Neutrophil infiltration is involved in 5-fluorouracil induced lipid peroxidative damage of the small intestine.

UNLABELLED: This study evaluated the mechanisms of 5-fluorouracil (5-FU) induced intestinal damage by investigating the lipid peroxide (LPO) level, myeloperoxidase (MPO) activity and disaccharidase activity in rats. Group I animals (n = 6) were sacrificed and served as a normal control group. Group II animals (n = 24) were given a single intraperitoneal injection of 5-FU (150 mg/kg) and every 8 rats were sacrificed on day 1, 3 and 5 after injection respectively. RESULTS: LPO concentration in blood and intestinal mucosa was significantly higher in the group II than in the group I on day 1 and 3 (P < 0.05). MPO activity was significantly higher in the group II than in the group I at different times (P < 0.01). Lactase activity on day 5 (P < 0.01); sucrase activity (P < 0.01 and P < 0.05 respectively) and maltase activity (P < 0.01) on day 3 and 5, were significantly lower in the group II than in the group I. CONCLUSION: The results indicate that neutrophil infiltration may be involved in 5-FU-induced lipid peroxidative damage of the small intestine which was reflected by the decreased disaccharidases activities.

[Preliminary studies on proliferation of hemorrhagic fever with renal syndrome virus in Leptotrombidium (L.) Scutellare].

In order to observe further the proliferation and kinetic changes of hemorrhagic fever with renal syndrome virus (HFRSV) in Leptotrombidium (L.) Scutellare infected with HFRSV, the raised Leptotrombidium (L.) Scutellare which were at larve stage or at nymph stage were grinded and sterilized by filtration at interval of 20 days, each batch of the filtrate was inoculated separately to Vero-E6 cells and the titre (TCID50/ml) of HFRSV was measured. The results confirmed: HFRSV were isolated from each batch of larve besides the batch at the 60th days, and the titre of HFRSV was -10(-1)-10(-4); HFRSV were also isolated from two batches of nymph. The isolated HFRSV were amplified by PCR technique and the HFRSV RNA were positive. The above results give rise to evidence that Leptotrombidium (L.) Scutellare might be the transmission vector of HFRSV.

[Cord blood plasma selectively stimulates the expansion of hematopoietic pregenitor cells in vitro].

OBJECTIVE: To explore the effect of cord blood (CB) plasma on the expansion of hematopoietic progenitor cells. METHODS: The effect of CB plasma on the expansion of CB and bone marrow (BM)-derived hematopoietic progenitor cells was studies and compared with those of adult's peripheral blood (PB) plasma and cytokines such as IL-3 and SCF. RESULTS: (1) CB plasma increased CFU yields of CB-derived progenitor cells by 2.23 +/- 0.52-fold and this expansion was dependent on CB plasma concentrations supplemented in the cultures. Greater expansion of progenitor cells was found when ABO was unmatched between the CB-derived cells and CB plasma. (2) Stimulating effect of CB plasma on BM-derived progenitor cells was also related to ABO type. CFU yields were increased by 1.55 +/- 0.43-fold when ABO was unmatched and not increased when ABO was matched. (3) Expansion of CB-derived progenitor cells by IL-3 and SCF was revealed. Significant increase in CFU yields occurred in the addition of both the cytokine and the CB plasma. (4) No expansion occurred in CB-and BM-derived cell cultures supplemented with PB plasma. CONCLUSION: There is factor(s) selectively stimulating expansion of progenitor cells in CB plasma. It may be beneficial that CB plasma was transfused with CB-derived stem cell transplantation.

[Luminescence properties and energy transfer between Ce3+ and Tb3+ ions in KA1F4: Ce, Tb].

Matrix KA1F4 was prepared through reaction of mixtures of solution K2CO3 upon certain solution of Al(OH)3 plus HF(40%),dehydrating at 100 degrees C and annealing for 48hr at 450 degrees C. A series of KA1F4 : Ce,Tb phosphors were synthesized by a solid reaction method. Their excitation and emission spectra have been investigated. It was observed that Ce3+ can effectively sensitize Tb3+ in KA1F4, so as to enhance the emission of Tb3+ significantly. KA1F4 :Ce,Tb might be a new high efficient green emitting phosphor.

Molecular studies on single cells harvested by micromanipulation from archival tissue sections previously stained by immunohistochemistry or nonisotopic in situ hybridization.

Molecular analysis of isolated single cells is a powerful tool for analyzing heterogeneity within a population of cells and for clarifying issues of cell origin and clonality. Current techniques are limited by the availability of suitable fresh tissue. To broaden the applicability of molecular techniques at single-cell level, we have developed an approach that uses routinely processed archival tissue. Immunoglobulin heavy chain (IgH) gene rearrangement was analyzed in large tumor cells from four cases of diffuse large cell B-non-Hodgkin's lymphoma and in small reactive T and B lymphocytes from three cases of lymphocytic predominance Hodgkin's disease. One case of Epstein-Barr virus (EBV)-encoded RNA (EBER)-positive angiocentric pulmonary T-cell lymphoma was assayed for the presence of the BamHI-W multiple-copy fragment of the EBV genome. T- and B-lymphoid cells were immunostained with anti-CD3 and CD20, respectively. The tissue sections from the EBER-positive T-cell lymphoma were stained by nonisotopic in situ hybridization. Single cells were mobilized after proteolytic treatment under an inverted microscope using a hydraulic micromanipulator at a magnification of 400 x. Isolated cells were aspirated into a micropipette fixed to a second micromanipulator and transferred into a PCR tube. The IgH complementarity determining region (CDR)3 was successfully amplified in 17 of 52 (33%) small B-lymphocytes from lymphocytic predominance Hodgkin's disease using a previously reported semi-nested PCR method, and the products from each case differed in size as expected of a polyclonal population. None of the 49 small T lymphocytes demonstrated any amplifiable IgH CDR3 products, indicating no significant cellular contamination. The IgH CDR3 sequence analysis of the PCR products indicated a clonal relationship among harvested cells. In the T-cell lymphoma case, the harvested EBER-positive cells were amplifiable for the multiple-copy fragment BamHI-W of the EBV genome. Our study indicates that single-cell analysis can be performed on paraffin-embedded archival tissue after being subjected to immunoperoxidase and in situ hybridization procedures.

Canine lung transplantation after more than twenty-four hours of normothermic preservation.

BACKGROUND: Consistent clinical results have not been achieved when lung preservation times exceed 6 hours. The aim of this study was to use an alternative normothermic autoperfusion technique for lung preservation and transplantation. METHODS: In six paired dogs, donor lungs were removed, along with the heart, liver, pancreas, duodenum, and both kidneys, and were preserved for 24 to 33 hours in a normothermic autoperfused multiple organ block. Orthotopic left lung transplantation was performed at the end of the preservation period. RESULTS: Lung function was good during the preservation period. With a gas mixture of 50% O2 + 3% CO2 + 47% N2 delivered to the multiorgan block, arterial oxygen tension ranged from 331 +/- 19 to 383 +/- 8 mm Hg; carbon dioxide tension ranged from 18 +/- 5 to 32 +/- 5 mm Hg; and pH ranged from 7.36 +/- 0.02 to 7.45 +/- 0.08. After transplantation, the dogs were kept anesthetized and ventilated for 24 hours with the same gas mixture. The opposite pulmonary artery was occluded 0 to 6 hours after transplantation. Arterial blood pressures were stable after surgery. Arterial oxygen tension was maintained between 205 +/- 39 and 320 +/- 57 mm Hg, and arterial carbon dioxide tension was maintained between 23 +/- 2 and 34 +/- 2 mm Hg. Lung tissue wet/dry weight ratio was 4.94 +/- 0.17 after preservation; this ratio did not differ from that found in normal controls (4.91 +/- 0.10). CONCLUSIONS: This study shows that the lungs were well preserved for more than 24 hours of preservation when the normothermic multiorgan block preparation was used. The transplanted left lung was able to support the anesthetized dog after the opposite pulmonary artery was occluded.

L-glutamine inhibits nitric oxide synthesis in bovine venular endothelial cells.

This study was conducted to test the hypothesis that L-glutamine has differential effects on nitric oxide (NO) synthesis from L-arginine in bovine venular endothelial cells (EC) stimulated by A23187 (a Ca++ ionophore) and receptor-mediated vasodilators (bradykinin and substance P). EC were cultured at 37 degrees C for 24 h in the presence of 0.4 mM L-arginine and 0.0 to 2.0 mM L-glutamine with or without 1 microM A23187, 1 microM bradykinin or 10 microM substance P. The release of nitrite and nitrate by EC was used as an indicator of NO synthesis. A23187, bradykinin or substance P increased NO synthesis from L-arginine by EC in the presence or absence of L-glutamine. The addition of L-glutamine (0.5 and 2 mM) markedly increased intracellular concentrations of L-glutamine, L-glutamate and L-aspartate and decreased NO synthesis by EC in a concentration-dependent manner in the presence or absence of A23187, bradykinin or substance P. L-Glutamine had no effect on L-arginine uptake by EC or on intracellular L-arginine concentration. Neither L-glutamine nor its glutaminase metabolites (ammonia, L-glutamate and L-aspartate) had any effect on endothelial NO synthase activity. Taken together, these results suggest that the inhibition by L-glutamine of NO synthesis from L-arginine is unlikely to result from an effect of L-glutamine on L-arginine transport or NO synthase activity. Although the mechanism involved remains unknown, regulation of the arginine-NO pathway by L-glutamine may have pharmacologic and therapeutic implications in such conditions as inflammation and septic shock by inhibiting NO generation from L-arginine in endothelial cells.

Predictions of serum vancomycin concentrations by means of six different equations for calculation of creatinine clearance.

Serum vancomycin concentrations were predicted in 59 patients by means of the PKS programme using six different equations to calculate creatinine clearance. These equations are (i) Chiou & Hsu equation, (ii) Cockroft-Gault lean body-weight equation, (iii) Cockroft-Gault total body-weight equation, (iv) Jelliffe equation, (v) Jelliffe & Jelliffe equation, and (vi) Siersback-Neilsen equation. The predictions were made by using (i) the implemented population pharmacokinetic parameters (IPPP), and (ii) the Bayesian method (BM). The results show that (i) if IPPP can lead to under-predictions, and Chiou & Hsu equation is most suited for predictions, and (ii) if IPPP can lead to over-predictions, the Cockroft-Gault total body-weight equation is the most suited for predictions.

Isotypic antibody responses of a population in an endemic area of schistosomiasis japonica and their epidemiologic significance.

The present study was designed to explore if there exists a correlation between predominant isotype-defined antibody levels and reinfection in low age groups of the population in an endemic area of schistosomiasis japonica in China. One hundred and thirty-eight individuals aged 3-25 years old were selected for serological investigations including the levels of IgG, IgG4, IgM and IgE, detected by ELISA with soluble egg antigen and soluble adult worm antigen. Results show that age is a determinant for SEA-specific IgG, IgG4, and IgE, and SWA-specific IgG and IgG4 antibody levels, which increased with age, and that SEA- and SWA- specific IgG4 antibody levels are risk factors of reinfection, ie, the risk of reinfection occurrence of the population with high level of SEA or SWA-specific IgG4 is 2.83 or 2.40 times, respectively, that with low level of SEA or SWA-specific IgG4, suggesting that in the endemic area of schistosomiasis japonica, there exists a possibility that in the population aged 3-25 years, SEA and SWA-specific IgG4 antibodies mediate a blocking immunity response.
