Proteins of the extracellular fluid of mouse brain: extraction and partial characterization.

An extraction procedure for the isolation of proteins from the brain extracellular fluid (ECF) was developed and applied to studies of the ECF components of mouse brain. Perfused intact brains were incubated in an isotonic medium for periods of up to 2 h at 0 degrees C to allow the release of ECF into the medium without disruption of the integrity of the tissue. The validity of the extraction procedure was established by (a) the fact that the total yield of ECF proteins was constant per unit weight of brain tissue, (b) the absence of tyrosine hydroxylase, an enzyme marker of the cytoplasmic fraction, from the extracts, and (c) the distinctive features of the one- and two-dimensional gel electrophoretic patterns of ECF proteins as compared with those of the cytoplasmic and membrane fractions. The results indicate that the extracellular fluid of mouse brain contains a mixture of proteins with a wide distribution of molecular weights (10,000-100,000 daltons) at a concentration level of about 0.3%.

[Use of evoked potentials for the intraoperative differentiation of motor and sensory fascicles].

The animal experiments demonstrate the possibility of differentiating intraoperatively between motor and sensory fascicles of the peripheral nerve. This differentiation is based on somatosensory evoked potentials, which are summarized with an averager. Peripheral nerves in four patients have been reconstructed with this method. The first (median nerve lesion) has complete motor function of the thenar eminence seven months after operation.

[Protein binding of the macrolide antibiotic turimycin. Methodologic effects on results].

A methodological investigation deals with the binding of the macrolide antibiotic turimycin to bovine serum albumin (BSA) and serum proteins. On this occasion, it is pointed out that, especially when serum is used, the critical evaluation of the analytical method is of the same importance as the utilization of standardized procedures for the quantification of the protein binding in the sense of general comparability. This concerns, for example, the chemistry of the reaction used for detection, the formation of degradation products of the active principle in the serum during the study of the binding and possible repercussions on the chemical determination, the electrolyte content of the sample in the punched hole of the microbiological test plate, losses of activity or synergistic effects of the serum-antibiotic combination during incubation of microbiological test plates after termination of the equilibrium dialysis. The determination of binding constants by means of a competitive fluorescence titration, the chemical analysis of equilibrium dialyses and their parallel assessment with the aid of the agar-diffusion plate test led to results which were not in agreement with each other. Turimycin which is very slightly soluble at pH = 7.4 and fairly soluble at pH = 5.0, is practically not bonded at the lower pH value of BSA and serum proteins (fluorescence titration of BSA: Kb approximately 20; equilibrium dialysis and chemical evaluation). The microbiological determination in serum on the basis of equilibrium dialyses yields higher values for the binding of turimycin.(ABSTRACT TRUNCATED AT 250 WORDS)

[Spectroscopic studies on the formation of metal complexes and on the protein binding of antiviral thiosemicarbazone derivatives (author's transl)].

The complexation of some thiosemicarbazones and isothiosemicarbazones of isatin and quinolin-2-aldehydes with Cu2+, Zn2+ and Mn2+ ions was spectrometrically investigated. Semiquantitative data, obtained from extinction values, about the relative complexing tendencies within some groups of homologous substances were brought in relation to their antiviral effects and binding to bovine serum albumin. The complexing tendencies were greatest in compounds with methyl substituents and decreased for higher alkyl substituents. whereas the binding to protein increased in the same order. The well-known maxima of the antiviral observed with medium alkyl groups may be explained by a superposition of these effects.

[On the methodology of the determination of alkylating activities of nitrogen mustards with nitrobenzylpyridine (NBP) in biological material (author's transl)].

It is dealt with modifications to the spectrophotometric determination of the alkylating behaviour of bifunctional nitrogen mustards with 4-(4'-nitrobenzyl)pyridine (NBP). The authors succeeded in characterizing spectrophotometrically, by means of a one-phase procedure, not only soluble, but also difficultly soluble, therapeutically known and newly synthetized mustards in regard to their alkylating behaviour. In the framework of in vitro studies, the authors report of experimental details of the proper preparation of biological material for the determination of residual activities, the analytical approach being always depending on the structure of the respective mustard. The differences in the time courses of the acylating attacks in serum, blood and suspensions of leukaemic cells are demonstrated by the example of some selected nitrogen mustards.

[Age and sexuality from the point of view of inhibitants of homes for the aged and geriatric nursing homes (author's transl)].

The paper is on the problem whether or not are the groupspecific particularities--produced by analyzing the reaction of elder and old inhabitants of homes for the aged to loveful and tender demonstrations at age and the desired characteristics by choosing a new partner corresponding with the social differentiation consisting of the social characteristics: age, sex, personal status, contents of work, length of residence in the home of aged and physical mobility. It is to be said that the value orientations of inhabitants of homes for the aged in loveful and tender demonstrations at age contain the whole range of the social differentiation without clear attachment. No type of the social differentiation of inhabitants of homes for the aged and geriatric nursing homes tends to a certain type of choosing a partner. There was no evidence for a well defined connection between social differentiation and attitude to sexuality at age and in the home of aged.

Effect of STS 557 on semen composition, fertility and sexual behaviour of male rabbits.

Fertility control of male rabbits was achieved by daily oral administration of STS 557 (17 alpha-cyanomethyl-17 beta-hydroxy-estra-4.9(10)-diene-3-one) over a period of 8 weeks. In doses of 10 and 20 mg STS 557 per animal per day, fertility inhibition was accompanied by a decrease of mean number of mounts and ejaculations. Furthermore, semen volume, number of sperm per ejaculate, and sperm motility were reduced. At a dose of 5 mg STS 557 per day, male sterility was associated with reduction of sperm motility and semen fructose content. Libido, semen volume, sperm number, and sialic acid content in semen remained unaffected. Development of male contraceptives on the basis of chronic progestin treatment without concomitant androgen supplement may thus be possible.

Effects of levonorgestrel and STS 557 on testis in rodents.

In immature male Wistar rats levonorgestrel and STS 557 (17 alpha-cyanomethyl-17 beta-hydroxyestra-4,9(10)-diene-3-one) reduced testicular growth, testicular DNA contents and plane of tubular cross-section in a dose dependent manner. In this respect, STS was less active than levonorgestrel. In immature castrated male rats the LH-suppressing effect of STS 557 was about 15 times lower than that of levonorgestrel. The results suggested that peripheral inhibitory effect of the two progestins was coupled with central LH suppression. On the other hand, in mating tests with male hybrid mice STS 557 possessed high fertility inhibiting activities in comparison to levonorgestrel and chlormadinone acetate. Limited dissociation has been shown between fertility inhibition and mating rate. All changes were reversible. Findings are discussed in view of development of male contraceptives.

[Age and sexuality from the point of view of inhabitants of homes for the aged geriatric nursing homes (author's transl)].

This work handles about the need of erotic relations by older and old inhabitants in homes of age and geriatric nursing homes, asks for their imaginations at loveful and tender demonstrations in age, for which characteristics they attach great importance at the election of a new partner. The investigation has the character of a niveau analysis - before we investigate the laws of phenomena, their change and development of phenomena must be known. The potential unity of all inhabitants presents in the open cited central questions groupspecific specialities, their possible differentiation remains reserved for a later information. The concentration of the different results lets perceive that inhabitants in homes of age and geriatric nursing homes loveful and tender actions in age answer slightly in affirmative, that each 5. inhabitant expects sexual readiness from a new partner, that two third of all inhabitants mean sexual relations in age and home contribute to well-being and were expressions of their succeeded integration in the society.

Enhancement of the antiviral activity of pyrimidine derivatives against mengovirus by visible light.

Eleven pyrimido-pyrimidine derivatives, seven with significant antiviral activity against Mengovirus, five against Coxsackie B1 virus and four antiviral negative compounds were tested for their photosensitizing ability. All seven compounds with antiviral activity in vitro showed an enhanced antiviral action against Mengovirus under irradiation with visible light, a fact that may be caused by photodynamic processes. It was tried to correlate the oxidation potentials of sensitizers with their photodynamic activity. By means of mass-spectrometric investigations, molecular fragmentation was examined following thin layer chromatography (TCL) before and after irradiation. Furthermore, binding affinity to biopolymers (BSA and RNA) was investigated to reveal conformity in differences of antiviral activity. The main results are the following: 1. Generally, strong antiviral activity can be correlated with strong binding affinity. 2. No significant correlation could be detected between oxidation potentials of antiviral compounds and their enhanced antiviral activity under irradiation conditions, although in some cases sensitizer with higher oxidation potentials are more effective than those with lower ones. 3. The lower the photostability of the compounds the higher was the light-induced antiviral activity. 4. No alteration of the molecular ion peak and fragmentation pattern before and after irradiation was indicated by means of mass-spectrometry and TLC using fairly comparable conditions.

[Essay of proteins after aldehyde treatment in biological objects (author's transl)].

Following fixation with formalin or glutaraldehyde, the protein content of bovine serum albumin, lysozyme, non-disintegrated yeast cells and liver nuclei of mice is determined by means of a brom-phenol blue staining procedure and the widely used Lowry-technique. The bromphenol blue technique permits determinations of soluble as well as particulate proteins or protein mixtures fixed with up to 6% formalin or 2% glutaraldehyde. Recovery rates differ no more than 20% as related to unfixed controls. Using the bromphenol blue method it is not necessary to separate aldehyde or any interfering material by additional steps prior to determination. Factors for correcting protein content of biological material after aldehyde treatment are available. In this way, comparative biochemical as well as cyto- and histochemical investigations of enzymatic activities after aldehyde fixation are possible. Some advantages of the bromphenol blue technique with special reference to the analysis of particulate and/or aldehyde-fixed specimens are discussed.
