The yeast genus Trichosporon spec. LS3;molecular characterization of genomic complexity.

The genomic structure of an industrial yeast strain Trichosporon spec. LS3 was compared with Trichosporon adeninovorans type strain CBS 8244. The cot values, the portion of single copy sequences (CBS 8244: 10.9 x 10(9) D, LS3: 10.6 x 10(9) D) as well as of repetitive sequences (CBS 8244: 6.0 x 10(9) D, LS3: 5.5 x 10(9) D) per haploid genome and genome complexity of these strains (CBS 2844: 16.9 x 10(9) D, LS3: 16.1 x 10(9) D) have been analysed. Both strains show a high genome complexity. The mitochondrial DNA content was measured and compared. No plasmidal DNA was identified for both strains. A survey of the data of genomic DNA made it possible to postulate for each of both strains a haploid set of chromosomes. The intraspecific reassoziation values of the nuclear DNA from the strain LS3 and CBS 8244 are interpreted to confirm classification of these strains based on physiological and genetical properties into one species Trichosporon adeninovorans.

Heterologous gene expression of the glyphosate resistance marker and its application in yeast transformation.

The E. coli aroA gene was inserted between yeast promoter and terminator sequences in different shuttle expression plasmids and found to confer enhanced EPSP synthase activity as well as resistance to glyphosate toxicity. Subsequently, a transformation system using these newly constructed vectors in yeast was characterized. The efficiency of the glyphosate resistance marker for transformation and selection with plasmid pHR6/20-1 in S. cerevisiae laboratory strain SHY2 was found to be relatively high when compared with selection for LEU2 prototrophy. The fate of the recombinant plasmid pHR6/20-1 in the transformants, the preservation of the aroA E. coli DNA fragment in yeast, mitotic stability, EPSP synthase activity, and growth on glyphosate-containing medium have been investigated. As this plasmid also allows direct selection for glyphosate resistant transformants on rich media, the glyphosate resistance marker was used for transforming both S. cerevisiae laboratory strain SHY2 and brewer's yeast strains S. cerevisiae var. "uvarum" BHS5 and BHS2. In all cases, the vector pHR6/20-1 was maintained as an autonomously replicating plasmid. The resistance marker is, therefore, suitable for transforming genetically unlabelled S. cerevisiae laboratory, wild, and industrial yeast strains.

Identification of a lys2 mutant of Candida maltosa by means of transformation.

We have isolated five mutants of Candida maltos, which lack the 2-aminoadipate reductase activity, an enzyme involved in the lysine biosynthesis. By means of complementation analysis using protoplast fusion, the isolated mutants were divided into two complementation groups. Thereof the C. maltosa strain G457 could be transformed by the plasmids pDP12 and pDP13, which contain the LYS2-coding gene of Saccharomyces cerevisiae. On the basis of our presented results obtained by studies on hybridization, stability, and recovery of plasmids from C. maltosa transformants, we suggest that transformation does proceed integratively.

Synthesis and cloning of a human beta-casomorphin-5 gene.

As so far a singularity among opioid peptides, bovine beta-casomorphin-5 has been shown to display pronounced cardiatic activities. The gene sequence of a human beta-casomorphin-5, including plasmid insertion and fusion protein expression and cleavage instrumentary, has been synthesized by a manual phosphoramidite procedure and cloned in a bacterial system.

Physical mapping and genome organization of mitochondrial DNA from Candida maltosa.

Mitochondrial (mt) DNA of the ascomycetous yeast Candida maltosa was isolated and characterized. The mtDNA is circular and the size estimated from restriction analysis performed with 7 endonucleases was 52 kb pairs. A restriction map was constructed, using the cleavage data of four endonucleases. Using mt genes from Saccharomyces cerevisiae, six structural genes (large rRNA, apocytochrome b, cytochrome c oxidase subunit I and subunit II, ATPase subunit 6 and subunit 9) were located on the C. maltosa chondriome by cross hybridization experiments. The comparison between the mt genomes of C. maltosa and six other yeasts showed differences in the overall genome organization.

Transformation of Candida maltosa and Pichia guilliermondii by a plasmid containing Saccharomyces cerevisiae ARG4 DNA.

Saccharomyces cerevisiae, Candida maltosa and Pichia guilliermondii have been transformed by the plasmid pYe(ARG4)411, which contains the S. cerevisiae ARG4 gene inserted into pBR322. In all transformants argininosuccinate lyase as well as beta-lactamase were detected. The ARG+ phenotype of transformants is mitotically unstable. Closed circular pYe(ARG4)411 DNA was detected in transformant DNA preparations by hybridization to pBR322 DNA and by transformation of E. coli to ampicillin resistance.

[Characteristics of mitochondrial DNA from Candida spec. EH15].

The restriction endonucleases EcoRI, PstI and BglII were used to digest the mitochondrial DNA from Candida spec. EH 15. Molecular masses of single digested DNA fragments were calculated. The mitochondrial DNA size (28-30 x 10(6) D) comprises 2/3 of the mitochondrial DNA from Saccharomyces cerevisiae.

[Direct sonographic detection of tubal pregnancy following a fertilization operation].

In a case of fallopian pregnancy after a microsurgical performance for fertilisation we obtained the diagnosis by means of ultrasound. We saw the intact gestational sac and the embryo directly and point out the importance of early sonographical screening of pregnancy after surgical treatment of infertility. Diagnosis and differential diagnosis by ultrasound are discussed.

[Purification and isolation of the arom aggregates of Schizosaccharomyces pombe].

The five enzymes that catalyzing steps two through six in the prechorismate polyaromatic amino acid biosynthetic pathway are physically associated and have been purified up to 400-fold from Schizosaccharomyces pombe. The native arom aggregate has a molecular weight of approx. 140,000-145,000 based on gel filtration, glycerol-density-gradient centrifugation, and polyacrylamide-gel electrophoresis in the presence of sodium dodecyl sulphate. Similarities between the S. pombe arom aggregate and that of Neurospora crassa and Euglena gracilis are discussed.

[Prenatal diagnosis of an intestinal atresia by means of ultrasound - case report and discussion (author's transl)].

An intestinal atresia was diagnosed by means of ultrasonography and amniofetography in the 34. gestional week. The discussion agendas are, how far the given ultrasonogramms are typical for fetal intestinal atresia and how much part was played by sonography and amniofetography by the search of diagnosis; further, whether already antenatal diagnosed intestinal abnormality justifies a fetal indicated cesarean section, which could be important for timing of the surgical performance and for the betterment of the perinatal situation.
