[Gene expression, metabolic regulation and aging].

The biological ageing comprises changes of of structure and function depending on time with the beginning of the autonomy of a living being up to death. On the one hand, the causes of this process are genetically fixed, on the other hand they depend on environmental factors and are not programmed. In the course of life the total genome and also the nucleic acid underlie influences, which must have consequences on the gene expression. In these cases the changed sensitivity of the genetic apparatus in comparison to hormones is impressionably changed. The decreased protein biosynthesis may be stimulated by increased amino acid concentrations and by SH-substances, Age-depending changes are also observed for enzymatically catalyzed reactions and in the regulation of chains of metabolism. At first the biologic ageing appears as a complex multiform process, which reaches the simplest integration level in the cellular metabolism.

[Molecular biology of aging. 8. Aging of intermitotic cells--relationship to protein synthesis in the mucosa of the small intestine].

1. The incorporation of 14C-leucine in a cell free system of intestine mucosa is diminished in aging. 2. The absence of 2-mercaptoethanol in the system of incorporation increases the age differences. 3. By combination of a system from intestine with liver preparation we observed a participation of both the microsomes and the postmicrosomal supernatant. 4. The activity of Ribonuclease is not changes in the aging process. Our results demonstrated the alterations of the intermitotic cells in aging.

[On the molecular biology of the ageing. VI. Information. Changes of the protein biosynthesis in the age (author's transl)].

1. Most reports in the literature describe a diminished protein synthesis in the old organism. 2. The diminished protein synthesis may be the result of changes of ribosomes, unequal concentration of endogen m-RNA, changes of the activity or concentration of cytosol factors, especially the influence of SH-substances. 3. The age-dependent changes of protein biosynthesis are dependent on the technique of preparation and on the nutrition state of the organism. 4. Addition of 2-mercaptoethanol during preparation or to the incubation medium decreases the age-dependent differences of protein synthesis.
