Lack of hepatitis B virus DNA sequences in sera from patients with acute and chronic liver diseases diagnosed as non-A, non-B-hepatitis.

The sera of 15 individuals with transfusion-associated acute or chronic non A, non B hepatitis, which lacked hepatitis B virus markers, were tested for hepatitis B virus DNA by dot blot hybridization test. Three sera of two patients positive in this test, however, also gave positive results when the labeled plasmid was used as probe instead of labeled HBV-DNA, indicating false positive results in the initial test. In conclusion, the data indicated that sera of patients with confirmed non A, non B hepatitis do not contain DNA-sequences in the serum hybridizing with HBV-DNA.

HBsAg clearance in patients with long-standing chronic active hepatitis B and hepatitis B virus-induced liver cirrhosis.

During a period of 3 years we observed 5 patients with chronic active hepatitis B and 2 with hepatitis B virus-induced liver cirrhosis who cleared HBsAg from their sera after 2-14 years of HBsAg carriership. 4 of them developed anti-HBs. After HBsAg clearance there was no evidence of persisting inflammatory activity within their livers. 5 of the 7 patients had been treated for 1-39 months with prednisolone, sometimes in combination with azathioprine. This therapy, however, had been stopped more than 3 years before these patients terminated their HBsAg carriership. Our observations indicate that even after long-standing chronic active hepatitis B or hepatitis B virus-induced liver cirrhosis HBsAg may be eliminated in a considerable number of patients.

[The effect of the cell separation procedure on neutrophilic granulocyte function. I. Research on the morphology and phagocytosis behavior of isolated granulocytes].

The investigations were aimed at detecting the impact of various cell separating techniques on the morphology and function of neutrophilic granulocytes. Methodically simple cell separating techniques were used, such as spontaneous sedimentation of ACD-AG blood and mechanically defibrinated blood and a flotation technique which makes use of cell isolation by means of a separating solution and centrifugation. Granulocytes which were isolated by the cell separator Haemonetics M 30 for therapeutic purposes and which, according to data in literature, are not damaged by cell separating techniques were used as reference cells. The electronic-optical evaluation of granulocyte morphology and the in vitro investigation of the phagocytic capacity for rice starch (ingestion rate) of differently isolated granulocytes were made after cell separation.

[The effect of the cell separation procedure on neutrophilic granulocyte function. II. Research on the bactericidal capacity and the NBT reduction ability of isolated granulocytes].

The investigations were aimed at examining the impact of methodically simple separating and enriching techniques for gaining granulocytes on the bactericidal capacity and NBT reduction in isolated granulocytes. For this purpose, granulocytes were gained by the technique of sedimentation and flotation. Granulocytes isolated by the cell separator Haemonetics model 30 were used as reference material. The findings of the investigations together with those of previous studies revealed no indication of the granulocyte function and morphology being damaged by the separating techniques used.

[The function of granulocytes stored in whole blood in relation to the storage time].

Whereas numerous investigations and extensive experiences have been available for some years on problems of storing erythrocytes and thrombocytes, it was only recently that the storage of granulocytes with the aim at maintaining their function for a longer period has been given proper regard as a problem, which, however, could not be brought to a satisfactory solution. Primarily, attention is focused on specific cell physiology. At the same time it is necessary to have at hand a number of approaches covering as many partial aspects of the granulocyte function as possible to identify the functional damages or the extent of cell damage in connection with storage. In the present paper the behaviour of granulocytes under conditions commonly used in transfusion medicine for storing blood are investigated for a period of 4 days by controlling the total leukocyte and granulocyte values, pH value, osmolality, vitality test in the colour excluding test as well as by function tests such as behaviour of phagocytosis and NBT capability of reduction. A report is presented on the results of quantitative and qualitative changes in the investigated parameters.

[Detection and significance of HBsAg associated receptors for polymerized human serum albumin in acute hepatitis B virus infection].

Receptors for polymerized human serum albumin (R-pHSA) may play a role in the attachment of hepatitis B virus (HBV) to hepatocytes. Therefore, we evaluated the incidence and prognostic value of R-pHSA in acute hepatitis B virus infection. High titers of R-pHSA were found in 12/12 patients with HBeAg positive acute hepatitis B (log2-Titer: 8.5 +/- 1.5). Titers for R-pHSA and HBsAg correlated closely. Furthermore, R-pHSA occurred (log2-titer: 6.5 +/- 1.5) in 10/10 patients with HBeAg positive chronic active hepatitis B (CAH-B). In the course of HBV infection, R-pHSA were eliminated earlier than HBsAg. 4/20 patients which were asymptomatic HBsAg carriers had high titers of R-pHSA in correlation to the histological findings of CAH-B. Asymptomatic HBsAg carriers which were R-pHSA negative had a normal liver histology. R-pHSA were found to be an early prognostic marker in acute hepatitis B infection.

[Acute fatty liver of pregnancy. A case report of the differential diagnosis of pregnancy-associated liver diseases].

Acute fatty liver of pregnancy (AFLP) is a rare, but often fatal disorder in the third trimester of pregnancy. We report on a patient with AFLP complicated by acute hemorrhagic diathesis, who survived after early diagnosis and adequate treatment. On electronmicroscopy, the characteristic microvesicular fatty deposits in the centrolobular hepatocytes could be demonstrated.

[The present epidemiological status of African swine fever].

At present, African swine fever (ASF) persists as an enzootic infection both on the African continent and in Europe (Portugal, Spain, and Sardinia). The recent outbreaks of ASF in Belgium and in the Netherlands have again demonstrated the threat of this disease to the swine population in Germany. The main reasons for this threat are the great tenacity of this virus and its stability in meat and meat products together with an immense tourism into these enzootic areas. Epizootiological peculiarities, such as virus replication in ticks and inapparent infections in wild boars are the reason why eradication of the disease has failed so far, especially when pigs are allowed to roam the countryside.

Association of hepatitis Be antigen (HBeAg) with the core of the hepatitis B virus (HBcAg).

Three substances (pronase E, sodium dodecylsulfate (SDS) and guanidine hydrochloride) with different chemical actions partially convert HBcAg to HBeAg. This process retains the integrity of the HBcAg particle, which was not different between HBcAg subpopulations, and does not generate HBcAg or HBeAg sub-units. DNA polymerase activity was destroyed by SDS and guanidine hydrochloride, but not by pronase E. Serum HBeAg could not be converted into HBcAg, suggesting that this might be an irreversible process. The data are consistent with the assumption that HBcAg and HBeAg are coded for by the same gene (C gene of the HBV-DNA).

[Evaluation of clinical efficacy of platelet substitution in acute leukemia using resonance thrombography].

For evaluating the clinical effectiveness of thrombocyte substitution, the resonance thrombogram was registered before and after thrombocyte substitution in 50 patients with acute leukemia and the increase of thrombocytes (corrected increment) determined. Thrombocyte substitution led to a significant diminution of the platelet amplitude (p less than 0.01) and platelet value. The highest value of these alterations of parameters was reached 1 hour after transfusion, it was somewhat lower after 24 hours, yet the initial value was not reached again. Correlations could be found to exist between the platelet amplitude and the corrected thrombocyte increase 1 hour and 24 hours after transfusion. The resonance thrombography enables the successful control of thrombocyte substitution to be made and it is suitable for monitoring hemostasis in severe thrombocytopenia due to production.

[Delta infection in HBsAg carriers].

Anti-delta as an indicator of chronic delta infection was found in 7/180 HBsAg carriers. This indicates a low incidence of delta infection in Germany. The following risk factors to acquire delta infection were established: Mediterranean origin (3 cases) and transfusion of blood products (4 hemophiliacs). All anti-delta positive individuals had a biopsy proven chronic active hepatitis and the serological pattern: HBsAg, anti-HBc and anti-HBe positive without indicators of Dane particle synthesis (negative tests for HBeAg and DNA polymerase activity).

[Amebic colitis - differential diagnosis of ulcerative colitis and Crohn disease].

Amoebic colitis rarely develops in patients who had not stayed in a country where amoebiasis is endemic. "Crohn's colitis" was diagnosed in a 33 year old female patient who never had stayed in such an area. The dramatic deterioration of the disease led to revise the early diagnosis. The reported case indicates that the conventional laboratory, endoscopic, histologic, and radiologic examinations don't prove the anamnestically unsuspected amoebic colitis. Since the correct diagnosis was verified by stool and serologic examinations, these should be performed in any doubt of a specific colitis.

Isolation and comparison of arylsulfatase A from rat liver and Morris hepatoma 7777.

Rat liver and Morris hepatoma 7777 arylsulfatase A were isolated from the soluble lysosomal extract by a procedure involving blue-Sepharose affinity chromatography, DEAE-cellulose chromatography, hydrophobic chromatography on phenyl-Sepharose and preparative polyacrylamide gel electrophoresis. The preparation obtained by this method was apparently homogenous in disc electrophoresis and in immunoelectrophoresis. The comparative studies revealed that the properties of arylsulfatase A from rat liver and Morris hepatoma 7777 are very similar, considering molecular weight of the native monomer and its subunits, the ability to form tetramers, isoelectric point, Michaelis constant and the anomalous kinetics of the reaction. The twofold elevation of arylsulfatase B activity found in Morris hepatoma 7777 suggests that the enzyme may have certain functions in tumor growth.

Simultaneous infection with non-A, non-B and B viruses in a patient exhibiting different histologic patterns of acute hepatitis.

We present the case of an elderly female patient who developed acute and severe hepatitis with two distinct peaks following blood transfusion. The serological markers and histopathology during the first attack were characteristic for hepatitis non-A, non-B(NANBH), whereas the second peak was due to hepatitis B virus (HBV) infection. Electron microscopy revealed morphologic equivalents for both hepatitis agents.
