Clinical trial on termination of early pregnancy with RU486 in combination with prostaglandin.

Termination of early pregnancy was performed in 1572 healthy women with RU486 (mifepristone, 600mg orally once), followed 36-60 hours later by administration of methyl ester of dl-15-methyl-PGF2 alpha (PG05, 1mg vaginal suppository). Complete abortion was accomplished in 91.2% (1433/1571), incomplete abortion in 4.8% (76/1571), and continued pregnancy in 3.9% (62/1571). The time elapsed between RU486 intake and complete expulsion was 2.4 +/- 1.3 days. Expulsion took place on the third day in 935 women (72%), and on the 4th day in 273 women (21.0%). Uterine bleeding occurred on the second or third day after RU486 intake in 1256 women (88.8%), and lasted 11.7 +/- 6.4 (SD) days, range 2-55 days. One subject had blood transfusion due to excessive bleeding. The main side effects were nausea/vomiting (22.3%), abdominal pain (10.2%), headache/dizziness (4.1%) and diarrhea (2.8%). Fatal side effects have not been reported in this study. About 73% of subjects with complete abortion assessed the treatment as good to excellent. Even in the failed cases, 25-42% of subjects considered the treatment as good. Further studies are needed to determine the optimal dose of the RU486 regimen. It should be emphasized that the treatment must be used under close medical supervision in order to monitor the uterine bleeding.

[Genetic profile of LIBP/1 inbred strain derived from the Kunming outbred stock of the mouse].

To make the genetic profile of the LIBP/1 inbred strain obtained from Kunming mice, the most widely used outbred stock in China, 26 loci were examined. The genotypes of four kinds of coat color genes were a/a, B/B, c/c and D/D. The results of testing 21 biochemical marker genes showed Akp-1b, Amy-1a, Car-2a, Ce-2a, Es-1b, Es-3a, Es-10a, Es-11a, Gpd-1a, Gpi-1a, Gus-1b, Hbbs, Idh-1a, Ldr-1a, Mod-1a, Mup-1b, Pep-3b, Pgm-a, Sep-1b, Tam-1c, and Trfb. The H-2 gene loci were Kb and Db.

Prevalence of vitamin B12 deficiency among geriatric outpatients.

BACKGROUND: Healthy people can have low levels of cobalamin (vitamin B12) without symptoms or signs of cobalamin deficiency. Early detection of deficiency is imperative for treatment to be effective. Development of radioimmunoassay tests has greatly improved accurate determination of cobalamin (Cbl) levels. Nevertheless, results of studies of Cbl deficiency vary widely because of the variety of populations studied. METHODS: In a prospective study, we tested 100 consecutive, unselected geriatric outpatients in a primary care setting to determine the prevalence of cobalamin deficiency. All patients, 65 years of age or older, who visited the office of one of the authors during a period of 11 consecutive working days, had their serum Cbl level checked. If the level was 299 pg/mL or lower, serum intrinsic factor and parietal cell antibodies, serum gastrin, part 1 Schilling test, serum methylmalonic acid, and total homocysteine were done, when possible, for the diagnosis of type A gastritis and intracellular Cbl deficiency. RESULTS: Sixteen percent of geriatric outpatients had serum Cbl levels of 200 pg/mL or below, and 21% had levels between 201 and 299 pg/mL. Among the 16 patients with levels < or = 200 pg/mL, 2 patients had macrocytic anemia, 3 patients had peripheral neuropathy, and 8 patients had type A gastritis. Among the 21 patients with levels of 201 to 299 pg/mL, 2 patients had peripheral neuropathy, 9 patients had type A gastritis, and none of the patients had macrocytic anemia. Among the patients whose methylmalonic acid and total homocysteine levels were determined, the results were high in 80% of those with Cbl levels < or = 200 pg/mL and in 33% of those with levels from 201 to 299 pg/mL. CONCLUSIONS: The prevalence of Cbl deficiency in geriatric outpatients was found to be higher than in any recent report. The lower limit of the normal range for Cbl level should be increased to 300 pg/mL.

Effect of vitamin C supplementations on iron deficiency anemia in Chinese children.

A total of 65 children with mild iron deficiency anemia (IDA) were divided into 5 groups, and received 0, 25, 50, 100 and 150 mg/day of vitamin C (VC) respectively every day for 8 weeks. Hemoglobin, serum ferritin, free erythrocyte and hematocrit were determined every week. At a daily average intake of about 30 mg of VC and 7.5 mg of Fe, the results of the study indicate that: (1) VC supplement alone could effectively control children's IDA, and a dose-dependent relationship was observed. (2) 50 mg/day of VC is the most efficient dosage and 6 weeks is the shortest time for an effective therapy. (3) With a diet predominantly comprised of plant foods, it is suggested that appropriate dose of VC should be supplemented for the children during winter and spring in northeastern areas of China.

The effects of crowding on adults of Echinostoma caproni in experimentally infected golden hamsters.

Fifty-nine of 60 (98%), 6-month-old male golden hamsters, Mesocricetus auratus, fed 15 (group A), 50 (group B), or 200 (group C) metacercarial cysts of Echinostoma caproni (Digenea: Echinostomatidae) were infected 7-34 days postexposure. The mean number of worms recovered in groups A, B and C were 9, 10, and 50, respectively. The percentage recovery was significantly different between group A (63%) and groups B (21%) and C (23%). The intestine was divided into three equal regions (I, II, III). Worms from group A were located in segments II and III of the small intestine whereas worms from groups B and C were distributed in all three segments. The body area, ovarian and testicular areas of worms from group A were greatest, followed in decreasing order by body and gonadal areas of worms from groups B and C. Echinostoma caproni eggs were found in the faeces of all the hamsters examined from groups A, B and C by days 9, 10 and 11, respectively. Physical damage occurred at the site of attachment of the echinostome. Pathological observations indicated the presence of enlarged lymphatic nodules with lymphocytes being the primary cellular infiltrate at the site of parasite attachment.

A comparison of modulation of proliferation of thymocyte by testosterone, dehydroisoandrosterone and androstenedione in vitro.

Although the relationship between androgens and the immune system has been well established, the immunomodulatory function of different kinds of androgens on the cellular immunity remains to be defined. In the present study, the in vitro influences of testosterone, dehydroisoandrosterone and androstenedione on the Con A induced proliferation of thymocyte were determined. The results showed that both dehydroisoandrosterone and testosterone suppressed the Con A induced proliferation of thymocyte, while dehydroisoandrosterone was less potent than testosterone. In contrast, relatively high doses of androstenedione augmented the proliferative activities of thymocytes. Taken together, these data implied that small structural changes of androgens result in markedly different biological effects. These findings suggest that the balance between these steroids may be a significant determinant in maintaining immune homeostasis.
