4+/CD8+ ratio, and blood IL-2 level increased after the treatment (P < 0.05). There was no significant change in CD8+ T subset after the treatment. These findings indicate that cimetidine can promote T lymphocyte proliferation and differentiation, especially TH1 lymphocyte activity.

[Effects of homoharringtonine on proliferation of human retro-ocular fibroblasts].

This is a study amied at the effects of homoharringtonine on the proliferation of human retro-ocular fibroblasts. The human retroocular fibroblasts (HROFb) were cultured and MTT colorimetry assay was used to determine the proliferation of HROFb. The influence of homoharringtonine on the proliferation of HROFb was observed in vitro. The results showed that MTT colorimetry assay could precisely reflect the proliferation of HROFb. The absorbance was directly proportional to the number of the cells (r = 0.996, P < 0.001). The proliferation of HROFb was inhibited by the homoharringtonine and the cell proliferation percentage was correlated positively to the homoharringtonine level (r = 0.6932, P < 0.001).

[Interaction between biofilm formed by Pseudomonas aeruginosa and antibacterial agents].

UNLABELLED: We hypothesized that bacterial biofilm formation could be an important factor that make some infection intractable. Interaction of P. aeruginosa biofilm with antibactial agents was examined in vitro. METHODS: P. aeruginosa incubated for 6 days in a saline-teflon system formed biofilm on the surfaces of the teflon piece. The biofilm were characteried by dense colonization of bacteria and thick membranous structure that covered the colonies, it was observed with a scanning electron microscopy. CPFX, CAM, ROX and Herba Andrographitis were added to the biofilm bacteria. RESULTS: The rate of survial of the floating bacteria decreased to 0.02% after 4 hours of contact with 2MIC of Ciprofloxacin (CPFX), but that of biofilms was 41% When 10 micrograms/mL of Clarithromycin (CAM per mL was combined with CPFX, the survial rate for the biofilm bacteria decreased to 0.2%. When 12 micrograms/mL of Roxithromycin (ROX) was combined with CPFX, the survial rate for the biofilm bacteria decreased to 0.5%. When 0.05 g of Burm. f. per mL was combined with CPFX the survial rate for the biofilm bacteria decreased to 2.7%. CONCLUSION: We conclude that biofilm bacteria are much more resistant to CPFX than the floating bacteria. The combination of CPFX and CAM, ROX HA were more effective in decreasing the biofilm bacteria survial rate than CPFX alone. The results suggest that the combination of CPFX and CAM, ROX, HA can be one of the therapeutic manauvers against biofilm bacteria.

[Ca II triplet equivalent widths of spiral galaxy M31 and elliptical galaxy M32].

The Calcium II triplet (Ca II T) lines in the near infrared spectral region is very important feature for late type stars, which serves as an important metallicity indicator for such stars. This is also true for stellar systems composed mainly of late type stars, such as old stellar clusters and galaxies. In this work, we made observations of the very famous spiral galaxy M31 and its dwarf elliptical companion M32, using the OMR spectrograph attached to the 2.16-meter telescope in Beijing Astronomical Observatory. Near-infrared spectra of the nuclear and disk regions on M31, as well as two slices along the major and the minor axis of M32, are derived. Equivalent widths of the Ca II triplet lines are measured using normal astrophysical spectrum analyzing techniques. The results are fundamental data for these two galaxies, which can be applied to researches for stellar population analysis of these galaxies.

Anti-CD154 or CTLA4Ig obviates the need for thymic irradiation in a non-myeloablative conditioning regimen for the induction of mixed hematopoietic chimerism and tolerance.

BACKGROUND: Thymic irradiation (TI) or repeated administration of T cell-depleting monoclonal antibodies (TCD mAbs) is required in a previously described non-myeloablative regimen allowing allogeneic marrow engraftment with stable mixed chimerism and tolerance. As both treatments might be associated with toxicity in the clinical setting, we evaluated whether T-cell costimulatory blockade could be used to replace them. METHODS: C57BL/6 mice received depleting anti-CD4 and anti-CD8 mAbs on day -5, 3 Gy whole body irradiation (day 0), and 15x10(6) fully MHC-mismatched, B10.A bone marrow cells. In addition, hosts were injected with an anti-CD154 mAb (day 0) and/or CTLA4Ig (day +2). Chimerism in peripheral blood was followed by flow cytometric (FACS) analysis, and tolerance was assessed by skin grafting, and also by mixed lymphocyte reaction (MLR) and cell-mediated lympholysis (CML) assays. The frequency of certain Vbeta families was determined by FACS to assess deletion of donor-reactive T cells. RESULTS: Chimerism was transient and tolerance was not present in animals receiving TCD mAbs on day -5 without costimulatory blockade. The addition of anti-CD154 and CTLA4Ig, alone or in combination, reliably permitted induction of high levels of stable (>6 months) multi-lineage chimerism, with specific tolerance to skin grafts and donor antigens by MLR and CML assays. Long-term chimeras showed deletion of donor-reactive CD4+ peripheral blood lymphocytes, splenocytes, and mature thymocytes. Administration of TCD mAbs only 1 day before bone marrow transplantation plus anti-CD154 also allowed induction of permanent chimerism and tolerance. CONCLUSIONS: One injection of anti-CD154 or CTLA4Ig overcomes the need for TI or prolonged host TCD in a preclinical model for the induction of mixed chimerism and deletional tolerance and thus further decreases the toxicity of this protocol. Achievement of tolerance with conditioning given over 24 hr suggests applicability to cadaveric organ transplantation.

Investigations into the thermodynamics of polypeptide interaction with nonpolar ligands.

In this paper, we describe a general procedure to evaluate the thermodynamics of the interaction between polypeptides and hydrophobic ligands in the presence of aquo-organic solvent mixtures. These studies address experimental requirements for the determination of the linear free energy relationships, derivation of partition coefficients or other extrathermodynamic parameters such as contact areas, or assessment of the conformational changes that may occur when polypeptides or proteins interact with immobilized nonpolar ligands. Not unexpectedly from thermodynamic arguments, the trends and magnitudes of free energy parameters, such as the enthalpy of association, as previously derived in many studies from gradient elution reversed-phase high-performance liquid chromatographic (RP-HPLC) measurements are often different from the data for the same parameters derived from equilibrium binding or microcalorimetric determinations. To reconcile these divergencies and to more closely examine the thermodynamic basis of the interaction of polypeptides with nonpolar ligands, the dependency of the logarithmic capacity factor, ln k', on temperature, T, for several polypeptides (bombesin, beta-endorphin, glucagon) have been investigated using a n-butylsilica and acetonitrile-water or methanol-water mixtures of defined solvent compositions. With low-pH, acetonitrile-water mixtures, the van't Hoff plots, i.e., the plots of ln k' versus 1/T, were nonlinear over the range of T = 278-358 K, although within a narrow temperature range, e.g., from T = 278-308 K, the experimental data for these polypeptides could be approximated by a linear relationship. This nonclassical van't Hoff behavior was associated with interactive processes that involved temperature-dependent enthalpic, entropic, and heat capacity changes. In contrast, with low-pH, methanol-water mixtures, the van't Hoff plots showed dependencies that were essentially linear over the range of T = 278-358 K. The slopes of the van't Hoff plots with acetonitrile-water and methanol-water mixtures at a defined T value and solvent composition were significantly larger than those found for the corresponding experiments carried out under gradient elution RP-HPLC conditions. From these plots of ln k' versus 1/T, the changes in the apparent enthalpy of association (delta H++assoc) and the apparent entropy of association (delta S++assoc) for the interaction of these polypeptides with the solvated n-butyl ligands at different T and solvent compositions have been determined. For these polypeptides, both delta H++assoc and delta S++assoc exhibited linear dependencies on the volume fraction, phi, of the organic solvent over a narrow range of T, but the slopes of these plots were dependent on the T range examined. The dependencies of the slope term, S, and the intercept term, ln ko, derived from the plots of ln k' versus phi as a function of T, have also been investigated. A new relationship linking the S values with delta H++assoc and delta S++assoc as a function of T and phi has been derived and validated. In addition, the relationship between S, delta H++assoc, delta S++assoc, the apparent change in heat capacity, delta C++assoc, and the accessible surface area, delta Atot, of these polypeptides has been examined, thus providing a linkage of these thermodynamic and extrathermodynamic parameters to the partition coefficient, P, and the molecular properties of these polypeptides. The results confirm that entropy-enthalpy compensation effects participate in the interaction of polypeptides with hydrophobic ligands. This investigation has confirmed that the use of solvent-water mixtures of defined composition, rather than the more convenient practice of using gradient elution methods, is essential if thermodynamically consistent values of the binding affinities and partition coefficients are to be quantitatively derived. Sequence analysis and expression of the aspartokinase and aspartate semialdehyde dehydrogenase operon from rifamycin SV-producing amycolatopsis mediterranei.

A approximately 4.8 kb KpnI fragment, from the upstream region of the methylmalonyl-CoA mutase gene (mutAB) of rifamycin SV-producing Amycolatopsis mediterranei, was cloned and partially sequenced. Codon preference analysis showed three complete ORFs. ORF2 is internal to ORF1, and encodes a polypeptide corresponding to 172 amino acids, whereas ORF1 encodes a polypeptide of 421 amino acids. They were identified as the encoding genes of aspartokinase alpha- and beta-subunits by comparing the amino acid sequences with those in the database. The downstream ORF3, whose start codon was overlapped with the stop codon of both ORF1 and ORF2 by 1 bp, was identified as the aspartate semialdehyde dehydrogenase gene (asd), encoding a polypeptide of 346 amino acids. Subclones containing either the ask gene or the asd gene were constructed, in which the genes could be expressed under Lac promoters. Two subclones could transform E. coli CGSC 5074 (ask-) and E. coli X6118 (asd-) to prototrophy, supporting the functional assignments. Southern hybridisation indicated that the approximately 4.8 kb sequenced region represented a continuous segment in the A. mediterranei chromosome. It is concluded that ask and asd genes are present in an operon in A. mediterranei, and therefore that organisation of these two genes is the same as in most gram-positive bacteria, such as Mycobacteria, Corynebacterium glutamicum and Bacillus subtilis, but is different from Streptomyces akiyoshiensis.

Determination of polyphenols by high-performance liquid chromatography with inhibited chemiluminescence detection.

A chemiluminescence reaction detector was developed for the detection of polyphenols separated by HPLC based on the inhibition of chemiluminescence from the luminol-potassium hexacyanoferrate(III) reaction by polyphenols. The separation was carried out on a RP-C18 column at 37 degrees C by using stepwise gradient elutions. The detection limits are in the range of 6.8 x 10(-7)-2.0 x 10(-9) g/ml for catechol, protocatechuic acid, chlorogenic acid, rutin, resorcinol, hydroquinone and p-tert.butylpyrocatechol. The method is sensitive, selective, fast and simple. It has been successfully applied to the determination of chlorogenic acid and rutin in real tobacco samples.
