[The relationship between p53 and PCNA in germ cell carcinoma].

To analyse the relationship between cellular proliferating status, p53 expressions and tumor biological characteristics, immunohistochemistry was used in 13 cases of testicular germ cell carcinoma paraffin section. The results show that PCNA-LI is higher significantly in invasive tumor group than in noninvasive group (P < 0.05). Although there were no significant differences of PCNA-LI, p53 between seminoma and nonseminoma, two parameters in the latter group were somewhat higher. There was a correlation of PCNA-LI with p53 in nonseminoma group (r = 0.64). It is concluded that PCNA-LI and p53 are both correlated positively with tumor differentiation.

[Radical operation for Budd-Chiari syndrome under extracorporal circulation with deep hypothermia and circulatory pause].

The authors designed a new operation for the treatment of Budd-Chiari syndrome under deep hypothermia anesthesia with extracorporeal circulation, opening the superhepatic inferior vena cava (IVC) when the circulation was paused. Then the diseased focuses of IVC and HVs were resected under direct bloodless vision. After the incision of IVC was sutured, the normal circulation restored. From September 1993 to November 1995, 11 cases were treated in our hospital. All patients are well postoperatively, and followed up from 5 to 33 months. Ultrasonography revealed patent IVC and hepatic veins in all cases.

[Allotransplantation of fetal adrenal capsules for treating steroid deficiency].

A man suffering from bilateral adrenal carcinoma was treated by total adrenalectomy and steroids substitution. One and a half months later, two fetal adrenal capsules were grafted to his greater omentum. No immunosuppressive agent was given. From the fourth month exogenous steroid dose was gradually reduced. Five months after transplantation, substitution therapy was discontinued. The patient showed no signs of steroid deficiency and all laboratory studies were normal. Seven months later, the patient died of metastasis of the carcinoma. Pathological examination showed that much adrenal cortex had regenerated in the greater omentum. The findings suggest that this method for treating chronic steroid deficiency is simple and successful.

[The pathogenetic influence of tubulointerstitial lesions on the glomerular function in IGg nephropathy: a study of 90 cases].

To evaluate the significance of tubulointerstitial lesions in IgA nephropathy, clinicopathological studies were performed on 90 cases of this disease. The degree of tubulointerstitial lesions was assessed semiquantitatively by light microscopic observation. The relation between glomerular filtration rate (GFR) and multiple clinicopathological factors was analysed by correlation analysis and logistic regression. The results showed that the degree of renal impairment at the time of biopsy was clearly correlated with and largely determined by the tubulointerstitial lesions, in particular the density of interstitial infiltration by lymphocytes and monocytes/macrophages. It is indicated that tubulointerstitial lesions play an important role in the pathogenesis of renal impairment in IgA nephropathy.

Sequences of the cDNAs encoding the heavy- and light-chain Fab region of an antibody to the phenylurea herbicide diuron.

The cDNAs from a hybridoma (mAb 481.1) specific for diuron, a widely used phenylurea herbicide, were cloned into the phage display vector pComb8. Antigen-binding clones were selected by panning on diuron-hapten-BSA conjugates. The nucleotide and deduced amino-acid sequences encoding the Fab regions of the light (kappa) and heavy (gamma) chains were determined. The light chain was from mouse kappa chain subgroup III and the heavy chain was a member of the mouse H chain subgroup III(d).

Tumor necrosis factor alpha increases tyrosine phosphorylation of a 23-kDa nuclear protein in U937 cells through ceramide signaling pathway.

In many tumor cell lines, tumor necrosis factor alpha (TNF alpha) causes profound alterations in nucleus including chromatin condensation and DNA fragmentation. However the mechanism is largely unknown. Here we demonstrate that TNF alpha treatment of U937 cells increased tyrosine phosphorylation of a 23-kDa nuclear protein (P23) maximally by 13-fold, which occurred after 2.5 h treatment concomitantly with occurrence of DNA fragmentation. Tyrosine kinase inhibitor, herbimycin A, inhibited either tyrosine phosphorylation of P23 or DNA fragmentation, suggesting that the P23 phosphoprotein may be involved in the TNF alpha-induced changes in nucleus. Increase of intracellular Cer level by hydrolysis of cell surface SM with bacterial sphingomyelinase or addition of either C2-Cer or natural Cer also resulted in increase of P23 tyrosine phosphorylation, which was detectable at 1.5 h treatment and reached maximum after 2 h treatment, indicating that P23 phosphoprotein may be a downstream effector of Cer signaling pathway. Our data provide further evidence that Cer signaling pathway may mediate signal transduction of TNF alpha.

Identification of a novel class of mammalian Fc gamma receptor.

A cDNA encoding an Ig receptor that conferred the ability to bind erythrocytes sensitized with IgG2, but not IgG1, was cloned by screening a cattle alveolar macrophage library, made in the vector pCDM8, expressed in COS-7 cells. A search of the PIR database indicated a greater level of similarity with human Fc alpha R than with any other FcR. The percentage of identical amino acids was 41% and nucleotides 56%. This high similarity is between the extracellular and transmembrane domains; the cytoplasmic tails are unrelated. Similarities with human Fc gamma RI, Fc gamma RII, Fc gamma RIII, Fc epsilon RI, or bovine Fc gamma RII were less than 28%. COS-7 cells transfected with the cloned plasmid bound erythrocytes specifically sensitized with IgG2 but not with IgG1. In tests with heat-aggregated bovine Igs, IgG2 purified from serum bound to transfected COS-7 cells but IgG1 from serum and IgA from tracheobronchial secretions did not. Human serum IgA also failed to bind to transfected COS-7 cells although it did bind to bovine neutrophils and monocytes. Only aggregated bovine IgG2 inhibited the binding of IgG2-sensitized erythrocytes to COS-7 cells transfected with the plasmid. These observations indicate that the FcR bound IgG2 specifically and was not a receptor for IgA. Thus, this receptor, which we have named bovine Fc gamma 2R, represents a novel class of mammalian Fc gamma R, the evolution of which is likely to have been influenced by the truncated hinge of the bovine IgG2 molecule. Analysis of genetic divergence of the genes encoding bovine and human Fc gamma R and Fc alpha R indicated that the novel bovine gene and the human Fc alpha R gene probably evolved from a common ancestor, which is not shared by other Fc gamma R.

Inhibition of tumor necrosis factor alpha- and ceramide-induced internucleosomal DNA fragmentation by herbimycin A in U937 cells.

In many tumor cell lines, tumor necrosis factor alpha (TNF alpha) causes apoptosis with characteristic internucleosomal DNA fragmentation. However, the mechanism is largely unknown. Here we examined the involvement of protein tyrosine kinases by using their inhibitors. Among various tyrosine kinase inhibitors tested, only herbimycin A was found to inhibit internucleosomal DNA fragmentation but not apoptotic morphological changes and cell death induced by TNF alpha in U937 cells. Herbimycin A was able to block DNA fragmentation when it was added to the cell culture as late as 1.5 h after TNF alpha treatment. These results demonstrate that herbimycin A selectively inhibits a later event involved in the process of apoptois that results in internucleosomal DNA fragmentation. Sphingomyelinase and ceramide (Cer) induced internucleosomal DNA fragmentation was also inhibited by herbimycin A, supporting the hypothesis that Cer may be a novel second messenger mediating the cytotoxic effect of TNF alpha.

Crystal structure of the cys2 activator-binding domain of protein kinase C delta in complex with phorbol ester.

Protein kinase Cs (PKCs) are a ubiquitous family of regulatory enzymes that associate with membranes and are activated by diacylglycerol or tumor-promoting agonists such as phorbol esters. The structure of the second activator-binding domain of PKC delta has been determined in complex with phorbol 13-acetate, which binds in a groove between two pulled-apart beta strands at the tip of the domain. The C3, C4, and C20 phorbol oxygens form hydrogen bonds with main-chain groups whose orientation is controlled by a set of highly conserved residues. Phorbol binding caps the groove and forms a contiguous hydrophobic surface covering one-third of the domain, explaining how the activator promotes insertion of PKC into membranes.

Plasmid DNA entry into postmitotic nuclei of primary rat myotubes.

These studies were initiated to elucidate the mechanism of DNA nuclear transport in mammalian cells. Biotin- or gold-labeled plasmid and plasmid DNA expression vectors for Escherichia coli beta-galactosidase or firefly luciferase were microinjected into the cytoplasm of primary rat myotubes in culture. Plasmid DNA was expressed in up to 70% of the injected myotubes, which indicates that it entered intact, postmitotic nuclei. The nuclear transport of plasmid DNA occurred through the nuclear pore by a process common to other large karyophilic macromolecules. The majority of the injected plasmid DNA was sequestered by cytoplasmic elements. This understanding of plasmid DNA nuclear transport provides a basis for increasing the efficiency of gene transfer.

Induction of apoptotic DNA fragmentation and cell death by natural ceramide.

We have found that a solvent mixture of ethanol and dodecane was able to disperse natural ceramide (Cer) into aqueous solution. In U937 cells, addition of natural Cer to medium caused a characteristic pattern of DNA fragmentation, which is indistinguishable from that caused by TNF alpha. The effective concentration of Cer is as low as 25 nM. The present study provides direct evidence that natural Cer functions as a second messenger mediating TNF alpha-induced DNA fragmentation. The use of this solvent to deliver hydrophobic natural Cer to cells will contribute to the elucidation of the biological function of Cer.

A morphometric study on the endometrial activity of women before and after one year with LNG-IUD in situ.

The endometrium was studied by morphometry in 34 healthy women before and after one year's use of an IUD releasing levonorgestrel at a rate 20 micrograms/24 hr (LNG-IUD-20). The effect was compared with that of an IUD releasing 2 micrograms levonorgestrel/24 hr (LNG-IUD-2). The appearance of suppressed endometrial proliferative activity and increased inflammatory reaction was similar in the two experimental groups, suggesting that the morphological alterations were independent of the LNG dosage. The result of this study, combined with clinical data, suggests that LNG-IUD-20 could replace LNG-IUD-2 without any additional side effects. The efficacy of LNG-IUD-20 is also likely to last for a longer period than the LNG-IUD-2.

Nucleotide sequence analysis of HLA-B*1523 and B*8101. Dominant alpha-helical motifs produce complex serologic recognition patterns for the HLA-B"DT" and HLA-B"NM5" antigens.

Assigning a precise serologic specificity to the class I HLA-B"NM5" and HLA-B"DT" molecules has proven difficult, with patterns of serologic cross-reactivity suggesting that NM5 is most like antigens in the B5 CREG and that DT is either B7 or B40 like. To better understand the relationship these antigens share with other HLA-B molecules we determined the nucleotide sequence of the alleles encoding HLA-B"NM5" and HLA-B"DT". Sequencing results show that NM5 shares the most overall sequence homology with the B70 antigens and that differences at the alpha-helical Bw4/Bw6 epitope preclude serologic cross-reactivity between NM5 and the B70 antigens. Accordingly, NM5 has been assigned the name B*1523. The strong serologic impact of helical sequence conservations and variations is reiterated for the class I HLA-B"DT" molecule. Comparative analysis demonstrates that sequence conservations in the first domain's alpha-helix stimulate cross-reactivity between HLA-B"DT" and HLA-B7, whereas epitopes conserved in the second domain's alpha-helix impel cross-reactivity between HLA-B"DT" and HLA-B48. To convey the unique lineage of this hybrid B7/B48 molecule the name HLA-B*8101 has been assigned to HLA-B"DT".

Excitation and inhibition by cisapride of spontaneous and evoked electrical responses in smooth muscles of guinea-pig stomach.

In isolated circular smooth muscles of the guinea-pig stomach, cisapride depolarized the membrane, increased the amplitude and interval of slow waves, and enhanced the cholinergic excitatory junction potential, with no change in the nonadrenergic noncholinergic inhibitory junction potentials. Methysergide mimicked the excitatory actions of cisapride on junction potentials. Results showed that cisapride has dual actions on gastric muscles; excitation by facilitating the release of ACh possibly via 5-HT receptor blockade and also by depolarizing the smooth muscle membrane, and inhibition by reducing the frequency of slow waves, possibly by acting directly on the pacemakers.

Characterization and distribution of a cloned rat mu-opioid receptor.

We have cloned and expressed a rat brain cDNA, TS11, that encodes a mu-opioid receptor based on pharmacological, physiological, and anatomical criteria. Membranes were prepared from COS-7 cells transiently expressing TS11 bound [3H]diprenorphine with high affinity (KD = 0.23 +/- 0.04 nM). The rank order potency of drugs competing with [3H]diprenorphine was as follows: levorphanol (Ki = 0.6 +/- 0.2 nM) approximately beta-endorphin (Ki = 0.7 +/- 0.05 nM) approximately morphine (Ki = 0.8 +/- 0.5 nM) approximately [D-Ala2, N-Me-Phe4,Gly-ol5]-enkephalin (DAMGO; Ki = 1.6 +/- 0.5 nM) uch much greater than U50,488 (Ki = 910 +/- 0.78 nM) > [D-Pen2,5]- enkephalin (Ki = 3,170 +/- 98 nM) > dextrorphan (Ki = 4,100 +/- 68 nM). The rank order potencies of these ligands, the stereospecificity of levorphanol, and morphine's subnanomolar Ki are consistent with a mu-opioid binding site. Two additional experiments provided evidence that this opioid-binding site is functionally coupled to G proteins: (a) in COS-7 cells 50 microM 5'-guanylylimidodiphosphate shifted a fraction of receptors with high affinity for DAMGO (IC50 = 3.4 +/- 0.5 nM) to a lower-affinity state (IC50 = 89.0 +/- 19.0 nM), and (b) exposure of Chinese hamster ovary cells stably expressing the cloned mu-opioid receptor to DAMGO resulted in a dose-dependent, naloxone-sensitive inhibition of forskolin-stimulated cyclic AMP production. The distribution of mRNA corresponding to the mu-opioid receptor encoded by TS11 was determined by in situ hybridization to brain sections prepared from adult female rats. The highest levels of mu-receptor mRNA were detected in the thalamus, medial habenula, and the caudate putamen; however, significant hybridization was also observed in many other brain regions, including the hypothalamus.

A plasmid-based self-amplifying Sindbis virus vector.

Sindbis virus was used as a self-amplifying eukaryotic expression vector. A recombinant cDNA genome of this (+)-strand RNA virus was placed under the transcriptional control of a Rous sarcoma virus LTR (RSV) promoter. Transfection of this plasmid construct into mammalian cell lines (3T3, HepG2, and 293 cells) resulted in expression of the luciferase reporter gene. High-expression levels were also measured after transfection into primary rat myoblasts. In differentiated myotubes, expression levels generated by the Sindbis virus vector were up to 200 times higher than those obtained with a conventional RSV expression vector. In vivo expression was detected after injection of plasmid DNA into mouse quadriceps. In vivo expression was transient and undetectable by day 16. This self-amplifying expression vector can be used for generating high-level expression of transgenes in vitro and in vivo. Its transient nature in vivo could allow for safe, short-term delivery of gene products in gene therapy protocols. It should facilitate the study of Sindbis and other RNA viruses.

The hydrolysis of cell surface glycosphingolipids by endoglycoceramidase reduces epidermal growth factor receptor phosphorylation in A431 cells.

This paper presents a new method to evaluate the biological significance of glycosphingolipids (GSLs) using a GSL-specific enzyme, endoglycoceramidase (EGCase), by which GSL-sugar chains are removed from the cell surface of living cells. In this report, the effects of EGCase on epidermal growth factor (EGF)-dependent tyrosine-specific EGF receptor (EGFR) phosphorylation of A431 cells are described. After treatment of A431 cells with EGCase II (20 mU/ml) in the presence of the activator for 12 h, all acidic GSLs tested were reduced to approximately 70% of control, but no hydrolysis occurred on Gal alpha 1,4Gal beta 1,4Glc beta 1,1Cer (Gb3Cer) and GalNAc beta 1,3Gal alpha 1,4Gal beta 1,4Glc beta 1,1Cer (Gb4Cer). In plasma membrane fractions of A431 cells, the reduction of gangliosides by EGCase II was found to be much faster than that of intact cells and reached 54.8% reduction of total gangliosides after 2 h incubation with the enzyme. EGF-dependent phosphorylation of EGFR of A431 cells was inhibited by the reduction of cell surface GSLs by EGCase when either intact A431 cells or their plasma membrane fractions were used, while EGF binding to their receptors was not changed. Neither hydrolysis of cell surface GSLs nor reduction of EGFR phosphorylation occurred when A431 cells were incubated with the activator or EGCase alone. The exogenous addition of ceramides or sphingosines, and treatment of cell membranes with sphingomyelinase, had no effect on the EGFR phosphorylation of purified membrane fractions, while the inhibitory effect of EGCase II on EGFR phosphorylation was restored by the addition of GM3-sugar chains, but not by lactose or sialic acid.(ABSTRACT TRUNCATED AT 250 WORDS)

HIV risk behaviors but absence of infection among drug users in detoxification centers outside Yunnan province, China, 1993.

OBJECTIVES: To date, HIV spread in China has occurred principally among injecting drug users (IDU) in remote border regions of Yunnan province. We therefore sought to better understand the risks for and prevalence of HIV infection among drug users in parts of China outside Yunnan province. METHODS: A behavioral survey of drug use and AIDS-related knowledge was conducted among all consenting drug users who entered treatment from 1 November to 31 December 1993 in seven provincial drug detoxification centers outside Yunnan province. After giving verbal informed consent, all drug users were tested for HIV. RESULTS: Of the 1293 study participants, 207 (16%) reported injecting drugs. The proportion of IDU among all drug users varied widely by region, from 1% in Sha'anxi and Gansu provinces in the northwest region to 58% in Guangxi province in the south. IDU were more likely than non-IDU to be single and unemployed or self-employed, but did not differ in other demographic aspects. Among all drug users, 2% reported sharing needles without cleaning equipment, while 5% shared with some cleaning. Although 1060 (82%) drug users had heard of AIDS and most knew about its sexual (79%), parenteral (77%), and perinatal (60%) modes of transmission, many had misconceptions about its spread by casual contact. Of the 207 IDU tested for HIV, none were HIV-positive (95% confidence interval, 0--1.4). CONCLUSION: The absence of HIV infections detected in this study suggests that the prevalence of HIV is currently low among IDU in China outside the Yunnan province. However, the behavior of these IDU puts them at high risk for HIV infection. Prevention efforts are needed to prevent the spread of HIV among IDU throughout China and to avoid the experience of neighboring countries in Asia.
