[Immuno-pathomorphological study of placenta with anticardiophospholipid antibody positive].

OBJECTIVE: To investigate the relationship between the anticardiophospholipid antibody (ACA)-positive cases and the poor pregnancy outcomes by observing the immuno-pathomorphology of the placenta. METHODS: 16 cases of ACA-positive placenta (8 normal and 8 poor pregnancy outcomes) were collected as the studied group, and 8 cases of ACA-negative placentas served as the controlled group, and immunofluorescence technique (IFT) was used. RESULTS: In the studied group, the deposits of immuno-complex were found in the cytoplasm of trophoblast and on the walls of villous blood vessels. IgA manifested as filiform, and IgG, IgM, C3 and C4 appeared as filiform, granular and tubercular as well. In contrast, all the immuno-globulins displayed negative reaction in the controlled group, and C4 demonstrated some small focal positive reaction. The frequency of ACA-positive in IUGR cases were 19.30% (22/144), and of the IUGR in all the ACA-positive pregnant women was 15.28%(22/144). There were 3 cases of IUGR complicated by severe pregnancy induced hypertension, 2 cases of prematurity and 5 newborns with serum ACA IgG positive. CONCLUSION: The poor pregnancy outcome in ACA-positive women is related to the dysfunction of autoimmuno-system, and ACA-positive may be one of the causes of IUGR. The immuno-injury occurred in the placenta may affect the normal function of placenta. Maternal antibody may be transmitted to the fetus, and its significance needs further study.

[Screening of proteins interact with FMR1 by yeast two-hybrid system].

OBJECTIVE: To establish yeast two-hybrid system for screening of protein(s) interacted with the fragile X metal retardation protein (FMRP). METHODS: Fragment of exon 11 to 15 of the FMR1 cDNA was recombined with DNA-binding domain of the pBTM116 vector as bait to screen a mouse embryo cDNA library. RESULTS: Thirteen clones were confirmed to be able to specifically interact with FMRP bait. Sequence analysis showed that 12 clones are overlapping ones containing cDNA fragments of the mouse ubiquitin-conjugating enzyme gene (mUBC9). CONCLUSIONS: The interaction between UBC9 and FMRP is supported both by similarity search of the amino acid sequences of the mouse and human UBC9 and by expression characteristics of the two proteins. The biological significance of the interaction is to be further studied.

[Accuracy and predictability of photorefractive keratectomy for astigmatism correction].

OBJECTIVE: To study the accuracy and predictability of photorefractive keratectomy (PKR) for astigmatism correction. METHODS: 53 eyes of 30 cases with compound myopia astigmatism and 33 eyes of 23 cases with simple myopia were treated with PRK and followed up for 6 months. The differences of corneal refractive power provided by corneal topography between the values at postoperative 6 months and that of the preoperative one were calculated with Holladay Ways to get real corrective degree and axis of astigmatism. The results were analyzed and compared with the preoperative predicted values. RESULTS: Six months after PRK, among the compound myopia astigmatism group, the ratio of over correction or under correction of real corrected astigmatic power within +/- 0.50D was 60.4%, the ratio of axis deviation between real and predicted correction within +/- 15 degrees was 58.5%. Among simple myopia group, 48.5% appeared more than -0.50 D myopia astigmatism in real correction, and the cylinder axis position was within 180 degrees +/- 44 degrees. The ratios of uncorrected visual acuity after PRK in the two groups equal to or better than 1.0 were 77.4% and 78.8%, respectively. There was no difference between these two groups (chi(2) = 0.024, P > 0.75). CONCLUSIONS: The method of myopia astigmatism correction using this PRK machine needs to be improved. After PRK treatment, the visual acuity of myopia astigmatism is similar to that of simple myopia group. It shows good efficacy. It is indicated that for myopia astigmatism lower than -2.00D, especially for the astigmatism with the rule, PRK could be performed according to equivalent spherical value.

[The determination of ligustilide in Ligusticum chuanxiong Hort. by supercritical fluid extraction].

In this study, a systematic method was used to optimize the supercritical fluid extraction (SFE) of ligustilide in Ligusticum chuanxiong Hort. Overall five variables (pressure, temperature, static extracting time, modifier concentration and CO2 dynamic extracting volume) were considered. To reduce the loss of volatile substance, a solid-liquid trap was developed for SFE collection. Comparisons were made on the solvent trap for SFE. The extracts were analyzed by high performance liquid chromatography (HPLC) with ultraviolet absorbance detection. The recoveries of 99.34% (RSD 1.70%) and 92.11% (RSD 5.72%) were achieved for solid-liquid trap and solvent trap, respectively. SFE conditions: temperature 40 degrees C, pressure 27.6 MPa, static extraction 3 min, dynamic extraction 7 ml and 0.1 ml CHCl3 as modifier. HPLC conditions: The HPLC column (3.9 mm x 150 mm) was packed with Nova-Pak C18(4 microns). Naphthalene was used as internal standard. The mobile phase was methanol--10% isopropyl alcohol solution (53:47 v/v). The detection wavelength was 280 nm and the flow rate was 0.8 ml.min-1.

[Studies on the pharmacokinetic-pharmacodynamic model of ketoprofen beta-CD inclusion complex in rabbits].

A simple HPLC-UV method was established to determine concentrations of ketoprofen (KP) in rabbit plasma following oral administration. Twelve rabbits were selected and divided into three groups. KP beta-CD inclusion complex suspension, KP entity suspension (in 0.5% CMC-Na) and 0.5% CMC-Na suspension (as a placebo group) were administered orally to the three groups of rabbits, respectively. Differences in the pharmacokinetics-pharmacodynamics (PK-PD) parameters between KP beta-CD inclusion complex and KP entity were examined. The results indicate that the established HPLC-UV method could be used to assay the concentrations of KP in rabbit plasma with good precision. The distribution phase T1/2 alpha of the complexated KP was 0.4 h, while that of the KP entity was 0.56 h, KP in beta-CD inclusion complex could be absorbed more rapidly. The early effect values of the KP inclusion complex were higher than those of the KP entity. The maximal antipyretic effect occurred after the peak of plasma concentration. The phenomenon indicates that the effect compartment of ketoprofen is in the peripheral compartment.

[Researches on immunogenicity identification of recombinant Sj22.6(rSj22.6) kDa antigen gene of schistosoma japonicum].

AIM: To identify the immunogenicity and the potentiality of rSj22.6 as a candidate vaccine antigen for schistosomiasis japonica. METHODS: rSj22.6 was tested by Western blot with specific antibodies to identify its immunogenicity. The rSj22.6 protein was isolated and purified by SDS-PAGE and injected twice into rabbits to raise antirSj22.6 serum that was tested by Western blot to recognize the native protein of adult S. japonicum and SEA. C57 mice were immunized with rSj22.6 and challenged with S. japonicum cercariae to identify the protective immunity raised by rSj22.6. RESULTS: rSj22.6 could be recognized by specific antibodies and could stimulate the rabbits to produce high level of anti-rSj22.6 antibodies. Anti-rSj22.6 serum could recognize an adult native protein with the same molecular weight as rSj22.6. The preliminary immune-challenge test resulted in significant reduction in adult worm burden. CONCLUSION: rSj22.6 possesses immunogenicity in stimulating rabbits to produce high level of specific antibody and has protective role as a candidate vaccine antigen against S. japonicum infection.

[Researches on the nucleotide sequencing of the recombinant Sj 22.6 kDa antigen gene of Schistosoma japonicum].

AIM: To sequence the cloned gene in pGSj24 and to identify the encoded protein. METHODS: The cloned gene in pGSj24 was digested from the recombinant plasmid by EcoRI, ligated into the M13mp19 vector and sequenced by automatic sequencer. The sequence was analyzed by Goldkey DNA and Protein Analytical Program and DNASIS Program. RESULTS: The pGSj24 cloned gene was demonstrated to be 840 bp containing one opened reading frame (ORF) with an initiation codon ATG at position 23 nt and a termination codon TAA at position 596 nt, encoding a protein with a molecular weight of 22.6 kDa. At the upstream and downstream of the ORF there were termination codons, so the encoded protein was unable to be larger. However, there was a termination codon TAA at position 11 nt, suggesting why the 22.6 kDa protein expressed separately. The nucleotide sequence of the pGSj24 cloned gene shared 95% identity with that of the corresponding part of S. japonicum 22.6 kDa antigen gene, and 99.7% identity in the encoding part. The deduced amino acid sequence analysis showed a sequence motif known as EF-Hand calcium binding domain, several endoplasmic reticulum targeting sequences and microbodies C-terminal targeting signals. The possible antigen determinants were predicted within the amino acid fragments of 29-32aa, 63-68aa and 87-101aa. CONCLUSIONS: The cloned gene in pGSj24 is the gene that encodes Sj22.6 kDa antigen.

[Screening and characterization of Schistosoma japonicum reinfection-related cDNA clones].

AIM: To obtain cDNA clones coding for S. japonicum (Sj) reinfection-related specific immunologic molecules for vaccination or diagnosis. METHODS: An adult S. japonicum cDNA library, of which the cloning efficiency is 3.13 x 10(6), with directional inserts in the vector lambda gt11, was screened with pooled serum from 43 reinfected human cases in a schistosomiasis-endemic area with high level of Sj specific-lgG4 antibodies. Primers located several bases away from the insert position on the lambda gt11 arm were used to amplify the insert fragments by standard PCR. SILVER SEQUENCE (PCR-silver) DNA sequencing system was then used to sequence selected positive clones with relatively large inserts and homology comparison with GenBank database was also carried out. RESULTS AND CONCLUSION: Eleven phage plaques were defined as positive out of the 8.6 x 10(4) phage plaques immunoscreened by human specific lgG4 antibody. GenBank database retrieval showed that 4 new Sj gene clones including Sj reinfection-related specific protein-encoding gene and Sj mitochondrial protein-coding gene have been obtained.

[Hemorheologic change after strong blood flow disturbance caused by gunshot and its influence on the prognosis of the wounded animals].

Three types of experimental models were used in this study: 1. Both thighs of dogs were wounded by 5.56 mm bullet. 2. Femoral artery and vein were tied at the time of wounding. 3. Shock caused by simply bleeding the dogs' femoral arteries. We observed the characteristics of change hemorheology and analyzed its effects on the wounded dogs. The results showed that strong blood flow disturbances always brought about hemorheologic changes such as rapidly increased blood viscosity and red cell's aggregation index and the simultaneous phenomenon of blood concentration. These may be related to extensive endothelium injury and destruction of a number of blood cells caused by disturbance wave. The above change may give rise to abnormality of microcirculation and disturbance of matter transmission; in that sense, it may provide the physiopathologic basis for the occurrence and development of the complication.

[The measurement of baroreflex sensitivity in stress-induced hypertensive rats by spectral analysis].

Simultaneous spectral analysis of short-term systolic blood pressure variability (SBPV) and heart rate variability (HRV) was applied to determine the change of baroreflex sensitivity (BRS) in Sprague-Dawley (SD) rats. The modulus of the transfer function between fluctuations in systolic pressure and beat-to-beat interval would be an appropriate quantification of BRS. The animal experiment was performed on two groups of rats: normotensive rats and stress-induced hypertensive rats (SIHR). V1-receptor antagonist d(CH2)5Tye(Me) AVP of arginine vasopressin (AVP) was microinjected into intracerebroventricle. The results showed that, in base-line condition before administration, although the BRS in very low frequency band (0-0.035 Cycle/Beat, VLF), low frequency band (0.035-0.12 Cycle/Beat, LF), high frequency band (0.12-0.32 Cycle/Beat, HF) and total BRS(the sum of the three bands) were all decreased, the BRS in VLF(P < 0.05) and LF(P < 0.01) decreased significantly by statistics; and that, to the SIHR, the BRS in VLF was significantly lower after AVP-V1 administration than in base-line condition, while to normotensive rats, the BRS did not change. It indicates that the facilitating effect of AVP on the beroreflex in SIHR is mainly due to V1-receptor in central nerve system. In summary, the transfer function between SBPV and beat-to-beat interval fluctuation could be an index of BRS. This method can be developed for future clinical application.

The effect of curcumin on human B-cell immortalization by Epstein-Barr virus.

Cyclosporine is a commonly used immunosuppressant in solid-organ transplantation. It is, however, associated with an increased incidence of Epstein-Barr virus (EBV)induced post-transplant lymphoproliferative disorder (PTLD). In this study, human B lymphocytes isolated from healthy volunteers were immortalized in vitro with EBV. The effect of oxidative stress mediated by cyclosporine A or hydrogen peroxide on in vitro B cell immortalization was studied by coculturing immortalized B cells with cyclosporine A and hydrogen peroxide. Curcumin, a phenolic extract of the spice turmeric, was then used to observe its effect on this process. We found that in vitro B-cell immortalization with EBV was promoted by the oxidative stress induced by cyclosporine A and hydrogen peroxide, with the maximum effect seen at concentrations of 500 ng/ml and 100 microM, respectively. Curcumin blocked the B-cell immortalization in a dose-dependent fashion with nearly complete inhibition at 20 microM. We conclude that, because both hydrogen peroxide and cyclosporine A strongly promote in vitro B-cell immortalization with EBV (the putative process responsible for PTLD) and curcumin, an extract of a common spice is an effective inhibitor of this process; curcumin may be an effective adjunct in the prevention of PTLD in the patients undergoing therapy with cyclosporine A.

Assessment of glomerular filtration rate by means of the three-compartment closed model after a single intravenous injection.

The two-compartment open model is currently used to assess glomerular filtration rate (GFR) with multiple blood samples. Numerous limited sampling models have been developed to reduce the number of blood samples. In the present study, the three-compartment closed model was used to assess GFR after a single intravenous injection. The three-compartment closed model includes blood, peripheral and urine compartments, thus the samples in urine can be used to estimate parts of pharmacokinetic parameters, thus to reduce the number of blood samples. Theoretically, the three-compartment closed model can assess GFR with one blood sample at any sampling time and three urine samples at any three sampling times.

Glomerular Filtration Rate↗

Assessment of glomerular filtration rate by means of the four-, and more compartmental closed models after a single intravenous injection.

This is the continuation of our study on the estimation of glomerular filtration rate (GFR) after a single intravenous injection using the closed compartmental model. In our previous study, a three-compartment closed model including blood, periphery and urine was developed to estimate GFR. Although the model developed in our previous study is an improvement of the two-compartment open model currently used in the assessment of GFR, there is a limitation in our previous model and the current two-compartment open model, i.e. these models are suitable for the tracers whose elimination is solely via the renal pathway. However, several tracers used to assess GFR are eliminated via both renal and non-renal pathways, therefore it is also important to develop the models to include the non-renal elimination pathways for these tracers. In the present study, all the possible non-renal elimination pathways were modelled by means of four-, five-, and n-compartment closed models after a single intravenous injection. Although more compartments were included into the modelling, the results show that the inclusion of more non-renal elimination compartments only leads to the increase in the similar analytical solutions for these compartments. Therefore, the equation in our previous study can be used to estimate GFR for the tracers whose elimination is not only via the renal pathway, but also via the non-renal pathway, even via an arbitrary number of the non-renal pathways.

The clonal relationship between nodular sclerosis Hodgkin's disease with a clonal Reed-Sternberg cell population and a subsequent B-cell small noncleaved cell lymphoma.

We evaluated the clonal relationship between a case of nodular sclerosis Hodgkin's disease (NSHD) and a small noncleaved cell (SNC) lymphoma that subsequently developed. Single Hodgkin and Reed-Sternberg (H-RS) cells were isolated from immunostained sections of the NSHD by micromanipulation, and the immunoglobulin heavy chain gene (IgH) complementarity determining region (CDR) III of the cells was amplified by the polymerase chain reaction (PCR). A clonal population of H-RS cells was found in the NSHD tumor. The nucleotide sequence of the clonal H-RS cells was compared with the clonal IgH CDRIII sequence from the SNC lymphoma. The two sequences were found to be unrelated. In addition, clone-specific primers and probes were designed from the two clonal IgH CDRIII sequences and used to investigate the presence of the respective clonal population in the NSHD tumor. The latter studies confirmed the presence of a major clonal H-RS cell population, as detected by the single cell assay, but cells corresponding to the SNC clone were not demonstrable by this highly sensitive technique. These findings suggest that the SNC arising in this case represents the development of the second neoplasm clonally unrelated to the preceding NSHD. They also support the recent findings that the H-RS cells in classical HD consist of a clonal population of B cells.
