Enzymatic characterization of hepatitis C virus NS3/4A complexes expressed in mammalian cells by using the herpes simplex virus amplicon system.

The hepatitis C virus (HCV) NS3 protein possesses three enzymatic activities: an N-terminal serine protease activity, a C-terminal RNA-stimulated NTPase activity, and an RNA helicase activity. To characterize them, the full-length NS3(631)/4A and three C-terminal truncated proteases (NS3(201)/4A, NS3(181)/4A, and NS3(155)/4A were expressed in mammalian cells with HSV amplicon-defective viruses. Our results revealed that all of the NS3/4A proteins produced in mammalian cells (except NS3(155)/4A) are active in processing both cis and trans cleavage sites. Temperature optimization studies revealed that the protease is more active at temperatures ranging from 4 to 25 degrees C and is completely inactive at 42 degrees C. The RNA-stimulated ATPase activity was characterized with a partially purified NS3(631)/4A fraction and has a higher optimal temperature at 37 to 42 degrees C. The effects of detergents on both NS3 protease and RNA-stimulated ATPase were similar. Nonionic detergents such as Triton X-100, Nonidet P-40 and Tween 20 did not affect the activities, while anionic detergents such as sodium dodecyl sulfate and deoxycholic acid were inhibitory. Zwitterionic detergent such as 3-[(3-cholamidopropyl)- dimethyl-ammoniol-1-propanesulfonate (CHAPS) inhibited protease activity at a concentration of 0.5% (8 mM), which had no effect on ATPase activity. Finally, RNA-unwinding activity was demonstrated in the NS3(631)/4A fraction but not in the similarly purified NS3(181)/4A and NS3(201)/4A fractions. NS(363)/4A unwinds RNA duplexes with 3' but not 5' single-stranded overhangs, suggesting that the NS3 RNA helicase functions in a 3'-to-5' direction.

[Intrapericardial administration of drugs in the animal experiment].

Intrapericardially supplied agents for instance cardioactive drugs were transferred via lymph vessels in the myocardium likely. In case of dye application the myocardium and also the cardiac valves were coloured additionally. The effect on the heart should be more intensive and faster than after systemic application.

[Experimental studies on the problem of improving the myocardial blood circulation in an intraoperatively resuscitated heart].

A pumping system for supporting an intraoperatively failured heart is presented. This system consists of a bell-shaped receptacle for the heart and a pump with a working volume of 49 ml and a working pressure of 220 Torr. The pumping power is transmitted to the heart by means of an external pneumatic pressure line. The efficacy of the pump in the high pressure system amounted to 20-30 per cent. An effective coronary perfusion in case of a cardiac failure is guaranteed by a venous underpressure pump of this pumping system. A favourable influence was also observed in case of the low pressure system.

[Experimental studies on the effect of the administration of oxygen into the pericardial cavity in acute myocardial ischemia].

Artificial myocardial ischaemia caused by ligation of the left anterior descending coronary artery was treated in minipigs by the application of oxygen into the pericardial cavity. The results show that in this way acute myocardial ischaemia may be reduced. The application of oxygen into the pericardial cavity brought about, within 15 min, an improvement in the haemodynamic parameters. Several electrolytes, which were estimated from the blood of the sinus venosus, returned to normal.

[The reaction of some enzymatic and coagulative physiologic parameters after electrically induced thrombosis in the arterial vascular system in dog and pygmy pig].

Changes of enzyme and coagulation parameters were observed after electrically induced thrombosis in the arterial vascular system. These changes where only significant if the thrombosis was induced in the coronary arteries and a myocardial infarction was following. On the contrary anodic direct current (D. C. 10 mA about 15 minutes) in the femoral artery of dog or pygmy pig did not produce any significant changes of the examined parameters.

[Prevention of postoperative phlebothrombosis in the femoral vein by means of external pneumatic massage of the extremity].

The postoperative thrombo-embolism belongs to the most dangerous complications in the postoperative phase. The frequency of the postoperative phlebothrombosis increased in the last years because more risk patients were operated. External pneumatic massage of the extremity produces acceleration of the blood flow in the veins and a increase of the fibrinolytic activity of the venous vascular wall. A significant retrogression of the thrombus size was proved in animal experiments in dogs by application of external pneumatic massage after artefactitiously induced thrombosis in the femoral vein.

[Prevention of postoperative phlebothrombosis by means of pneumatic massage of the extremities in animals].

Thromboembolism may be considered as the most serious complication in the postoperative period. Due to improvements in anaesthesia and postoperative supervision even those patients who are in a bad state of health may be operated upon. The frequency of postoperative phlebothrombosis, however, has increased in recent years. External pneumatic massage of the extremities will speed up the blood flow in the veins and increase the fibrinolytic activity in the vein wall. In animal experiments we could observe a significant decrease of the thrombus size in artificially produced thrombosis in the vena femoralis of dogs, if after thrombus induction the external pneumatic massage was applied.

[Experimental model of coronary thrombosis in the closed thorax in swine].

Coronary thrombosis was induced in 14 miniature pigs by means of direct current. The induction electrode was introduced via the common carotid artery into the coronary artery. The intensity of current (1 mA) and the duration of current flow (60 min) were kept constant in all the experimental animals. 6 miniature pigs survived the period of observation (48 h). In all instances, an injury to the intima was the morphopathological location factor. With the exception of one animal, which died after an induction time of 60 min, coronary thrombosis was stated in all the miniature pigs. The experimental arrangement which produces closed-thorax coronary thrombosis is suited for cardiosurgical, physiopathological and pharmacological experiments, especially because the anatomy and the degree of development of the porcine heart are most similar to those of the human heart.

[Experiences in the determination of thyroxine iodine in serum in various functional conditions of the thyroid gland].

A report is given on the method of the thyroxine iodine determination in the serum. Of 213 patients the result of the thyroxine iodine determination was compared with the clinically and radiologically established functional condition of the thyroid gland. The findings are discussed. Similar results were also reported of other in-vitro-tests (T3-test, T4-test, BEJ and PBJ). According to our experiences we consider the thyroxine iodine determination in the serum siutable as screening test which - when there is a suspicion of a hypo- or hyper- thyroidism - needs the supplementation by the radio-iodine functional and localisation tests.
