Streptococcus sanguis on media containing 3% sucrose gives rise to characteristic hard cohesive colonies (designated Spp+). Populations of Spp+ bacteria (strain Challis) on sucrose media switch to a soft noncohesive phenotype (designated Spp-) at a frequency of 10(-4) to 10(-3). Spp- bacteria switch back to Spp+ bacteria at a similar frequency. Successive rounds of Spp variation were observed. The Spp phenotypic switch was associated with changes in extracellular glucosyltransferase activity.
Increased sensitivity to killing by human serum complement occurs in Salmonella typhimurium strains in which the 95-kilobase virulence plasmid is integrated into the chromosome. This phenotypic change appears to be due to alterations in plasmid gene expression and is reversed by the presence of an autonomous plasmid bearing a cloned region of the virulence plasmid. Accordingly, this region has been termed rsk for reduced serum killing. Sequence analysis of the region reveals that rsk is composed of a series of direct 10-base-pair (bp) repeats with a 21-nucleotide periodicity. Two adjacent repeats are identical, but increasing loss of conservation is apparent with increased distance both 5' and 3' of these highly conserved 10-mers. The smallest isolated sequence which restores the serum-resistant phenotype is only 66 bp long and contains the two identical 10-mers and one degenerate 10-mer (8 of 10 bp conserved) 3' of these. The minimal rsk region of 66 bp does not appear to contain a coding sequence, or a promoter, for a structural gene. It is proposed that the minimal rsk is an isolated regulatory site involved in the regulation of the serum resistance of S. typhimurium. Integration of the 95-kilobase plasmid disrupts the normal regulation of virulence plasmid genes, resulting in an increase in the killing of the bacteria by complement activated by the classical pathway. The introduction of the minimal rsk on a multiple-copy plasmid restores resistance to serum killing, possibly through the titration of a trans-acting regulatory factor.
Integration of the Salmonella typhimurium virulence plasmid into the chromosome reduces mouse virulence, serum resistance, and HeLa cell adhesion-invasion while prolonging lag time in minimal medium. The proposed virulence plasmid regulatory element, rsk, partially restores virulence and fully restores the other three phenotypes to wild-type levels. Plasmid curing reduces virulence without affecting the other phenotypes. rsk has no apparent effect on the cured strain.
"The apparent Southeast Asian paradox of fairly slow urbanization but rapidly growing urban populations is due to continued high rates of natural increase, especially in rural areas. Southeast Asian countries differ greatly in the nature of their urban hierarchies, and the appropriate policy goals and strategies therefore also differ. In countering growing urban primacy, indirect approaches emphasizing macro-economic and sectoral policies conducive to more dispersed patterns of urban growth have greater potential impact than direct attempts to slow the growth of large city populations. Greater decentralization of power and decision making over resource allocation is also needed."

A large and symptomatic idiopathic renal arteriovenous (AV) fistula closed over a two-year period of clinicoradiologic observation. This feature of idiopathic renal AV fistulas has not been reported previously, to our knowledge. If the presenting symptoms permit nonintervention, not only the trauma-related but also the idiopathic renal AV fistula can be conservatively managed and may heal spontaneously.
The development of coccidial infections in 21 free-ranging, unweaned beef calves from birth to 8 months was investigated by examining faecal samples for oocysts. Most calves commenced shedding oocysts within a month of birth, and had shed all nine species identified by 3-4 months. Oocysts were shed by three calves as young as 12 and 13 days. The oocysts species shed earliest, and the commonest thereafter, were Eimeria bovis, E. ellipsoidalis and E. zuernii. Although several oocyst counts between 5.0 X 10(3) and 114.0 X 10(3)g-1 of faeces of these potentially pathogenic species were recorded there was no clinical disease. Some calves shed oocysts approximately four times more frequently than others, and individual species counts greater than or equal to 5.0 X 10(3)g-1 were confined to 10 of the calves. Intermittent oocyst shedding continued throughout the study with a similar distribution of oocyst species from all calves.

This is a one-to-one, age- and race-matched case-control study involving 55 histologically confirmed black prostate cancer patients and 55 controls who were seen at three major hospitals in Washington, DC from 1982 to 1984. Personal interviews were conducted to obtain the number of times food items of specified serving size were consumed per week by cases and controls; the subjects were grouped according to the age periods 30-49 and 50 years and older. We then calculated the average daily consumption of each of 18 nutrients per 1,000 calories. There was a significant negative association between linoleic acid (p less than 0.04) for the 50 years and older group, thiamin (p less than 0.05) for those 30-49 years old, riboflavin (p less than 0.03) for the 50 and older group, and iron (p less than 0.05) for those 30-49 years old. The results of this study suggest that the intake of thiamin and iron (in subjects 30-49 years old), linoleic acid and riboflavin (in subjects 50 years and over) could be protective because control subjects consumed more of these nutrients than did the cases.
A field investigation of marine coastal waters revealed that the frequency of pigmented bacteria and the occurrence of bacterial antibiotic resistance were higher at the air-water interface than in the bulk water. The differences in the frequency of pigmented colonies at the surface and in the bulk-water samples could not be explained by the degree of cell surface hydrophobicity or by bacterial adhesion to air-water interfaces. Pigmented strains exhibited a higher degree of multiple drug resistance than did nonpigmented strains. However, the frequency of multiple drug resistance in nonpigmented strains was also substantial. An average of 91% of all strains were resistant to more than one antibiotic, and 21% of the bacteria isolated were resistant to five of the eight antibiotics tested. High numbers of plasmid-carrying strains were found among selected surface isolates, but the presence of detectable plasmids could not be correlated with either pigmentation or multiple drug resistance. Furthermore, selected surface isolates were significantly more resistant to mercury than were bulk-water bacteria. The higher frequency of pigmented, antibiotic-resistant, and mercury resistant strains at the air-water interface than in the bulk water are discussed in terms of various forms of selective pressure and genetic exchange at the surface.
The association of the virulence plasmid of Salmonella typhimurium with resistance to the bactericidal activity of human serum was studied in chromosomally isogenic pairs of strains differing in their virulence plasmid status. The presence of the plasmid correlated in three pairs of strains with resistance to serum. The absence of the plasmid correlated with increased sensitivity to serum, whereas the reintroduction of the plasmid to the cell resulted in the restoration of resistance to serum. Complement was activated by the classical and alternative pathways equally well by both strains of a pair, but the differential bactericidal action of serum was apparent only after the activation of complement by the classical pathway. No differences in the chemical compositions or in the molecular weight ranges of the lipopolysaccharides were apparent between paired strains. This work confirms the presence of a virulence plasmid-associated mechanism of resistance to serum and distinguishes it from lipopolysaccharide-mediated resistance.

Post-weaning Eimeria zuernii coccidiosis in beef calves was treated prophylactically with monensin from intra-ruminal continuous release devices. The evaluation of four treatments during three consecutive years also allowed observations on the epidemiology of this disease to be made. Monensin was an effective prophylactic and the intra-ruminal devices were a convenient method of continuous administration over a one to two month period. The three years of the trial included two characteristically very dry and one unusually wet weaning period. Coccidiosis was more severe in the dry years suggesting that oocyst challenge was less important than the immunodepressive effect of weaning and dietary stress in precipitating disease. Severe disease also occurred in calves subsequent to elimination of oocyst shedding by monensin during the initial treatment trial. The occurrence of disease in conditions inimical to oocyst survival and development and after treatment suggested a role for arrested endogenous stages.
A series of (aminophenyl)pyrrolidine-2,5-diones has been prepared that bear structural similarities to aminoglutethimide (1, 3-(4-aminophenyl)-3-ethylpiperidine-2,6-dione). The inhibitory activity of these compounds was evaluated toward human placental aromatase and bovine adrenal cholesterol side chain cleavage (CSCC) enzyme assay systems. Selective, competitive inhibition of the aromatase enzyme system was demonstrated by 5 (3-(4-aminophenyl)-1-methyl-pyrrolidine-2,5-dione, Ki = 1.75 microM), 6 (3-(4-aminophenyl)-1,3-dimethylpyrrolidine-2,5-dione, Ki = 1.75 microM), 7 (3-(4-aminophenyl)-3-methylpyrrolidine-2,5-dione, Ki = 0.8 microM), and 8 (3-(4-aminophenyl)-3-ethylpyrrolidine-2,5-dione, Ki = 1.0 microM). Compound 15 (3-(4-aminophenyl)pyrrolidine-2,5-dione) proved unexpectedly difficult to prepare following standard methods and was only moderately inhibitory toward aromatase (IC50 = 20 microM). Compound 16 (3-(4-aminophenyl)-3-ethyl-1-methylpyrrolidine-2,5-dione) was weakly inhibitory toward testosterone aromatization and totally inactive toward androstenedione aromatization. These compounds were either weak or ineffective inhibitors of the CSCC enzyme systems, while 1 gave Ki values toward aromatase and CSCC enzymes of 0.68 and 14 microM, respectively. The unsubstituted phenylpyrrolidinediones were inactive in either system, and the 4-nitrophenyl derivatives exhibited weak, nonselective inhibition, indicating the importance of the primary amine moiety for potent inhibition of aromatase activity.
This one-to-one, age- and race-matched case-control study involved 181 histologically confirmed black prostate cancer patients and 181 controls seen at three major hospitals in Washington, DC, during the period 1979-1982. Personal interviews were conducted to obtain the number of times food items of specified serving size were consumed per week by cases and controls during the age periods 30-49 and 50 years and older. Then the average daily consumption of each of 18 nutrients per 1,000 calories was calculated. There was risk enhancement associated with increased intake of proteins, total fat, saturated fat, oleic acid, and vitamin A during the age period 30-49 years. The association was highly significant for vitamin A and approached statistical significance for the other four nutrients. A hypothesis based on disturbance of the zinc-retinol binding protein-vitamin A axis was put forward to explain the relative risk enhancement effect of vitamin A on prostate cancer.
