Physiopathological aspects of body overweight in the female climacteric.

The correlation between overweight and the climacteric was studied in 550 menopause clinic patients by investigating certain clinical and sociocultural parameters (age, marital status, educational level, occupation and type of work, calorie intake, smoking habits, parity, blood pressure, previous hormonal therapy and climacteric symptoms), evaluating plasma levels of various hormones (17 beta-oestradiol, follicle-stimulating hormone (FSH), luteinizing (LH), testosterone, hydrocortisone, adrenocorticotrophic hormone (ACTH), triiodothyronine (T3), thyroxine (T4), growth hormone (GH) and insulin), glucose and various lipid fractions (total lipids, total cholesterol, nonesterified fatty acids (NEFA), triglycerides and phospholipids) and exploring the blood-clotting pattern ( Owren 's test, euglobulin lysis time, antithrombin III and prothrombin agglutination time (PAT). The subjects were classified as normal weight or overweight by reference to Broca's Index, as modified by Brusch , and the degree of overweight was determined by means of the Body Mass Index (BMI). Of the subjects examined, 49% were overweight and, in successive years following the menopause, there was a growing bipolarization of the weight increase. The correlation between overweight in the climacteric and the parameters considered was found to be significant only in regard to calorie intake, age and educational level. Post-menopausal gonadotrophin levels in blood were significantly lower in the overweight than in the normal-weight women. With the onset of menopause, the plasma level of testosterone fell in the normal-weight women, while it increased, along with that of hydrocortisone, in the overweight women. In the normal-weight women at menopause, it was found that there was a tendency towards a substantial increase in lipid fractions and glycaemia, as well as a state of hypercoagulability. In the overweight women, the tendency was towards an even more marked increase in both glycaemia and the various lipid fractions, and, besides the hypercoagulative state, there was an associated reduction in fibrinolytic activity. It is concluded that the menopause not only causes metabolic changes but also aggravates the metabolic and endocrine tendencies which characterize overweight subjects and thus, clinically, constitutes an obesity risk factor in those women who already demonstrate a tendency towards overweight in the pre-menopausal phase.

Inverse relationship between anti-SV40 TASA and anti-H-2 cytotoxic responses.

The in vitro cytotoxic response against H-2d and H-2b SV40-transformed fibroblasts was studied in a 40-h 3H-proline assay. A very low response against SV40 TASA is associated with the H-2d antigens on target cells: however, SV40-transformed H-2d cells are as immunogenic as SV40-transformed H-2b cells and prime against H-2b target cells. The data concerning in vitro amplification of the anti-SV40 TASA response and the involvement of cyclophosphamide-sensitive suppressor populations confirm the comparable immunogenicity of SV40-transformed H-2d AND H-2b cells and cannot account for the haplotype-related behavior observed with SV40-transformed target cells. The study of the response against allogeneic SV40-transformed cells shows the reverse situation: the lower cytotoxic response is now associated with the H-2b antigens on SV40-transformed cells. As suggested by the data presented here, an interaction between SV40 TASA and H-2 antigens might be postulated.

A study of thyroid function in the pre- and post-menopause.

Serum levels of thyroid-stimulating hormone (TSH), triiodothyronine (T3), thyroxine (T4), free triidotthyronine (FT3), free thyroxine (FT4) and thyroxine-binding globulin (TBG) were measured in 382 healthy women who were attending a menopause clinic. Of these women, 140 were in the pre-menopausal phase, 162 had been post-menopausal for less than 3 yr and 60 had been post-menopausal for greater than 3 yr. Analysis of the results as a function of climacteric situation, revealed that the only significant difference was a lower serum concentration of FT3 in the women who had been post-menopausal for over 3 yr than in the women who were in the pre-menopausal phase. Statistical analysis of the results as a function of both climacteric situation and age showed that the serum level of TBG increases with age, while serum levels of circulating thyroid hormones decrease, and that although the menopause does not bring about marked changes in thyroid function, it nevertheless tends to reduce that function, especially when menopause occurs at the expected time. Finally, by correlating values with T4 values on an individual basis, it was possible to ascertain that when physiological changes in thyroid function do occur in the climacteric, they are not caused by primary changes in TSH secretion.

Comparative susceptibilities of organisms isolated in Sicily to cefotetan and other antibiotics.

In recent years there has been a gradual increase in the number of infections caused by Klebsiella, Enterobacter and Serratia and non-fermentative bacilli such as Pseudomonas, Alcaligenes and Acinetobacter. The object of this investigation was to evaluate the antibacterial properties of the cephalosporins cefoxitin, cefoperazone, cefotaxime and of gentamicin, in comparison with cefotetan, a new cephamycin with a high degree of stability to beta-lactamases. The cefotetan MIC was not affected either by size of the bacterial inoculum, or the culture medium and the serum protein binding of cefotetan was 85%. Cefotetan showed only moderate antibacterial activity, similar to that of the other cephalosporins tested, against the Gram-positive cocci, whereas it was very active against fermentative and non-fermentative Gram-negative bacilli, except for Pseudomonas aeruginosa. The remarkable activity of cefotetan against Klebsiella, Enterobacter and Serratia is of particular interest.

Impaired H-2 expression in B16 melanoma variants.

We studied the expression of H-2b alloantigens in three different B16 melanoma lines cultures in vitro. Cell lines were B16-F1 and two cell cultures (named B16-A and B16-B) newly derived from two different in vivo sublines of B16 melanoma. The assays used were in vivo tumour growth in allogeneic (BALB/c and B10.BR) as compared to syngeneic mice, in vitro cell-mediated cytotoxicity by anti-H-2b immune lymphocytes and absorption of anti-H-2b antisera activity. The B16-F1 line was able to efficiently kill allogeneic hosts, could not be lysed by anti-H-2b cytotoxic effectors and did not express any serologically detectable amount of H-2b alloantigens. The B16-A line was H-2 positive during the early in vitro passages, then, at the 8th-10th passages, it acquired the capacity to kill allogeneic hosts, lost the sensitivity to anti-H-2b cytotoxic effectors and the H-2Kb antigens became undetectable The expression of H-2Db was reduced, although at a lower degree. Similar data were obtained with B16-B cells, which after 10 in vitro passages grew and killed allogeneic hosts, showed a decreased sensitivity to cytotoxic anti-H-2b effectors and a very low expression of the K region antigens. The results indicate that H-2 expression is altered in B16 melanoma lines and this may influence the different metastatic capacity of such cells.

Glucocorticoid receptor and in vitro sensitivity to steroid hormones in human lymphoproliferative diseases and myeloid leukemia.

The glucocorticoid receptor (GR) quantitation by a whole-cell assay and/or cytosol technique and the in vitro sensitivity to steroids have been assessed in peripheral blood cells from normal donors and patients with chronic lymphatic leukemia (CLL), acute lymphoblastic leukemia (ALL), lymphosarcoma cell leukemia (LSCL), acute nonlymphatic leukemia (ANLL), and chronic myeloid leukemia (CML). Within the lymphoproliferative diseases, ALL cells exhibited the highest GR concentration (regardless of the method used) and the highest in vitro inhibition of spontaneous [3H]thymidine ([3H]TdR) uptake by glucocorticoids. A significant relationship between GR concentration (whole-cell assay) and in vitro sensitivity to dexamethasone was also found. On the contrary, CLL cells presented the highest sensitivity to glucocorticoids in PHA-stimulated cell cultures. Cells from the only two ALL patients who did not undergo a remission after glucocorticoid-inclusive chemotherapy had both the lowest in vitro sensitivity to dexamethasone and the lowest GR concentration with whole-cell assay. Concerning myeloid leukemia, ANLL patients had GR concentrations slightly higher than those found in the ALL group but exhibited the lowest degree of inhibition of spontaneous [3H]TdR uptake by dexamethasone (stimulatory effects occurred in some cases). CML cells exhibited an inhibition degree by in vitro glucocorticoids significantly higher than that of ANLL cells but not different from that of lymphoproliferative diseases. No clear relationship among GR pattern, in vitro cell sensitivity to glucocorticoids, and clinicohematologic parameters was observed in myeloid leukemia-bearing patients.

The clinical use of arginine aspartate in male infertility.

130 male subjects (61 with asthenospermia, 69 with oligoasthenospermia) classified according to spermiologic patterns have been treated with different therapeutic outlines (gonadotrophins, antibiotica, arginine aspartate high levels, their associations); 89 subjects have been undergone with arginine aspartate, 41 without arginine aspartate. The results have been critically examined in order to different therapeutic outlines, to spermiogram, to diagnosis, to use of arginine aspartate in different spermiologic alterations, to use of arginine aspartate with respect to control groups, to arginine aspartate levels.

The occurrence of multiple steroid hormone receptors in disease-free and neoplastic human ovary.

The cytoplasmic receptors for 17 beta-estradiol (ER), 5 alpha-dihydrotestosterone (AR), progesterone (PR), and cortisol (GR) have been quantified in 36 specimens from the human ovary (13 disease-free, 5 benign, and 18 malignant) by a dextran-coated charcoal (DCC) technique. The occurrence of receptor-positive biopsies were: ER 46%, AR 85%, PR 54%, GR 92%, in normal tissue; ER 40%, AR 100%, PR 20%, GR 50%, in benign tumors; and ER 67%, AR 72%, PR 50%, GR 88%, in malignant lesions. Furthermore, the simultaneous occurrence of ER and PR in malignant tumors was 50% yet all four receptors were found to be present only in 44% of the cases. The findings reported here on the strong correlation existing between ER and PR presence or amount agree with previous observations on normal and neoplastic specimens from human breast and endometrial tissues.

Multiple steroid hormone receptors in normal and abnormal human endometrium.

The cytoplasmic concentrations of ER, AR, PR, and GR were determined in 124 specimens of normal and abnormal endometrium and other uterine human tissues by the DCC technique. In the endometrial carcinoma group, we observed that pretreatment with MAP leads to low cellularity, higher amount of AR, lower amounts of detectable ER, GR, and PR: the last receptor was almost always absent. A positive correlation between ER presence and tumor grade of differentiation was found in endometrial tumors from hormone-untreated patients. With the value of 142 fmol/mg DNA as the cut off point between high and low binding capacity, the frequency of the single receptors within the hormone-untreated cancer group ranged from 61% to 88%; ER and PR were simultaneously present in 55% of cases (they are tightly correlated in the different biopsies with respect to frequency and amount); ER-AR-PR were present in 45% and all the four receptors in 40% of cases. Slightly higher values were found in normal endometrium collected from hormone-untreated patients.

[Measurement of cytoplasmic receptors specific for steroid hormones. IV) Glucocorticoid receptor].

Binding of 3H-cortisol (3H-C) and 3H-dexamethasone (3H-DX), a fluorinated glucocorticoid which does not bind to Corticosteroid Binding Globulin (CBG), was investigated in two different target tissues, human breast cancer and human leukemic lymphocytes, using a modified DCC assay method. Maximal binding to cytosol occurred, when increasing concentrations of 3H-C or 3H-DX were incubated in the presence or in the absence of an excess of cold cortisol or dexamethasone, at 0 degrees C for 1 hr or 4 hr, respectively. A 50 sec exposure to DCC was sufficient to separate bound from free labelled steroid. Data presented suggest that two different classes of glucocorticoid exist, which are differently distributed in target tissues, and may be identified on the basis of affinity for the tracer, binding specificity and thermolability.

HI rubella antibody determination. (A comparison of methods proposed for the removal of non-specific inhibitors).

The Authors consider the different methods so far proposed for the selective removal of non-specific inhibitors for HI rubella antibodies. They show that the best results are obtained by dextran sulphate and acetone treatments, while the kaolin and heparin-MnCl2 procedures should be avoided especially in the serological diagnosis of acute rubella infection. The presence of specific IgM was demonstrated by 2-mercaptoaethanol.
