[Cellular immunity in sarcoidosis].

The characteristics of the circulating lymphocytes of 24 patients suffering from sarcoidosis were studied. The surface markers and the in vitro lymphocytic activity, both spontaneous and after stimulation with PHA, were assessed. Each culture was carried out in both homologous and autologous plasma. The stimulating ability of the plasma of sarcoidosis patients on normal lymphocytes was also evaluated. The patients were divided into two groups, i.e. acute sarcoidosis and sarcoidosis in remission. From this study it can be stated that depression of T-dependent immunity is particularly evident in patients with active disease. This depression seems to be mainly due to the presence in the plasms of factors which inhibit the cellular immune response.

Mitogenic action of neuraminidase.

A new effect of NCV on lymphocytes is demonstrated. This property is the capacity to act as a mitogen in and of itself. The possible mechanisms of this phenomenon are discussed.

Spontaneous lymphocyte transformation in chronic lymphocytic leukaemia.

In ten patients with CLL and in eight controls T lymphocytes were separated out by the rosetting method. The in vitro reactivity of these cells and of unseparated lymphocytes was studied in cultures without stimulants. A net increase of spontaneous lymphocyte transformation was found in the populations enriched with T cells in patients with CLL with respect to analogous populations in the controls. This increase is interpreted as a cell-mediated immunological reaction and due to T-cell sensitization towards the neoplastic B cells.

[Study of cellular immunology in malignant and benign monoclonal gammapathies].

In 25 cases of myeloma and 15 cases of benign paraproteinemia the functional situation of circulating lymphocytes was studied. The technique used was the incorporation of 3H-thymidine after PHA stimulation. E rosette formation and immunoglobulin bearing cells were evaluated. A reduction of 3H-thymidine incorporation in the blastic cells was seen in the patients with myeloma (the difference was statistically significant: p less than 0.001). In benign monoclonal gammopathies the reduction was moderate and was not statistically significant. The different blastic response in vitro in the two group of patients tested confirms that the respective immunocompetent systems clearly differ.

Peripheral lymphocytes in paraproteinemia.

In 22 cases of myeloma and 12 cases of benign paraproteinemia the functional situation of circulating lymphocytes was studied. The technique used was the incorporation of 3H-Thymidine after PHA stimulation. E rosette formation and immunoglobulin bearing cells were evaluated. A reduction of 3H-Thymidine incorporation in the blastic cells was seen in the patients with myeloma (2,942 +/- 421 cpm/10(6) lymphocytes in respect to 13,182 +/- 920 of the control group). The difference was statistically significant: p less than 0.001. In benign monoclonal gammopathies the reduction was moderate and was at the limits of the significance: 9,482 +/- 1,100. The percentage of E rosettes and of immunoglobulins bearing cells was normal. The different behaviour of the blastic response in vitro in the two groups of patients tested confirms that the respective condition of the immunocompetent system is clearly different. A possible interpretation of this type of behaviour is discussed.

[Stimulating effect of B-lymphocytes in mixed cultures on autologous T-lymphocytes in chronic lymphatic leukemia].

Two different cellular populations among the circulating lymphocytes were separated by using a technique based on the T lymphocytes capacity to form nonimmune rosettes with sheep red blood cells. The first population (P) is essentially composed of T lymphocytes, the second (population S) is made up mainly of type B lymphocytes. This technique was applied in the study of circulating lymphocytes in 11 patients with chronic lymphocytic leukaemia and was satisfactory both for good reproducibility and for its ability to obtain vital separate lymphocytes. Normal reactivity to PHA and PWM of population P and normal capacity to form E rosettes with SRBC were found. In mixed culture of T lymphocytes stimulated by B cells of the same patient significant blastic response was observed.

Immunobiology of primary neoplasms of the C.N.S. Preliminary results.

A preliminary report on the immunological monitoring of primary intracranial neoplasms is presented. The most important findings concerning B-cells and T-cells mediated immunity and delayed hypersensitivity reactions in this kind of patients, are discussed. Among the fairly significant immunobiological pictures, the Authors stress the "normal behaviour" of the B-cells dependent pool, the undebatable depression of the delayed hypersensitivity reactions, and the noticeable failure of the T-cell dependent pool, particularly marked in malignant gliomas, which are also characterized by the presence of cells with natural cytotoxic activity.
