Molecular characteristics of uronic-acid-rich protein, a strong inhibitor of calcium oxalate crystallization in vitro.

Uronic-acid-rich protein (UAP) is a new urinary macromolecule which strongly inhibits calcium oxalate crystal formation. It is a glycoprotein with a molecular weight of about 35,000 Da, and its carbohydrate content is 8.5%. This inhibitor is composed of two polypeptidic chains crosslinked by chondroitin sulfate. It exhibits partial structural homology with alpha 1-microglobulin. The inhibitory activity seems to be supported by peptidic chains as determined by enzymatic assay.

[Use of a Ca(++) antagonist in noise-induced hearing loss in animal experiment].

Hearing thresholds at 1-2-4-6 and 8 kHz were determined by brainstem evoked response audiometry (BERA) in 24 guinea pigs. The animals were then divided into 6 groups. Group 1 (controls) received no drugs and were not exposed to noise. Group 2 were given 15 mg/24 h/kg diltiazem s.c. for 13 days. Group 4 was exposed to the same noise as group 3 but was also treated with diltiazem 15 mg/kg/24 h from three days before noise exposure until the end of the experiment. Group 5 was exposed to a 110 dB SPL broad-band noise for 3 h daily for 10 days. Group 6 was exposed to the same noise as group 5 and was treated with diltiazem 15 mg/kg/24 h from three days before noise exposure until the end of the experiment. After the last noise exposure hearing thresholds were determined again by BERA and were found to be unchanged in those animals treated with the drug alone and the controls. In those groups with drug administration and noise exposure hearing loss was less than in the animals exposed to noise alone. This difference was more pronounced in the 90 dB groups than in the 110 dB groups.

Complete 1H- and 13C-resonance assignments for D-mannooligosaccharides of the beta-D-(1-->2)-linked series released from the phosphopeptidomannan of Candida albicans VW.32 (serotype A).

D-Mannooligosaccharides (dp 1 to > 17) were released by mild acid hydrolysis from the phosphopeptidomannan of a Candida albicans strain of A serotype (VW.32). Among these, mannooligosaccharides ranging from bi- to hepta-ose, which were obtained in appreciable amounts, were structurally investigated and found to belong to the beta-D-(1-->2)-linked series. The occurrence of such compounds has already been reported in other Candida albicans strains. The complete 1H- and 13C-resonance assignments for manno-tri- to manno-hepta-ose are reported and general rules applicable for the 1NMR spectrum analysis of linear mannooligosaccharide of the general structure, beta-D-Man p-(1-->2)-[beta-D-Man p-(1-->2)]n-beta-D-Man p are proposed.

Structural analysis of the carbohydrate chains isolated from mistletoe (Viscum album) lectin I.

Two glycopeptide fractions prepared from mistletoe (Viscum album) lectin I by Pronase digestion were fractioned by affinity chromatography on a concanavalin A-Sepharose column. With 400-MHz 1H NMR spectroscopy, in conjunction with sugar analysis, the following oligosaccharide structures could be determined: two oligomannose-type glycans in the ratio 4:1, one containing six mannose and the other containing five mannose units, both with two 2-acetamido-2-deoxyglucose units. In addition, a mannotriosyl-->N,N'-diacetylchitobiose glycan containing a xylosyl group and an alpha-fucosyl group (1-->3)-linked to the 2-acetamido-2-deoxyglycosyl-1 residue, a common core element of many plant glycoproteins, was also observed.

Heterogeneity of bovine lactotransferrin glycans. Characterization of alpha-D-Galp-(1-->3)-beta-D-Gal- and alpha-NeuAc-(2-->6)-beta-D-GalpNAc-(1-->4)- beta-D-GlcNAc-substituted N-linked glycans.

Lactotransferrin isolated from a pool of mature bovine milk has been shown to contain N-glycosidically-linked glycans possessing N-acetylneuraminic acid, galactose, mannose, fucose, N-acetylglucosamine, and N-acetylgalactosamine. The glycopeptides obtained by Pronase digestion were fractionated by concanavalin A-Sepharose affinity chromatography into three fractions: slightly retained (A), retained (B), and strongly retained (C). The structure of the glycans of the three fractions has been determined by application of methanolysis, methylation analysis, fast atom bombardment-mass spectrometry, and 1H NMR spectroscopy. Diantennary structures without GalNAc were present as partially sialylated and partially (1-->6)-alpha-L-fucosylated structures in Fractions A and B. Sequences containing alpha-D-Galp-(1-->3)-beta-D-Gal on the alpha-D-Man-(1-->6) antenna, and beta-D-GalpNAc-(1-->4)-beta-D-GlcNAc and alpha-NeuAc-(2-->6)-beta-D-GalpNAc-(1-->4)-beta-D-GlcNAc on the alpha-D-Man-(1-->3) antenna were characterized in the oligosaccharide-alditols obtained by reductive cleavage of Fraction B. A series of Man4-9-GlcNAc structures were identified in Fraction C after endo-N-acetyl-beta-D-glucosaminidase digestion. These results show that the structures of bovine lactotransferrin glycans are more heterogeneous than those of previously characterized transferrin glycans.

1H-and 13C-n.m.r. spectroscopy of 2-oxo-3-deoxy-D-glycero-D- galactononulosonic acid-containing oligosaccharide-alditols bearing Lewis X, Lewis Y and A-Lewis Y determinants isolated from the jelly coat of Pleurodeles waltl eggs.

Three acidic oligosaccharide-alditols carrying Lewis X, Lewis Y and A-Lewis Y determinants were isolated from the jelly coat of Pleurodeles waltl eggs. These compounds possess the following structures. Gal beta 1-4(Fuc alpha 1-3)GlcNAc beta 1-3[2-oxo-3-deoxy-D- glycero-D-galactononulosonic acid (KDN)alpha 2-6] GalNAc-ol; Fuc alpha 1-2Gal beta 1-4(Fuc alpha 1-3)GlcNAc beta 1-3(KDN alpha 2-6) GalNAc-ol and Fuc alpha 1-2(GalNAc alpha 1-3)Gal beta 1-4(Fuc alpha 1-3)GlcNAc beta 1-3(KDN alpha 2-6)GalNAc-ol. The complete 1H-n.m.r.-spectrum assignment for the three compounds and the 13C-n.m.r. analysis of the A-Lewis Y determinant-containing heptasaccharide are reported.

Primary structure of neutral and acidic oligosaccharide-alditols derived from the jelly coat of the Mexican axolotl. Occurrence of oligosaccharides with fucosyl(alpha 1-3)fucosyl(alpha 1-4)-3-deoxy-D-glycero-D-galacto-nonulosonic acid and galactosyl(alpha 1-4)[fucosyl(alpha 1-2)]galactosyl(beta 1-4)-N-acetylglucosamine sequences.

Six major neutral and acidic oligosaccharide-alditols were prepared from the jelly coat of Mexican axolotl eggs. These compounds were demonstrated to contain 3-deoxy-D-glycero-D-galacto-nonulosonic acid (dNloA) and L-fucose (Fuc). The broad diversity of neutral and sialylated oligosaccharides derived from human salivary mucins.

Mucin glycopeptides were prepared from the salivary mucins of 20 healthy donors with blood group O. The carbohydrate chains of the high-molecular-weight mucins were released by alkaline borohydride treatment. Neutral and monosialylated oligosaccharide-alditols were purified by ion-exchange chromatography, gel filtration, and HPLC. The structures of the oligosaccharide-alditols were determined by high-resolution 1H-NMR spectroscopy in combination with fast atom bombardment mass spectrometry and methylation analysis. Thirty-seven oligosaccharide-alditols were characterized and illustrate the extreme diversity of the salivary mucins carbohydrate chains. This diversity might represent a mosaic of bacterial adhesion sites and be involved in the early events of the nonimmune defense of the oral cavity. Among these 37 oligosaccharide-alditols, 31 have not been previously described in human saliva and five of these are novel structures: [formula: see text]

Comparative study of asparagine-linked glycans of plasma T-kininogen in normal rats and during acute inflammation.

Rat T-kininogen has been separated into two molecular variants by affinity chromatography on concanavalin A (ConA): a ConA-reactive (ConA+) and a ConA-non-reactive (ConA-) fraction, from which carbohydrate chains were quantitatively released by hydrazinolysis. On the basis of high-resolution 400 MHz 1H-n.m.r. spectroscopy of the re-N-acetylated hydrazinolysates, the carbohydrate structures of the two ConA molecular variants of rat T-kininogen were established. The ConA-non-reactive species contains a single type of carbohydrate chain with the following structure: [formula: see text] The ConA-reactive fraction contains the same structure and the following additional one: [formula: see text] The relative abundance of the two molecular forms is profoundly affected during inflammation (ratio ConA+/ConA-: 44% in normal and 95% in inflamed T-kininogen), but no structural modification of the carbohydrate chains was observed.

Primary structure of four human milk octa-, nona-, and undeca-saccharides established by 1H- and 13C-nuclear magnetic resonance spectroscopy.

The structures of two octasaccharides, one nonasaccharide, and one undecasaccharide, isolated from human milk, have been investigated by 1H- and 13C-nuclear magnetic resonance spectroscopy. The structures of these oligosaccharides are: beta-D-Galp-(1----4)-[alpha-L-Fucp- (1----3)]-beta-D-GlcpNAc-(1----3)-beta-D-Galp-(1----4)-[alpha-L-Fucp+ ++- (1----3)]-beta-D-GlcpNAc-(1----3)-beta-D-Galp-(1----4)-D-Glc; beta-D-GALp-(1----3)-[alpha-L-Fucp-(1----4)]-beta-D-GlcpNAc-(1---- 3)-beta-D - Galp-(1----4)-[alpha-L-Fucp-(1----3)]-beta-D-GlcpNAc-(1----3)-beta -D-Galp- (1----4)-D-Glc; beta-D-Galp-(1----4)-[alpha-L-Fucp-(1----3)]-beta-D-GlcpNAc-(1---- 6)-(alpha - L-Fucp-(1----2)-beta-D-Gal-(1----3)-[alpha-L-Fucp-(1----4)]- beta-D-GlcpNAc- (1----3))-beta-D-Galp-(1----4)-D-Glc; and alpha-L-Fucp-(1----2)-beta-D-Galp-(1----3)-beta-D-GlcpNAc-(1----3) -beta-D- Galp-(1----4)-[alpha-L-Fucp-(1----3)]-beta-D-GlcpNAc-(1----6)-[alp ha-L- Fucp-(1----2)-beta-D-Galp-(1----3)-beta-D-GlcpNAc-(1----3)]-beta-D -Galp- (1----4)-D-Glc. The two octasaccharides have been previously isolated from human milk as a mixture, and in a pure form from new-born feces, but the n.m.r. data were not provided. These two octasaccharides display the di-Lewis X and the composite Lewis A-Lewis X antigenic determinant, previously described as neo-antigens of adenocarcinoma cell lines.

Characterization of Le(x), Le(y) and A Le(y) antigen determinants in KDN-containing O-linked glycan chains from Pleurodeles waltlii jelly coat eggs.

Novel acidic oligosaccharides were isolated in large amounts by reductive alkaline treatment of the jelly coat of Pleurodeles waltlii (Michah) eggs. The oligosaccharides were found to contain the newly described KDN as acidic monosaccharide and possess either the Le(x), Le(y) and A Le(y) antigenic determinants. Occurrence of Le(x) and Le(y) determinants previously recognized as tumor-associated antigen (TAA) demonstrates that mucins of lower animals may represent a rich and easily available source for preparing TAA. Moreover, it reinforces the hypothesis according to which TAA are evolution markers.

Peptide-N4-(N-acetylglucosaminyl)asparagine amidase (PNGase) activity could explain the occurrence of extracellular xylomannosides in a plant cell suspension.

We have previously isolated mannoside and xylomannoside oligosaccharides with one or two terminal reducing N-acetylglucosamine residues from the extracellular medium of white campion (Silene alba) suspension culture. We have now demonstrated the presence of peptide-N4-(N-acetylglucosaminyl)asparagine amidase (PNGase) activity in cell extracts as well in the culture medium that could explain the production of those compounds. An additional xylomannoside, (GlcNAc)Man3(Xyl)GlcNAc(Fuc)GlcNAc, was characterized, and 1H- and 13C-NMR assignments for the oligosaccharide Man3(Xyl)GlcNAc(Fuc)GlcNAc were obtained using homonuclear and heteronuclear spectroscopy (COSY).

1H and 13C-NMR assignments for sialylated oligosaccharide-alditols related to mucins. Study of thirteen components from hen ovomucin and swallow nest mucin.

Four hundred MHz 1H-NMR and 100 MHz 13C-NMR spectra of thirteen sialylated oligosaccharide-alditols isolated from hen ovomucin and swallow nests (Collocalia mucin) were studied. The resonance assignments were determined by combining multiple-relayed coherence-transfer chemical-shift-correlated spectroscopy (multiple-Relay-Cosy) and 1H/13C chemical-shift-correlated 2-D experiments.

Presence of human antibodies reacting with Candida albicans O-linked oligomannosides revealed by using an enzyme-linked immunosorbent assay and neoglycolipids.

In order to study the presence of antibodies directed against Candida albicans O-linked oligomannosides (oligomannosides O) in patient sera, we have developed an enzyme-linked immunosorbent assay (ELISA) involving neoglycolipids constructed with these residues (NGLO). Oligomannosides O released by mild alkaline degradation of the C. albicans cell wall phosphopeptidomannan (PPM) contained one to seven mannose residues, among which the quantitatively major components, mannobiose and mannotriose, were shown by 1H nuclear magnetic resonance to contain exclusively alpha (1-2) linkages. The pool of oligomannosides was converted to neoglycolipids by coupling them to 4-hexadecylaniline in an equimolar reaction checked by thin-layer chromatography. We have tested against these neoantigens, coated on ELISA plates, 15 pairs of sera corresponding to individual seroconversions observed in 15 patients during the course of a mycological and serological survey of candidiasis. For all patients, seroconversions resulted in an increased level of antibodies against NGLO. A significant correlation was observed between the results of ELISA-NGLO, ELISA involving the original PPM molecule, and routine antibody detection tests, indirect immunofluorescence assay, and cocounterimmunoelectrophoresis. These results therefore demonstrate the synthesis of human antibodies reactive with oligomannosides O constitutive of the C. albicans mannan molecule which have been previously described as exhibiting an inhibitory effect on human lymphocytic proliferation.

1H-NMR spectroscopy of manno-oligosaccharides of the beta-1,2-linked series released from the phosphopeptidomannan of Candida albicans VW-32 (serotype A).

Manno-oligosaccharides (DP 2 to greater than 15) were released by mild acid hydrolysis from the phosphopeptidomannan of a Candida albicans strain of A serotype (VW-32). Manno-oligosaccharides ranging from biose to heptaose were obtained in appreciable amount. Structural investigation of these oligosaccharides showed them to be of the beta-1,2-linked series. The occurrence of such compounds has already been reported in other strains of Candida albicans. We here report the assignment of the structural reporter groups of each of them, and general rules applicable for the 1H-NMR spectrum analysis of linear manno-oligosaccharide of general structure: Man(beta 1-2) [Man(beta 1-2)]nMan

Substrate specificity of rat liver cytosolic alpha-D-mannosidase. Novel degradative pathway for oligomannosidic type glycans.

The substrate specificity of rat liver cytosolic neutral alpha-D-mannosidase was investigated by in vitro incubation with a crude cytosolic fraction of oligomannosyl oligosaccharides Man9GlcNAc, Man7GlcNAc, Man5GlcNAc I and II isomers and Man4GlcNAc having the following structures: Man9GlcNAc, Man(alpha 1-2)Man(alpha 1-3)[Man(alpha 1-2)Man(alpha 1-6)]Man(alpha 1-6) [Man(alpha 1-2)Man(alpha 1-3)]Man(beta 1-4)GlcNAc; Man5GlcNAc I, Man(alpha 1-3)[Man(alpha 1-6)]-Man(alpha 1-6)Man(alpha 1-3)] Man(beta 1-4)GlcNAc; Man5GlcNAc II, Man(alpha 1-2)Man(alpha 1-2)Man(alpha 1-3) [Man(alpha 1-6)]Man(beta 1-4)GlcNAc; Man4GlcNAc, Man(alpha 1-2)Man(alpha 1-2)Man(alpha 1-3)Man(beta 1-4)GlcNAc. The different oligosaccharide isomers resulting from alpha-D-mannosidase hydrolysis were analyzed by 1H-NMR spectroscopy after HPLC separation. The cytosolic alpha-D-mannosidase activity is able to hydrolyse all types of alpha-mannosidic linkages found in the glycans of the oligomannosidic type, i.e. alpha-1,2, alpha-1,3 and alpha-1,6. Nevertheless the enzyme is highly active on branched Man9GlcNAc or Man5GlcNAc I oligosaccharides and rather inactive towards the linear Man4GlcNAc oligosaccharide. Structural analysis of the reaction products of the soluble alpha-D-mannosidase acting on Man5-GlcNAc I and Man9GlcNAc gives Man3GlcNAc, Man(alpha 1-6)[Man(alpha 1-3)]Man(beta 1-4)GlcNAc, and Man5GlcNAc II oligosaccharides, respectively. This Man5GlcNAc II, Man(alpha 1-2)Man(alpha 1-3)[Man(alpha 1-6)]Man(beta 1-4)GlcNAc, represents the 'construction' Man5 oligosaccharide chain of the dolichol pathway formed in the cytosolic compartment during the biosynthesis of N-glycosylprotein glycans. The cytosolic alpha-D-mannosidase is activated by Co2+, insensitive to 1-deoxymannojirimycin but strongly inhibited by swainsonine in the presence of Co2+ ions. The enzyme shows a highly specific action different from that previously described for the lysosomal alpha-D-mannosidases [Michalski, J.C., Haeuw, J.F., Wieruszeski, J.M., Montreuil, J. and Strecker, G. (1990) Eur. J. Biochem. 189, 369-379]. A possible complementarity between cytosolic and lysosomal alpha-D-mannosidase activities in the catabolism of N-glycosylprotein is proposed.
