Chemistry of the sulfate groups in a sulfated glycoprotein from rabbit submandibular gland.

A 35S-labeled sulfated glycoprotein was isolated from rabbit submandibular glands. Acid stability studies on the 35sulfate groupings present in the intact glycoprotein gave a half-life of 45 min. Partial acid hydrolysis of the 35S-labeled glycoprotein in 0.1 M HC1 for 90 min at 100 degrees C liberated a radioactive fraction which was free from peptide and fractionated in the monosaccharide range of a Sephadex G-15 column. Examination of this fraction by paper chromatography revealed the presence of a major component having the characteristics of N-acetylglucosamine 6-0-sulfate and a minor component having the properties of N-acetylgalactosamine 6-0-sulfate. The presence of ester sulfate groups in the intact glycoprotein was confirmed by infrared spectroscopy.

Hyaluronic acid in supragingival dental calculus.

A method is described for the isolation of heteropolysaccharides from human supragingival calculus. One component was identified as hyaluronic acid, by electrophoretic mobility, testicular hyaluronidase digestion and cetylpyridinium chloride profiles. No sulphated glycosaminoglycans were detected.

Glycosaminoglycans of human dental pulp.

Dental pulps were obtained from sound human permanent molars and premolars. The pulps were treated to remove lipid, protein and nucleic acid and glycosaminoglycans were finally isolated by ethanol precipitation. Electrophoretic examination and CPC-cellulose profiles revealed the presence of hyaluronic acid, dermatan sulphate, chondroitin 4 sulphate and chondroitin 6 sulphate. Hyaluronic acid accounted for only a low percentage of the total glycosaminoglycan content while dermatan sulphate was present in unusually high amounts. Chondroitin 4 and 6 sulphates represented the major glycosaminoglycan fraction.

Immunochemical localization of the small leucine-rich proteoglycan lumican in human predentine and dentine.

The immunocalization of the small interstitial leucine-rich keratan sulphate proteoglycan lumican was studied in human teeth using a polyclonal antibody towards lumican and a standard indirect peroxidase technique. There was intense labelling for lumican in predentine, with immunopositivity also observed in and around the tubules, around the odontoblasts and in the pulp with a fibrillar distribution. The previous reported ability of lumican to control collagen fibrillogenesis and it putative function in collagen spacing suggest potential roles for lumican in predentine in relation to mineralization.

Levels of glycosaminoglycans in peri-implant sulcus fluid as a means of monitoring bone response to endosseous dental implants.

The presence of certain glycosaminoglycans in peri-implant sulcus fluid may be an effective means of monitoring changes in bone metabolic activity following initial loading of implant abutments. This study has investigated levels of chondroitin 4 sulphate and hyaluronan in peri-implant sulcus fluid from titanium osseointegrated implants following initial abutment placement and exposure to masticatory stresses. Abutments were placed after a 3-month osseointegration period post-initial surgical placement of the interosseous stage. 10 edentulous patients, each with 5 mandibular implants were reviewed at 2, 4, 6 and 8 days after abutment placement. Clinical details were assessed and recorded and sulcus fluid collected in microcapillary tubes for a 5-min period for each abutment. Levels of glycosaminoglycans were assessed using cellulose acetate electrophoresis and densitometric scanning of alcian blue stained strips against known glycosaminoglycan standards. Maximum levels of sulcus fluid (0.3-1.25 microliters/5 min) were evident at 4 days with a decrease towards 8 days. Levels of sulphated glycosaminoglycans were also maximal at 2-4 days (range 0.03 - 0.126 micrograms/5 min) and decreased at 6-8 days. Hyaluronan was detected within a similar range of values reaching maximal levels at 4 days and decreasing by 8 days. The results indicate that glycosaminoglycan levels of peri-implant sulcus fluid is an effective means of measuring and maintaining changes in bone metabolism. The absence of proteodermatan sulphate precludes soft tissues being a source of these markers.
