Conjugation of capsular polysaccharide to alpha-haemolysin from Staphylococcus aureus as a glycoprotein antigen.

The capsular polysaccharides of two pathogenic strains of Staphylococcus aureus 8914 and 31 were isolated and purified. These polysaccharides were conjugated to alpha-haemolysin prepared from the same strains. Amongst the various coupling procedures which were tested the best results were obtained with sodium cyanoborohydride and glutaraldehyde. The conjugates were purified and their immunologic properties were tested. They gave a positive response against antisera from whole bacterial cells.

Ciclopirox nail lacquer 8%: in vivo penetration into and through nails and in vitro effect on pig skin.

This report presents original methods to assess the bioavailability of an antifungal drug from a varnish preparation in finger nails. For the studies with human volunteers a ciclopirox 8% nail lacquer was used to determine its efficacy in the treatment of onychomycoses. In vivo studies were performed on the fingernails of healthy volunteers by determining the total amount of ciclopirox penetrated per milligram of nail and the partition of the drug in the plate of the nails (technically divided into four layers). Ciclopirox concentrations were evaluated by measuring the inhibition of Candida pseudotropicalis growth in vitro. The ciclopirox concentration after 30 days treatment was determined as 3.35 +/- 0.82 micrograms/mg nail material. This is a sufficient amount to kill the fungal pathogens. In addition, in vitro penetration experiments were carried out with excised pig skin. Lacquer formulations from 0.5 to 8% were used to inhibit the growth of Trichophyton mentagrophytes. Formulations from 2 to 8% led to a strong to total inhibition of the dermatophyte after 30 min treatment time.

Ciclopiroxolamine cream 1%: in vitro and in vivo penetration into the stratum corneum.

Studies were conducted to assess the penetration of 1% ciclopiroxolamine cream, establishing the concentrations of the antimycotic compound in different layers of the stratum corneum with two skin models. Results of in vitro studies using skin from domestic pigs indicate that ciclopiroxolamine has the ability to penetrate fast into the epidermis, by inhibiting and killing inoculated Trichophyton mentagrophytes. In vivo investigation of the stratum corneum has been performed in healthy human volunteers by 20 strippings collected in four layers. Fungicidal concentrations of ciclopiroxolamine were determined after extraction from the strippings. These studies demonstrate that the concentrations reached levels which are sufficient to inhibit and kill pathogenic fungi.

Protooncogene expression in rat liver by polychlorinated biphenyls (PCB).

1. The expression of 10 protooncogenes was studied in control rat liver and at various times after exposure to polychlorinated biphenyls (PCB), a known tumour promoter. 2. The expression of protooncogenes in liver is more pronounced in those rats treated with PCB beginning at weaning ('weanlings') than in adult rats. 3. The RNA levels of c-Ha-ras, c-raf, c-yes, c-erbA and c-erbB are elevated after PCB feeding. 4. Nuclear run-on transcription analysis revealed that the altered expression of the protooncogenes is transcriptionally regulated. 5. In one group the prompt rise of the protooncogene transcription rate is followed by a decline (c-Ha-ras, c-raf c-yes). In a second group a further increase in transcription at later feeding times (c-erbA, c-erbB) was observed. 6. A correlation between the altered expression of these protooncogenes and the action of PCB as a tumour promotor remains to be determined.

[Adaptative differential maturation of neurohypophyseal provasotocin in amphibians: presence of hydrin 2(vasotocinyl-glycine) in anura but not in urodela].

Occurrence of hydrin 2 (vasotocinyl-glycine), an intermediate in the provasotocin processing, has been shown in the neurointermediate lobe of the pituitary of semi-aquatic or terrestrial amphibian species belonging to the order Anura, namely Rana esculenta, R. temporaria, R. pipiens (Ranidae), and Bufo bufo, B. marinus, B. ictericus (Bufonidae). In contrast hydrin 2 appears absent in species belonging to the order Urodela, namely the axolotl Ambystoma mexicanum (Ambystomidae) and the pleurodele newt, Pleurodela waltlii (Salamandridae). Whereas water transport through the skin and the urinary bladder is stimulated by vasotocin and hydrin 2 in anuran species, this stimulation is known not to occur in urodele species.

Studies on the antifungal antibiotics: bacillomycin D and bacillomycin D methylester.

Bacillomycin D, an antifungal compound of Bacillus subtilis, was produced during the stationary phase of growth when all the glucose of the medium was exhausted and the pH reached about 8. In addition to bacillomycin D, bacillomycin D methylester was characterized by its antifungal properties and its haemolytic activity.

[Histologic studies of the postnatal development of the uterus of dwarf goats].

Thirty-four Syrian goats were investigated for postnatal uterus development, from the first day after parturition to the age of six months. Structures were found to undergo conspicuously early differentiation. Full morphological uterus development was established at the age of three to four months. Syrian goat thus is sexually mature at that early age. This is considered to be essential to good suitability of Syrian goat as experimental animal.

[Tumors of the facial bones. Current imaging: x-ray computed tomography versus MRI].

This study evaluates the comparative efficacy of MRI and CT in evaluating facial tumors. MRI, with its excellent contrast on T2 weighted and post-gadolinium sequences, is superior to CT for delineating tumor extension, the information essential for appropriate surgical intervention. Both modalities can sometimes be helpful for tumor characterisation.

Influence of divalent ions on the solubility of iturin and bacillomycin L, antifungal peptidolipids of Bacillus subtilis.

When calcium chloride was added to the culture medium of strains producing iturin or bacillomycin L, the antibiotic, normally excreted in the supernatant of the culture medium, was found in the cell pellet. This apparent inhibition of the antibiotic excretion was studied and it was demonstrated that CaCl2 precipitated the antibiotic after its excretion in the medium. The ability of various chloride salts to precipitate iturin and bacillomycin L was tested and the most effective salts were CaCl2 and MnCl2. Comparison of the compounds obtained by CaCl2 precipitation and by acid precipitation showed that, in the latter case, major antibiotics were accompanied by minor congeners resulting from modifications of genuine antibiotics.

[Hodgkin's disease: epidemiological and anatomopathological aspects].

Only seventy cases of Hodgkin's disease were reported in Cameroon over a twelve-year period (1979-1989), which represents about six cases a year. Hodgkin's disease therefore seems to be rare in Cameroon where it will be useful to follow its epidemiological course. At the present time, 81.5% of patients are male and children aged 10 to 19 are mostly affected. Type I (lymphocytic predominance) is the more frequent, representing 40% of cases. Biopsy leading to the diagnosis is mainly performed on cervical lymph nodes (58% of cases).

[The value of nuclear magnetic resonance in the study of iron overload in thalassemia patients].

In patients with thalassemia treated by long-term chronic blood transfusions who survive beyond the first ten or twenty years of life but received no or inadequate chelating therapy during the first years, evaluation of iron overload and its consequences on tissues may prove an arduous task. MRI is a non-invasive means of measuring the amount of iron in the liver and the consequences of the iron overload on the heart and other tissues. For this purpose, MRI is more satisfactory than CT scan studies. In this investigation, 20 patients with thalassemia major underwent MRI. Multiple spin echos were used to allow determination of the transversal relaxation constant T2. This constant, expressed in ms, is related to the concentration of iron in the liver as in the following expression: (C) = 5 410/T2-110. MRI studies disclosed a discrepancy between the severity of hepatic hemosiderosis and development of decompensated iron overload cardiomyopathy. In a unique case, in which a heart transplant and two MRI studies were performed, the severe iron overload that failed to respond to several years of subcutaneous chelating therapy was more than halved by intensive intravenous chelation through a central catheter. MRI studies of the heart provide valuable morphologic and functional data. Although the amount of iron in the myocardium cannot as yet be quantified, modifications of the transversal relaxation time provide information on the severity of the overload and the presence of other myocardial alterations.

[Evaluation of the role of MRI at 1.5 T in the staging of the initial extension of cancer of the uterine cervix].

To evaluate the sensibility and specificity of MRI in the initial staging of carcinoma of the uterine cervix, a retrospective study of 41 patients (48 MR scans) was undertaken. The sensitivity of T2-weighted images in the detection and determination of tumor location was 90% and specificity was 100%. On T2-weighted images, the signal intensity of the tumor was high in 64%. Determination of tumor size was correct in 80%. The ability of MRI to demonstrate the extension to the uterine corpus was acceptable with a sensitivity of 63% and a specificity of 100%. The extra-uterine extension was difficult to appreciate especially for parametrial, vaginal and bladder involvement with a sensitivity of 38%, 43 and 67% respectively. Concerning the lymph nodes involvement, the sensitivity was 38%. The results of this study point out the ability of MRI to evaluate the size and location of the tumor. The accuracy of MRI to appreciate the involvement of the parameters, vagina and bladder is disappointing.

Studies on the biosynthesis of iturin, an antibiotic of Bacillus subtilis, and a lipopeptide containing beta-hydroxy fatty acids.

The biosynthesis of iturin, an antibiotic containing a beta-amino fatty acid, was studied by incubating Bacillus subtilis in the presence of various 14C-labelled precursors. Sodium acetate or palmitic acid were incorporated into the beta-amino acids of iturin. Among the alpha-amino acids (asparagine, glutamine, serine, proline and tyrosine) in the peptidic part of iturin, asparagine appears to be the best precursor. In the presence of sodium [14C]acetate or [14C]asparagine, there was a synthesis of radioactive compound (compound X) before the synthesis of radioactive iturin. Compound X contained asparagine and/or aspartic acid, glutamine and/or glutamic acid and beta-hydroxy fatty acids.

Biochemical and immunochemical studies of proteolytic fragments of exotoxin A from Pseudomonas aeruginosa.

Limited proteolysis of Pseudomonas aeruginosa exotoxin A by four proteases (chymotrypsin, Staphylococcal serine proteinase, pepsin A and subtilisin) resulted in the formation of polypeptides having a molecular mass of approximately 25 kDa. They possessed both enzymatic activity and residual antigenicity. Their N-terminal sequence analysis showed that the different proteases cleaved exotoxin A in a very restricted area within domain Ib (amino acids 365-404). As a result, the polypeptides contained a large portion (13-34 amino acids) of domain Ib linked to the adjacent C-terminal domain III (amino acids 405-613). The major fragment derived from subtilisin cleavage, at a final yield of 35% (S-fragment; residues 392-613; 24201 Da; pI 4.7) possessed the same level of ADP-ribosyltransferase activity as uncleaved exotoxin A (by mass), and a 37-fold higher NAD-glycohydrolase activity. Polyclonal antibodies from rabbits against exotoxin A completely inhibited the ADP-ribosyltransferase activity of both exotoxin A and the S-fragment, but not the NAD-glycohydrolase activity of the S-fragment. Antibodies against the S-fragment neutralized the ADP-ribosyltransferase activity of exotoxin A. These data determine the primary proteolytic cleavage site of exotoxin A, suggest that some residues in the amino acid sequence 392-404 of exotoxin A seem to have a role in binding or positioning elongation factor 2 (EF-2) and show that antibodies recognize the EF-2-binding site but not the NAD(+)-binding site.

Structural investigations and biological activity of inositol sphingophospholipids from Phytophthora capsici.

Inositol sphingophospholipids that protect pepper (Capsicum annuum c.v. Yolo Wonder) against pathogen have been isolated by chromatographic methods from the mycelium of Phytophthora capsici. The structure of the major compound was determined by chemical methods and mass spectrometry. Phosphodiester bond cleavage of the phospholipid by mild alkaline hydrolysis liberated a ceramide which contained a C16-sphingosine. This long-chain base was identified by gas chromatography and mass spectrometry of its trimethylsilyl derivative. One of the amide-linked fatty acids was found to be 4-hydroxy-2 docosenoic acid. Fast-atom-bombardment mass spectrometry and fast-atom-bombardment collison-induced tandem mass spectrometry were used to characterize the ceramide as N(4-hydroxy-2-docosenoyl)C16-sphingosine. These sphingolipids have a protective effect on cotyledons of young peppers against necrotic lesions induced by the pathogen P. capsici.

Evolutionary specificity of hydrins, new hydroosmotic neuropeptides: occurrence of hydrin 2 (vasotocinyl-Gly) in the toad Bufo marinus but not in the viper Vipera aspis.

Hydrin 2 (vasotocinyl-Gly), a hydroosmotic peptide resulting from differential processing of provasotocin and recently identified in frog neurohypophysis, has been looked for in the pituitary gland of an exotic toad (Bufo marinus) and of a reptile (Vipera aspis). Hydrin 2 has been found in the amphibian but not in the reptile. This result confirms the evolutionary specificity of hydrin 2 that has been identified only in frogs and toads but not in birds and reptiles. Occurrence of hydrin 2 is explained by its regulatory function on the water permeability of the skin of anurans.

The cytotoxicity of Pseudomonas exotoxin A, inactivated by modification of the cell-binding domain I, is restored when conjugated to an erythroid cell-specific targeting agent.

To be capable of selective killing of tumor cells, the non-selective Pseudomonas aeruginosa exotoxin A must have its cell-binding domain inactivated or removed and then be chemically linked to, or genetically fused with, a specific targeting agent. In the present study, epsilon-NH2 groups of lysine residues of the cell-binding domain of exotoxin A were extensively propionylated with N-succinimidyl-3-propionate (NSP). The NSP-treated exotoxin retained its cytocidal ADP-ribosyltransferase activity, but it could no longer bind to, and inhibit the proliferation of, Friend murine erythroleukemia cells. Cytotoxicity (i.e., the ability to inhibit proliferation) for the Friend erythroid cells was restored completely to the NSP-inactivated exotoxin by conjugating it to ADIF, an autocrine factor secreted by chicken erythroleukemia cells which selectively inhibits the differentiation of erythroid cells such as Friend erythroleukemia cells without inhibiting their proliferation.
