Comparison of RNases and toxins upon injection into Xenopus oocytes.

Several toxins abolish cellular protein synthesis by attacking specific sites in 28 S RNA. One of these toxins, alpha-sarcin, is an RNase that also cleaves nonspecifically on the 3' side of purines in deproteinized RNA. Several other RNases were injected into Xenopus oocytes, examined for their ability to abolish protein synthesis, and compared with alpha-sarcin and ricin. Surprisingly, pancreatic RNase A or B abolished oocyte protein synthesis at concentrations (approximately 0.03 nM) comparable to, or lower than, the amount of alpha-sarcin (approximately 2 nM) or ricin (approximately 0.07 nM) required to abolish protein synthesis. RNases S and T1 only inhibited oocyte protein synthesis when used at concentrations approximately 10 x higher than RNase A whereas RNases C, T2, U2, and nuclease P1 required concentrations approximately 100 times higher than RNase A to abolish protein synthesis. There was a direct correlation between the degradation of oocyte RNA and the inhibition of protein synthesis. The RNase inhibitors RNasin and Inhibit-Ace injected into the oocyte both prevented RNase A from hydrolyzing oocyte rRNA and abolishing protein synthesis. Enzymatically inactive oxidized RNase A did not inhibit protein synthesis when injected into the oocyte. None of the RNases or alpha-sarcin abolished protein synthesis when added to oocyte extracellular medium. Angiogenin is a human plasma protein that induces blood vessel formation in chick embryos, has 35% amino acid identity with RNase A, and cleaves 18 S and 28 S RNA in rabbit reticulocyte lysates (St. Clair, D. K., Rybak, S. M., Riordan, J. F. & Vallee, B. L. (1988) Biochemistry 27, 7263-7268, and references therein). Recombinant angiogenin injected into oocytes abolished protein synthesis, and this toxic effect was inhibited by RNasin but was not inhibited by Inhibit-Ace. Unlike RNase A and the other nucleases that hydrolyzed cellular rRNA, no cleavage of 18 or 28 S RNA by recombinant angiogenin was seen at concentrations 100 x greater than necessary to abolish protein synthesis. Recombinant angiogenin must selectively attack specific RNA(s) or another target in the cell.

[Is it permissible to treat young diabetics with oral antidiabetics?].

Authors analyzed the case history of 25 young diabetic patients, whose disease has been diagnosed before the age of thirty. The question that has been raised: is it allowed to treat young diabetics with oral drugs? By classifying the patients, they stated followings: In the 1. group they classified 16 verified MODY/NIDDY patients. In the second group they classified 3 young diabetics, whose disease had been evaluated as slowly progressing IDDM (autoimmune form). 3 patients belonged to the 3 group. They had been classified as MODY/NIDDY patients, however an extremely long lasting remission period--due to the short observation time--can not be excluded. The remaining 3 diabetic patients belonged to the IDDM group, with a long remission period. They were treated incorrectly with oral hypoglycemic drugs. Young diabetics can be treated with oral drugs only in case, when they are proven MODY/NIDDY patients. The precise differential diagnosis between this form and autoimmune IDDM, as well as long lasting remission periode, is extremely important.

[Differences in nutrition behavior between weekends and work days: results of the nutrition survey in 1984/85 of the MONICA project in Augsburg].

Seven-day food records of the MONICA project Augsburg dietary survey 1984/85 in 899 men aged 45-64 years were used to compare food and nutrient intake of weekdays and weekends. Differences were found in food intake. The mean intake of energy, macronutrients and especially alcohol was higher on weekends, whereas the dietary constituents independent of total energy intake, as percentage supply of energy from macronutrients, did not show huge differences.

Analysis of subsite preferences of HIV-1 proteinase using MA/CA junction peptides substituted at the P3-P1' positions.

The residues P3, P2, P1, and P1' of a peptide corresponding to the matrix/capsid protein junction in the HIV-1 gag protein (Ser-Gln-Asn-Tyr-Pro-Ile-Val) were systematically replaced and the effect of these single amino acid substitutions on the hydrolysis of each peptide by HIV-1 proteinase was studied. Subsites S1 and S1' of the enzyme showed explicit preference for hydrophobic moieties, but beta-branched amino acids and proline are not tolerated in S1. The S2 subsite shows a preference for small polar and apolar amino acids; it may be occupied by Asn, Asp, Glu, Cys, Ala, or Val, other substitutions, especially by Gln and Ser, prevent hydrolysis of the peptides. In subsite S3 all amino acids except proline can be accommodated.

CD4-Pseudomonas exotoxin hybrid proteins: modulation of potency and therapeutic window through structural design and characterization of cell internalization.

Replacing the Pseudomonas exotoxin A (PE) cell binding domain with the human immunodeficiency virus (HIV) gp120 binding domain from CD4 yields a hybrid toxin (CD4-PE) with potential therapeutic use in treating acquired immunodeficiency syndrome (AIDS). To find the most therapeutically potent combination of CD4 and PE four different hybrid toxins composed of one [CD4(122)] or two [CD4(181)] Ig-like CD4 domains and sequences of PE where the binding domain was partially [PE(392)] or completely [PE(364)] removed were constructed and expressed in Escherichia coli. The number of CD4 domains determined the binding affinity to gp120 and in cell viability assays the window between specific and nonspecific cytotoxicity. The length of PE determined the potency of the drug. The optimal hybrid toxin was composed of two Ig-like domains of CD4 and PE(392). Investigation of the internalization mechanism of CD4-PE revealed that the hybrid toxin binds to target cells and is endocytosed within one hour. However, more than 6 hours are required for maximum translation inhibition. In contrast to PE which is inhibited by ammonium chloride treatment, cell toxicity of CD4-PE is not affected by ammonium chloride. Further investigations showed that the acid-induced hydrophobicity change which is required for membrane translocation is also observed with CD4-PE but at significantly higher pH than with PE.

Selected nutrient intakes of middle-aged men in southern Germany: results from the WHO MONICA Augsburg Dietary Survey of 1984/85.

In the WHO MONICA Augsburg survey of 1984/85 dietary intake was assessed in an age-stratified cluster sample of 899 men aged 45-64 years by 7-day records. The mean energy intake was 2,609 kcal (10.9 MJ), 15.9% of which came from protein, 38.1% from fat, and 36.6% from carbohydrate. A remarkably high proportion of 9.4% was derived from alcohol. Saturated fatty acids accounted for 14.6% of total energy, monounsaturated fatty acids for 13.2% and polyunsaturated fatty acids for 5.2%. The results showed that current dietary habits do not conform with the national nutritional guidelines.

Characterization of a DL-dityrosine-containing macromolecule from yeast ascospore walls.

We have shown previously that the outer layers of yeast ascospore walls contain dityrosine and that this amino acid is a major component of the cross-linked peptides present in the spore wall (Briza, P., Winkler, G., Kalchhauser, H., and Breitenbach, M. (1986) J. Biol. Chem. 261, 4288-4294). We now present evidence that dityrosine is located in the outermost layer and that it is in the DL-configuration. Although the proteins (peptides) of the spore wall are insoluble, the macromolecule containing dityrosine can be solubilized by partial acid hydrolysis of spore walls. Analysis of this macromolecule indicates that it contains more than 50 mol% dityrosine and a very limited number of other amino acids. Interestingly, part of the dityrosine of spore walls is present in the DL-configuration. We speculate that not only the high degree of cross-links in the outermost layer but also the D-configuration of part of the alpha-C-atoms of dityrosine could contribute to the spores' resistance to lytic enzymes.

[Long-term remission in diabetes mellitus diagnosed at an early age].

The authors deal with the clinical picture of total remission in diabetes, among young patients (below 30 years). In their interpretation "complete remission" means total withdrawal of insulin treatment for at least 2 months. Out of 14 patients with complete remission, the classified 7 patients--by clinical and immunogenetical parameters--as noninsulin-dependent diabetes in the young (MODY-NIDDY). 1 diabetic patient belongs to the autoimmune-subgroup of IDDM. The remaining 6 patients could be classified as IDDM-s. However their clinical and immunogenetical parameters were rather atypical. In conclusion they raised the possibility that this subgroup is heterogenous with in IDDM.

Acidotropic amines inhibit proteolytic processing of flavivirus prM protein.

Treatment of flavivirus-infected mammalian and mosquito cells with acidotropic amines (such as chloroquine, ammonium chloride, or methylamine) inhibited the normal proteolytic processing of the virus prM protein to M. As a result, virions from infected cells which had been treated with acidotropic amines late in the virus replication cycle contained prM protein rather than M protein. Identification of the prM protein was based on molecular weight, glycosylation, and reactivity with an anti-prM monoclonal antibody. Infected cells which had not been treated with acidotropic amines did release, along with virions which contained the mature M protein, variable amounts of virus containing the prM precursor. The relative amounts of these two types of virions were influenced both by the virus and the host cell type. Virions containing the prM protein had a lower specific infectivity than virions containing the M protein; however, in experiments with a macrophage cell line this low specific infectivity was significantly increased if the anti-prM monoclonal antibody was used to facilitate virus entry via Fc receptors. Our findings indicate that the proteolytic cleavage of prM requires an acidic environment and is necessary to generate fully infectious virus. We suggest that the cleavage of prM occurs in the acidic post-Golgi vesicles.

Products of Allicin Transformation: Ajoenes and Dithiins, Characterization and their Determination by HPLC*.

Constituents of crushed garlic bulbs ( ALLIUM SATIVUM L.), such as allicin, ( E/Z)-ajoene, and the vinyldithiins, were synthesized and purified by an LC method. Purity was proved by TLC, HPLC, and GC systems. Identity was confirmed by comparison with spectral data from the literature. In order to establish external standards, silica gel adsorbates of the respective substances were prepared. For quantitative determination of allicin and its transformation products, an isocratic HPLC method was developed. Calibration was performed by the use of these external standards, and by comparison to an internal standard. The influence of different media on allicin transformation, including the ( E/Z)-ratio of the ajoenes, has been elaborated.

Quantitative Determination of Allicin and Alliin from Garlic by HPLC*.

The procedure for allicin synthesis could be improved. The time for HPLC analysis of allicin was shortened by the application of different isocratic elution systems. Calibration was performed by use of an allicin/silica gel adsorbate as external standard and its allicin content could be confirmed by different methods. L-(+)-Alliin was synthesized and applied as external standard for the quantitative determination of alliin by HPLC. Diastereoisomers had been separated by repeated recrystallization. Fresh ALLIUM SATIVUM bulbs from different origins were analyzed with respect to allicin content after complete enzymatic conversion of alliin; allicin contents found were in the range of 0.4%. The corresponding alliin contents were in the range of 0.9%. For a comparative evaluation of the alliin- and the allicin-HPLC determination methods, commercially available garlic preparations were analyzed, demonstrating that both methods are appropriate. However, the application of the HPLC system for allicin determination, in addition to providing information on the alliin-dependent allicin-generating capacity, enables the simultaneous quantification of allicin transformation products, such as ajoenes, dithiins, and alkyl sulfides. It was found that, for quantitative GLC analysis of allicin, allicin has to be used as an external standard.

Mental and behavioural disorders due to psychoactive substance use (section F1): results of the ICD-10 field trial.

The ICD-10 field trial of section F1 (mental and behavioural disorders due to psychoactive substance use) shows comparatively high acceptance of the new classification in German-speaking countries. Goodness of fit and subjective confidence of the main diagnosis are above average. Analysis of interrater reliability with regard to main diagnostic categories reveal certain problems. The reliability values are flow average within the ICD-10 field study. Possible causes for low interrater reliability are inadequate differentiation from section F0 (organic disorders) and uncertainties in the differential assessment of psychopathological symptoms between F1 and F2, as also between F1 and F4. Interrater reliability could be improved by introducing "psychoactive substance use" as a separate axis in a forthcoming multiaxial scheme for ICD-10.

CD4-Pseudomonas exotoxin conjugates delay but do not fully inhibit human immunodeficiency virus replication in lymphocytes in vitro.

The CD4 molecule is a high affinity receptor for the human immunodeficiency virus (HIV) envelope glycoprotein (gp160 or gp120). This glycoprotein is expressed on the surface membrane of cells infected with HIV. It has, therefore, been suggested that a soluble form of CD4 might be used as a targeting agent to deliver toxins selectively to cells infected with HIV. We demonstrate that CD4-Pseudomonas exotoxin A (PE) conjugates inhibit the proliferation of gp160-transfected Chinese hamster ovary cells and block HIV replication in virus-infected H9 cells. However, this inhibition of HIV replication appears to be incomplete since virus replication occurs following removal of the toxin conjugates from these cultures. Moreover, CD4-PE conjugates delay but do not inhibit HIV replication in human peripheral blood lymphocytes. These studies suggest that such conjugates should be assessed only as potential adjunctive therapies in the acquired immunodeficiency syndrome.

[Feline immunodeficiency virus in Switzerland: clinical aspects and epidemiology in comparison with feline leukemia virus and coronaviruses].

Serum samples from 1421 domestic cats (561 healthy, 860 sick) were tested for FIV-, FeLV- and coronavirus infection. The results were stored in a computer data base and compared with epidemiologic data and clinical findings. All 3 infections were significantly more prevalent in sick than healthy cats: FIV was found in 0.7% of healthy and 3.4% of sick cats. For FeLV the prevalence was 3.0% and 13.0% and for coronavirus 21% and 36.2%, respectively. FIV-infected cats were mostly male (73%); no sex predilection was observed in FeLV- and coronavirus infection. In sick cats FIV-infection was significantly more prevalent in cats greater than 2 years of age; no age-dependence was found in FeLV- and coronavirus infections. The prevalence of FIV-infection increased significantly with the number of animals per household. In contrast, the frequency of FeLV infection decreased with the number of animals per household. Prevalence of coronavirus infection did not vary with group size or living conditions. The following clinical symptoms were associated with infection: FIV: general depression, diseases of the urinary tract; FeLV: general depression, fever, rough hair coat, lymphadenopathy, impaired functions of heart and circulation and muscle atrophy; coronavirus: lymphadenopathy and alterations in the abdomen. It was concluded that based on the clinical symptoms alone FIV-infection could not be diagnosed nor differentiated from the other 2 infections.

Purification, assay and kinetic features of HIV-1 proteinase.

1) The aspartic proteinase of the human immunodeficiency virus type 1 (HIV-1) was purified from cultures of recombinant E. coli. The enzyme preparation is homogeneous as judged by SDS-polyacrylamide gel electrophoresis and isoelectric focusing. 2) A rapid assay procedure for the proteinase was established which makes use of the cleavage of a radiolabeled decapeptide and the separation of substrate and labeled product by ion-exchange resin. 3) Activity of the enzyme is optimal at an ionic strength of 2.5-3.5M; also, the inhibitor pepstatin is a more potent inhibitor at higher ionic strength. This can be attributed to a tighter binding of both substrate and inhibitor in high-salt buffer. 4) The Km value of the decapeptide substrate is independent of the pH in the range of 3.5-7.5, while kcat shows a bell-shaped curve with a maximum at pH 5.2. The shape of the curve can be attributed to pKa values of 4.2 and 6.2 of groups on the enzyme. Pepstatin inhibition is optimal below pH 5.5, but becomes weak above pH 6.

[The effect of intravenous administration of arginine and glycine on the blood plasma content of glucose, insulin, free fatty acids and total alpha-amino-N-compounds in lambs of different ages].

Intravenous infusion of 0.5 mmol/kg B.W. to lambs aged eight to 14, 15 to 21, 22 to 28, and 29 to 35 days led to higher blood plasma levels of insulin and total alpha-amino-N. Rises were stronger and longer lasting after infusion of 1.25 mmol/kg B.W. of arginine. Blood plasma levels of glucose and free fatty acids were temporarily decreased in response to infusion. Infusion to lambs aged between eight and 14 days of 0.5 mmol/kg B.W. of glycin did not change insulin, glucose nor free fatty acids, though total alpha-amino-N was very slightly increased for very short time. Infusion of 1.25 mmol/kg B.W. of glycin proved less effective than that of arginine with regard to higher levels of insulin and total alpha-amino-N.

[Clinical and laboratory chemical studies of the incidence and extent of diabetic cheiropathy in childhood and adolescence].

Diabetic cheiropathy is often not diagnosed because of a paucity of symptoms. In the present study, 100 type I diabetic patients and 20 controls were investigated. The age of patients ranged from seven to 26 years, the duration of diabetes was one to 23 years. Extension of hands by "prayer position", a hand-print and angle determinations of joints were considered for the determination of diabetic cheiropathy. The presence of cheiropathy was unrelated to the duration of diabetes. However, it was well correlated with the quality of blood sugar control and it was often associated with diabetic retinopathy: Diabetic cheiropathy was detected in 53 (53%) of the diabetic patients, and in 26 (49%) of them diabetic retinopathy was also diagnosed. Only three patients had a retinopathy without cheiropathy. On the basis of our results, diabetic cheiropathy is a surprisingly common feature in young type I-diabetics. It is not to be considered as a late complication but rather a sequel of badly controlled diabetes mellitus.

Awareness and treatment of hypercholesterolemia: results of the first survey of the MONICA Project Augsburg.

In the first survey of the MONICA project Augsburg (1984/1985), awareness and treatment of hypercholesterolemia was determined on a two-stage cluster sample of 5069 25-64 year old citizens (participation 79.6%). Hypercholesterolemia was defined according to the Conference of the European Atherosclerosis Society (EAS) and of the National Cholesterol Education Program (NCEP). Awareness (elevated lipid levels found during the last 12 months), and drug and dietary treatment were determined by an interview. The awareness was low and varied between 4 and 24% in the different age-sex groups with no significant differences between sexes, with one exception: The awareness in women aged 55-64 years with cholesterol levels greater than or equal to 300 mg/dl was 38% and differed significantly from that found in men in the same age group (21%). Older participants showed a higher degree of awareness than younger; in men the awareness was independent from the cholesterol level, in women those with higher levels showed a better awareness than those with lower levels (results from a logistic regression analysis for cholesterol levels greater than or equal to 240 mg/dl). The use of lipid-lowering drugs was low in the study population (2% in men, 1% in women). The drug treatment of hypercholesterolemia was low and in no age-sex group higher than 7%, despite the definition of hypercholesterolemia; the dietary treatment was not higher than 20%. In conclusion it is shown, that there is a great need for the identification and management of hypercholesterolemia in the examined population in Southern Germany.
