[Synthesis and serum cholesterol-lowering and uterotropic effects of 15,16,17-trisubstituted oestratriene 3-methyl ethers. Part 63: On steroids (author's transl)].

The preparation of the 17 beta-carbamoyl derivatives 4 and of the 17 beta-carbonic acid esters 5 from 15 beta,16 beta-epoxy-3-methoxyoestra-1,3,5(10)-triene-17 beta-ol (3a), and the subsequent opening of the 15 beta,16 beta-epoxy group resulting in 15 beta,16 alpha,17 beta-trisubstituted oestratrienes 6 are described. The influences of several substitutents in the positions 15 and 16 and of the 17 beta-hydroxy group and derivatives thereof on the uterotropic and cholesterol-lowering activities were investigated in rats after oral application. A correlation between the cholesterol-lowering and the uterotropic activity could be established. Activity dissociation was not achieved.

[Immunsuppression by vitamin B6-antagonists. Prolonged survival of inbred rats with renal transplants (author's transl)].

The combined effect of Vitamin B6 free diet and 4-Desoxipyridoxin (a potent Vitamin B6-Antagonist) on the rejection reaction of inbred rats after renal transplantation was investigated. More than 90% of the control animals without Vitamin B6-deficiency died between 7 and 13 days with uremia due to an acute transplant rejection, where as the Vitamin deficient and successfully transplanted animals (n=38) showed a significantly prolonged survival time. In 17/38 rats survival was longer than 35 days. Vitamin B6-deficiency, induced by Desoxipyridoxin and Vitamin B6-deficient diet offers the possibility of an effective and reversible immunosuppression in renal transplantation.

Transplantation-induced immune complex kidney disease in rats with unilateral manifestation in the allografted kidney.

Kidney transplantation between major histocompatibility system-identical rat strains LEW.1N (donor) and BN (recipient) is regularly followed by serious immune complex (membranous) glomerulonephritis. The disease localizes in the transplant only and spares the recipient's own contralateral kidney. The recipients develop both circulating immune complexes, as well as circulating antibodies against an allogeneic, tubular epithelial antigen of the donor. Antibodies eluted from the diseased kidneys display the same specificity. The transplant disease, therefore, is not autoimmune glomerulonephritis but an alloimmune, organ-specific illness unrelated to the usual histocompatibility system.

[Hyaluronic acid in pleural fluids: an additional parameter for clinical diagnosis on diffuse mesotheliomas (author's transl)].

187 pleural fluids of various etiology are examined with the intention to find hyaluronic acid. The colorimetric method of Morgan-Elson was used for this purpose. The mucopolysaccharid was identified in 34 cases: 18 in 30 cases of diffuse mesotheliomas 1 in 74 cases of secondary tumours of the pleura 8 in 20 cases of tuberculous pleurisy 5 in 37 cases of nonspecific pleurisy 2 in 6 cases of purulent pleursy. A level from more than 212 mg/litre was detected in 10 cases of mesotheliomas only. Such a concentration is consequently the proof for diffuse mesothelioma. The diagnostical improvement in other cases of mesotheliomas resulted from a combination of the following parameters: coagulation of protein by the method of Weltmann, cytological investigation of cellsedimentation and measurement of hyaluronic acid. Our experience with the diagnosis of diffuse mesotheliomas is discussed also in account to the results which are published since 1965 from a scientific team working in Lille, France, reporting the value of detection and measurement on hyaluronic acid in pleural fluids.

[Immunsuppression under vitamin B6 deficiency (author's transl)].

The influence of a Vitamin B6 free diet and the effect of Desoxipyridoxin (a potent Vitamin B6 Antagonist) on the immunesystem of mice was investigated in several experimental setups. Rejection times of skintransplants from inbred strains (129 J) on C57bl/6J mice served as a model for immunosuppression. Rejection times under alimentary Vitamin B6 deficiency did not exceed those under hypocaloric nutrition associated with comparable weight loss. Immunosuppression with the Vitamin B6 antagonist Desoxipyridoxin was much more pronounced. The effect was related to the duration of treatment. Desoxipyridoxin offers the possibility of effective immunsuppression, which is readily reversible.

[Topographic phase analysis of urinary concrements using crystal-optic methods].

In the present paper was reported on the results of the topographic crystal-optic analysis of urinary calculi of 560 concrements. 59% of all urinary calculi had a different phase content in the nucleus and in the calyx. All frequent minerals of the urinary calculi could be proved in the nucleus of the calculus. It could be shown that nucleus and calyx may have a monomineral as well as a polymineral structure. It is referred to the importance of the topographic analysis of the urinary calculi for the metaphylaxis of the urolithiasis.

Single nephron study of intrarenal urea handling in experimental pyelonephritis.

Micropuncture experiments were carried out in 20 rats with bilat. experimental chronic pyelonephritis. Inulin and urea concentrations were estimated in the late proximal, early distal and latte distal nephron segment. Proximal urea reabsorption did not differ from that in control animals. Fractional amount of filtered urea in the early distal segment was significantly lower in the pyelonephritic rats, urea reabsorption in the distal tubule approcimated zero. The data indicate that intrarenal urea recirculation is abolished by the experimental lesion, thus determining maximal urine urea concentration. It is suggested that the concentrating defect of experimental pyelonephritis is not only a result of an adaptational increase in individual nephron GFR; an additional intrinsic lesion seems to be present, located in the renal medulla.
