The extent of the Aznalcóllar pyritic sludge spill and its effects on soils.

This paper presents some initial results from the Instituto Tecnológico Geominero de España's (ITGE) study of the Aznalcóllar mine spill. The spatial distribution of the pyritic sludge released was surveyed by using remote sensing data, aerial photography, and more than 700 field measurements on the sludge thickness. Initial estimation of the extent of the sludge was provided by radar data. Maps at 1:10,000 scale, drawn on the basis of field data and interpretation of aerial photos, show the distribution of the sludge, divided into 168 subsections on the basis of average thickness. GIS analysis provided estimates of the area and volume of the sludge. Three approaches were followed in order to survey the effects of the spill on the Guadiamar river alluvial soils: (1) Mineralogical and chemical characterization of the sludge and its evolution until its removal. Alteration products of the pyritic sludge were also analyzed. (2) Determination of geochemical background of soils in the Guadiamar river basin, in order to establish the content of heavy metals and other elements in the soil before the spill. (3) Assessment of the sludge effect on soils caused by the acid water and the deposited sludge, by comparison of the heavy metal content of soil under the sludge layer with that of background soil. Finally, an airborne multispectral survey was carried out over the Aznalcóllar-Doñana area to evaluate its efficiency for monitoring soil condition during and after sludge removal.

Efflux pump-mediated quinolone resistance in Staphylococcus aureus strains wild type for gyrA, gyrB, grlA, and norA.

Fluoroquinolone efflux was studied in 47 Staphylococcus aureus clinical strains with MICs of ciprofloxacin (CFX) of < or = 2 micrograms/ml. Forty-three strains were wild type for gyrA, gyrB, and grlA quinolone resistance-determining regions and for norA and its promoter region. Forty of these strains (MICs of CFX, 0.1 to 0.2 microgram/ml) did not show efflux of fluoroquinolones. Three strains (MICs of CFX, 1 to 2 micrograms/ml) showed efflux. These results suggest that efflux can appear in S. aureus clinical strains in the absence of mutations in norA and its promoter.

Reproductive function in male rats with chronic nephrosis.

Endocrine dysfunction has been associated with renal diseases. The present study was conducted to explore reproductive function in male rats with chronic nephrosis. Experimental chronic nephrosis was induced by the administration of 7.5, 5.0 and 5.0 mg per 100 g body weight of puromycin aminonucleoside on days 0, 21 and 35, respectively. Reproductive function was evaluated on the basis of hormonal concentrations, mass of accessory sex organs and fertility during an 84 day period. Circulating LH, FSH, testosterone and oestradiol concentrations were measured by specific radioimmunoassays, while fertility was estimated by the rate of pregnancy induction. Samples were collected on days 7, 14, 28, 56 and 84. The results showed an important endocrine dysfunction characterized by low concentrations of LH and FSH during the first month, after which concentrations were similar to control values or even increased on days 56 and 84. Testosterone and oestradiol decreased significantly at all time points evaluated. The mass of the testes did not alter. However, the mass of the prostate and seminal vesicle decreased only during the first 2 weeks, and became essentially normal thereafter. The reproductive capacity of nephrotic males was eliminated on day 7, whereas on day 14, 16% of the group was able to mate successfully and subsequently most animals recovered their normal reproductive function. This study demonstrates for the first time that rats with experimental chronic nephrosis develop an important endocrine dysfunction, characterized mainly by persistent reduction in testosterone concentrations, which impairs reproductive capacity only transiently.

Failure of zafirlukast to prevent ibuprofen-induced anaphylaxis.

BACKGROUND: Anaphylaxis to nonsteroidal anti-inflammatory drugs is thought to depend on cycloxygenase inhibition coupled to upregulation of 5-lipoxygenase-dependent pathways. The introduction of leukotriene-receptor antagonists afforded the opportunity to test this hypothesis. These agents provide at least partial protection against aspirin-induced anaphylaxis during controlled challenges but we did not know whether the level of protection was high enough to block symptoms from ingestion of a full dose of aspirin. METHODS: We report a patient with moderately severe asthma who experienced an episode of anaphylaxis following ingestion of 400 mg of ibuprofen while under therapy with 20 mg of zafirlukast given twice a day. RESULTS: No further episodes of anaphylaxis have been noted following institution of complete avoidance to all nonsteroidal anti-inflammatory drugs. CONCLUSIONS: Patients who are sensitive to cycloxygenase inhibitors should practice complete avoidance of these drugs even while under therapy with leukotriene modifiers.

The effects of RGD (Arg-Gly-Asp) peptides on glaucoma filtration surgery in rabbits.

BACKGROUND AND OBJECTIVE: Integrins are the main mediators of the interaction between fibroblasts and extracellular matrix (ECM) during scar formation. The adhesion motive RGD (Arg-Gly-Asp) is contained in the ECM and is recognized by the integrin receptor. Soluble peptides containing the RGD sequence can compete with -RGD- contained in the ECM for binding to the integrin receptor and thus prevent cell adhesion and scar formation. MATERIALS AND METHODS: The peptides RGD (p602), GRGDSP (p603), GRGDSPCA (p604), and GGRGDSPCA (p605) were used in 25 glaucoma filtering surgeries in rabbits (five eyes per peptide and five with saline). RESULTS: Postoperative subconjunctival injections of peptides were given at days 0, 4, 8, 12, and 16. Bleb size, bleb survival, and signs of toxicity were examined. The GRGDSPCA and GGRGDSPCA showed an increase in bleb formation, size, and survival with no clinical signs of toxicity compared with controls (P < .008). Histopathologic evaluation confirmed inhibitory effects in scar formation with bleb formation, and transmission electron microscopy demonstrated that there was no toxicity to the ciliary body. CONCLUSION: These peptides were effective in controlling scar formation in glaucoma filtering surgery.

Assessment of multiple coagulase-negative staphylococci isolated in blood cultures using pulsed-field gel electrophoresis.

One of the criteria used to determine the clinical importance of coagulase-negative staphylococci (CNS) is the isolation of the bacteria from sequential blood cultures. Sequential isolates of CNS obtained from five immunocompromised patients over three months were genetically characterized by pulsed-field gel electrophoresis (PFGE). This typing method was compared to two first-line typing methods: determination of the species and of antibiotic susceptibility. In four patients the initial clinical evaluation changed because of the PFGE results several episodes of bacteremia would have been wrongly assessed if only the biotype and the antibiotype had been determined. Pulsed-field gel electrophoresis should therefore be used for CNS strains from immunocompromised patients or those suffering from chronic diseases with non-concordant biotype and anti-biotype.

Heart disease education and prevention program targeting immigrant Latinos: using focus group responses to develop effective interventions.

Although research has provided considerable knowledge concerning the positive effects of behavioral change on morbidity and mortality from heart disease and related risk factors, some segments of the population have not benefited equitably from this information. In April 1995, the National Heart, Lung, and Blood Institute (NHLBI) conducted seven focus groups to determine knowledge and attitudes about heart disease and associated risk factors, identify media usage and preferences, and assess publications usage and preferences among Spanish-speaking Latino immigrants residing in the Washington, D.C., metropolitan area. This information was gathered to assist in the development of key messages and strategies for the NHLBI Latino Community Cardiovascular Disease Prevention and Outreach Initiative, Salud para su Corazón--a heart disease prevention and education campaign. Findings from these focus groups indicate that Latinos may not benefit from heart disease prevention messages developed for the general population because of language and cultural differences. The researchers concluded that health education and disease prevention programs targeting the Latino community should develop educational materials and interventions that address language preferences and cultural values. Furthermore, to be effective, these programs should show people how to make positive behavioral changes based on their current circumstances, while remaining sensitive to the fact that Latino immigrants face major life adjustments and many are still greatly influenced by their country of origin.

Characterisation of the novel junctions of two minichromosomes of Leishmania major.

The minichromosomes M210 and M230 of Leishmania major are linear amplicons which have an inverted repeat structure. We report here the molecular cloning of the megabasic chromosome region where M210 and M230 recombination points are located. The analysis of this chromosomal region revealed a 374 bp fragment directly repeated 3 times, which was also repeated elsewhere in the genome. DNA fragments containing the novel junctions of the minichromosomes M210 and M230 have been likewise cloned. The nucleotide sequence of these fragments was determined and compared with their corresponding DNAs in the source chromosome. It revealed that M210 contains a central 226 bp non-repeated fragment, whereas the central fragment of M230 was 5 kb long. A 9 bp sequence which was repeated in inverted orientation around the recombination points of M230 was found. M210 showed instead an A + T rich DNA which could be implicated in its generation.

Location in the source chromosome of the 180-kb minichromosome of Leishmania major and characterization of the novel junction.

The 180-kb LD1 minichromosome of Leishmania major (M180) is a large inverted duplication which arises spontaneously from a megabasic chromosome. In this work this locus has been located in the source chromosome at a telomeric position. Both the novel junction created in the middle of the minichromosome, as well as its counterpart region in the source chromosome were cloned and sequenced. Two inverted repeats, which could give rise to two imperfect stem-loops, and an A + T-rich DNA sequence were the only particular features observed in this region. A putative model for the formation of the minichromosome is proposed.

Complement-mediated lysis and infectivity for mouse macrophages and sandflies of virulent and attenuated Leishmania major promastigotes varying in expression of the major surface protease and lipophosphoglycan.

The infectivity to mouse macrophages and sandflies, the expression and enzymatic activity of the major surface glycoprotein (gp63), the developmental modification of lipophosphoglycan (LPG) and three metacyclogenesis markers (promastigote body size, lectin agglutination and complement resistance) were compared in four related Leishmania major promastigote lines. The lines, which differed in their virulence for BALB/c mice, were examined in both logarithmic and stationary phase. Although the two non-virulent lines were unable to survive and multiply within the macrophages, they were better at attaching to the macrophages and infecting sandflies than the two virulent lines, which were highly infective for macrophages. Except for the higher resistance of the attenuated parasites to complement-mediated lysis, there were no clear differences between the metacyclogenesis markers of the four lines. The amount and enzymatic activity of surface gp63 was relatively high in the attenuated promastigotes and this appears to be related to a higher expression of gp63 genes. In terms of LPG, cells of all the lines had approximately twice the number of galactose and mannose residues per molecule when in logarithmic phase than when in stationary phase. LPG of the virulent lines also contained approximately twice the mannose and galactose residues of the attenuated line. Although L. major gp63 could therefore be important for promastigote survival in the sandfly and for the resistance to complement-mediated lysis, there was no apparent correlation between gp63 expression and promastigote survival in the macrophage. A very elongated LPG could be necessary for the survival and proliferation of the parasite in macrophages.
