Respiratory syncytial virus infection of human respiratory epithelial cells up-regulates class I MHC expression through the induction of IFN-beta and IL-1 alpha.

CD8+ T cells mediate some of the damage to the lung epithelium following respiratory syncytial virus (RSV) infection. Since CD8+ T cells recognize antigen-laden class I MHC molecules on the target cells, we examined in this study the expression of class I MHC by RSV-infected respiratory epithelial cells. Respiratory epithelial cell lines and bronchial epithelial cells from normal human tissue responded to RSV infection with an increased expression of class I MHC as determined by flow cytometry and immunoprecipitation of class I MHC from metabolically radiolabeled cells. The increase in class I MHC expression was dependent on infectious, replicating virus. UV-irradiated culture supernatants from RSV-infected A549 cells, when added to fresh A549 cell cultures, induced an increase in class I MHC expression by those cells. The class I MHC increasing activity within supernatants from A549 cells was due, in large part, to IFN-beta, and to a lesser extent to IL-1 alpha. The addition of neutralizing Abs to both cytokines completely blocked the increase in class I MHC expression by cells treated with the above-mentioned supernatants. These results demonstrate that RSV infection elicits IFN-beta production by respiratory epithelial cells, which in turn leads to an increase in their synthesis of class I MHC, which would facilitate their recognition and lysis by RSV-specific CD8+ T cells.

An improved green fluorescent protein gene as a vital marker in plants.

A synthetic green fluorescent protein (GFP) gene (pgfp) was constructed to improve GFP expression in plants. Corn and tobacco protoplast transient assays showed that pgfp gave about 20-fold brighter fluorescence than the wild-type gene (gfp). Replacement of the serine at position 65 with a threonine (S65Tpgfp) or a cysteine (S65Cpgfp) yielded 100- to 120-fold brighter fluorescence than wild-type gfp upon excitation with 490-nm light. Incorporation of a plant intron into the coding region yielded an additional 1.4-fold improvement, for a cumulative improvement of about 150-fold in fluorescence at 490-nm excitation. Various versions of pgfp were also stably introduced into corn, wheat, tobacco, and Arabidopsis plants. Bright-green fluorescence was observed with a fluorescence microscope in virtually all examined tissues of transgenic monocots and dicots. In the case of Arabidopsis, expression of the pgfp gene under the enhanced 355 promoter of the cauliflower mosaic virus produced green fluorescence that was readily detectable by eye using a hand-held, long-wave ultraviolet lamp and/or a black-light source.

Sexually transmitted disease control in China (1949-1994).

This paper summarizes the historical experiences in venereal disease control in China during the 1950s. Venereal diseases had been all but eliminated in the whole country till 1964. However, along with the implementation of open-door policy and economic reform in the 1980s, the social environment was changed to a great extent in this country. Sexually transmitted diseases (STDs) were re-introduced in the Chinese mainland and new foci of infection established themselves in some cities. During the recent 8 years the national STD case-reporting and sentinel surveillance systems have been set up. The results of surveillance show that the annual incidence of STD has been on the increase. The existing factors associated with the increasing incidence of STD mainly are; population movement, increasing affluence in a part of population, the availability of multiple sexual partners (including the prostitution) and asymptomatic STD increased. Finally, the strategies for STD control are discussed in detail.

Integration of traditional and modern methods in the identification of AFB cultures isolated from clinical specimens of patients with skin diseases.

This article reports the identification of 57 AFB cultures isolated from clinical specimens by using traditional methods (TM, including biochemical and cultural methods) and modern ELISA with monoclonal antibody (McAb-ELISA) and nested primer gene amplification assay (NPGAA). The representive AFB culture M. A1, A7, A19, A21 and A22) isolated from human lepromas were identified as new species by TM and it was shown that they were not identical to M. leprae by McAb-ELISA and NPGAA. Among another set of samples (M. S17, S1, S2, S2R, S7, S29), M. S17 was identical to M. scrofulaceum as assessed by TM only, while the others were found to be similar to M. tuberculosis and different from M. leprae using TM and McAb-ELISA, and identical to M. tuberculosis with NPGAA. The authors conclude that TM and MM are very useful for identifying mycobacteria, while MM was much more sensitive and specific than TM. The selection and use of these methods depends on practical need.

[Revelations of the origin of Chinese nation from clustering analysis and frequency distribution of HLA polymorphism in major minority nationalities in mainland China].

This paper is a joint report on the analysis of polymorphism of HLA class I, II and III antigens in Chinese major minority nationalities (Miao, Buyi, Mongol of Inner Mongolia, Man, Hui, Tibetan and Uygur), and southern and northern Han samples as a part of the workshop organized by the 11th International Histocompatibility Workshop and Conference (IHWC) held on 6-13 November 1991 in Yokohama. Clustering analysis showed that Miao and Buyi cluster with the southern Han and Mongol, Man, Hui and Tibetan cluster around the northern Han, suggesting that the Chinese nation comprises northern and southern two major populations. Uygur clusters with a large group of Caucasoid samples from very far point. Frequencies of HLA alleles of Caucasoid origin (A3, B8 and others) decrease eastwards and southwards, and those of HLA alleles of Southeast Asia Mongoloid origin (B46 and others) decrease northwards. The gradient distribution of HLA allele frequencies suggests the unceasing migration and mutual amalgamation of our ancestors since the remote pre-historic times. From the fact that Chinese nation comprises northern and southern two major populations, this paper discussed the possible revelations of the origin of Chinese nation.

An epidemiological survey of deformities and disabilities among 14,257 cases of leprosy in 11 counties.

This study was planned and conducted in Yang Zhou Prefecture, covering 11 counties that were formerly areas with a high prevalence of leprosy. Out of 14,257 leprosy patients, 8122 (56.97%) cases with deformities and disabilities were found. The disability rate is much higher in patients with MB leprosy (81.15%) than in PB leprosy (53.04%). The statistical data and the type of deformities and disabilities are presented. The influences of various host factors and disease factor which cause disability and deformity are discussed.

Response of the canine bladder base during reflex micturition.

The response of the bladder base to filling and voiding was studied in a surgical dog model in which the bladder base was separated from the main body and closed to form a chamber. In the bladder distension phase there was a reduction in pressure in the bladder base chamber, which implies relaxation of the bladder base. In the expulsion phase the reflex bladder body activity took place with an increase in bladder base pressure, indicating contraction. Relaxation and contraction were blocked by propranolol and phentolamine respectively and both responses were abolished by trimetaphan. It was concluded that the bladder base activity in the micturition cycle is a reflex action mediated via the sympathetic nervous system.

Effect of three-year multidrug therapy in multibacillary leprosy patients.

The feasibility and effects of a 3-year treatment using rifampicin (RFP), clofazimine (B663) and dapsone (DDS) in multibacillary leprosy patients in Yangzhou Prefecture and Dongtai County (1983-1986) are reported. Among 591 active multibacillary leprosy patients in the two areas, 569 (96.30%) were treated with this regimen. Of 303 cases available for analysis, 196 (64.7%) cases showed negative skin smears and clinical inactivity. The rest showed different degrees of improvement. The average reduction of BI was 0.78. The intensity and frequency of ENL and neuritis decreased markedly with treatment. The main side-effects were pigmentation and ichthyosiform changes of the skin, but these did not influence treatment.

CD2 and other surface molecules in the regulation of non-MHC-restricted cytolytic function.

The effect of anti-CD2 and Fc receptor binding molecules on the cytolytic function of a highly enriched population of CD3- large granular lymphocytes (LGL) was studied. These cells could mediate natural killer (NK) activity, antibody-dependent cellular cytotoxicity (ADCC) and lectin-dependent cellular cytotoxicity (LDCC). Both ADCC and LDCC were enhanced by anti-CD2. The enhanced LDCC could also be observed with IL-2-activated LGL. However, NK cell activity was usually slightly diminished or unaffected by anti-CD2 binding. Immune complex and aggregated human IgG had no effect on ADCC but an anti-CD16 showed a dose-dependent inhibition of ADCC, reversible by anti-HLA-ABC and anti-CD2. Cross-linking of LGL surface-bound anti-CD2 caused an almost complete inhibition of LDCC and ADCC but had much less effect on NK activity. These experiments show that ADCC and LDCC mediated by CD3- LGL can be influenced by perturbing the CD2 molecule. NK activity was, however, affected differently, suggesting some basic differences in the pathway of ADCC and NK function.
