[The effect of a synthetic tripeptide nervous tissue cultured in vitro].

Explants from chick embryo PNS (ganglion trigeminale) and from CNS of embryonal rats (hippocampus) and dissociated cells from chick embryo cerebral hemispheres were cultivated in maximow chambers in the presence of various concentrations of placental serum and of a chemically synthesized tripeptide Gly-His-Lys. 1. The presence of tripeptide in the nutrient medium with a low concentration of serum did not compensate the outgrowth of nerve fibers, that take place in the growth medium. 2. In the presence of tripeptide in the nutrient medium with low concentration of serum the index of growth area increased significantly. 3. Within the first days in cell cultures 0,01 microgram tripeptide pro ml medium stimulated the outgrowth of neuronal processes. 4. The experiments indicated, that the tripeptide did not replace the serum. The possible role of tripeptide as a system in controlling neuron-glial ratio in vitro is discussed.

[The effect of Solcoseryl on explant cultures of the hippocampus].

Explants of hippocampus from fetal rats were cultivated in Maximow chambers in semisynthetic medium up to 12 days in vitro. The cultures were fixed Bouin, slided 15 micron, coloured with Klüver-Barrera and some morphological parameters were tested. 1. The nerve fiber index increased by influence of 1% Solcoseryl in relation to control cultures, which growed in minimal medium. An essential stimulation was observed by application of placentar serum and embryonal extract into the culture medium. 2. Die decrease of the number of neurons and glial cells per unit of area and a small decrease of the area of neuron nuclei was discussed in relation to the effect of the pharmacon Solcoseryl on O2- consumption. 3. Solcoseryl (firm Solco AG, Base) is an extract of calf blood. It can not substitute other tissue extracts.

[The role of the pupil's size in the perception of other persons].

Studies by Hess (1975) show that the pupil serves as a source of information in perception of the emotional state of another person. Large pupils are interpreted in a positive sense. Basing on the results of Hess, the hypothesis that pupil size does not become a relevant variable in person perception until after puberty, was formed. The following result would be interpreted as supporting the hypothesis: since girls reach puberty before boys, more girls should react to pupil size than boys of the same age. 121 students between the ages of 11 and 16 years judged in several statements which of two face photos of a teacher expressed a greater degree of positive attention and esteem towards the students. The two face photos were identical with the exception of pupil size. All age groups rated the photos with larger pupils more positively. Sex differences occurred in the ratings of the 11 to 12-year-olds. Some basic differences between this experiment and that of Hess are discussed.

[Action of cyclic N6-monobutyryladenosine-3':5'-monophosphate (mb-cAMP) on cell on cell and explant cultures of the PNS and CNS in vitro].

Chick embryo trigeminal ganglia of nine days and hippocampus excised from fetal rats between the sixteenth and eighteenth day of gestation were cultivated in cell- and explantcultures in Maximow-chambers. The cultures were placed in a medium containing 2 mM, 1 mM, 0.2 mM or 0.02 mM cyclic N6-Monobutyryladenosin-3':5'-monophosphat (mb-cAMP). The cultures of the PNS, the trigeminal ganglai show after 48 h by 1 mM mb-cAMP longer nerve fibers and a increased nerve fiber index. Smaller concentration did not change significant nerve fiber growth. Cell- and explantcultures of the hippocampus show a stimulation of the nerve fiber growth and the nerve fiberindex by concentration of 0.02 mM mb-cAMP. After ten days in vitro the nerve cells degenerated by higher concentrations.

[The effect of Solcoseryl on in-vitro cultured cells].

Explants of peripherical nervous system (PNS), skin and ventriculus cordis from chick embryo were cultivated in Maximow chambers and the effect of Solcoseryl, Fa. Solco Basel AG, on some morphological parameters was tested. 1. The growth of tissue cultures is influenced by Solcoseryl in relation to concentration and time of application. The index of area in cultures of PNS and cor increased within the first days. By long time application up to 6 days in vitro the index of area decreased and the index was the same than in controls. Explants of skin showed no essential stimulation of growth. 2. The number of cells per unit of culture in the outgrowth of PNS, cor and skin was different influenced. The density of cells in cultures of PNS and skin decreased (signif. difference). In explants of heart we could not observe a difference between the inside and outside of the outgrowth. An influence of Solcoseryl on the degree of migration is discussed. 3. The area of cell nuclei from heartcells was observed. The area decreased under the influence of Solcoseryl. The difference is significant. 4. The mitotic index of heart cells increased by application of Solcoseryl within the first 2 and 3 days in vitro. 5. The number of nucleoli per nucleus of heart cells under experimental conditions increased significant. It is discussed, Solcoseryl influenced in vitro metabolic processes in suitable systems; stimulation of cell proliferation and migration and rns-synthesis was observed within the first days of cultivation. In-vitro-systems are important objects and they are suitable for tests of pharmaca in vitro.

[The effect of orotic acid and orotic acid derivatives on the in vitro differentiation of hippocampus neurons-morphometric and electron microscopic studies of ribosomes].

1. By means of electron microscopic and quantitative methods at the explantate cultures of the fetal hippocampus in vitro the influence of the orotic acid, sodium orotate and methylglucamine orotate on the neurogenesis was investigated. 2. After three days of the in vitro cultivation the neuroblasts influenced by these drugs show a smaller respectively not different degree of differentiation compared to the controls. 3. Under the influence of the orotic acid and of its derivatives the neurogenesis is significantly stimulated. The drugs produce a significant increase of the membrane-bound ribosomes and polysomes. The total number of ribosomes increases following the application of orotic acid by 20%, of sodium-orotate by 48% and of methylglucamine-orotate by 23% compared to the controls (alpha = 0,1%). 4. Sodium-orotate shows with reference to the neuronal development the clearest stimulatory effect. After 20 days in vitro the total number of ribosomes is by 60% higher at the treated cultures than in the controls. 5. The results might suggest, that an enlarged supply of the pyrimidine nucleotide via a raising of the RNA- and the protein synthesis might stimulate the development of the neuroblasts even under the in vitro conditions.

[Effect of Solcoseryl on nerve tissue under in vitro conditions].

Explants from trigeminal ganglia and skin of chick embryos and hippocampus from fetal rats were cultivated in Maximow assembly in the presence of Solcoseryl (Solco AG, Basel), a blood extract of calf. Solcoseryl in vitro did not influence the regeneration of nerve fibers from CNS explants. A stimulatory effect of Solcoseryl in vitro by 1% concentration on the outgrowth of new processes in explants of PNS was demonstrated. It is discussed: under optimal concentration Solcoseryl may be important for the influence of the composition of the medium in which explants of the nerve system and skin are cultivated.

[Effect of orotic acid on the in vitro cultured nerve tissue].

Cell- and explantcultures of the hippocampus of 18 days old fetal rats and explantcultures of the ganglion trigeminal of 9 days old chick embryos were cultivated in Maximow-chambers. Orotic acid, Sodiumorotate and methylglucaminorotate by concentrations of 10(-3)...10(-7) g/ml medium were added to the cultures. By concentrations of 10(-7) g/ml medium in the CNS-cultures and 10(-6) g/ml medium in the PNS-cultures is stimulated the nerve fiber growth and is increased the nerve fiber index. After three weeks in vitro is a higher number of neurons in the cultures compared to the controls.

Action of ethanol and other solvents on the nerve tissue under in vitro conditions.

Explants and cells of nervous tissue were cultivated in the presence of aethanol, tween 80, dimethylformamid (DMF) and dimethylsulfoxid (DMSO) and the influence upon the index of area, the growth rate and fiber index was observed. They are important to solutions of drugs for tests in vitro. At the beginning of the cultivation aethanol in vitro influenced the regeneration of nerve fibers from explants and cells. A significant increase of the index of growth was observed. After a long term influence of tween 80, DMF and DMSO an inhibition of differentiation of neurons in vitro was observed.

[Effects of brain extract and hydrolysate on nerve tissue in vitro].

Explants of PNS and CNS of chick embryos or fetal rats were cultivated and brainhydrolysat cerebrolysin or a synthetic aminoacidsolution or a brain extrakt was applied to the cultures. Cerebrolysin exercised in vitro an effect, which is related to charge only trifling. The cerebrolysin effect is essential related to the concentration/medium. High concentration of cerebrolysin decreases the growth of nerve fibers. Migration of non-neuronal cells increases. A stimulation of nerve fibers growth resembling to NGF was not seen. The density of cells or number of cells/volume unit, rate of mitosis and phases of mitotic non-neuronal cells is changed by cerebrolysin. The effect of cerebrolysin in vitro is compared to the tested aminoacidsolution and brainextract resembling. NGF may be important selectiv for stimulation of the activity of sympathetic and sensoric ganglia. Brainextract and brainhydrolysat and aminoacidsolution influenced in vitro PNS- and CNS-tissue cultures.

[Morphological study of hippocampal development in cell and explant cultures].

Cell-and explantatcultures from hippocampal region of embryonal rats and newborn rats were cultivated up to 4 weeks. Pyramidal neurons, multiformed neurons and granule cells were observed in the explantatcultures. In the cellcultures were observed small and big Neurons after 4 weeks in vitro. Mantenance and differentiation of cellcultures are possible only by showing in at least 60 000 cells/ml. The Morphology of neurons in vitro was related to in vivo.

[Cell differentiation in the hippocampus of the fetal rat in cell culture].

Cell cultures from hippocampus of 16 and 17 days old embryonal rats were cultivated up to 4 weeks. After 24 hours in vitro on 18.4 percent of cells and after 5 days in vitro on 70 percent of cells processes could be recognized. These are neuroblasts. The cells reaggregated. Nerve fibers after 4 weeks in vitro are 200 to 300 mum long. Small and big neurons with 12 mum to 26 mum diameters of perikarya, bi- and multipolar neurons after 4 weeks in vitro were observed. In cultures and meningothel-monolayer developed. Maintenance and differentiation of cultures are possible only by sowing in at least 60,000 cells/ml medium. The advantage of cell culture opposite to organ culture exists in experiments with immediate selective influence.
