Transport and hydrolysis of maltose by Schwanniomyces castellii.

Hydrolysis and transport of maltose into Schwanniomyces castellii was studied under aerobic and anaerobic conditions. Amylase and glucosidase were not synthetized in presence of maltose in anaerobic conditions. Maltose permease was synthesized in anaerobiosis and its functioning is not inhibited. Glucose strongly repressed induction. The half-saturation constant for uptake of maltose was 0.06 mM. The rate of uptake of maltose was decreased by 2,4-dinitrophenol, antimycin or sodium azide. The significance of these results in relation to the Kluyver effect is discussed.

Pharmacokinetics of spiramycin after intravenous, intramuscular and subcutaneous administration in lactating cows.

Spiramycin is a macrolide antibiotic that is active against most of the microorganisms isolated from the milk of mastitic cows. This work investigated the disposition of spiramycin in plasma and milk after intravenous, intramuscular and subcutaneous administration. Twelve healthy cows were given a single injection of spiramycin at a dose of 30,000 IU/kg by each route. Plasma and milk were collected post injection. Spiramycin concentration in the plasma was determined by a high performance liquid chromatography method, and in the milk by a microbiological method. The mean residence time after intravenous administration was significantly longer (P less than 0.01) in the milk (20.7 +/- 2.7 h) than in plasma (4.0 +/- 1.6 h). An average milk-to-plasma ratio of 36.5 +/- 15 was calculated from the area concentration-time curves. Several pharmacokinetic parameters were examined to determine the bioequivalence of the two extravascular routes. The dose fraction adsorbed after intramuscular or subcutaneous administration was almost 100% and was bioequivalent for the extravascular routes, but the rates of absorption, the maximal concentrations and the time to obtain them differed significantly between the two routes. Spiramycin quantities excreted in milk did not differ between the two extravascular routes but the latter were not bioequivalent for maximal concentration in the milk. However, the two routes were bio-equivalent for the duration of time the milk concentration exceeded the minimal inhibitory concentration (MIC) of various pathogens causing infections in the mammary gland.(ABSTRACT TRUNCATED AT 250 WORDS)

Production of food and fodder yeasts.

A decade or so ago, there was considerable interest in developing single cell protein production from raw materials. Many factors have influenced the development of fodder yeast technology, notably the biochemistry and physiology of the yeast. It is shown that those considerations have led to the choice of a continuous fermentation technology.

Animal Feed

[Acquired atrophic pigmented band-like lesions following Blaschko's lines].

Over a seventeen years period we observed 5 patients presenting with very similar lesions the equivalent of which we could not find in the literature. These were pigmented and more or less atrophic bands which exactly followed Blaschko's lines. The lesions appeared during childhood or adolescence, between the ages of 6 and 20 years and always were unilateral. In all cases they were located on the trunk where they formed a recumbent "S" pattern characteristic of Blaschko's lines. They often started 3 to 6 cm away from the posterior midline and ended precisely on the anterior midline. These bands varied in number and in 3 out of 5 cases they were associated with pigmented atrophic bands or lines on the limbs of the same side. The intensity of pigmentation and atrophy was variable. The lesions were asymptomatic and only caused a cosmetic prejudice. They were perfectly fixed, and during a 2 to 30 years observation period they remained unmodified. The five skin biopsies performed on 3 patients showed no abnormality of the epidermis other than irregular and moderate hyperpigmentation of its deep part. In the dermis, there was no distinct pigment incontinence, no inflammation and no alteration of connective tissue texture; the elastic network was invariably normal. The impression of skin atrophy was not due to true dermal hypoplasia and perhaps corresponded to atrophy of the subcutaneous cellular tissue. No biological disturbance was observed. These lesions can easily be distinguished from epidermic naevi, incontinentia pigmenti and areas of hypoplastic dermis.(ABSTRACT TRUNCATED AT 250 WORDS)

[Ioversol 300 and iopamidol 300 in "whole body" computed tomography. A double-blind clinical trial].

The authors compared ioversol 300 to iopamidol 300 by double blind study in patients undergoing enhanced computed body tomography. There was no significant difference in clinical tolerance and efficacy between the two products. Only minor reactions were recorded in both groups. Contrast media were not considered to be responsible for the poor diagnostic quality of a few explorations. Ioversol 300 appears as a well tolerated and effective contrast medium for computed body tomography.

[Epidermal cyst and osteolysis of the cranial vault].

In a 40-year old man undergoing, under local anaesthesia, excision of an epidermal cyst located in the frontal region, at the border of the scalp, the operator had difficulties in removing the deep part of the cyst and perceived an underlying bone depression. The depression was caused by a 2 x 1.3 cm wide lacuna in the calvarium, which was subsequently treated by neurosurgeons. Histology showed only fragments of a simple epidermal cyst wall and no evidence of dermoid cyst. The causes of osteolysis associated with congenital or acquired skin lesions are reviewed. In this case, the old age and volume of the cyst may explain the osteolysis by mechanical compression. This case is exceptional since we were unable to find other examples in the literature, apart from dermoid and trichilemmal cysts.

[Extrapleural hematoma].

The authors report a case of a voluminous extra-pleural hematoma in relation to a fracture of the body of the eleventh thoracic vertebra on a 74 year-old man after craniothoracic injury. Thoracic CT Scan showed a homogeneous posterior parietal mass of tissue density (40 HU) outlined by a fatty lining associated with a fracture of the eleventh thoracic vertebra. This allowed a modification in the diagnosis of a hemothorax established previously on conventional radiography. The authors insist on the gravity of this affection when the volume of the hematoma is important. But this is very rare (6 cases out of which 5 were autopsic findings). If present it requise an urgent surgical treatment without forgetting the fiability of CT Scan which relies on putting into evidence the pleural lining sign.

[Facial hemiatrophy, homolateral cervical linear scleroderma and thyroid disease].

A case of facial hemiatrophy and homolateral cervical band of scleroderma, complicated by hypothyroidism is reported. This case raises two problems: one is the problem of distinction between Romberg's disease and facial hemiatrophy due to a genuine localized scleroderma; the other concerns the relationship between localized scleroderma and dysthyroidism. The generalized scleroderma-dysthyroidism association has now been recognized, but the coexistence of thyroid disease and localized scleroderma has not yet been reported. Several pathogenetic hypotheses on this association are discussed.

Facial Dermatoses

The normal uterus on magnetic resonance imaging and variations associated with the hormonal state.

Magnetic Resonance Imaging of the pelvis has lead to a new radioanatomical description of the uterus. Variations in the anatomical zones of the uterus were noted with different sequences used in MRI. There was also structural and signal variation depending on hormonal impregnation. Twenty seven MRIs of pelvices were carried out on 16 female volunteers. Eight of them were on oestroprogesterone as the method of contraception (Group I) and had a single MRI. Six were not taking contraceptives (Group II) and had different MRI's during the same cycle. Two women already had already gone through the menopause (Group III). Results showed differences in the size of the uterus (uterine height, length of the uterine body and cervix, diameter of the cervix) between groups I and II but no cyclic changes were observed within group II. The myometrium was found to be thicker in group I and in the luteal phase of group II females in comparison to the follicular phase in the same group. The endometrial cavity was seen in 40% of cases on T1 weighted sequences; on T2 weighted sequences it was seen in all the cases. It was thicker during the second phase in group II than in group I and III. The myometrial signal compared to that of the bladder content (urine) showed a significant increase between early and late echoes on T2 weighted sequences in group I and during the second phase of the menstrual cycle in group II. No variations, or at times a decrease in signal, were observed during the first phase of the cycle in group II.(ABSTRACT TRUNCATED AT 250 WORDS)

Characterization of alternative respiratory pathways in the yeast Schwanniomyces castellii by the study of mutants deficient in cytochromes a+a3 and/or b.

After a general review of the proposed mechanisms and physiological roles of the alternative respiratory pathways found in various organisms, the studies are focussed on the amylolytic yeast Schwaniomyces castellii. In addition to the cytochrome chain, the wild type presents two alternative pathways insensitive to antimycin A. One is salicylhydroxamic acid (SHAM)-sensitive and azide-insensitive; the other is SHAM-insensitive and sensitive to high azide concentration. Conditions for mutagenesis and screening are described, which allow isolation of mutants deficient in cytochromes a+a3 and/or b in this yeast previously classified as petite negative. The relative proportions of the alternative respiratory pathways are compared in the wild type and mutant strains following inhibition by SHAM and azide at optimal concentration as determined by iso-inhibition curves. The growth of the cytochrome deficient mutants on citrate, a non-fermentable carbon source, and the ability of the wild type to grow on citrate+antimycin A, after a lag of about 10 h, indicate an involvement of the alternative pathway(s) in energy production. Rotenone sensitivity of respiration and ATP level confirm the presence of a functional phosphorylation site 1. The role of each alternative respiratory pathway in energy production is discussed.
