Angiogenin activates human umbilical artery smooth muscle cells.

Angiogenin stimulates proliferation of human umbilical artery smooth muscle cells. This activity of angiogenin depends on the cell density and requires nuclear translocation of the ligand as well as activation of SAPK/JNK MAP kinase. Angiogenin binds to a 170-kDa putative receptor on the cell surface and induces phosphorylation of SAPK/JNK. It also undergoes nuclear translocation in a time and concentration dependent manner. Neomycin inhibits nuclear translocation of angiogenin and abolishes angiogenin-induced cell proliferation but does not inhibit SAPK/JNK phosphorylation. The data demonstrate that smooth muscle cells are targets for angiogenin and that both SAPK/JNK phosphorylation and nuclear translocation of the ligand are required for angiogenin to activate smooth muscle cells.

The scaffolding protein CASK mediates the interaction between rabphilin3a and beta-neurexins.

CASK, a member of the membrane-associated guanylate kinase (MAGUK) superfamily, binds to the carboxyl-terminus of beta-neurexins on the intracellular side of the presynaptic membrane. The guanylate kinase-like (GUK) domains of MAGUKs lack kinase activities, but might be important for mediating specific protein-protein interaction. By a yeast two-hybrid approach, we identified an interaction between the GUK domain of CASK and the C2B domain of rabphilin3a, a presynaptic protein involved in synaptic vesicle exocytosis. The interaction was confirmed by in vitro GST pull-down and co-immunoprecipitation assays. It was proposed that presynaptic vesicles might be guided to the vicinity of points of exocytosis defined by beta-neurexins via the interaction between rabphilin3a-CASK-beta-neurexins.

Phosphorylation of RGS9-1 by an endogenous protein kinase in rod outer segments.

Inactivation of the visual G protein transducin, during recovery from photoexcitation, is regulated by RGS9-1, a GTPase-accelerating protein of the ubiquitous RGS protein family. Incubation of dark-adapted bovine rod outer segments with [gamma-(32)P]ATP led to RGS9-1 phosphorylation by an endogenous kinase in rod outer segment membranes, with an average stoichiometry of 0.2-0.45 mol of phosphates/mol of RGS9-1. Mass spectrometry revealed a single major site of phosphorylation, Ser(475). The kinase responsible catalyzed robust phosphorylation of recombinant RGS9-1 and not of an S475A mutant. A synthetic peptide corresponding to the region surrounding Ser(475) was also phosphorylated, and a similar peptide with the S475A substitution inhibited RGS9-1 phosphorylation. The RGS9-1 kinase is a peripheral membrane protein that co-purifies with rhodopsin in sucrose gradients and can be extracted in buffers of high ionic strength. It is not inhibited or activated significantly by a panel of inhibitors or activators of protein kinase A, protein kinase G, rhodopsin kinase, CaM kinase II, casein kinase II, or cyclin-dependent kinase 5, at concentrations 50 or more times higher than their reported IC(50) or K(i) values. It was inhibited by the protein kinase C inhibitor bisindolylmaleimide I and by lowering Ca(2+) to nanomolar levels with EGTA; however, it was not stimulated by the addition of phorbol ester, under conditions that significantly enhanced rhodopsin phosphorylation. A monoclonal antibody specific for the Ser(475)-phosphorylated form of RGS9-1 recognized RGS9-1 in immunoblots of dark-adapted mouse retina. Retinas from light-adapted mice had much lower levels of RGS9-1 phosphorylation. Thus, RGS9-1 is phosphorylated on Ser(475) in vivo, and the phosphorylation level is regulated by light and by [Ca(2+)], suggesting the importance of the modification in light adaptation.

Expression profiling suggested a regulatory role of liver-enriched transcription factors in human hepatocellular carcinoma.

By using a cDNA array representing 14,000 cDNA clusters, we studied the expression profiles in paired clinical hepatocellular carcinoma (HCC) samples and the distal nontumorous liver tissues from the same patients. Despite the significant heterogeneity among the clinical samples, 72 genes (including 30 novel genes) were down-regulated and 84 genes (including 48 novel genes) were up-regulated in >50% of the cancer samples that were identified. The alterations in gene expression levels were confirmed by Northern blot and reverse-transcription PCR in all of 4 randomly selected genes. It was conspicuous that 21 of 38 hepatocarcinoma (HCC) down-regulated genes studied previously were reportedly regulated by a group of liver-enriched transcription factors (LETFs), and 12 of 36 HCC up-regulated genes studied previously were involved in protein translation. Reexamination of the cDNA array data further revealed that most of the genes known to be regulated by LETFs were down-regulated in at least a portion of the HCC samples. Among the LETFs, the expression level of CCAAT/enhancer-binding protein (C/EBP) alpha was down-regulated in cancer, whereas hepatocyte nuclear factor 1 (HNF-1), HNF-3beta, HNF-4alpha, and HNF-4gamma were up-regulated. The expression profiling thus suggested multiple regulatory pathways involved in HCC, especially that related to LETFs.

Chaoslike behavior in nonchaotic systems at finite computation precision.

The dynamical behavior of a two-dimensional map is investigated numerically. A chaoslike behavior, i.e., a nonsmooth distribution of the attractor and seemly sensitive dependence of the motion on initial condition is found as the system state is nonchaotic (both Lyapunov exponents are nonpositive). The key point for this strange behavior is that the mode corresponding to the second negative Lyapunov exponent contains positive local Lyapunov exponent segments. It is argued that this kind of behavior may be typical and easily observed in practical numerical computations and experiments where small noise is inevitable.

Collective directional transport in coupled nonlinear oscillators without external bias.

Directed collective motion in a circular array of unidirectionally coupled oscillators with symmetric potential is obtained numerically in the absence of external bias. This striking feature is interpreted as the effect of the spontaneous breaking of temporal symmetry of the coupling. It is revealed that a proper match of various control parameters is important in generating an optimal coherent global transport. Noise-sustained directed transport is also observed, and the related stochastic resonance in an autonomous system is identified.

Fudenine, a C-terminal truncated rat homologue of mouse prominin, is blood glucose-regulated and can up-regulate the expression of GAPDH.

Messenger RNA differential display was applied to screen for the blood glucose-regulated genes in SD rat skeletal muscle. The rat homologue of the mouse prominin was thus identified. Comparing to its mouse and human homologues, fudenine was C-terminal truncated due to a single nucleotide deletion. However, its mitochondrial energy transfer signature peptide PQDLVKKLI remained intact. Fudenine, an 592-amino acid containing, 66-kDa glycoprotein, is a novel plasma membrane protein with four transmembrane segments flanking by two large glycosylated extracellular domains. Although it is devoid of the last transmembrane domain comparing to its homologues, fudenine also locates in cell membrane by transfection of fusion plasmid pFudenine-EGFP into CBRH7919 cell and L-6TG cell. Overexpression of fudenine in CBRH7919 cell line up-regulated the mRNA level of GAPDH (3-phosphate glyceraldehyde dehydrogenase), while long-term glucose exposure resulted to reduced GAPDH expression. Since high blood glucose level induced the expression of fudenine in skeletal muscle, which in turn up-regulated the expression of GAPDH, we propose that fudenine might be a candidate gene for diabetes mellitus.

Generalized splay state in coupled chaotic oscillators induced by weak mutual resonant interactions.

Dynamic behavior of coupled chaotic oscillators is investigated. A transition from high-dimensional hyperchaos to a generalized periodic splay state is found for extremely weak coupling. Chaotic nature of a single oscillator and mutual resonant interactions are regarded to be responsible for this self-organized ordering. The functional phase distribution of the generalized splay state, which is essentially different from the equal-phase-separation distribution of the conventional splay states, can be well predicted by analyzing a single periodically forced oscillator.

Partial synchronization and spontaneous spatial ordering in coupled chaotic systems.

A model of many symmetrically and locally coupled chaotic oscillators is studied. Partial chaotic synchronizations associated with spontaneous spatial ordering are demonstrated. Very rich patterns of the system are revealed, based on partial synchronization analysis. The stabilities of different partially synchronous spatiotemporal structures and some dynamical behaviors of these states are discussed both numerically and analytically.

Plasminogen activator inhibitor-1 regulates cell adhesion by binding to the somatomedin B domain of vitronectin.

Plasminogen activator inhibitor-1 (PAI-1) binds to the somatomedin B (SMB) domain of vitronectin. It inhibits the adhesion of U937 cells to vitronectin by competing with the urokinase receptor (uPAR; CD87) on these cells for binding to the same domain. Although the inhibitor also blocks integrin-mediated cell adhesion, the molecular basis of this effect is unclear. In this study, the effect of the inhibitor on the adhesion of a variety of cells (e.g., U937, MCF7, HT-1080, and HeLa) to vitronectin was assessed, and the importance of the SMB domain in these interactions was determined. Although PAI-1 blocked the adhesion of all of these cells to vitronectin-coated wells, it did not block adhesion to a variant of vitronectin which lacked the SMB domain. Interestingly, HT-1080 and U937 cells attached avidly to microtiter wells coated with purified recombinant SMB (which does not contain the RGD sequence), and this adhesion was again blocked by the inhibitor. These results affirm that PAI-1 can inhibit both uPAR- and integrin-mediated cell adhesion, and demonstrate that the SMB domain of vitronectin is required for these effects. They also show that multiple cell types can employ uPAR as an adhesion receptor. The use of purified recombinant SMB should help to further define this novel adhesive pathway, and to delineate its relationship with integrin-mediated adhesive events.

Transformants of Metarhizium anisopliae sf. anisopliae overexpressing chitinase from Metarhizium anisopliae sf. acridum show early induction of native chitinase but are not altered in pathogenicity to Manduca sexta.

Extracellular chitinase activity has been implicated in the pathogenesis of several fungal infections. Following induction with chitin, the insect pathogens Metarhizium anisopliae sf. acridum ARSEF strain 324 and Metarhizium anisopliae sf. anisopliae ARSEF strain 2575 secrete 44-kDa basic and acidic isoforms of endochitinase, respectively. The gene from strain 324 (Chit1) was cloned and inserted into the genome of strain 2575 under the control of Aspergillus regulatory elements such that transgenic 2575 (2575-Chit(+)) expressed CHIT1 in a noninducing medium (i.e., not containing chitin). Isoelectric focusing followed by a zymogram technique revealed that neither wild-type 2575 nor 2575-Chit(+) produced significant amounts of the native 2575 acidic chitinase in a noninducing medium. However, in a chitin-containing medium, 2575-Chit(+) produced the native chitinase earlier than strain 2575, soon after secretion of CHIT1. We hypothesize that this is due to the production of soluble inducers following chitin hydrolysis by CHIT1 and that M. anisopliae uses enzymes expressed at low levels to sense the nature of the polymeric nutrient present in the immediate environment. However, the chitinase overproducers did not show altered virulence to caterpillars (Manduca sexta) compared to the wild-type fungus, suggesting that wild-type levels of chitinase are not limiting for cuticle penetration.

Production of poly-3-hydroxybutyrate by Bacillus sp. JMa5 cultivated in molasses media.

A strain of Bacillus sp. coded JMa5 was isolated from molasses contaminated soil. The strain was able to grow at a temperature as high as 45 degrees C and in 250 g/l molasses although the optimal growth temperature was 35-37 degrees C. Cell density reached 30 g/l 8 h after inoculation in a batch culture with an initial concentration of 210 g/l molasses. Under fed-batch conditions, the cells grew to a dry weight of 70 g/l after 30 h of fermentation. The strain accumulated 25-35%, (w/w) polyhydroxybutyrate (PHB) during fermentation. PHB accumulation was a growth-associated process. Factors that normally promote PHB production include high ratios of carbon to nitrogen, and carbon to phosphorus in growth media. Low dissolved oxygen supply resulted in sporulation, which reduced PHB contents and dry weights of the cells. It seems that sporulation induced by reduced supply of nutrients is the reason that PHB content is generally low in the Bacillus strain.

A comparison of dietary and non-dietary factors of hypertension and normal blood pressure in a Chinese population.

The aim of this study was to compare the differences of dietary and non-dietary factors in hypertension and normal blood pressure (BP) of Chinese urban people. Two cross-sectional population surveys were carried out in Tianjin, one of the three largest cities in China. A total of 2068 subjects aged 35-64 years were selected by a random stratified cluster sampling, 367 men and 395 women were hypertensive, and 623 men and 683 women had normal BP. The diet was assessed by food weighing plus consecutive individual 3-day food records. Health status, health behaviours and anthropomentry were assessed. The whole population had a high sodium and low potassium consumption level. Hypertensive men and women had significantly higher daily dietary sodium intake (P < 0.05) and hypertensive women had higher dietary sodium-to-potassium ratio (P < 0.01) than men and women with normal BP. Age, body mass index, overweight and family history of hypertension were positively related to high BP. dietary sodium intake and dietary sodium-to-potassium ratio were positively associated with high BP. Education level in women and commuting physical activity in men were inversely associated with high BP.

Single nucleotide polymorphism identification in candidate gene systems of obesity.

We have constructed a large panel of single nucleotide polymorphisms (SNP) identified in 68 candidate genes for obesity. Our panel combines novel SNP identification methods based on EST data, with public SNP data from largescale genomic sequencing, to produce a total of 218 SNPs in the coding regions of obesity candidate genes, 178 SNPs in untranslated regions, and over 1000 intronic SNPs. These include new non-conservative amino acid changes in thyroid receptor beta, esterase D, acid phosphatase 1. Our data show evidence of negative selection among these polymorphisms implying functional impacts of the non-conservative mutations. Comparison of overlap between SNPs identified independently from EST data vs genomic sequencing indicate that together they may constitute about one half of the actual total number of amino acid polymorphisms in these genes that are common in the human population (defined here as a population allele frequency above 5%). We have analyzed our polymorphism panel to construct a database of detailed information about their location in the gene structure and effect on protein coding, available on the web at http://www.bioinformat ics.ucla.edu/snp/obesity. We believe this panel can serve as a valuable new resource for genetic and pharmacogenomic studies of the causes of obesity.

Antibiotic susceptibility of Helicobacter pylori in the Chinese population.

AIM: To assess antibiotic susceptibility of Helicobacter pylori (H. pylori) strains to metronidazole, clarithromycin and tetracycline in the Chinese population, and to test the stability of antibiotic resistance in H. pylori 1 year after storage at -80 degrees C. METHODS: Fifty H. pylori strains isolated from patients with peptic ulcer disease were recovered from storage at -80 degrees C. Susceptibility of these strains to metronidazole, clarithromycin and tetracycline was determined by using validated disk diffusion tests, which was repeated 1 year after storage at -80 degrees C. The DNA profiles of each strain were determined by using the polymerase chain reaction-based-random amplified polymorphic DNA fingerprinting technique (PCR-RAPD). This was repeated if any change in antibiotic susceptibility pattern was noticed. RESULTS: The resistance rate was 50% to metronidazole and 8% to clarithromycin. None of the strains was resistant to tetracycline. A dual resistance to metronidazole and clarithromycin was demonstrated in three H. pylori strains. The antibiotic susceptibility test reproduced itself in 92% (36 of 39) of the strains 1 year later; the three strains with dual resistance exhibited susceptibility to both antibiotics. Variation in antibiotic susceptibility pattern in the three H. pylori strains was associated with change in the RAPD fingerprint. CONCLUSION: The prevalence of resistance in H. pylori is high to metronidazole but low to clarithromycin in the Chinese population. The disk diffusion test appears to be a simple and reliable test, while antibiotic resistance in some H. pylori strains may disappear after long-term storage at -80 degrees C.

Visually induced reorientation illusions.

It is known that rotation of a furnished room around the roll axis of erect subjects produces an illusion of 360 degrees self-rotation in many subjects. Exposure of erect subjects to stationary tilted visual frames or rooms produces only up to 20 degrees of illusory tilt. But, in studies using static tilted rooms, subjects remained erect and the body axis was not aligned with the room. We have revealed a new class of disorientation illusions that occur in many subjects when placed in a 90 degrees or 180 degrees tilted room containing polarised objects (familiar objects with tops and bottoms). For example, supine subjects looking up at a wall of the room feel upright in an upright room and their arms feel weightless when held out from the body. We call this the levitation illusion. We measured the incidence of 90 degrees or 180 degrees reorientation illusions in erect, supine, recumbent, and inverted subjects in a room tilted 90 degrees or 180 degrees. We report that reorientation illusions depend on the displacement of the visual scene rather than of the body. However, illusions are most likely to occur when the visual and body axes are congruent. When the axes are congruent, illusions are least likely to occur when subjects are prone rather than supine, recumbent, or inverted.

Different worlds, different tasks for health promotion: comparisons of health risk profiles in Chinese and Finnish rural people.

The aim of this study was to compare cardiovascular risk factors of working-aged people in Chinese and Finnish rural villages. The surveys were carried out in 1989 in Tianjin, China, and in Kuopio, Finland. Altogether, 897 Chinese inhabitants and 795 Finnish subjects participated in the surveys. Health behaviours were recorded, and height, weight, blood pressure, heart rate and serum lipids were measured. Generally Finns had a significantly higher mean body-mass index, systolic and diastolic blood pressures, and serum total cholesterol, low-density lipoprotein cholesterol, and total cholesterol/high-density lipoprotein ratio than the Chinese. However, no difference was seen between Chinese and Finnish women in diastolic blood pressure and serum triglycerides. Lower high-density lipoprotein cholesterol levels were observed in Finnish men than in Chinese men, whereas a higher mean level was shown in Finnish women than in Chinese women. There were significantly higher mean heart rates and prevalence of smoking in Chinese than in Finnish populations. More people who were overweight, obese and hypertensive were found in the Finnish than in the Chinese populations. Most of the Finns had two or more cardiovascular risk factors compared with the Chinese, the majority of whom were in the group with less than two risk factors. In conclusion, the risk profiles are clearly somewhat different in these two countries. A major task for the Chinese health policy and health care system is to decrease smoking and to prevent obesity and hypertension. In Finland, the biggest task seems to be the reduction of weight and lipid abnormalities, and the prevention of hypertension.

Cranial reossification with absorbable plates.

The purpose of this study was to examine the effect of Lactosorb absorbable plates on bone healing across cranial bone defects in the rabbit skull. Two 10-mm diameter parietal skull defects were created in each of 20 rabbits, with one defect being placed on either side of the sagittal suture. In 10 rabbits, an absorbable plate was placed across both the inner and outer cortices of the left defect, and in the other 10 rabbits, an absorbable plate was placed across the outer cortex only of the left defect. The right defect always served as the control side, with no plate being placed across it. Rabbits were killed an average of 25 weeks postoperatively. Areas of reossification in the experimental and control defects of each rabbit were then measured, examined histologically, and compared. Growth across defects spanned by one plate was also compared with growth across defects spanned by two plates. Histologic and statistical analyses revealed no significant differences in reossification between the control and experimental defects in each animal and between the defects spanned by one versus two plates. This study suggests that these copolymer absorbable plates neither inhibit nor facilitate reossification across 10-mm diameter rabbit cranial defects.
