Early cardiovascular changes in adolescents with high blood pressure values.

To assess the type and degree of cardiovascular involvement in the early phases of hypertension, 46 adolescents with casual blood pressure (BP) greater than the 95th percentile for age and sex (23 males and 23 females, mean age 14 years) were studied by M-mode echocardiography. Twenty-seven adolescents with casual BP about the 50th percentile, 17 males and 10 females, matched for age, were studied as controls. Adolescents with casual high BP values showed an increase in left ventricular mass (P less than 0.01) and wall thickness to radius (h:R) ratio (P less than 0.01) in comparison to controls. Cardiac index was increased and was correlated with left ventricular mass in hypertensive subjects (P less than 0.05). The correlation between cardiac output and left ventricular mass suggests that cardiac output is a factor in the development of left ventricular hypertrophy.

Bromocriptine reduces plasma noradrenaline and 3,4-dihydroxyphenylacetic acid in normal and hypotensive subjects.

The effect of a single oral dose of bromocriptine 2.5 mg was evaluated in 11 normotensive and 6 hypertensive volunteers. 150 min after drug administration, a significant decrease in plasma noradrenaline concentration from 202 to 124 pg/ml in normotensive and from 197 to 119 pg/ml in hypertensive patients was observed. Plasma 3,4-dihydroxyphenylacetic acid, a major metabolite of dopamine, fell from 1132 to 956 pg/ml in normal subjects and from 1242 to 807 pg/ml in hypertensives. No change in plasma adrenaline was found. At the same time, mean arterial pressure showed a significant decrease from 90 to 81 and from 132 to 111 mmHg in normotensive and hypertensive subjects, respectively. Bromocriptine also inhibited the increase in noradrenaline level that occurred when the subjects changed from the supine to the standing position. The inhibition was more evident in hypertensive subjects. It is suggested that the hypotensive effect of bromocriptine is mediated by the inhibition of noradrenaline release due to the stimulation of dopamine receptors on noradrenergic nerve terminals.

A study of the parameters affecting flow gradient analysis of catecholamines, DOPA and DOPAC by ion pair liquid chromatography with electrochemical detection.

A general method for the determination of norepinephrine, epinephrine, dopamine, 3,4-dihydroxyphenylalanine and 3,4-dihydroxyphenylacetic acid is described. It employs the generation of a flow gradient during the analysis by ion-pair reversed phase liquid chromatography. The method allows a reduction of the analysis time maintaining high resolution of the peaks close to the solvent front. The effect of the flow gradient on column efficiency, detector response and retention are evaluated. The application to specimens of amniotic fluid, urine, human plasma and rat brain tissue is shown.
