Antigenic, molecular and functional heterogeneity of Clara cell secretory proteins in the rat.

In an earlier publication we had reported the preparation of a rabbit antiserum specific for rat Clara cell secretory proteins. This rabbit anti-rat Clara cell serum was found to react with two proteins in rat lung lavage by crossed-immunoelectrophoresis. Immunoblotting of rat lung lavage proteins, after sodium dodecylsulphate (SDS) polyacrylamide gel electrophoresis, disclosed three bands of reactivity with anti-Clara cell serum. The relative molecular masses of these three proteins were about 200 (protein A) 55 (protein B) and about 12 kDa (protein C). Anti-Clara cell antibodies eluted from Sepharose-4B-linked protein C (as well as the antiserum raised by immunizing rabbits with protein C) reacted with proteins A and C. Anti-Clara cell antiserum unbound to proteins A and C (as well as antiserum raised by immunizing rabbits with protein B) reacted with protein B only. In non-SDS polyacrylamide gel electrophoresis, protein B migrated as a single band, slightly cathodic to albumin; protein C resolved into three bands, all anodic to albumin. Immunoblots of isoelectric focusing gels showed three bands (pI 5.2-5.7) that reacted with antibody to protein C, and four bands corresponding to protein B were seen in the pI range 4.6-5.0. As determined by immunoperoxidase staining of paraformaldehyde fixed methacrylate embedded 1 micron thick sections of rat lung, protein(s) A (and protein C) and protein B were present in the same cells and in the same granules. Protein B was resistant to trypsin digestion, whereas proteins A and C were readily degraded by trypsin. Rat Clara cell secretory proteins consist of at least two antigenic types that appear to be functionally distinct, and each antigenic type displays charge microheterogeneity.

In vitro translation of rat lung surfactant apoprotein mRNA.

Rat pulmonary surfactant contains apoproteins of molecular weights 38,000, 32,000, 26,000 and 10,000-12,000. The structural and metabolic interrelationships of these proteins are not clear as yet. In order to investigate if they arise from a single or multiple precursor protein (s), we isolated total poly(A)RNA from rat lungs, performed its translation in vitro in the presence of [35s]-methionine and reticulocyte lysate, immunoprecipitated the translation products with anti-rat surfactant antibody, and analyzed them by SDS-polyacrylamide gel electrophoresis and autoradiography. A single translation product of molecular weight 35,000 was detected. Since the antibody used in the immunoprecipitation recognizes the 38,000, 32,000 and 26,000 dalton proteins, it is concluded that at least these three proteins arise from the 35,000 dalton precursor by post-translational modifications.

Malignant fibrous histiocytoma of solitary jejunal diverticulum.

A rare case of malignant fibrous histiocytoma arising from a solitary jejunal diverticulum leading to its perforation is described in a 37-year-old male. The patient presented with peritonitis and on exploration revealed perforation of a jejunal diverticulum containing a soft, friable mass. Biopsy from the mass revealed a malignant fibrous histiocytoma. This appears to be the first description in literature of a malignant fibrous histiocytoma with that particular localization.

Immune status in advanced upper gastrointestinal cancers.

Thirty-eight cases of advanced upper gastrointestinal cancer were assessed for their immune status prior to any form of therapy. Cell-mediated immunity, as tested by absolute lymphocyte count, T-cell count and delayed cutaneous hypersensitivity reactions to purified protein derivative and phytohemagglutinin showed severe depression. Immunoglobulins A and M were elevated, while immunoglobulin G reduced. No correlation could be established between the immune status and the site of cancer or its extent, the performance status of the patients, or their response to chemotherapy.

Nutritional status in advanced upper gastrointestinal cancer.

Fifty-four patients with advanced upper gastrointestinal cancer were assessed for their nutritional status prior to any form of therapy. Anthropometric parameters--weight loss, arm muscle circumference, skin-fold thickness--and biochemical parameters--serum albumin and delayed cutaneous hypersensitivity reactions to purified protein derivative (PPD) and phytohemagglutinin (PHA)--were used for nutritional assessment. Using standard values for comparison, about 80% of all patients were found to be nutritionally depleted. Immunocompetence did not show significant correlation with other nutritional parameters.

Persistence of aldrin in flooded soil under the cover of rice.

The persistence of aldrin in soil and its translocation into rice plants and grains were tested under two different field conditions. Treatment T1, which consisted of aldrin application at the rate of 1.5 kg active ingredient/ha at nursery sowing, did not leave aldrin residues in rice plants 40 days after application (10 days after transplanting). Dieldrin was present in plants to the extent of 0.04 ppm at this stage but completely disappeared (below detectable limit) within 70 days (30 days after transplanting). Treatment T2 involved aldrin application 10 days after transplanting in flooded soil at the same rate and left 0.518 ppm residues in soil just after application. These residues dissipated to their half in 90 days. At this stage its conversion to dieldrin was also observed. Remaining residues dwindled to below the detectable limit in 270 days. Plant samples analyzed from this treatment at 7 and 30 days contained 0.079 and 0.169 ppm aldrin and 0.025 and 0.08 ppm dieldrin, respectively. Rice grains also had aldrin and dieldrin residues (each at 0.001 ppm level) at harvest time but these were below the Maximum Tolerance Limits. Both the treatments were found safe from the toxic residue point of view.

Enzymes of isolated brush border membrane of Cotugnia digonopora, and their insensitivity to anthelmintics in vitro.

Purified brush border membrane of Cotugnia digonopora showed the presence of a number of phosphohydrolases. Among these, alkaline phosphatase was extremely active. Other enzymes such as glucose-6-phosphatase, fructose-1,6-diphosphatase, cAMP-phosphodiesterase, 5'-nucleotidase and adenosine-triphosphatase were also active. Observations were made on the activities of various ATPases; whereas the enzyme was activated by Ca++ and Mg++ in an additive manner, its sensitivity to ouabain was negligible. Furthermore, in the presence of EDTA the enzyme activity was quite significant. The treatment of isolated brush border membrane with mebendazole, niclosamide and praziquantel in vitro did not alter the activity of these enzymes. However, treatment of intact worms drastically affected the integrity of the membrane.

Clinical pattern of vitiligo in Libya.

Patients with vitiligo attending the polyclinics of Benghazi during a 9-month period were reviewed. The proportionate case ratio was 0.33% of all skin cases. The mean age at onset of the disease was 19.38 years; mean age at reporting the disease was 23.06 years. There was more frequent involvement of exposed parts in men compared with women. Vitiligo areata responded better to treatment. The history did not confirm the genetic predisposition of the disease. Association with known autoimmune disease is reported.

Secretory proteins of the lung in rodents: immunocytochemistry.

The reactivity of rabbit antisera to rat lung secretory proteins with other rodent species was evaluated by immunocytochemistry. Rabbit anti-rat surfactant apoprotein antiserum reacts with the cytoplasm of rat, mouse, and hamster type II pneumocytes and is specific for these cells. Rabbit antiserum to rat Clara cell secretory proteins stains rat, mouse, and hamster Clara cells. Rabbit antisera specific to the two antigenic types of rat Clara cell antigens were also both reactive with rat, mouse, and hamster Clara cells. An antiserum to the non-serum proteins of hamster lung lavage was also prepared and shown to be specifically reactive with hamster Clara cells. The availability of specific reagents for secretory proteins of rodent lungs is expected to facilitate studies of the respective cell types in various pathologic states.

Mixed lymphocyte reaction supernatants exert varied effects upon gingival fibroblasts in vitro.

Supernatants from mixed lymphocyte cultures (MLRs) were tested for modulation of gingival fibroblast growth by both dye and 3H-thymidine uptake assays. Significant levels of inhibition of growth were observed with both assays with 24 and 72 hr MLR supernatants, but stimulation of growth was seen to occur, consistently, with 48 and 96 hr MLR supernatants as assessed by the dye uptake method. Fractionation of 24 hr supernatants by gel-filtration, on Ultrogel ACA 54 yielded fractions with enhanced growth inhibitory activity. The MLR supernatants would appear to contain mediators with opposing growth effects at different times during the culture.
