Chemistry of the sulfate groups in a sulfated glycoprotein from rabbit submandibular gland.

A 35S-labeled sulfated glycoprotein was isolated from rabbit submandibular glands. Acid stability studies on the 35sulfate groupings present in the intact glycoprotein gave a half-life of 45 min. Partial acid hydrolysis of the 35S-labeled glycoprotein in 0.1 M HC1 for 90 min at 100 degrees C liberated a radioactive fraction which was free from peptide and fractionated in the monosaccharide range of a Sephadex G-15 column. Examination of this fraction by paper chromatography revealed the presence of a major component having the characteristics of N-acetylglucosamine 6-0-sulfate and a minor component having the properties of N-acetylgalactosamine 6-0-sulfate. The presence of ester sulfate groups in the intact glycoprotein was confirmed by infrared spectroscopy.

A comparison of the plaque-inhibiting effect of stannous fluoride and chlorhexidine.

A double blind clinical trial was undertaken in order to compare the plaque inhibiting effect of SnF2 (0.2% and 0.3%) with chlorhexidine (0.1%) applied as mouthrinses in a test panel of 12 dental hygienist students. The experimental series were performed in periods of four days. The mean Plaque Index values were 0.35 and 0.2 when employing SnF2 and 0.1 when chlorhexidine was used, whereas distilled water resulted in a mean value close to 1.0. The long-term effect of a 0.3% SnF2 solution was tested in another group of five students for a period of three weeks. The two trials confirmed previous reports claiming that SnF2 is effective as a plaque inhibitor and, furthermore, that this effect could be maintained for a period of at least three weeks. The staining problem was less than in similar studies using chlorhexidine.

Chemical analysis of a sulphated macromolecule isolated from rabbit submandibular gland.

Chemical analysis of a sulphated macromolecule isolated from the rabbit submandibular gland showed that about 70 per cent of the dry weight could be accounted for in terms of amino acids of which the acidic amino acids, proline and alanine were the most abundant. Total carbohydrate was 11%. Galactose was the main neutral sugar but galactosamine, glucosamine, neuraminic acid, glucose and mannose could also be demonstrated. Sulphate accounted for 1,4%. No uronic acid was found. The data indicate that the sulphated macromolecule from the rabbit gland is in fact a sulphated glycoprotein.
