[Levels of lead on the livers of dogs from the Paris region].

Lead level has been determined in the liver of 51 Dogs of different ages, sex and breed from districts of Paris. This level is from 0,16 ppm (fresh weight) up to 5,4 ppm. 23 p. cent of Dogs examined have more than 3 ppm in their livers. The level is not correlated with sex, breed and size. It increases with age of animals.

[Curing of the congenital osteopetrosis of "op" rats by thymus grafts].

A graft of normal thymus performed after thymectomy can cure the lethal osteopetrotic condition of the "op" Rat. The beneficial effect of normal bone marrow injection can be suppressed by prior thymectomy of the "op" Rat. Thymus is likely to play a role in the pathogenesis of osteopetrosis.

[The effects of somatostatin in acromegaly].

Somatostatine is the hypothalamic factor which inhibits the secretion of growth hormone. The administration of a synthetic form decreased growth hormone levels by 50 to 75% in 5 acromegalic patients. The action is rapid but the effect is not prolonged. Prolactin was reduced in only case with galactorrhea. Thyreostimulin, as well as gastrin, are unaffected. Plasma insulin levels, and to a lesser extent those of glucagon, are decreased by somatostatine which causes no variation in either cortisol or blood glucose. Somatostatine, by correcting the pathological secretion of hormone, opens the way to medical treatment of acromegaly.

Comparative in vitro biological activity of 1-34 N-terminal synthetic fragments of human parathyroid hormone on bovine and porcine kidney membranes.

1-34 N-terminal fragments of human parathyroid hormone with sequences according to Brewer et al (hPTHB) and to Niall et al (hPTHN) were synthesized and compared for their ability to activate bovine and porcine kidney cortex membrane adenylate cyclase. Results show that these two hormone sequences are able to activate these membranes but at least in this in vitro assay, hPTHN is about 10 times more active than hPTHB on bovine as well as on porcine membranes. These "apparent potencies" with respect to the potency of bPTH 1-34 in bovine and porcine membrane assay systems are respectively 4% and 11% for hPTHB and 39% and 156% for hPTHN. These relative potencies may be interpreted as corresponding to species specificity of the hormone receptor structural relationships.

[Tests of stimulation of calcitonin secretion. Value in medullary cancer of the thyroid].

The detection of high circulating levels of calcitonin is a most valuable procedure to diagnose advanced cases of medullary carcinoma of the thyroid. However, the diagnosis of a primitive tumor in the early stages of development (as in cases of the familial form of the disease) or of a metastasis following ablation of the tumor is more difficult. In the latter cases, the levels of circulating calcitonin may be within normal limits, and for diagnosis one must then resort to tests to stimulate the secretion of calcitonin. We are reporting from our personal experiences the advantages, disadvantages and inconveniences of the three most well-known tests. All patients responded positively to administration of calcium or pentagastrin. The alcohol test, however, produced inconsistent results.

[Heterogeneity of human calcitonin].

Chromatographic analysis of plasma, urine and tumour tissues extracted from medullary cancer patients demonstrates the existence of immunoreactive calcitonins with higher molecular weight than that of the monomer hormone.

[Multidimensional analysis in clinical investigation. Application to the pathogenic classification of idiopathic hypercalciuria].

Multidimensional analytical techniques enable any number of variables to be studied simultaneously. These techniques have been described and a few examples of their application to biomedical research given. A classification of so called "idiopathic hypercalciuria" was made using this approach which demonstrated the existence of three possible mechanisms of pathogenesis.

[Analysis of disorders of the principal pathways of calcium metabolism in Paget's disease. Effects of calcitonin administration. 26 cases].

Analysis of the principal routes of calcium metabolism by means of calcium-45 in 26 patients suffering from Paget's diseases sometimes showed, in agreement with data from the literature, a considerable increase in the common stocks of calcium in 25 cases, and an elevation in the anabolism (Vo+) in bone and in the catabolism (Vo-) in bone in all cases. The calcium balance was sometimes equal, but often clearly positive (10 cases) or negative (10 cases) and the level intestinal absorption of calcium was usually within the normal limits. An elevation in the endogenous fecal calcium (Vf) noted in half of the cases, although no value was below normal, seemed to represent an metabolic anomaly of abnormally high frequency in patients with Paget's disease, even though this fact has never been emphasized before. Statistically significant correlations were shown between the values of different kinetic and metabolic parameters in the group of patients studied. A second kinetic study carried out after several weeks of treatment with porcine calcitonin showed a statistically significant diminution in the values of Vo +, Vo -, endogenous fecal calcium, calciuria, hydroxyprolinuria, and in alkaline phosphatasaemia, as well as significant variations in the calcium balance in a positive direction. The common calcium stocks did not show a significant diminution, contrary to the findings of other authors. The level of intestinal absorption of calcium increased more often than it decreased, but the magnitude of the variations was slightly less than the level of statistical significance.

[Heterogeneity of immunoreactive calcitonin in the plasma of patients with bone marrow cancer].

Plasma from patients with medullary carcinoma containing very high levels of immunoreactive calcitonin were fractionated by filtration on Sephadex gel. In all cases the elution gave four immunoreactive fractions. Two of these fractions correspond to the volume of elution of the monomere and of the dimere of human calcitonin. The two other fractions emerge at an elution volume corresponding to much higher molecular weights. After stimulation of calcitonin secretion in vivo, by dynamic tests, the fractions corresponding to the monomere and dimere increase more strongly than the two other fractions. Preliminary studies of secretion, in vitro, of calcitonin by medullary carcinoma tissue, show the presence in the incubate of four immunoreactive forms having the same elution characteristics as those found in the plasma. The significance of these results is discussed.

Calcitonin treatment of post-menopausal osteoporosis. Evaluation of efficacy by principal components analysis.

The efficacy of long term treatment of senile osteoporosis by low doses of calcitonin was established using five parameters of calcium kinetics and a quantitative pain scale. Under treatment the calcium balance improved, due predominantly to a decrease in bone resorption associated with an increase in bone accretion and intestinal absorption of calcium. In addition, the hormone had a marked analgesic effect, which increased with the length of the treatment. Principal components analysis enables to establish the value of a therapeutic agent for the management of a progressive disease with period of regression like osteoporosis, for which the eficacy of previously advocated treatments had never been proven.
