kdgREcc negatively regulates genes for pectinases, cellulase, protease, HarpinEcc, and a global RNA regulator in Erwinia carotovora subsp. carotovora.

Erwinia carotovora subsp. carotovora produces extracellular pectate lyase (Pel), polygalacturonase (Peh), cellulase (Cel), and protease (Prt). The concerted actions of these enzymes largely determine the virulence of this plant-pathogenic bacterium. E. carotovora subsp. carotovora also produces HarpinEcc, the elicitor of the hypersensitive reaction. We document here that KdgREcc (Kdg, 2-keto-3-deoxygluconate; KdgR, general repressor of genes involved in pectin and galacturonate catabolism), a homolog of the E. chrysanthemi repressor, KdgREch and the Escherichia coli repressor, KdgREco, negatively controls not only the pectinases, Pel and Peh, but also Cel, Prt, and HarpinEcc production in E. carotovora subsp. carotovora. The levels of pel-1, peh-1, celV, and hrpNEcc transcripts are markedly affected by KdgREcc. The KdgREcc- mutant is more virulent than the KdgREcc+ parent. Thus, our data for the first time establish a global regulatory role for KdgREcc in E. carotovora subsp. carotovora. Another novel observation is the negative effect of KdgREcc on the transcription of rsmB (previously aepH), which specifies an RNA regulator controlling exoenzyme and HarpinEcc production. The levels of rsmB RNA are higher in the KdgREcc- mutant than in the KdgREcc+ parent. Moreover, by DNase I protection assays we determined that purified KdgREcc protected three 25-bp regions within the transcriptional unit of rsmB. Alignment of the protected sequences revealed the 21-mer consensus sequence of the KdgREcc-binding site as 5'-G/AA/TA/TGAAA[N6]TTTCAG/TG/TA-3'. Two such KdgREcc-binding sites occur in rsmB DNA in a close proximity to each other within nucleotides +79 and +139 and the third KdgREcc-binding site within nucleotides +207 and +231. Analysis of lacZ transcriptional fusions shows that the KdgR-binding sites negatively affect the expression of rsmB. KdgREcc also binds the operator DNAs of pel-1 and peh-1 genes and represses expression of a pel1-lacZ and a peh1-lacZ transcriptional fusions. We conclude that KdgREcc affects extracellular enzyme production by two ways: (i) directly, by inhibiting the transcription of exoenzyme genes; and (ii) indirectly, by preventing the production of a global RNA regulator. Our findings support the idea that KdgREcc affects transcription by promoter occlusion, i.e., preventing the initiation of transcription, and by a roadblock mechanism, i.e., by affecting the elongation of transcription.

[Limited pleuro-pneumonectomy for bronchogenic carcinoma associated with cancerous pleural effusion].

OBJECTIVE: To evaluate the effects of limited pleuro-pneumonectomy for bronchogenic carcinoma associated with cancerous pleural effusion. METHODS: From January. 1994 to December 1997 we performed limited pulmonary lobectomy wedge resection of lung, resection of the metastases of the parietal pleura, visceral pleura, diaphragmatic pleura and pericardiac pleura, and electro incision of the micormetastases of the above position as well as cleaning the mediastinal lymph nodes in 14 cases. RESULTS: The 14 cases were followed-up to December 1998. Eight patients of them died, the other 6 patients survived with a survival period of 7 - 30 months (median survival period 16.5 months). No pleural effusions recurrence was seen. The longest survival period was 30 months. There were no operative mortality and no severe postoperative complication. Chest distress, dyspnea and pain of chest or abdomen were relieved. CONCLUSIONS: This simple method has such advantages as, less traumatic, rapid rehabilitation and effective control of pleural effusion. The postoperative survival period is longer than that of palliative operation and medical therapy. The pain caused by parietal pleural metastasis can be relieved.

[Clinical and immunological studies on transplantation of total cornea with circular lamellar sclera].

OBJECTIVE: To observe the clinical results of total cornea with circular lamellar sclera transplantation (CST) and study its immunological mechanisms by animal experiments. METHODS: 109 patients with corneal transplantation were retrospectively analyzed, and according to the methods of operation they were divided into groups to compare their rate of rejection and the time of occurrence of corneal neovascularization. The methods of grouping and operation in animal experiments were similar to that in the clinical practice. RESULTS: The time of occurrence of neovascularization and rejection of CST were later than that in the total corneal transplantation (TCT), and the postoperative rate of rejection at 3 to 6 months was obviously increased in comparison with that in the partial penetrating keratoplasty (PKP). CONCLUSION: CST is an important method for rebuilding ocular anterior segment, and the donor limbus is active in the immunological of rejection.

[Effect of decompression or transplantation of the facial nerve on serious advanced facial paralysis].

OBJECTIVE: To investigate operative effect on serious advanced facial paralysis. METHOD: 28 cases (28 sides) with serious facial paralysis underwent decompression or transplantation of facial nerve. RESULT: After following up 0.5-2 years postoperatively, the total, most or partial and no recovery were 14 cases (50%), 12 cases (42%) and 2 cases (7.1%) respectively. CONCLUSION: The operation performed as early as possible can be an effective procedure for correcting serious advanced facial paralysis if conservative treatment could be confirmed to be helpless.

Sequence analysis of the MSP 1 gene of Plasmodium falciparum isolates from Hainan, China.

AIM: To obtain the complete sequence and analyze the diversity of the MSP 1 molecule from the Chinese isolates of Plasmodium falciparum. METHODS: Genomic DNA was prepared directly from blood samples spotted on filter papers from 2 malaria patients from Baoting County, Hainan Province. PCR amplification of the target gene was carried out using 5 pairs of oligonucleotides specific for the MSP 1 gene. Direct sequencing of the target gene fragments was performed using ABI PRISM Dye Terminator Cycle Sequencing Ready Reaction Kit (Perkin Elmer) in a automatic ABI PRISM 310 Genetic Analyzer. RESULTS: For the first time, two complete sequences of the MSP1 gene from two Chinese isolates of Plasmodium falciparum were obtained. A comparison with the previously reported sequences identified them as members of the MAD20 allelic family. The deduced amino acid sequences of the MSP1 from this two Chinese isolates were identical with each other except for Blocks 2, 4 and 8. CONCLUSION: The sequences of the MSP 1 from two Chinese isolates of P. falciparum belong to the MAD20 allelic family. Minor variations through the whole sequences exist compared with the MAD20 sequence. The results provide the first evidence of the diversity of the MSP 1 molecule from Chinese isolates of Plasmodium falciparum.

[Spectrophotometric determination of trace amount of cadmium in marine products].

The colour reaction of Cd(II) on xylend orange in the presence of hexamethylenetetramine-KNO3-HNO3 has been studied. Cd(II) forms the complex at pH = 6.2-6.4 (lambda(max) = 578.4 nm) with an apparent molar absorptivity of 1.6 x 10(4) L x mol(-1) x cm(-1). Beer's law is obeyed for Cd(II) in the range of 0.5-6 microg x mL(-1). The detection limit is 0.007 microg x mL(-1) with the recovery of 90%-105%. The method is simple and highly sensitive, and has been applied to the analysis of sample with satisfactory results.

[Determination of inorganic and organic tin in water samples by graphite furnace atomic absorption spectrometry].

This paper proposed a method for the determination of inorganic tin and organotin species. Total tin contents can be directly detected by zirconium-coated graphite furnace atomic absorption spectrometry,while the organotin was detected by hydride generation combined with solvent extraction and the difference between the total tin and organotin was inorganic tin. The detection limits were 58 pg and 27 pg for organotin and inorganic tin,respectively. The relative standard deviations of this procedure, were 2.13% and 2.04%, respectively. Several water samples were analyzed by this method. The recoveries ranging from 90.5% and 98.5% were obtained.

[Study on magnesium ionization in cathodic sputtering glow discharge plasma].

A method, based on the Doppler broadening-dependent absorption width and the ionization degree measured by the ratio of relative atomic and ionic absorbance in cathodic sputtering glow discharge (CSGD) plasma with aluminum-magnesium alloys as cathodes, is proposed. The experimental reveals that the plentiful magnesium ions in the plasma under the conventional discharge conditions of the atomizer do influence the analytic working curves. Corrected method is derived and factors which result in magnesium ionization degree are disscused.

Melanopsin: An opsin in melanophores, brain, and eye.

We have identified an opsin, melanopsin, in photosensitive dermal melanophores of Xenopus laevis. Its deduced amino acid sequence shares greatest homology with cephalopod opsins. The predicted secondary structure of melanopsin indicates the presence of a long cytoplasmic tail with multiple putative phosphorylation sites, suggesting that this opsin's function may be finely regulated. Melanopsin mRNA is expressed in hypothalamic sites thought to contain deep brain photoreceptors and in the iris, a structure known to be directly photosensitive in amphibians. Melanopsin message is also localized in retinal cells residing in the outermost lamina of the inner nuclear layer where horizontal cells are typically found. Its expression in retinal and nonretinal tissues suggests a role in vision and nonvisual photoreceptive tasks, such as photic control of skin pigmentation, pupillary aperture, and circadian and photoperiodic physiology.

Coating of hydroxyapatite on highly porous Al2O3 substrate for bone substitutes.

A thin film of hydroxyapatite (HA) was successfully coated onto a highly porous substrate of alumina by using a novel solution technique. The coated HA bonded strongly to the substrate through a glass sintering aid with an intermediate thermal expansion coefficient. The coating was also found to be microstructurally and chemically uniform on the surfaces of the interconnected pores in the matrix of the substrate. Mechanical property testing results showed that the strength of the HA-coated composite was significantly improved, indicating a high potential for structural bone substitutes in hard tissue prosthetics.

The observation of transplanted embryonic motoneurons in the denervated muscles of adult rats.

OBJECTIVE: To observe the survival of embryonic motoneurons after they were transplanted into the denervated skeletal muscles and to find a new method to retard the atrophy of denervated muscles. METHODS: Dissociated embryonic motoneurons prelabled with 5-bromo-2'-deoxyuridine (Brdur) on the embryonic days 12 were injected into the denervated gastrocnemius muscles of adult rats. Then gastrocnemius muscles were processed with Nissl staining, acetylcholinesterase staining and Brdur immunocytochemical staining to show the implanted motoneurons at 9 and 22 weeks post-transplantation. Myofibrillar ATPase staining was used to show the morphology of muscle fibers. The rats in experimental group were implanted with embryonic motoneurons in the predenervation muscles, while the rats in control group were injected with just culture medium without motoneurons. RESULTS: Embryonic motoneurons survived, developed and extended long axons to form neuromuscular junctions with the denervated muscles. The differentiation of muscle fibers and fiber type grouping occurred among bigger fibers in experimental group. The transverse area was smaller and there was no apparent fiber type grouping in control group. CONCLUSIONS: Embryonic motoneurons can survive, develop and reinnervate denervated muscles after being transplanted into denervated muscles. It is worth further investigating on ameliorating the atrophy of denervated muscle.

[The effects of clenbuterol on intramuscular collagen metabolism in denervated muscle].

In order to explore the effects of clenbuterol on intramuscular collagen metabolism in denervated skeletal muscles, a randomized, double-masked and placebo-controlled group were studied. Seventy-one patients with complete function loss in muscularcutaneous nerve resulted from brachial plexus injury were administered clenbuterol or placebo 60 micrograms Bid for more than 3 months. Biopsies of the biceps brachia muscle were performed at the beginning and end of this study. The biopsied muscles were processed with anti-collagen I and IV immunohistochemical stains and image analysis as well. The result showed that the collagen proliferation of both type I and IV was much reducible in the clenbuterol-treated group than that of the placebo-treated group (P < 0.05). It was concluded that clenbuterol could inhibit partially the proliferation of intramuscular collagens in denervated skeletal muscle.

[Embryonic motoneurons transplanted into denervated skeletal muscles].

OBJECTIVE: To observe the living situation of homografting embryonic motoneurons in denervated skeletal muscles. METHODS: We transplanted dissociated ventral spinal cord cells from 12 embryonic days rats prelabeled with 5-bromo-2-deoxyuridine(Brdur) into denervated skeletal muscles of adult rats. The denervated skeletal muscles were collected at 9 and 22 weeks respectively, and Nissl, acetylcholinesterases immunocytochemistry and ATPase staining were used to show the transplanted embryonic motoneurons and the morphography of muscle fibers. RESULTS: Transplanted embryonic motoneurons could survive, extend out long axons, and form neuromuscular junctions with muscle fibers. Type grouping fibers with bigger transverse area were also observed in the experimental group. Muscle fibers were much smaller and did not form fiber type grouping in the control group. CONCLUSION: Homografting embryonic motoneurons could survive well and reinnervate the denervated skeletal muscles.

[Isotope tracer combined with trichloroacetic acid precipitation method for determination of nerve growth factor concentration in mouse plasma].

OBJECTIVE: To establish the radioisotope (125I) tracer method combined with trichloroacetic acid (TCA) precipitation for the determination of nerve growth factor (NGF) in mouse plasma. METHODS: Radioisotope tracer labelling method combined with TCA precipitation was used to determine NGF in mouse plasma. RESULTS: The detection limit of the method for NGF in plasma was 2.0 ng/ml. The intra-assay cv (CV) and the inter-assay CV were all less than 10%. The assay linearity was determined over the range of 12.5-400 ng/ml in plasma and the correlation coefficient was 0.9990. The average recovery of the method for NGF was 95.21%. CONCLUSIONS: The radioisotope tracer method combined with TCA precipitation is stable, sensitive and reliable. It is suitable for the determination of NGF concentration in mouse plasma.

[Observation of clinical effect of acupuncture on vascular dementia and its influence on superoxide dismutase, lipid peroxide and nitric oxide].

OBJECTIVE: To observe the clinical effect of electroacupuncture and acupuncture on vascular dementia (VaD) and the influence on superoxide dismutase (SOD), lipid peroxide (LPO) and nitric oxide (NO) levels. METHODS: Forty-six cases of VaD were randomly divided into two groups, the electroacupuncture group (EA group) and the acupuncture group (AP group). Assessment of Hasgawa's dementia scale (HDS), functional activities questionnaire (FAQ) and neurologic deficit scoring were done before and after treatment, and the changes on SOD, LPO and NO levels were observed. RESULTS: After treatment, in the EA group, the HDS elevated, the FAQ lowered, SOD increased and LPO and NO decreased significantly, as compared with before treatment, P < 0.01. But these parameters were not changed significantly in the AP group after treatment. The clinical symptoms were improved in both groups, but the reduction on neurologic deficit score was not significant. The total effective rate of the EA group was higher than that of the AP group. CONCLUSION: The immediate effect of electroacupuncture was superior to that of acupuncture, suggesting that the electroacupuncture was more effective in promoting the intelligence recovery than acupuncture.
