Structure and expression of oncogenes in surgical specimens of human breast carcinomas.

We have performed an analysis of ras, c-myc, c-myb, c-erbB1 and c-erbB2 oncogenes in 100 surgical samples of human breast carcinomas. No point mutations have been detected at the 12th codon of c-Ha-ras and c-Ki-ras in 40 and 65 breast cancer DNAs, respectively. One out of 65 samples showed a 50-fold amplification of c-Ha-ras that, however, was not overexpressed. Alterations in the structure of c-myc, c-myb c-erbB1 and c-erbB2 oncogenes were sporadically observed. In 20 tumour samples, the study of expression of a series of oncogenes revealed that c-Ha-ras was the predominantly transcribed gene among the ras gene family whereas c-fos appeared the most constantly and significantly expressed nuclear oncogene.

Activated c-K-ras and c-N-ras oncogenes in 3-methylcholanthrene-induced BALB/c fibrosarcomas.

DNAs from fourteen fibrosarcomas induced by 3-methylcholanthrene (MCA) in BALB/c mice were analyzed for the presence of transforming oncogenes following transfection on NIH3T3 cells. Six transfection-positive DNAs contained an activated ras gene: four c-K-ras and two c-N-ras. These results demonstrate that c-K-ras is not the only oncogene of the ras family activated in MCA-induced murine fibrosarcomas as was previously indicated.

Characterization of papillomavirus polypeptides from bovine cutaneous fibropapillomas.

Virions of bovine papilloma virus (BPV) were isolated from a pool of cutaneous bovine fibropapillomas and purified by CsCl gradient centrifugation. SDS-PAGE revealed several polypeptides with an Mr ranging from 76K to 19K. Western blot analysis of the viral isolate identified additional polypeptides when a rabbit anti-BPV serum was used, but only the main capsid component of 57K when a rabbit antiserum raised against human papillomavirus was used. The viral preparation was then 125I-labelled and further purified by gel filtration. SDS-PAGE of immunoprecipitates of the anti-BPV serum with different fractions from the chromatographic column revealed the polypeptides of 76K, 57K and 28K to be viral structural components. The 28K polypeptide, not previously characterized, was shown to be composed of several molecular forms, migrating over a pH range of 3.5 to 4.6 when analysed by two-dimensional PAGE. Following SDS-PAGE performed under non-reducing conditions, the 28K and 76K polypeptides and the main capsid component of 57K appeared to be linked by disulphide bridges to form hetero- or homopolymers.

Modulation of the human Harvey-ras oncogene expression by DNA methylation.

A clone of the human transforming Ha-ras oncogene (pT24-C3) was methylated "in vitro" at its HpaII and HhaI sites and co-transfected with a selectable marker into NIH/3T3 cells. In the resulting colonies the normal or transformed morphology correlated respectively with a methylated or unmethylated status of the Ha-ras 1 promoter. A transfected, morphologically normal NIH/3T3 colony treated with 5'-azacytidine acquired a transformed phenotype growing in agar and in nude mice and showed demethylation of the promoter region of Ha-ras 1 gene. The cells of the reactivated colony produced human Ha-ras 1 mRNA and p21, and their DNA displayed transforming activity for NIH/3T3 recipient cells that was due to the original human Ha-ras 1 oncogene. We conclude that the methylation status of the promoter region of the human Ha-ras 1 modulates the expression and consequently the transforming activity of the oncogene.

CD1c but neither CD1a nor CD1b molecules are expressed on normal, activated, and malignant human B cells: identification of a new B-cell subset.

The CD1 cluster of monoclonal antibodies (MoAbs) CD1a, CD1b, and CD1c, identifies molecules that are differentially expressed on hematopoietic and nonhematopoietic tissues. Our earlier finding that the mantle zone (MZ) but not the germinal center (GC) of normal lymph nodes (LN) is CD1c+, CD1a-, and CD1b- prompted us to further investigate the expression of these molecules on normal, activated, and malignant B cells. We report that blood and spleen contain CD1c+ B cells that account for 49% +/- 20.4% (mean +/- SD) and 50.9% +/- 4.4% of the total B cell population, respectively. CD1a- and CD1b-specific MoAbs are unreactive with both B and T cells; these latter are CD1c- as well. When CD1c+ and CD1c- B cells are activated in vitro, the CD1c molecule is upregulated in the former subset and induced de novo in the latter. Conversely, activated blood T cells remain CD1c-. Neither CD1a nor CD1b molecules are detected on activated T and B lymphocytes. At ultrastructural level, the CD1c+ B cells exhibit distinctive features, namely, condensed chromatin with or without a nucleolus and a unique cluster of cytoplasmic vesicles and organelles; the number of nucleolated cells is higher in the spleen (95%) than in the tonsil (40%) or blood (5%). These findings further confirm the similarity between blood and MZ B cells. The CD1c expression assessed on 27 B-cell chronic lymphocytic leukemias (B-CLL) and 46 B non-Hodgkin's lymphomas (B-NHL) was detected on 41% and 32% of cases, respectively; the latter comprised four follicular and 11 diffuse histotypes. The Burkitt's lymphomas were CD1c-negative. The B-cell neoplasms were all CD1a- and, except for four with a weak cytoplasmic staining, all CD1b- as well. The clear-cut CD1c distribution in normal LN (MZ+, GC-) contrasted with the evidence that some B-NHL cells of GC origin (eg, follicular with predominantly small cleaved cells) were CD1c+. Overall, the finding that CD1c expression is restricted to a fraction of B cells present in lymphoid organs and in peripheral blood indicates that CD1c is a powerful marker for the identification and dissection of B-cell subsets whose functional properties can now be evaluated.

Early lymphocyte activation molecule defined by the monoclonal antibody MLR-3: biochemical and functional studies.

The MLR-3 monoclonal antibody reacts with activated but not with resting lymphocytes. We report that MLR-3 identifies an early activation molecule since its binding is detectable on T cells 1.5-2 hr after in vitro activation. Its expression, therefore, does not require DNA synthesis and precedes, by many hours, that of the receptors for interleukin-2 (IL-2R) and transferrin (TF-R). The MLR-3 antigen is also found on activated thymocytes (including the large early thymic CD3- subset) and B cells. The majority of T- and B-lymphoblastoid cell lines, as well as the myeloid and erythroid cell lines HL60, GM1 and K562, are MLR-3+; conversely, non-haemopoietic cell lines are MLR-3 negative. Seventy percent of B-cell chronic lymphocytic leukaemia and 15% of B non-Hodgkin's lymphomas (B-NHL) are MLR-3+. On tissue sections MLR-3 is reactive with epithelia, sweat glands, hair follicles and Henle's loop but not with vessels, connective, endothelium and many other tissues. In vitro studies show that MLR-3 (1-100 micrograms/ml) significantly alters the thymidine uptake of mitogen-treated lymphocytes:augmentation is found when T and B cells are induced with TPA-Ionomycin and reduction when induced with phytohaemoagglutinin (PHA) or Staphylococcus aureus Cowan strain 1 (SAC), respectively. On SDS-PAGE, MLR-3 immunoprecipitates a disulphide-linked heterodimer of MW 29,000-35,000: both subunits are glycosylated, phosphorylated and exhibit a pI of 4.1 and 5.0, respectively. Our data, particularly the in vitro results, suggest that the MRL-3 molecule could have an important role in the early hours of activation for the progression of resting lymphocytes into mitosis.

Carcinogenic effects of infantile and long-term 2,3,7,8-tetrachlorodibenzo-p-dioxin treatment in the mouse.

An infantile carcinogenesis assay was carried out with 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) injections administered intraperitoneally at 0, 1, 30 and 60 micrograms/kg b.w. doses to (C57BL/6J X C3Hf)F1 (B6C3) and to (C57BL/6J X BALB/c)F1 (B6C) mice, starting from the 10th day of life, once weekly repeated 5 times. Animals were then observed until 78 weeks of age. The induction of thymic lymphomas was related to treatment at 60 micrograms/kg dose level in both sexes of both hybrids, and at 30 micrograms/kg dose level in both sexes of B6C mice and in male but not female B6C3 mice. The incidence of hepatocellular adenomas was increased by TCDD treatment at 60 micrograms/kg dose level in B6C3 of both sexes but not in B6C mice. Hepatocellular carcinomas were seen at increased incidence at 30 and 60 micrograms/kg doses in B6C3 males but not in B6C3 females or in B6C mice of both sexes. The incidence of other tumor types was not related to treatment in both hybrids. A long-term carcinogenesis bioassay with TCDD was carried out in B6C3 mice treated by gavage at 0, 2.5 and 5.0 micrograms/kg b.w. doses from 6 weeks of age, once weekly for 52 weeks. The animals were observed until 110 weeks of age. An increased incidence of hepatocellular adenomas and carcinomas was related to treatment, at both doses and in both sexes. The incidence of other tumor types was uniformly low in treated and control groups, without any association with treatment, in both sexes.

Factors influencing the expression of endogenous retrovirus-related sequences in the liver of B6C3 mice.

The expression of RNA transcripts from three families of endogenous retrovirus-related sequences was investigated during liver cell proliferation in B6C3 mice. Treatment with a single dose of the liver mitogen and promoter of mouse hepatocarcinogenesis 1, 4-bis[2-(3,5-dichloropyridyloxy)]benzene (TCPOBOP), or with carbon tetrachloride (CCl4), induced liver cell proliferation at days 2 and 3 after treatment. Both of these treatments led to a marked increase in Moloney murine leukemia virus-related 6 kilobase RNAs, which were most abundant at day 1 after TCPOBOP treatment and at day 2 after CCl4. Intracisternal A particle-related 6 kilobase RNAs were markedly increased at days 1 and 2 after TCPOBOP and at days 1, 2, and 3 after CCl4. VL30-related transcripts were slightly decreased after TCPOBOP, but they were markedly increased at days 1 and 2 following CCl4. The livers of 15-day-old untreated mice contained about a 3-fold higher level of Moloney murine leukemia virus-related RNAs than adult liver. Intracisternal A particle-related 6-kilobase transcripts were present at 3-fold higher abundance in 7-day-old than in 15-day-old or adult liver. RNAs homologous to VL30 were detected at about the same levels in infant as well as adult livers. Inhibition of protein synthesis by the administration of cycloheximide to adult mice caused a marked increase in the amount of Moloney murine leukemia virus-, intracisternal A particle-, and VL30-related RNAs in the livers of the treated mice, suggesting the existence of labile proteins that normally regulate the abundance of these transcripts. We postulate that the amounts of these putative regulatory proteins vary during both normal development and carcinogenesis and also in response to specific agents that induce liver cell proliferation.

DNA methylation affecting the transforming activity of the human Ha-ras oncogene.

A plasmid containing the transforming Ha-ras gene and designated pT24-C3 was methylated in vitro using the sequence-specific bacterial methyltransferases HpaII and HhaI. Aliquots of the plasmid were methylated by the single enzymes or by the two enzymes simultaneously (double methylation). The transforming activity of the treated plasmids was assayed in the standard transfection assay on NIH-3T3 cells. Double methylation reduced the transforming activity of pT24-C3 about 80%, whereas treatment with the single methylating enzymes did not significantly affect the oncogene activity. Southern blot analysis of the transformants obtained with the methylated or mock-methylated pT24-C3 plasmids indicated in all the examined DNAs the presence of human Ha-ras sequences with methylation degrees consistent with the treatment of the plasmids. The Mr 21,000 oncogene protein p21 was also detected in several examined transformants. The DNA-demethylating agent 5-azacytidine restored the transforming activity of the double-methylated pT24-C3 upon 24 h incubation of transfected NIH-3T3 cells. Southern blot analysis showed integration of human Ha-ras with a methylation profile intermediate between the double-methylated and mock-methylated plasmids. It is suggested that DNA methylation of specific CG-containing target sites can affect the transforming activity of a human oncogene.

Genetic susceptibility to murine hepatocarcinogenesis is associated with high growth rate of NDEA-initiated hepatocytes.

The murine hybrids (C57BL/6J X C3Hf)F1 (B6C3) and (C57BL/6J X BALB/c)F1 (B6C), which have a high and low spontaneous and induced incidence of hepatocellular tumors, respectively, were treated with a single dose of NDEA at 1 week of age followed by TCPOBOP, a phenobarbital-like promoter of liver carcinogenesis, or by vehicle, and sacrificed at 30 weeks of age. The frequency per liver of hepatocellular nodules was similar in the two hybrids. However, in male mice the mean volume of nodules was about 10-fold greater in B6C3 than in B6C mice receiving NDEA followed by vehicle, and the treatment with TCPOBOP after NDEA stimulated nodule growth, with a much greater response in B6C3 mice. In female mice no differences in the mean volume of nodules were seen between hybrids after NDEA and vehicle, whereas upon NDEA and TCPOBOP treatment the mean volume of nodules was 25-fold greater in B6C3 than in B6C females. In addition, a few hepatocellular adenomas and carcinomas were observed, mostly in animals treated with NDEA and TCPOBOP, and they were 3-fold more numerous among B6C3 than B6C mice. TCPOBOP alone induced the same biochemical and hyperplastic effects in the liver of both hybrids. Using DNA probes homologous to Moloney murine leukemia virus, intracisternal A particle and virus-like 30S sequences, no correlation was apparent between the expression of any of these endogenous retroviral families and the strain susceptibility to hepatocarcinogenesis. We hypothesize that the different susceptibility to hepatocarcinogenesis between B6C3 and B6C mice is related to a higher growth rate of B6C3 than B6C initiated liver cells.

Effects of phenobarbital and 1,4-bis[2-(3,5-dichloropyridyloxy)]benzene on differentiated functions in mouse liver.

The promoters of murine hepatocarcinogenesis phenobarbital (PB) and 1,4-bis[2-(3,5-dichloropyridyloxy)]benzene (TCPOBOP) given to adult C3Hf female mice increased the content of total liver DNA by 1.6-1.8-fold each week after the beginning of treatment. Both compounds increased the aminopyrine-N-demethylase activity, decreased the glucose 6-phosphatase (G6Pase), alkaline phosphodiesterase I and alkaline phosphatase specific activities, but did not modify the gamma-glutamyltransferase levels. Both compounds decreased the abundance of tyrosine aminotransferase- and metallothionein I-related RNA transcripts. These findings confirmed the PB-like activity of TCPOBOP and showed that both chemicals had a pleiotropic effect on mouse liver, that was not limited to stimulation of drug metabolism, but also affected other hepatocyte functions.

Chromosomal abnormalities in a primary small cell lung cancer.

A cytogenetic analysis of a fresh primary tumor specimen of small cell lung cancer showed a del(3)(p14p23) in the majority of metaphases. Additional clonal changes were found in the karyotype. No abnormalities for Ha-ras, Ki-ras, N-ras, myb, or myc were detected by Southern blot analysis of the tumor DNA.

A cytogenetic, phenotypic, and molecular study of an immunoblastic lymphoma with a 14q + translocation.

An uncultured immunoblastic lymphoma, obtained from an untreated patient, was examined from a cytogenetic, immunophenotypic, and molecular viewpoint. The B-cell lineage, immunoglobulin light-chain type, and percentage of neoplastic cells were determined immunologically. Karyotyping showed the presence of a 14q + marker and suggested that the donor chromosome was chromosome 8. Southern-blot analysis of DNA from normal and lymphoma cells, using a molecular probes sequences related to the IGHJ and IGK immunoglobulin genes, confirmed the immunophenotype. A similar analysis, using probes homologous to IGHAC and MYC genes, showed that the t(8;14) detected by cytogenetic analysis resulted in a IGHAC-MYC rearrangement.

Two-stage liver carcinogenesis in the mouse.

A two-stage protocol for studying liver carcinogenesis was applied to the mouse. The protocol includes the treatment of 7-day-old mice with a single low dose of an initiating agent (diethylnitrosamine, NDEA), promotion starting after weaning and lasting about 20 weeks, and histologic analysis, at 30 weeks of age, of hepatocellular nodules on H&E stained sections. A stereologic analysis of results allows the evaluation of nodule frequency and size. Using this protocol in B6C3 and B6C mice, we have identified the promoting activity of the phenobarbital-like enzyme inducer, 1,4-bis[2-(3,5-dichloropyridyloxy)]benzene (TCPOBOP), which was found to be a strong hyperplaseogenic agent for mouse liver. These studies also indicated that the different susceptibility to hepatocarcinogenesis in B6C3 and B6C mice may be related to a higher susceptibility of B6C3 than B6C initiated liver cells to growth stimulation. A long-term study showed that B6C mice have a low incidence of spontaneous liver tumors but are susceptible to chemical hepatocarcinogenesis and, therefore, they may be an alternative model to B6C3 mice in carcinogenesis bioassays.

HRAS1 proto-oncogene polymorphisms in human malignant melanoma: TaqI defined alleles significantly associated with the disease.

We have analysed the DNA of peripheral blood leukocytes (PBL) from 55 melanoma patients and 53 healthy individuals and failed to find any significant association between melanoma and rare HRAS1 alleles defined by MspI/HpaII digestion. However, the analysis of the same DNAs for a different polymorphism based on the presence of additional TaqI sites in the variable tandem repeat region of HRAS1 showed that the total frequency of a group of allelic variants, named Tp, was significantly higher in melanoma patients than in normal donors.

Enhancing effects of carbon tetrachloride in mouse hepatocarcinogenesis.

The enhancing effects of 4 or 8 necrogenic doses of carbon tetrachloride (CCl4) on hepatocarcinogenesis were studied in female B6C3F1 mice previously treated with a single dose of N-nitroso-N-diethylamine (NDEA) at 7 days of age. The CCl4 treatments were given every other week and started either after weaning or at 18-26 weeks of age, and the animals were observed until 36 weeks of age. The number and size of hepatocellular nodules were then scored and analysed. Hepatocellular nodules were not observed in mice receiving either 4 or 8 CCl4 treatments without NDEA initiation. Both 4 and 8 CCl4 treatments promoted NDEA-initiated hepatocarcinogenesis, increasing nodule size but not nodule frequency. Eight CCl4 doses were more effective than 4 and gave the greatest increase in nodule size when administered from 18 to 32 weeks of age. In an additional experiment, 3 mice bearing hepatocellular nodules induced by a single infantile NDEA treatment were administered CCl4 once and killed 52 h later. Nine neoplastic nodules were examined and found to be resistant to the CCl4 induced necrosis, although cells in the nodules were actively dividing at the same rate as in the regenerating surrounding normal liver parenchyma. The ability of a few necrogenic CCl4 doses to enhance hepatocarcinogenesis may be related to a differential cytotoxicity on neoplastic and normal hepatocytes and to a similar mitogenic stimulus on both cell types.
