The effect of thymopoietin32-36 and ubiquitin on spontaneous immunopathology of New Zealand mice.

The effects of synthetic thymopoeitin32-36 and purified ubiquitin, in daily doses of 1 or 10 microgram, were studied in New Zealand mice. The most striking result was a 4-month delay in the appearance of Coombs' positive tests or Coombs' antibodies in New Zealand black (NZB) mice treated from 4 wk of age with ubiquitin. Ubiquitin also reduced the spleen weight in the animals and stimulated suppressor T cell activity in shorter term studies of older NZB and black and white (B/W) mice. Thymopoietin was also active in this assay and improved the mitogen responsiveness of lymph node cells in older treated NZB and B/W mice. Thymopoietin injections, from 4 wk of age, reduced the titer of Coombs' antibodies and thymocytotoxic antibodies in NZB mice and caused an increase in Thy 1+ spleen cells in these animals. In correspondingly treated B/W mice, thymopoietin reduced the anti-DNA antibody titer. While our present injection protocol did not result in clinical cure of the genetically determined autoimmune diseases of NZB and B/W mice, they do point to the feasibility of selective immunoregulation by these peptides in diseases associated with altered states of immune reactivity.

Evaluation of T cell subsets with monoclonal antibodies in synovial fluid in rheumatoid arthritis.

Monoclonal antibodies that react with all T cells (OKT3), the inducer-helper T cell subset (OKT4), and the suppressor-cytotoxic T cell subset (OKT8) have been used to evaluate T cell and T cell subpopulations in synovial fluid (SF) from patients with rheumatoid arthritis (RA). We found that the immunoregulatory ratio of SF lymphocytes ranged from normal values to extremely low values indicating normal to excess numbers of OKT8+ cells. The rheumatoid synovium in patients with RA is characterized by the presence of lymphoid aggregates in which HLA-DR+ interdigitating cells form close contacts with a large number of OKT4+ T cells, while the OKT8+ T cells are sparse. It may be that the localizing mechanisms for OKT8+ suppressor cells in lymphoid aggregates are deficient in RA and the cells are thus permitted to pass into synovial exudates, resulting in an increase of OKT8+ cells in the SF.

Biochemical changes in humans upon exposure to sulfuric acid aerosol and exercise.

This study evaluated the effect of a single exposure to sulfuric acid aerosol on six human biochemical blood parameters. A total of 18 human subjects were exposed to ambient air for 4 hr on the first day of exposure and to 100 microgram/m3 (0.033 microM) sulfuric acid aerosol (0.5 micrometer mean mass diameter) for 4 hr on the second day. A total of 17 human subjects were exposed for 4 hr to ambient air on both exposure days. On each exposure day, at 30 min and 90 min after exposure was initiated, all subjects engaged in a 15-min exercise period (walking 4 mph on a treadmill inclined at 10 degrees). Six biochemical blood parameters were measured before and after exposure: glutathione, lysozyme, glutathione reductase, serum glutamic oxaloacetic acid transaminase, serum vitamin E, and 2, 3-diphosphoglyceric acid. The results indicate no significant effect of one 4-hr exposure of humns to 100 microgram/m3 sulfuric acid aerosol. One significant post-exposure effect did occur--an increase in glutathione reductase--for both the control and acid-exposed group. It is concluded that at the levels studied, sulfuric acid aerosol is neutralized in the upper airways; consequently, no cellular damage is measurable.

Effect of die spacers on precementation space of complete-coverage restorations.

The authors measured the precementation space achieved in vivo with varying layers of die spacer. Two hundred and ninety-one castings were fabricated in the Advanced Education Program in Prosthodontics at New York University College of Dentistry, New York, New York. Each of four clinician groups applied one layer of Taub Die Hardener and from one to four layers of die spacer to within 0.5 to 1.0 mm of the dies' finishing line. The resultant castings were adjusted using GC Fit-Checker. After final seating of the casting, the Fit-Checker was removed, sectioned buccolingually, and measured using a micrometer at three locations (midocclusally, midaxially, and on the bevel) for a total of 657 readings. Only after data collection were the specimens identified by group. Analysis of variance demonstrated a significance for group and location. Post hoc one-way analysis of variance and Sheffe tests (P < .1) demonstrated that: (1) one or two layers of die spacer demonstrated smaller precementation spaces than three or four layers; (2) the midocclusal precementation space was greater than the midaxial precementation space; (3) on the bevel area where no die spacer was used, the most consistent and minimal precementation space was measured.
