Formation of alpha-1,2- and alpha-1,3-linked mannose disaccharides from mannosyl retinyl phosphate by rat liver membrane enzymes.

Mannosyl retinyl phosphate (MRP) was an active substrate for the transfer of mannose to methyl-alpha-D-mannose (CH3-alpha-man), p-nitrophenyl-alpha-D-mannose, and free mannose. The products formed during MRP incubation with CH3-alpha-man or with mannose were alpha-linked. The disaccharides formed by incubation of MRP with mannose were identified by paper chromatography and electrophoresis as mannose-alpha-1,2-mannose and mannose-alpha-1,3-mannose. Triton X-100 greatly inhibited mannose-alpha-1,3-mannose synthesis. In the absence of detergent, MnCl2, NiCl2, and ZnCl2 inhibited synthesis of both products. Formation of mannose-alpha-1,3-mannose was more sensitive to preincubation of the enzyme at 40 degrees C then was synthesis of mannose-alpha-1,2-mannose. No differences in membrane mannosyltransferase activity with MRP, compared to DMP, could be demonstrated.

[Fibrin glue for otorhinolaryngology (author's transl)].

A tissue glue easy to produce and made of the patient's own blood plasma could be synthesized. By the addition of different mixtures its characteristics and its applicability were tested. The most qualified glue combination for otorhinolaryngology was determined. Excellent results were obtained upon application on dura plastics and in microsurgery. The advantages of the glue are: no danger of infection by virus hepatitis, flexible intervals of adhesion, no storage problems, easy to synthesize, few expenses.

[Comparative determination of the vesico-urethral angle in incontinence via ultrasound and lateral urethro-cystogram (author's transl)].

Pre-operative diagnostic procedures in evaluating stress urinary incontinence in women became increasingly important within the last years. In many clinics the measurement of the posterior vesico-urethral angle using lateral urethro-cystograms has become an eminent criterion for the selection of the operative method applied. Until now, this angle could be measured by employing X-ray examination techniques. In this paper, an ultrasonic examination with the same accuracy as in X-ray. The technique of both methods and their advantages and disadvantages are discussed.

Effect of vitamin A deficiency on glycosylation of rat serum alpha 1-macroglobulin.

We previously reported that synthesis of the serum glycoprotein alpha 1-macroglobulin (alpha 1-M) was decreased in vitamin A-deficient rats. In a reexamination of this conclusion we have shown that the primary synthetic change, reflected by labeled glucosamine uptake, was an underglycosylation which initially has no effect on the glycoprotein's concentration in serum. The severity of this lesion developed progressively and was independent of the glycosylation of total serum glycoprotein, at least up to 11 days after onset of the deficiency. Less glucosamine was demonstrated chemically in a alpha 1-M purified from rats in the first week of deficiency; alpha 1-M from more severely deficient rats showed significant depressions of mannose, galactose, and glucosamine.

Synthesis in vitro of corticosteroid-binding globulin from rat liver messenger ribonucleic acid.

To show that corticosteroid-binding globulin (CBG), a plasma glycoprotein, is synthesized in the liver as well as to develop a tool with which to study the hormonal factors which control its synthesis, we undertook to translate rat CBG mRNA in vitro. Rat liver mRNA was translated in the presence of [35S]methionine in both a rabbit reticulocyte lysate (cell-free) System and a Xenopus laevis oocyte (whole cell) system. A monospecific antibody against highly purified rat CBG was used to precipitate a single 35S-labeled product from the mixture of newly synthesized hepatic proteins in the two systems. In the rabbit reticulocyte lysate system, the protein migrated with an apparent molecular weight of 41,000 on sodium dodecyl sulfate-gel electrophoresis; CBG isolated from rat serum migrated as a doublet with apparent molecular weights of 56,000 and 62,500. Since proteins synthesized by a cell-free system are not glycosylated, the difference in molecular weight can probably be accounted for by the carbohydrate content of plasma CBG. Translation of rat liver mRNA in a Xenopus laevis oocyte system again resulted in immunoprecipitation of a single 35S-labeled protein, but this time with an apparent molecular weight of 53,000 on sodium dodecyl sulfate-gel electrophoresis; this probably represents glycosylated CBG and corresponds to the apparent molecular weight of one of the species isolated from rat plasma.

[Behavior of reticulocytes of the peripheral blood after whole-body irradiation and stimulation of the bone marrow - a contribution for the detection of radiation-induced changes in cells of the peripheral blood].

The relative reticulocyte content and the average Fe-uptake of peripheral reticulocytes were investigated in rats after a blood loss and whole body irradiation as well after a combined treatment for a time of 15 days. The acute loss of blood caused a rapid increase of cellular uptake within 24 hours, whereas after irradiation a considerable diminution could be observed. In addition to a direct stimulation or inhibition a considerable diminution could be observed. In addition to a direct stimulation or inhibition of bone-marrow activity a direct impact of reticulocytes by blood loss and irradiation is discussed.

[Analysis of lymphatic node shadows in the axilla via mammography (author's transl)].

1. There exist a great number of lymphnode-shadows. 2. In senium you will see more lymphnode shadows than in the youth. 3. Evaluation of the axillary lymphnodes permits the differentiation between fatty infiltration and normal. A differentiation between inflammation and metastases is not allowed. 4. The existence of lymphnode shadows is not of importance for the result of the report.

Vitamin A and galactosyl transferase of tracheal epithelium.

We developed an assay for galactosyl transferase (uridine diphosphategalactose:glycoprotein galactosyltransferase, EC 2.4.1.22) in microsomes of rat tracheal epithelium and characterized the properties of this enzyme system. We found that vitamin A deficiency greatly depressed galactosyl transferase activity. Activity could be restored within 48 h by dosing the animal with retinyl acetate. Adding retinol to the microsomes from tracheal epithelium of vitamin A-deficient rats also restored galactosyl transferase activity to some extent. Full restoration was achieved by pre-incubating retinol with the microsomal preparation for 30 min. Optimal concentration of pre-incubated retinol was 10(-8) M. Pre-incubation with retinyl phosphate and retinoic acid stimulated galactosyl transferase activity in microsomes from tracheas of deficient rats; pre-incubation with dolichyl phosphate, anhydroretinol and the dimethylacetylcyclopentenyl analog of retinoic acid did not. We concluded that vitamin A is involved in the galactosyl transferase system of rat tracheal epithelium, possibly by being linked to galactose.

Quantitative assessment of brain lesions.

The availability of precisely measurable dependent variables such as neurochemical activity, homeostatic changes, and behavioral patterns calls for more precise methods of measuring the independent variables of brain lesion research. In this study a method of measuring volumes of internal brain lesions and estimating amounts of damage to specific nuclei is described. Enlarged pictures of histologic sections through the lesion area are used. The structures to be measured are cut out of the pictures and weighed, and a weight-volume conversion formula is applied to yield the volumes of the structures. Two variations of this volumetric method are compared and evaluated. Various applications and advantages of quantified lesion data are discussed and appropriate statistical procedures are described.
