Work in pigs has shown that malignant hyperpyrexia during anaesthesia may occur without suxamethonium having been given. A virtually constant feature in reported cases and in our own observations is that all subjects developing hyperpyrexia had received nitrous oxide and halothane.

The influence of vitamin C on the pharmacokinetics of antipyrine was investigated in eleven elderly men aged 65-74. Antipyrine (15 mg kg-1) was administered intravenously on three separate occasions over a 7-week period: (a) before dietary vitamin C restriction, (b) after approximately 5 weeks of dietary vitamin C restriction, and (c) after 2 weeks of vitamin C supplementation (500 mg orally twice daily). The mean plasma and leucocyte vitamin C levels (+/- S.D.) before vitamin C restriction were 1.26 +/- 0.31 mg dl-1 and 26.6 +/- 6.7 micrograms 10(-8) leucocytes, respectively. These values decreased and then increased significantly following vitamin C restriction and supplementation, respectively. The mean plasma half-life of antipyrine was 10.2 h and the mean plasma clearance was 2.561 h-1 1.73 m-2 before vitamin C restriction. No significant changes in the clearance, volume of distribution, or half-life of antipyrine occurred during the study, indicating that short-term alterations in vitamin C intake do not affect the pharmacokinetics of antipyrine in elderly males.
The total body clearance (CL), renal clearance (CLR), and nonrenal clearance (CLNR) of caffeine from plasma were determined following the intravenous administration of caffeine (4 mg kg-1) to ten healthy men (aged 66-86 years) on three separate occasions. Positive correlations were observed between CL and urine flow rate (UFR), between CLR and UFR, and between CLNR and UFR (r = 0.8947, p = 0.0002; r = 0.8832, p = 0.0003; and r = 0.8920, p = 0.0002, respectively). Previous studies have established similar relationships between CLR and UFR for caffeine and its initial dimethylxanthine metabolites; theophylline, theobromine, and paraxanthine. A relationship between CL and UFR has not been reported previously.
Growth, estimated composition of weight gained, and stool patterns of term infants who were fed either a whey-predominant formula or a casein-predominant formula in a random design and of breast-fed infants were compared. All infants (N = 111) were healthy, singleton products of uncomplicated pregnancies. Birth weights and other anthropometric measures in the first few days of life were not different among the three feeding groups. Formula or breast milk was the infants' principal source of energy from birth to age 16 weeks. Average energy intakes of formula-fed infants and change of intakes with age were similar in both groups at all ages. Feeding groups were not significantly different at any age in weight, length, weight or length gain, head circumference, skinfold measurements, upper arm fat area and muscle area, or estimated total body fat. Stools of infants on the whey-predominant formula differed from both the breast-fed and casein-predominant formula groups.
Fluroxene is highly toxic to several animal species. This toxicity is enhanced by induction of raised levels of hepatic microsomal enzymes. Experiments in rats are described which seek to assess the rleative contribution to this toxicity of the individual component groups of the fluroxene molecule. Though results point to the trifluoroethyl moiety of fluroxene as that aspect of the molecule most responsible for the observed mortality, reduction of the vinyl group modifies the pattern of liver injury. That the liver necrosis, manifest following fluroxene anesthesia in the presence of microsomal induction, is alone the direct cause of the acute death of experimental animals is questioned.
