Evoked release of [14C]norepinephrine from the rat hypothalamus during feeding.

The pattern of catecholamine release was studied at sites adjacent to the lateral ventricle or in the anterior, dorsomedial or ventromedial hypothalamus of the rat as it was feeding. Endogenous stores of norepinephrine (NE) were first labeled by the microinjection of [14C]NE into these circumscribed sites. Subsequently, [14C]NE release was examined by repeated perfusions of an artificial cerebrospinal fluid through a push-pull cannula at the rate of 23 mul/min for 5-10 min every 30 min. After successive control perfusates were collected, food or water was given to the animal. During an interval of feeding, a significant efflux of [14C]NE and its metabolites occurred reliably from midline structures of the hypothalamus at the level of the ventromedial nucleus. Although the feeding-related output of [14C]NE detected within the anterior hypothalamus was lower, [14C]NE was also released from this region when the rat pressed a lever to obtain food pellets. These results support the view that endogenous catecholamines underlie, at least partially, diencephalic mechanisms controlling food intake, including sensory, motor, or motivational components.

Cyclo (Leu-Gly) + haloperidol: effects on dopamine receptors and conditioned avoidance responding.

Behavioral effects of cyclo (Leu-Gly) (cLG), administered either acutely or chronically, were assessed in combination with haloperidol in the rat. cLG administered chronically, produced a significant reduction in the increase in apomorphine-induced stereotypy produced by chronic haloperidol infusion. On the other hand, the same dose of cLG which reduced this induction of dopamine receptor supersensitivity due to chronic haloperidol treatment, failed to produce a change in the potency of haloperidol in blocking conditioned avoidance responding in the rat. Furthermore, degeneration-induced supersensitivity of dopamine neurons, produced by unilateral destruction of the nigrostriatal pathway, was not reduced by acute or chronic treatment with cLG as measured by apomorphine-induced rotation. These data suggest that cLG may decrease motor system side effects thought to be caused by chronic antipsychotic administration without affecting the therapeutic efficacy of the antipsychotic agent.

Isolation of the cembranolide diterpenes dihydrosinularin and 11-epi-sinulariolide from the marine mollusk Planaxis sulcatus.

Marine mollusks have been identified as a rich source of novel compounds. The diversity of the chemical nature of these compounds can be partially attributed to the dietary sources inasmuch as mollusks are known to concentrate selectively compounds present in their diet. Although cembranolide diterpenses have been isolated in ever-growing numbers from a variety of marine as well as terrestrial sources, there is only one report about their presence in a marine mollusk. In this communication, we report the isolation of dihydrosinularin and 11-epi-sinulariolide from the mollusk Planaxis sulcatus Born. Dihydrosinularin and 11-epi-sinulariolide acetate have been isolated previously from soft corals which, together with gorgonians, are the richest sources of marine cembranoids. It should be pointed out that this is the first report on the occurrence of the parent 11-epi-sinulariolide in a marine invertebrate. It is expected these diterpenes are of dietary origin although the possibility of biotransformation of a precusor by the mollusk cannot be ruled out at this time. Both dihydrosinularin and 11-epi-sinulariolide have been reported to have marginal antineoplastic activity against P-388 lymphocytic leukemia.

Controlling nitrate leaching in irrigated agriculture.

The impact of improved irrigation and nutrient practices on ground water quality was assessed at the Nebraska Management System Evaluation Area using ground water quality data collected from 16 depths at 31 strategically located multilevel samplers three times annually from 1991 to 1996. The site was sectioned into four 13.4-ha management fields: (i) a conventional furrow-irrigated corn (Zea mays L.) field; (ii) a surge-irrigated corn field, which received 60% less water and 31% less N fertilizer than the conventional field; (iii) a center pivot-irrigated corn field, which received 66% less water and 37% less N fertilizer than the conventional field; and (iv) a center pivot-irrigated alfalfa (Medicago sativa L.) field. Dating (3H/3He) indicated that the uppermost ground water was <1 to 2 yr old and that the aquifer water was stratified with the deepest water approximately 20 yr old. Recharge during the wet growing season in 1993 reduced the average NO3-N concentration in the top 3 m 20 mg L(-1), effectively diluting and replacing the NO3-contaminated water. Nitrate concentrations in the shallow zone of the aquifer increased with depth to water. Beneath the conventional and surge-irrigated fields, shallow ground water concentrations returned to the initial 30 mg NO3-N L(-1) level by fall 1995; however, beneath the center pivot-irrigated corn field, concentrations remained at approximately 13 mg NO3-N L(-1) until fall 1996. A combination of sprinkler irrigation and N fertigation significantly reduced N leaching with only minor reductions (6%) in crop yield.

Radiocarbon 14C differentiation of sparkling and carbonated wines.

Specific 14C-activities, percent of modern 14C-activity, and calculated percent of fermentation CO2 are presented for CO2 contained in commercial sparkling wines, labeled as champagne or produced by the bulk (charmat) process. These data are given for the production years 1976-1982. The survey encompassed effervescent wines produced in Spain, Italy, West Germany, California, and New York. Addition of synthetic CO2 to approximately 40 samples represented as sparkling wines was indicated by low 14C-activities of CO2 in these wines. Data for 14C-activity were also presented for the ethanol distilled from sparkling wines for the years 1977-1980. In all cases, the 14C-activity of ethanol was appropriate to the year of vintage.
