New neural cell lines derived from experimentally induced rat tumors and from human neuroblastomas.

We have isolated a series of new cell lines from tumors of the nervous system. Two approaches have been used. In the first, tumors were induced in neonatal rats with the potent carcinogen ethylnitrosourea. Tumors were put into cell culture and cloned after several passages. The resultant cell lines were characterized for a variety of presumptive neuronal and glial functions. Our second approach, designed to identify lines with neuronal characteristics, has been to culture and analyze cells derived from human neurolbastomas. Five lines have been characterized for the Na+ action potential ionophore and several enzyme markers.

Prevalence of Coxiella burnetii (Q fever) and Toxoplasma gondii among dairy goats in California.

The prevalence of antibodies against Coxiella burnetii and Toxoplasma gondii, the causative agents of Q fever and toxoplasmosis, was examined in goats to determine the extent of the potential of goats to tansfer these infections to persons. Of 1,054 goats from 234 premises in California, 248 goats (24%) were seropositive for C burnetii and 246 (23%) were seropositive for T gondii. Of 29 dairy goats tested, 2 (7%) were found to be shedding the Q fever rickettsia in their milk; T gondii was not detected. Factors of sex, age, location, and herd size are discussed.

Initial comparative response to peak pions and x-rays of normal skin and underlying tissue surrounding superficial metastatic nodules.

Given the limitations of available material and methods for measuring skin response, the relative biological effectivenss (RBE) for the development and healing of skin reaction to pions in this experiment is 1.43. This is based on data obtained from a patient with malignant melanoma, in whom multiple skin nodules and the surrounding normal skin were randomized into three dose levels for pions and x-rays. The RBE for skin reaction was obtained while the skin tumor nodules appeared to regress at least as rapidly with pion therapy as with x-rays. Without benefit of adequate observation of time required for nodule regrowth, any estimate of tumor RBE is speculative.

Effects of nerve growth factor administration of N-ethyl-N-nitrosourea carcinogenesis.

We attempted to determine whether the sympathetic nervous system in rodents is susceptible to co-carcinogenesis, employing murine salivary nerve growth factor (NGF) as a co-carcinogenic agent. NGF had no co-carcinogenic effect with either methylcholanthrene or ethylnitrosourea (ENU) on the sympathetic nervous system of the mouse, whether administered transplacentally, postnatally, or both transplacentally and postnatally. At a dose of ENU of 30 mug/g body weight, NGF did not shorten the latent period for tumor induction of BD-IX rats. In contrast, a 25% reduction in latent period was brough about by NGF for tumor appearance in BD-IX rats receiving 90 mug/g ENU. In both cases the frequency of urogenital tumors in rats was increased as a result of NGF administration, at the apparent expense of neural tumors.
