[Occurrence of Aspergillus fumigatus in West Berlin - contribution to the epidemiology of aspergillosis (author's transl)].

The increasing incidence of human A. fumigatus infections particularly such of the respiratory tract has led to this study of the epidemiological situation. During the period between October 1968 and December 1977, A. fumigatus was isolated from clinical material in 425 cases. These findings which were established at the Mycology Unit, Robert Koch Institute, Federal Health Office, exhibited the following distribution by regions of the body or type of specimen: respiratory tract 277 (65.1(); venous blood 58 (13.6%); urine 3 (0.7%); stools 9 (2.1%) and others 78 (18.3%). Approximately two thirds of these findings were made during the period from October to March and about one third between April and September. The presence of A. fumigatus in the air both indoors and outdoors and also in the environment of patients suffering from aspergillosis was studied with the aid of the sedimentation-method. This method was chosen because the occurrence of A. fumigatus conidia in the air is of epidemiological interest. The number of isolations of A. fumigatus from outdoors samples was low, so the search for inhalative conidia concentrated upon sites near A. fumigatus habitats. These studies revealed that aspergillosis patients, clinical material sampled from them, decaying plant material (agriculture, horticulture), and used clothes and linen may form foci for the spread of A. fumigatus conidia. The control of aspergillosis in the hospital environment involves in particular a control of aspergillosis patients by means of culture and serology (preferably by ghe immunodiffusion test). Numerous recommendations are made on how to prevent A. fumigatus infections in hospitals, at the working site, and in the laboratory.

[Action of cyclic N6-monobutyryladenosine-3':5'-monophosphate (mb-cAMP) on cell on cell and explant cultures of the PNS and CNS in vitro].

Chick embryo trigeminal ganglia of nine days and hippocampus excised from fetal rats between the sixteenth and eighteenth day of gestation were cultivated in cell- and explantcultures in Maximow-chambers. The cultures were placed in a medium containing 2 mM, 1 mM, 0.2 mM or 0.02 mM cyclic N6-Monobutyryladenosin-3':5'-monophosphat (mb-cAMP). The cultures of the PNS, the trigeminal ganglai show after 48 h by 1 mM mb-cAMP longer nerve fibers and a increased nerve fiber index. Smaller concentration did not change significant nerve fiber growth. Cell- and explantcultures of the hippocampus show a stimulation of the nerve fiber growth and the nerve fiberindex by concentration of 0.02 mM mb-cAMP. After ten days in vitro the nerve cells degenerated by higher concentrations.

[Familial translocation 3/22 MAT with partial trisomy 3q (author's transl)].

Two mentally retarded brothers with partial trisomy 3q show clinically similar malformations and deformities : dwarfism, bushy eyebrows, eversion of the nostrils, low inserted ears, high palate, microgeny, low hair insertion, short and broad hands with proximally inserted thumbs, clinodactylia of the 5th finger, syndactylies, mostly arch patterns on the digital pulps, muscular hypotonia, joint relaxation and cryptorchism. Both children had fits of convulsions. The younger boy showed, moreover, a perception deafness. The mother, the maternal grand-mother as well as the phenotypically normal sister of the patients revealed a balanced translocation 3/22 with a karyotype : 46,XX,t(3;22) (q25;p11).

[The effect of Solcoseryl on in-vitro cultured cells].

Explants of peripherical nervous system (PNS), skin and ventriculus cordis from chick embryo were cultivated in Maximow chambers and the effect of Solcoseryl, Fa. Solco Basel AG, on some morphological parameters was tested. 1. The growth of tissue cultures is influenced by Solcoseryl in relation to concentration and time of application. The index of area in cultures of PNS and cor increased within the first days. By long time application up to 6 days in vitro the index of area decreased and the index was the same than in controls. Explants of skin showed no essential stimulation of growth. 2. The number of cells per unit of culture in the outgrowth of PNS, cor and skin was different influenced. The density of cells in cultures of PNS and skin decreased (signif. difference). In explants of heart we could not observe a difference between the inside and outside of the outgrowth. An influence of Solcoseryl on the degree of migration is discussed. 3. The area of cell nuclei from heartcells was observed. The area decreased under the influence of Solcoseryl. The difference is significant. 4. The mitotic index of heart cells increased by application of Solcoseryl within the first 2 and 3 days in vitro. 5. The number of nucleoli per nucleus of heart cells under experimental conditions increased significant. It is discussed, Solcoseryl influenced in vitro metabolic processes in suitable systems; stimulation of cell proliferation and migration and rns-synthesis was observed within the first days of cultivation. In-vitro-systems are important objects and they are suitable for tests of pharmaca in vitro.

Ocular cryptococcosis - experimental and clinical observations.

During the previous studies, we could demonstrate that there are strains of Cryptococcus neoformans which after intraperitoneal inoculation, are capble of surviving only in the brain of mice without causing any apparent clinical symptom; only in about 4% of the animals the fatal involvement of the CNS occurred. The present investigations suggest that the selective involvement of eye, i.e., formation of intraocular cryptococcoma, with subsequent blindness is also possible under similar experimental conditions. It was observed that a short-lived uremia caused by intramuscular injection of 0.2 ml glycerine could enhance the rate of the selective involvement of the CNS including the eye. The uremia was controlled by the auxanographic method using STAIB's technique of serum-residual-nitrogen-auxanogram and employing the strain used for infecting the mice. These animal experiments are discussed in connection with a clinical case of intra-ocular cryptococcosis in which the only known basic disease was uremia of unknown orgin. The spontaneously healing cryptococcosis detected in this case after the treatment of uremia, has been discussed. Cases of ocular cryptococcosis described in the world literature have been also briefly discussed inconnection with our results. These microbiological observations will be supplemented by the histopathological findings to be published separately.

[Candida tropicalis as causative agent of septicemia (author's transl)].

In connection with microbiological findings in two patients, a report on septicemia caused by C. tropicalis is given. C. tropicalis is second in frequency to C. albicans in man. Because for the different biological properties of Candida species in each case of isolation of such fungi from clinical specimens an identification of the species is obligatory, because the generalizing diagnosis of "Candida mycosis" may be insufficient for the detection of epidemiological and pathogenetic associations. The possible primary resistance of C. tropicalis against the antimycotic 5-Fluorocytosine (Ancotil) is discussed. It is shown that the antigen similarity within the most important Candida species as e.g. between C. albicans and C. tropicalis may cause a false interpretation of serological results concerning the causative agent of the infection. For a safe inactivation of C. tropicalis on human skin, cleaning with 70% isopropyl alcohol for ten seconds is proposed. Because C. tropicalis is entering the human body preferably through food it is discussed whether a microbiological control of food for patients predisposed for such infections should be performed.

[The effect of orotic acid and orotic acid derivatives on the in vitro differentiation of hippocampus neurons-morphometric and electron microscopic studies of ribosomes].

1. By means of electron microscopic and quantitative methods at the explantate cultures of the fetal hippocampus in vitro the influence of the orotic acid, sodium orotate and methylglucamine orotate on the neurogenesis was investigated. 2. After three days of the in vitro cultivation the neuroblasts influenced by these drugs show a smaller respectively not different degree of differentiation compared to the controls. 3. Under the influence of the orotic acid and of its derivatives the neurogenesis is significantly stimulated. The drugs produce a significant increase of the membrane-bound ribosomes and polysomes. The total number of ribosomes increases following the application of orotic acid by 20%, of sodium-orotate by 48% and of methylglucamine-orotate by 23% compared to the controls (alpha = 0,1%). 4. Sodium-orotate shows with reference to the neuronal development the clearest stimulatory effect. After 20 days in vitro the total number of ribosomes is by 60% higher at the treated cultures than in the controls. 5. The results might suggest, that an enlarged supply of the pyrimidine nucleotide via a raising of the RNA- and the protein synthesis might stimulate the development of the neuroblasts even under the in vitro conditions.
