Simultaneous production of IFN-gamma, IFN-alpha/beta and nitric oxide in peritoneal macrophages from TDM-treated mice.

Peritoneal macrophages (PM) were isolated from mice treated with Dimycolate of Trehalose (TDM), a glycolipid extracted from the cell wall of Mycobacterium tuberculosis. PM from TDM-treated mice (TDM-PM) were shown to secrete consistent amount of IFN-gamma, which was not detectable in control Resident-PM (Res-PM), as revealed by ELISA. In addition, biologically active IFN was detected in the supernatants of TDM-PM, whereas no IFN production was found in those of control Res-PM. The addition of specific antisera to PM cultures revealed the simultaneous production of both type I and II IFNs in TDM-PM cultures. No reciprocal regulation in the production of IFN-gamma and IFN-alpha/beta was found in these cultures. In parallel, nitric oxide (NO) production was measured in TDM-PM cultures by detecting nitrites (NO2-). TDM-PM cultures accumulated high amounts of NO2- which decreased to the level of Res-PM in the presence of NMMA, an inhibitor of NO-synthases. In vitro, neither type I nor type II IFNs were involved in the stimulation of NO production. The capacity of macrophages to simultaneously secrete IFN-gamma, IFN-alpha/beta and NO upon in vivo TDM-treatment could be of particular relevance for the defense process of innate immunity in which macrophages play a crucial role.

Arachidonate metabolism triggered in primed macrophages by signals inducing antitumor activity.

The acquisition of antitumoral functions by mouse peritoneal macrophages is controlled by the addition of activating agents (lipopolysaccharide (LPS), muramyldipeptide (MDP) or A23187), on appropriately primed macrophages. The release of eicosanoids during this activation step was examined by radio-HPLC. We demonstrated that the induction of antitumor activity in primed macrophages by LPS or MDP was associated with the release of 20:4 derivatives; arachidonic acid was metabolized predominantly via the cyclooxygenase pathway to PGE2 and thromboxane. The production of PGE2, quantified by an enzyme immunoassay, was sustained and important (up to 20 ng/ml/h/10(6) macrophages). However, PGE2 and thromboxane did not seem essential to the activation process: induction of antitumor activity took place and was even enhanced in the presence of indomethacin, whereas it was decreased by exogenous PGE2. During culture in vitro, primed macrophages released spontaneously significant amounts of 20:4 metabolites and became unresponsive to activation stimuli. Again indomethacin had a positive effect: it protected primed macrophages against this loss of activability. Cyclooxygenase metabolites released in response to activating stimuli or spontaneously seem to trigger deactivation pathways.

[Acceptability of vaccination against influenza].

From 1984 to 1986 the general practitioners of Braine-le-Château (Belgium) have studied the frequency of influenza vaccination in people aged more than 65 years and its results. Without any particular promotion, 17% of patients were vaccinated in 1984. Following an information campaign conducted by general practitioners, the figure was 39% in 1985. The next year the information was completed by posters, editorials in newspapers and lectures organized for groups of retreated people. The number of vaccinated patients reached 41%. This study demonstrates that it is possible in a short time to improve the vaccination protection of elderly people through the action of practitioners convinced of efficacity of the influenza vaccine.
