Electro-acupuncture (EA) induced attenuation of immunosuppression appearing after epidural or intrathecal injection of morphine in patients and rats.

In order to investigate the effect of epidural morphine and electroacupuncture (EA) on immune function of postoperative patients, peripheral blood for NK (natural killer) cell activity assay were obtained from patients before and on the 1st, 3rd, 7th day after cholecystectomy. 1 mg morphine injected into epidural space after surgery significantly suppressed NK cell activities via prolonged inhibition and increased the inhibitory intensity of it. EA of "Zusanli (St. 36), Sanyinjiao(Sp. 6), Hegu(L.I.4) and Neiguan(P.6) points enhanced the NK cell activities after epidural injection of morphine. It was further observed that microinjection of 1 microgram/1 microliter naloxone into periaqueductal gray (PAG) partially antagonized the suppression of NK cell activity induced by intrathecal (ith) injection of 40 micrograms/50 microliters morphine, and so does EA of "Zusanli(St. 36)". When EA applied after naloxone microinjected into PAG, the NK cell activity was further augmented as compared to that using EA or PAG microinjection of naloxone alone. The above results suggest epidural morphine combined with EA is a better method to relieve postoperative pain, and opiate receptor in PAG might partially mediate the suppression of NK cell activity induced by intrathecal injection of morphine. Yet, the mechanisms of the attenuation of EA on morphine induced immunosuppression needs further study.

Hyphema in the newborn: report of a case.

This is a case report of an anterior chamber hemorrhage noted two days after spontaneous delivery in a full-term infant with perinatal asphyxia. There was probably vitreous hemorrhage in the same eye and extensive retinal hemorrhages in both eyes. By two weeks, the hyphema and vitreous hemorrhage had resolved and the retinal hemorrhages largely cleared.

Muscular asthenopia and eccentric ablation after photorefractive keratectomy.

PURPOSE: To investigate the relationship between muscular asthenopia and eccentric ablation after photorefractive keratectomy (PRK). METHODS: Sixteen eyes of eight myopic patients whose muscular asthenopia was corrected by subjectively accepted spectacle prism after PRK, with visual acuity better than 0.8, were followed for 6 to 14 months. On the basis of preoperative and post-operative data and corneal topography, we calculated the total corrected corneal power using the Holladay formula and then measured the ablation eccentricity (h, millimeters) and its direction. According to the formula delta approximately equals Dh, the prism effective value (delta) caused by the eccentric ablation was computed and compared with the objectively accepted spectacle prism. RESULTS: The subjectively accepted spectacle prisms were similar to calculated values from the formula. Mean difference was 0.10 +/- 0.25delta. The direction of the subjectively accepted prism was in the direction of the ablation deviation. CONCLUSIONS: Eccentric ablation is an important cause of muscular visual asthenopia after PRK. The asthenopia may be corrected by spectacle prism. The spectacle prism value may be estimated by the formula delta approximately equals Dh.

As2O3 induces apoptosis of the human B lymphoma cell line MBC-1.

OBJECTIVE: To see how arsenic trioxide (As2O3) affects proliferation of the human B lymphoma cell line MBC-1. METHODS: We studied the effect of As2O3 on MBC-1 cells and its mechanism by morphological observation, flow cytometry assay and DNA gel electrophoresis. RESULTS: As2O3 could upregulate p53 gene expression at protein level, inducing cell apoptosis and inhibiting the proliferation of MBC-1 cells. Upregulation of p53 expression appears to be important in the apoptosis of MBC-1 cells. CONCLUSIONS: As2O3 can inhibit the proliferation of MBC-1 cells by upregulating p53 gene expression, thus inducing apoptosis.

Effects of some natural products as components of Chinese herbal medicines on mammary gland growth and function in mice.

As a possible step to evaluate the role in the mammary glands of the scales of Guanshanjia, the fruits of Lulutong and the seeds of Wangbuliouxing, which are the natural products used widely as the components of herbal medicine in China for the improvement of lactation and the therapy of breast disorders, the effects of these agents on mammary gland growth and function were studied in female mice. The chronic administration of each agent in drinking water improved several parameters; however, the mode of action differed markedly among agents. The results indicate the necessity of acquiring enough knowledge of the characteristics of each agent for the efficient prescription of herbal medicine as well as the single use of the agent.

[The immunobiological characteristics of a Salmonella typhimurium strain acquiring a plasmid that controls antibiotic resistance and antibactericidal activity].

The influence of Rdf (resistance defense factor) plasmid inherited by Salmonella typhimurium recipient strain 189, previously deprived of its own virulence plasmid of 60 MD, on the immune status of F1 (CBA x C57BL/6) mice was studied. Experimental infection induced by genetically related pairs of S. typhimurium strains, differing by the presence of Rdf plasmid, revealed the stereotypical character of changes in the number of thymic, splenic, marrow and blood cells with some variations. In mice infected by transconjugant carrying Rdf plasmid immunological shifts were, on the whole, less pronounced than those in the animals infected with clone Rdf.

[The adhesive activity of Klebsiella pneumoniae controlled by plasmid 55 MD].

In K. pneumoniae clinical strains a nonconjugative plasmid, denoted as pAdh-55 and responsible for their adhesive activity with respect to HEp-2 cells, was detected. The comparison of genetically related pairs of K. pneumoniae strains differing in the presence of pAdh-55 revealed that the loss of this plasmid by bacteria was accompanied by a decrease, and its acquisition by a considerable increase, in the number of Klebsiella-affected cells in the monolayer. The correlation between the presence of pAdh-55 in K. pneumoniae strains and their adhesive activity was established both in experiments aimed at the morphological study of preparations stained with azure eosin and in experiments with isotope-labeled bacteria.

Effects of aristoloside, a component of guan-mu-tong (Caulis aristolochiae manshuriensis), on normal and preneoplastic mammary gland growth in mice.

Aristoloside, an extract of Guan-mu-tong (Gmt; Caulis aristolochiae manshuriensis) showing a marked anticancer potential, was dissolved with tap water (0.05%) and given to 4 month-old SHN mice for 30 days as drinking water. The treatment induced the inhibition of preneoplastic mammary gland growth and the stimulation of the excretion of urinary component. Ovarian follicles were dominant and the oestrous stage was elongated by the treatment. Aristoloside had little affect on normal mammary gland growth and serum levels of prolactin and free fatty acid. All findings were in agreement with those obtained from studies on chronic treatment with Gmt, and aristoloside is thus strongly suggested as being one of the major components of Gmt.

N-acetylglucosamine changes permeability of peritoneum during chronic peritoneal dialysis in rats.

OBJECTIVE: To evaluate the effect of supplementation of dialysis fluid with N-acetylglucosamine (NAG) on the permeability of peritoneum during chronic peritoneal dialysis in rats. DESIGN: Experiments were performed on rats with surgically implanted peritoneal catheters. Dialysis solution [Dianeal 1.5% (Baxter, Deerfield, IL, U.S.A.) supplemented with either NAG 50 mmol/L or glucose 50 mmol/L (control)] was infused intraperitoneally twice, every day, for 8 weeks. Peritoneal equilibration tests (PET) were performed in all animals at the beginning of the study and after 8 weeks of dialysis. Additionally, at the end of each week, dialysis solution infused in the morning was drained after 4 hours of intraperitoneal dwell. White blood cell count, creatinine, and total protein concentrations were measured in the effluent dialysate. After 8 weeks of dialysis, the morphology of the peritoneum was studied. RESULTS: In rats exposed to dialysis fluid supplemented with NAG, peritoneal permeability to creatinine and proteins was reduced when compared to animals dialyzed with glucose solution. In NAG treated animals, staining with alcian blue for polyanions in the peritoneal interstitium was significantly stronger than in rats dialyzed with glucose solution. CONCLUSIONS: Chronic peritoneal dialysis with dialysis solution supplemented with N-acetylglucosamine causes accumulation of glycosaminoglycans in the peritoneal interstitium, which results in a change of peritoneal permeability.

Quantitative variation of free amino acids in the central nervous system of MoMuLV-ts1-infected mice.

A temperature-sensitive mutant of Moloney murine leukemia virus (MoMuLV-ts1) induces polioencephalomyelopathy and hind limb paralysis in highly susceptible FVB/N strains of neonatal mice. This disease is characterized by progressive motor neurons loss and severe gliosis within specific target areas of the central nervous system (CNS). The mechanism(s) of this neurodegeneration is unknown. In the neonatal infection of the CNS, the MoMuLV-ts1 virus was reported to replicate within the endothelial, ependymal, astrocytes and microglial cells. Since no virus or viral products were recognized in the degenerating neurons, it is postulated that an indirect mechanism(s) caused the loss of neurons in the neonatally infected mice. This study was undertaken to investigate the possible pathogenic role of excitatory amino acids (EAAs) such as glutamate and other nonneurotransmitters amino acids (NAAs) in this animal model. The free amino acids concentration was analysed by a fluorometric HPLC method. The temporal measurements of the free amino acids concentration, glutamate, glutamine and arginine from the brain stem and spinal cord of MoMuLV-ts1-infected mice was significantly decreased when compared with the control non-infected mice. The concentration of EAAs during the course of this infection indicated a sharp decline in glutamate and its precursor, glutamine with early infection (10 days post infection-dpi). This deficiency persisted (20 and 30 dpi) in the spinal cord, where the neuronal loss was most severe, but not in the brain stem. A similar pattern occurs with the amino acid arginine. These observations suggest that an astrocyte-induced metabolic disturbance of glutamate and arginine in the CNS of developing mice, could be, in part responsible for the loss of motor neurons observed in this model.
