[Submicroscopic detection of oriented protein filaments in the plasmoditrophoblast of the human placenta (author's transl)].

In this paper is described by histochemical, polarization optical and electron microscopical techniques resp. an up to now unknown protein structure in the basal plasmoditrophoblast of the human placenta. This structure consists of noncollagenic filaments containing cystin or cystein. They are arranged in the plain in the form of a network and parallel to the surface of the chorionic villi. They occur in connection with Langhans cells, they regularly existed in young placentas, where they completely surround the chorionic villi, and they occur in the villi of termborn placentas only incomplete in parts of the plasmoditrophoblast. Differences of the molecular structure of the network of protein filaments, which are depended on the age, are not demonstrable.

[On the birefringence of formazans (author's transl)].

The enzyme succinate dehydrogenase was demonstrated histochemically in organs of rats, especially in experimentally infarcted hearts. 88 tetrazolium salts were used as indicators, the resulting formazans were examined for birefringence. Formazans with cristalline shapes show birefringence, but formazans of amorphous character do not. In the atypic species of nitrogroup-substituted monoformazans it is possible to form an oriented association to cell structures in spite of an amorphous precipitation. Therefore, anomalous colours of polarization, and dichroism is observed. Relations are discussed between shape of formazan deposits, polarity of formazans, and the occurrence of optical anisotropism.

[The use of N,N'-diethylpseudoisocyaninchloride for the polarization optical detection of polyanions in the histological section (author's transl)].

In the paper a technique of the polarization optical qualitative and quantitative detection of polyanions with N,N'-diethylpseudoisocyaninchloride in the histological section is described. It is possible to prevent depolymerisation after precipitation with potassium ferricyanide and to intensify the birefringence of the histological structures. In vitro attempts and measurements of the dispersion of the double refraction and of the dichroism indicate connections between the optical appearances, furthermore it is possible to make statements about the mechanism of the arrangement of the tissue-dye-ferricyanide-complex.

[Quantitative changes of the cytotrophoblast in underweight and normalweight newborn after risk pregnancy and antepartal therapy (author's transl)].

A histochemical-morphometric method was used to determine at the cytotrophoblast of 73 placentas, changes in the composition of the syncytium and in the Langhans' cell count in underweight and normalweight newborn with or without antepartal maternal therapy in comparison to newborn after uneventful pregnancy. The reduction of the percentage share of mature syncytium when the share of predegenerative and degenerative syncytium is increased, as weil as the increase in the average Langhans' cell count in the placentas of premature infants, hypotrophic and eutrophic infants delivered at term after risk pregnancy, compared with normal controls, is statistically significant. Significant differences in the percentage share of mature syncytium between the placentas of premature infants and hypotrophic infants delivered at term, as well as between placentas of eutrophic infants delivered at term, depending of the nature and severity of the risk, are discussed in connection with the significance of the syncytium for the processes of material transport. No correlation with the Apgar score and the occurrence of the respiratory distress syndrome of newborn, nor with the duration of the first and second stage of lavor with macroscopic placenta peculiarities, have been found. The good correlation between placenta findings and classification characteristics of newborn, and clinical aspects of the course of pregnancy, allows the possibility of diagnostic evaluation. Our results indicat also that the morphological performance must be included in the considerations explaining the success of treatment of placental insufficiency.

[Relationships between the HPL determination in maternal serum and the condition of the associated placental villous trophoblast].

Relationships between the determination of human placental lactogen in materna serum and the condition of associated placental villous trophoblasts.--Taking as a starting point both qualitative and quantitative differences in the composition of placental villous trophoblasts subsequent to risk pregnancies, the proportions of different qualities of syncytium and average numbers of Langhans' cells per field of view in 24 placentas of neonates of the 37th to 41st weeks of pregnancy are compared with the corresponding maternal serum HPL (human placental lactogen) values. The significantly lower proportion of mature syncytium determined in the case of a low HPL value as well as the result of linear regression analysis are indicative of a dependence of maternal serum values for HPL upon the proportion of mature syncytium. This particular result is also interpreted by the authors as confirming the view that "mature" syncytium can be considered to be "functional" syncytium. Like other investigators, the present authors should also point out the importance of the total mass of placenta which can compensate for lower proportions of functional syncytium in individual resorption villi. Whereas there are clearly evident relationships between the maternal estriol values and the condition of placental villous trophoblasts, such relationships could not be observed in our material for oxytocinase and HSAP.

[Submicroscopic structure of collagen--a basis for the interpretation of corresponding polarization-optical and histochemical findings].

In view of new possibilities of acquiring relatively quantitative information about the molecular structure of collagenous fibres in histological section with the help of polarization and histochemical methods a survey is given of the most important aspects of the molecular structure of collagen in connection with the formation of "cross-links", giving due attention to the role of functional groups. These create the basis for interpreting morphological findings covering free amino-groups of lysin and hydroxylysin, guanidine groups of arginine and carboxyl groups of glutamic acid and asparaginic acid, gained by adding phenol or toluidine blue to collagenous fibres and by performing path difference measurements. References made to the applicability of this polarization-histochemical method in clinical research and diagnostics, in gathering reproducible data on processes of development or growth and aging and pathological changes in collagen are intended to encourage wide use of this examination technique in practice.

Interference-microscopical thickness measurements of isolated nephrogenic basement membranes.

The results of interference-microscopical thickness measurements of isolated nephrogenic basement membranes are reported and discussed. Tubular and glomerular basement membranes were examined by the shearing technique with total image splitting, the kidneys used having obtained from pigs as well as from human beings. It was not possible to determine a statistically significant difference in thickness between tubular and glomerular basement membranes obtained from pigs. Human glomerular membranes were found to be much thicker in long-time diabetics than in normal subjects. Also, glomerular membranes of young persons are thinner than the membranes of adults.

[Metachromasia, double refraction and dichroism caused by Toluidine Blue reaction (author's transl)].

The present paper is concerned with those optical phenomena which may be caused in histological structures by the toluidine blue reaction. They relate to absorption as well as refraction properties. The paper gives a detailed description of how the toluidine blue reaction is carried out on histological preparations. The analysis of absorption results leads, in connection with physicochemical considerations, to a new explanation of the cause of toluidine blue metachromasia. In this, the assumption is made that the conditions of attachment of protons to the hydrogen-bearing nitrogen of dye molecules are largely responsible for the occurrence of metachromasia, the way in which the attachment of the proton to this quaternary nitrogen is influenced being of no consequence in this particular connection. It may result from both a combination of homogeneous toluidine blue molecules into associates and a shift to the alkaline region of the pH value of the dye solution. However, the term metachromasia is used to describe only that shift of the absorption spectrum which is produced by associates. The thus produced metachromasia ("concentration metachromasia") involves the development of a so-call "gamma-band" (with an absorption peak of gamma = 546 nm), there being a causal rather than a fundamental difference between the latter band and the normal metachromatic mu-band (with an absorption peak of gamma = 540 nm). Whereas in the case of concentration metachromasia and for an aqueous solution there may be observed an association of dye molecules having a maximum intermolecular distances of less than 5 angström units, which is due to the large number of toluidine blue molecules per unit volume, mu-metachromasia is usually observed in connection with chromotropic structures. This association results in the lone-pair electrons of the hydrogen-bearing nitrogen atom being included in the intermolecular resonance system, which in turn results in the attachment of a proton to this nitrogen being weakened, which leads to the assumption of a red color. The difference to gamma-metachromasia lies in the fact that even by treatment with dilute solutions, in which the dye is present in a monomeric form, such a metachromasia may be produced on account of the sufficiently dense charge pattern of the chromotropic structure. This particular pattern tends to induce an association of dye molecules which in turn are associated, through hydrogen bridges, with the anionic groups. The orthochromatic toluidine blue reaction was found to be a possible method of detecting bridge-bond-active hydrogen in histological structures. The cause of the assumption of a blue color, which is observed in this connection, is a monomeric attachment of the dye. Additional factors leading to ortho-chromatism are also discussed...
