Effects of shape-discrimination training on the selectivity of inferotemporal cells in adult monkeys.

Through extensive training, humans can become "visual experts, " able to visually distinguish subtle differences among similar objects with greater ease than those who are untrained. To understand the neural mechanisms behind this acquired discrimination ability, adult monkeys were fully trained to discriminate 28 moderately complex shapes. The training effects on the stimulus selectivity of cells in area TE of the inferotemporal cortex were then examined in anesthetized preparations. Area TE represents a later stage of the ventral visual cortical pathway that is known to mediate visual object discrimination and recognition. The recordings from the trained monkeys and untrained controls showed that the proportion of TE cells responsive to some member of the 28 stimuli was significantly greater in the trained monkeys than that in the control monkeys. Cell responses recorded from the trained monkeys were not sharply tuned to single training stimuli, but rather broadly covered several training stimuli. The distances among the training stimuli in the response space spanned by responses of the recorded TE cells were significantly greater in the trained monkeys than those in the control monkeys. The subset of training stimuli to which individual cells responded differed from cell to cell with only partial overlaps, suggesting that the cells responded to features common to several stimuli. These results are consistent with a model in which visual expertise is acquired through the development of differential responses by inferotemporal cells to the images of relevant objects.

Survival of enterohemorrhagic Escherichia coli O157:H7 in water.

Several recent Escherichia coli O157:H7 outbreaks associated with both drinking and recreational water raise concerns about waterborne illness caused by this pathogen. The survival characteristics of a mixture of five nalidixic acid-resistant E. coli O157:H7 strains (10(3) CFU/ml) in filtered and autoclaved municipal water, in reservoir water, and in water from two recreational lakes were determined for a period of 91 days at 8, 15 or 25 degrees C. Greatest survival was in filtered autoclaved municipal water and least in lake water. Regardless of the water source, survival was greatest at 8 degrees C and least at 25 degrees C. E. coli O157:H7 populations decreased by 1 to 2 log10 by 91 days at 8 degrees C, whereas the pathogen was not detectable (> or 3 = log10 decrease) within 49 to 84 days at 25 degrees C in three of the four water sources. SDS-PAGE of surface antigens of surviving cells revealed that there was no major alteration in lipopolysaccharide pattern, but outer membrane protein composition did change. These studies indicate that E. coli O157:H7 is a hardy pathogen that can survive for long periods of time in water, especially at cold temperatures. However, direct viable counts of E. coli O157:H7 determined by acridine orange staining remained essentially the same for 12 weeks at 25 degrees C, whereas viable counts on tryptic soy agar plates decreased to undetectable levels within 12 weeks. Results suggest that E. coli O157:H7 can enter a viable but nonculturable (VBNC) state in water.

[Determination of copper, iron and zinc in hair of mentally retarded children by oscillopolarography].

The oscillopolarographic method (proposed by Ding Jianwen)for the simulltaneous determination of copper, iron and zinc in hair was further studied and applied to the investigation of copper, iron, zinc content in hair samples from 46 mentally retarded children and 49 healthy children. For copper, iron, zinc, the detection limits were 0.03, 0.03, 0.01 microgram/ml, dynamic range 0.05-15, 0.05-2.5, 0.2-10 micrograms/ml, the RSD of sample analysis 3.2%, 3.3%, 2.4%, the recoveries 96: 0%-105.2%, 2%, 97.3%-109.0%, 94.7%-107.7%, respectively. The difference between the results for the same sample by this method and the atomic absorption spectroscopic method was no statistical significance. The results showed that the copper content and iron content of hair samples from mentally retarded children were significantly lower than those of healthy children.

[Study on the antagonistic action of selenite on fluoride-induced lipid peroxidation and on the changes of trace elements in rats].

Five groups of SD male rats were provided with deionized drinking water containing 0 and 150 mg/L NaF, and containing both 150 mg/L NaF and 0.5, 2.0 or 4.0 mg/L Na2SeO3 respectively for 10 weeks, in order to find out the optimal level of selenite in drinking water against fluoride toxicity. The results showed that fluoride could cause significant increase of lipid peroxides (LPO) and metabolic disorder of trace elements in the serum and kidney of rats. The antagonistic effect of 2.0 mg/L Na2SeO3 drinking water on the lipid peroxidation induced by 150 mg/LNaF was the most evident, whereas those of 0.5 and 4.0 mg/L Na2SeO3 were not obvious. It is concluded that selenite possesses significant antagonistic effects on renal damages induced by fluoride and 2.0 mg/L Na2SeO3 is the optimal concentration for the antagonistic effect on renal impairments induced by 150 mg/L NaF in drinking water.

[The study of endothelin and endothelin receptors in cyclic human endometrium].

OBJECTIVE: To determine the distribution of endothelin (ET) peptide and endothelin as well as their receptors (ETaR and ETbR) mRNA in human cycling endometrium in order to assess possible role of ET in endometrial reproductive physiology. METHODS: Twenty-three endometrial specimens from gynecological surgery for benign conditions were studied by means of immunohistochemistry, in situ hybridization and computer quantitative image analysis. RESULTS: The expression of ET peptide and ET mRNA in human endometrium varies throughout the menstrual cycle, increasing gradually from proliferative phase to secretory phase, with maximal levels in the pre-menstrual phase. Stromal expression is lower than in glandular epithelium and remains relatively constant in the menstrual cycle. ETaR and ETbR mRNA are detected in normal human endometrium, higher in glandular epithelial cells than in stromal cells. ETaR mRNA expression has no obvious cyclic changes, but ETbR mRNA expressions are higher in secretory endometrium than in proliferative endometrium. The ratio of ETaR/ETbR has also cyclic changes. CONCLUSION: Changes of Expression of ET and ET receptors in the endometrium throughout the menstrual cycle suggest that ET might be important in endometrial regeneration and repair following menstruation, and probably plays a role as the paracrine control of the uterine vascular bed.

[Influence of fetal distress on neonatal behavior neurological assessment and its prognosis].

OBJECTIVE: To study the short and long term prognosis of fetal distress. METHODS: 60 newborns with fetal distress (study group) and 60 normal newborns (control group) were assessed by neonatal behavior neurological assessment (20 items) in neonatal period and by reformed wechsler intelligence scale children-revised at 2 years of age. RESULTS: The total scoring of study group was significantly different from that of control group (P < 0.01). In the study group, there were 41 cases without neonatal asphyxia whose scoring was significantly different from that in the normal group (P < 0.05), whereas the scoring of other 19 cases with neonatal asphyxia was significantly lower than that of the control group (P < 0.001). We also found that the scoring of 38 acute fetal distress cases was significantly different from that of 22 chronic cases (P < 0.01). In two years follow-up study, the scoring of children's intelligence test in acute distress cases was not different from that of controls, but the scoring of intelligence test in chronic distress cases was markedly different from that of controls (P < 0.01). CONCLUSION: Fetal distress could affect neonatal behavior and their intellectual development.

[The changes of sex steroid receptors and morphology in Norplant treated endometrium].

OBJECTIVE: To study the relationship between the changes of sex steroid receptors as well as endometrial morphology and abnormal uterine bleeding induced by Norplant. METHODS: Immunohistochemistry, Western Blot, in situ hybridization and computer image analysis were employed to demonstrate the expression of estrone receptor(ER) progesterone receptor(PR) protein and mRNA as well as morphometric analysis of the endometrium in Norplant users. RESULTS: The content of DNA and the numbers of spiral arterioles decreased in Norplant treated endometrium. The percentage of glandular area in Norplant endometrium was much lower than that in cyclic endometrium. A statistically significant correlation was observed between the percentage of area of glands and abnormal uterine bleeding, as well as serum estrogen(E2) level, but E2 level had no direct relationship with the bleeding abnormality. As the time growing, the percentage of glandular area increased. Immunostaining and relative concentration of glandular and stromal ER in Norplant treated endometrium were significantly weaker and less than all the stage of cyclic endometrium. However, PR, especially stromal PR immunostaining was very intense, which was the same or more than that in all the stage of cyclic endometrium. In situ hybridization detected that the ER and PR mRNA was lower in Norplant treated endometrium than that in cyclic endometrium. The ER mRNA expression was more less abundant than PR mRNA expression. CONCLUSION: These results suggested that poor repair and regeneration of endometrium(especially glands and spiral arterioles) and abnormal expression of ER and PR may be related to pathogenesis of abnormal uterine bleeding induced by Norplant implant.

[Therapeutic effect of EGFR antisense cDNA on rat C6 gliomas in vivo].

OBJECTIVE: To study the therapeutic effect of EGFR antisense cDNA on rat C6 glioma in vivo and find out the feasibility of using EGFR gene as a target in gene therapy of gliomas. METHOD: Wild type C6 cells and C6 cells transfected with EGFR antisense cDNA were implanted stereotaxically to the caudate nucleus of right cerebrum of Wistar rats (control and transfected group, 8 and 6 rats respectively), and rats with cerebral tumor foci were treated with EGFR antisense cDNA mediated by Lipofectamine (treated group, 9 rats). The general manifestation, survival time, MRI features, histopathological change, proliferation activity and apoptosis of the gliomas in each group of rats were observed. RESULTS: The mean survival time of control animals was 17.3 days. That of the 6 transfected and 6 treated animals was significantly prolonged. Four transfected and 3 treated rats were killed on day 28-90 after implantation for histopathological examination. The surviving time of the 4 remaining rats (1 transfected and 3 treated rats) was over 200 days. MRI demonstrated a distinct cerebral tumor in control rats, but the tumor foci were not found in the transfected rats and disappeared almost completely in the treated rats. The cerebral gliomas of rats after treatment with EGFR antisense cDNA showed decrease in proliferation activity and apoptosis. CONCLUSION: The results of EGFR antisense cDNA in treatment of rat C6 glioma is encouraging. EGFR gene can be used as the target of gene therapy for malignant gliomas.

[Magnetic resonance imaging with multiplane histomorphometry in avascular necrosis of femoral head: analysis of trabeculae].

OBJECTIVE: To distinguish the differences of changes of bone trabecular structure between high and low signal area which could directly reflect the mechanical strength and estimate the risk of collapse in avascular necrosis of femoral head (ANFH). METHODS: After total hip replacement, the specimen of ANFH was scanned by MR imaging. Accurate localization, consecutive slicing, multiplane histomorphometry, correspondence of MR images with slices were used. Mean trabecular width (MTW micron) and mean trabecular space (MTS micron) were selected as observation indexes. RESULTS: MTW in the low signal area was lower than that in the high signal area. MTS in the low signal area was higher than that in the high signal area. The results indicated that the dispersive degree of bone trabeculae was higher in the low signal area. CONCLUSION: Bone trabeculae in the low signal area necrosed and atrophied so that the bone volume and mechanical strength of trabeculae declined and bone trabeculae in the low signal area were liable to microfracture and collapse.

[The effects of nimodipine on cerebrovascular function in acute cerebral infarction].

OBJECTIVES: To use cerebrovascular hemodynamics indexes (CVDI) to evaluate the effects of nimodipine on cerebrovascular function in acute cerebral infarction (ACI), and to investigate the changes of blood pressure (BP), heart rate (HR), and electrocardiography (ECG). METHODS: 30 patients with ACI in internal carotid artery system within 72 h from the oneset of symptoms were given nimodipine (10 mg, 0.25-0.5 microgram/kg/min) by intravenous drip (i.v.) every day for 14 days. CVDI, BP, HR and ECG were examined at various times during treatment. RESULTS: Nimodipine improved cerebrovascular function significantly. Cerebral blood velocity and flow increased, cerebrovascular resistance and criticle pressure decreased, and cerebrovascular elasticity and autoregulation improved. BP dropped significantly at the third day, and mainteined stable afterward. HR dropped remarkably at the fourteen day (P = 0.011). It was reasonable that the treatment of nimodipine (i.v.) lasted 10 days or so. CONCLUSIONS: Nimodipine improves cerebrovascular function significantly, but monitoring of BP and ECG is necessary during its application.

Pathologic changes, mechanisms and diagnosis in renal bone disease.

OBJECTIVE: To investigate the incidence rate, pathologic changes, mechanisms and diagnostic methods in renal bone disease. METHODS: The blood levels of carboxyterminal parpthyriod hormone (C-PTH), 1,25(OH)2D3, calcium and phosphate, aluminum in serum and bone tissue were measured. The bone biopsy and bone scan with 99m technetium methylene diphosphonate (99mTC-MDP) were performed in 51 uremic patients. RESULTS: One hundred per cent of the patients had varying degree of pathologic changes in bone, in which 50.9% of the patients presented high-turnover bone disease, 7.8% of the patients presented low-turnover bone disease and 41.8% of the patients had mixed-type bone disease. The levels of serum C-PTH were predominently high in high-turnover bone disease while the levels of serum 1,25(OH)2D3 were significantly decreased in low-turnover bone disease. There was a high positive rate for the diagnosis of renal bone disease by bone scan with 99mTC-MDP. CONCLUSIONS: The examination of bone pathology is the most valuable method for the diagnosis of renal bone disease. Bone scan with 99mTC-MDP has reference value when clinical conditions do not allow to make bone biopsy.

MR imaging in rat glioma model and gene therapy using EGFR antisence RNA.

OBJECTIVE: To study the value of 1.5T MRI in long-term follow-up of rat C6 glioma model and the efficacy of EGFR antisence RNA therapy in vivo. METHODS: Forty-three male Wistar rats, weighing 150 to 200 g, were used for this study. They were divided into four groups: group I (7 normal rats); group II (16 rats inoculated C6 cells in the right caudate nucleus); group III (12 rats with the C6 gliomas treated with EGFR antisence RNA by in site injection); and group IV (8 rats inoculated C6 cells transfected with EGFR antisence RNA in the right caudate nucleus). Group I was examined by plain and enhanced MR scanning. Group II-IV were followed up by plain and enhanced MR scanning and were sacrificed in variable time points for pathological examination. RESULTS: Cerebral hemispheres of normal rat were shown clearly on the MR image. The tumor could be seen about 1 week after inoculation. According to the findings on the follow-up MR scan, we could observe growth of the tumor or its regression after treatment. The tumor growth was significantly inhibited in group III and group IV as compared to those in group II. CONCLUSION: The growth of the rat C6 glioma model and its change after treatment could be shown clearly in 1.5T MR imaging; EGFR antisence RNA significantly inhibited the growth of glioma in vivo.

Lidocaine improving the cerebral protection by retrograde cerebral perfusion.

OBJECTIVE: To determine whether lidocaine can improve the cerebral protection provided by retrograde cerebral perfusion. METHODS: Sixteen mongrel dogs were placed on cardiopulmonary bypass and cooled to 20 degrees C. Retrograde cerebral perfusion was then carried out for 120 minutes, with the external jugular venous pressure kept at 3.33 kPa. Cardiopulmonary bypass was resumed, and the animals were rewarmed to 36 degrees C. The animals were divided into two groups. In the lidocaine group (n = 8), lidocaine was administrated continuously throughout the experiment. In the control group (n = 8), normal saline was given at the same rate. RESULTS: In both groups, cerebral tissue creatine phosphate and adenosine triphosphate concentrations and energy charge increased by the end of hypothermic cardiopulmonary bypass, decreased continuously during retrograde cerebral perfusion, and recovered gradually after resuming cardiopulmonary bypass. Nevertheless, they recovered to significantly higher levels in the lidocaine group than in the control group (creatine phosphate: 2.44 +/- 0.53 versus 1.61 +/- 0.49 mumol/g wet weight, P = 0.006; adenosine triphosphate: 0.71 +/- 0.18 versus 0.50 +/- 0.17 mumol/g wet weight, P = 0.029; energy charge: 0.59 +/- 0.10 versus 0.48 +/- 0.09, P = 0.044) by the end of the experiment. There was no significant difference between the two groups in the cerebral tissue water content (control group: 77.6% +/- 1.9%; lidocaine group: 77.6% +/- 1.3%). CONCLUSION: Continuous lidocaine infusion accelerates the recovery of cerebral tissue high energy phosphate contents after resuming cardiopulmonary bypass, but it has no effect on the formation of cerebral edema after retrograde cerebral perfusion.
