[Cytological assessment of buccal mucous membrane in newborns (author's transl)].

Karyopycnotic and keratinisation indices were determined in buccal smears which had been taken from newborns. An assessment was made of 239 smears which had been obtained from newborns in their first eight weeks of age, with the probands being subdivided into two groups, depending on breast feeding and feeding on powdered milk (Ki-Na).--Both indices were extremely high on the first day after birth, but they declined up to the eighth or 14th days of age which was attributed to postnatal reduction of high foetal oestrogen levels. Both indices then went up, with some difference being recordable between the two groups, depending on the modes of feeding. The keratinisation index of the breast-feeding group was much higher than that of the Ki-Na group, on account of very high oestrogen levels in breast milk, between the fourth and seventh days from delivery, with consequences on mucous membrane proliferation. The keratinisation index declined gradually along with oestrogen drop of breast milk and turned to be equal to that of the Ki-Na group, eight weeks after delivery.--Proliferation of buccal mucous membrane and its importance to efficient defence against infection during the first four weeks of age are discussed.

In vivo hamster skeletal muscle preparation: morphine sensitive - naloxone insensitive.

The hamster cheek pouch retractor muscle preparation has been evaluated as an assay model for morphine and the opiate antagonist, naloxone. The preparation is in vivo and stimulation of the muscle is reflex. The higher the concentration of morphine injected into the pouch the greater was the average percent reduction in contractile force. Unilateral injections of morphine did not affect the contralateral muscle and so action of the drug was judged to be peripheral and local. The results with naloxone suggested that the opiate receptors in this preparation were relatively insensitive to the drug. The model has potential as an in vivo assay system for testing opiates, opiate agonists and antagonists. It may also prove to be valuable in studying tolerance to opiates and the withdrawal syndrome, as well as serving as a model for pain modulation studies.

Lysozyme (muramidase) and hemolytic complement (CH50U) in sera of beagle dogs and minipigs after longterm application of different preparations of estrogens and gestagens.

In studies for the chronic toxicity of estrogens and gestagens, lysozyme and hemolytic complement (CH50U) in sera of beagle dogs and minipigs were measured. Various doses of the sexsteroids were given: 0.01 mg, 0.1 mg and 1.0 mg/kg respectively. The influence of the steroids on the tested parameters after an application for 6 months was not significant, only in a single series with unphysiologically, high dosages of the preparations were some values changed significantly.

Transcortin as an indicator of estrogenic potency in oral contraceptives.

Serum cortisol-binding globulin (CBG) was measured before, during and following a 21-day application period of 5 oral contraceptives used in the GDR. In a second group of patients determinations were carried out at the end of a 24-month treatment period and the end of the first post-treatment cycle. From a third group of volunteers blood samples were taken at the end of the first, second and third treatment cycles. All the drugs tested produced a marked increase of CBG in serum, reaching its maximum at the end of the second treatment cycle. Within four weeks following termination of application the level fell to the control range. There were no significant differences between the various contraceptives.

[Effects of hormonal contraceptives on parameters of lipid metabolism (author's transl)].

Constant rise of triglycerides and low but regular increase of phospholipid concentrations in serum were observed as a consequence of intake of single-phase hormonal contraceptives. This has been paralleled by inconstant, moderate rise in cholesterol. Those effects are likely to demonstrate dependence on oestrogen amounts of the preparations in question as well as on both quality and quantity of the progestagenic component. Rising concentrations of low density lipoproteins were recordable with regularity. However, variegated results so far have been obtained from studies into the behaviour of high-density lipoproteins. -The mechanism of origin of those alterations, which usually did not surpass normal limits, and their relevance to clinical practice are discussed.

[Comparative studies into effects of synthetic oestrogens and gestagens on certain serum protein fractions under standardised conditions (author's transl)].

Initial effects of the following hormonal contraceptives on certain serum proteins in 29 women were checked under standardised conditions through the first three cycles on the pill: Deposiston (ethinyloestradiol-sulphonate/norethisterone-acetate), Gravistat (ethinyloestradiol/norgestrel), Non-Ovlon (ethinyloestradiol/norethisterone-actate).--Effects were found to differ strongly by fractions. Differences were recorded also from action over time. Changes and deviations were confirmed to depend on oestrogen levels.

[Reversibility of alterations induced by sexual steroids in various serum protein fractions, following application of hormonal contraceptives (author's transl)].

Reversibility of changes induced by sexual steroids was studied in 19 different serum protein fractions of 20 women. The following preparations were available for testing: Gravistat (ethinyl-oestradiol/norgestrel), Non-Ovlon (ethinyloestradiol/norethisterone-acetate), Ovosiston (mestranol/chlormadinone-acetate), and Deposiston (ethinyloestradiol-sulphonate/norethisterone-acetate). The tests were made towards the end of the 24th cycle on the pill and in the first cycle thereafter.--The proteins tested were found to be affected in a differentiated way and, throughout, depending on oestrogen levels. Reversal was rapid, but the phase of restitution usually was longer than the period of testing and follow-up. The long-lasting action of Deposiston was visualised also in its "reversal effect".

Effects of an intranasally administered low dose progestogen on the pituitary and gonadal functions.

This study was undertaken to investigate possible mechanisms of action of a daily intranasally administered low dose of norethisterone acetate (40 microgram) in regularly menstruating young women. Besides measurement of basal body temperature, serum concentrations of FSH, LH, estradiol and progesterone were determined over one treatment cycle. An endometrial biopsy was taken on day 27 or 28 of the cycle. In spite of a considerable individual variation in the respective values the following effects could be observed: In 8 of 9 subjects the midcycle LH peak was abolished, while basal FSH and LH levels remained within the control range of the follicular and luteal phases. A variable reduction of progesterone levels below the range indicative of a normal luteal phase function suggested a tendency to atypical corpus luteum formation. In 8 of 9 endometrial biopsies, however, secretory transformation could be found.

[Experience with the anti-androgen cyproterone acetate in the management of hirsutism].

Use of the antiandrogen cyproterone acetate in the treatment of hirsutism.--25 women with hirsutism or hypertrichosis faciei were subject to a "reverse sequential therapy" with cyproterone acetate (Hammerstein's regimen) for 1 to 3 years. In 8 patients, a limited, in 14 patients a satisfactory and in one woman a good result could be recorded. Cycle irregularities and subjective side effects were within a tolerable range. Studies of the hair pattern in 15 patients revealed a higher telogen rate as well as increased diameters of the hair stalks, compared with controls, in the temporal region of the head. These alterations disappeared in the course of the treatment. In the parietal region of the head no significant differences of the hair pattern were found in comparison with the controls and during application of the steroids.

[The effect of hormones on the periodontal condition--clinical studies on 300 female patients].

In this study 300 women (100 pregnant women, 100 women taking contraceptives and 100 women without contraceptive medication) were examined, recording the periodontal condition (using a computer-orientated chart) and the periodontal index according to Kötzschke. The values for the total index and the superficial and deep components of the index according to Kötzschke, the pocket depth and the degree of tooth mobility (determined for each tooth) were significantly higher in women under contraceptive medication than in the control subjects.

[Determination of renal function during pregnancy via renal sequence scintigraphy (author's transl)].

Following a detailed description of the technique of examination and evaluation used in nuclear medicine, the article reports on first results obtained with renal sequence scintigraphy in pregnant women with normal kidneys. Attention is drawn to the diagnostic possibilities offered by this method, basing on a discussion with reference to the published literature, with particular emphasis on pregnancy and on the monitoring of risk pregnancies.

[Examination of hormonal contraceptives by enzyme induction (author's transl)].

The influence of drug stimulated biotransformation on the biological effectiveness of two different contraceptives (Deposition and Gravistat) was observed.--Phenobarbital, a potent enzyme inducer, is known to increase the metabolism of steroid hormones, including estrogens and progrestogens. -- The clinical symptoms of drug mediated enzyme induction of hormonal steroid contraceptives are dysfunctional bleedings. This phenomenon is discussed.
