Nucleotide sequence of the bovine cyclic-AMP responsive DNA binding protein (CREB2) cDNA.

The bovine cyclic AMP responsive binding protein cDNA (CREB2) was isolated from a lambda-gt11 cDNA expression library using a 32P labelled oligonucleotide corresponding to the 21 bp enhancer sequence present in the BLV LTR. The deduced amino acid sequence revealed that CREB2 contains a leucine zipper structure (residue 295 to 316), a basic amino acid domain (residue 268 to 291) and several potential phosphorylation sites.

A Bayesian graphic method for predicting individual phenytoin dosage schedule.

A simple and practical graphic method based on Bayesian feedback theory for predicting individual phenytoin dosage was evaluated. Compared with the mathematical calculation, the graphic method only needs one pair of steady-state phenytoin concentration-dose data to predict individual phenytoin dosage. The observed drug levels were compared with predicted ones in 12 epileptic out-patients with a correlation coefficient of 0.94. The mean error between the observed and predicted phenytoin levels was 0.62 mg.L-1. The mean kinetic parameters Vm and Km obtained from the graph were 7.01 +/- SD 0.88 mg.kg-1.d-1 and 4.93 +/- 0.91 mg.L-1, respectively.

[Pharmacokinetic-pharmacodynamic model of magnesium sulfate in treatment of pregnancy-induced hypertension].

Ten pregnancy-induced hypertension gravidas aged 24-36 (28.1 +/- 3.31) yr were infused iv with fast (7.5 g in 1 h) and slow (7.5 g in 5 h) rates consecutively. Blood pressure were measured and blood samples were collected at 0, 1, 6, 7, 9, 11, 15 h. The drug concentration of each sample in the serum was determined by colorimetric method. The integrated pharmacokinetic and pharmacodynamic model was used to analyze the data. The serum data were fitted to a biexponential equation and the effect data were fitted to a sigmoid Emax model. Their pharmacodynamic parameters were: Kco = 0.52.h-1, T1/2(kco) = 1.33 h, Emax = 20.5% Cc (50) = 6.15 micrograms.ml-1), y = 3.64. kco was not equal to k21 of the mammillary model. It shows that the site of action is situated in the effect compartment. The predicted effect could last 24 h.

[Radionuclide bone scan using single-photon emission-computed tomography in detecting oral and maxillofacial tumor infiltrating to the jaws].

83 cases (including 53 cases of patients and 30 cases of normal volunteer) of SPECT bone scan had been studied for detecting oral and maxillofacial tumor infiltrating to the jaws. The results was contrasted with X-ray, operation and pathology, and indicated that SPECT bone scan could reveal the early tumor infiltrating to the jaws and accurate extent of tumor lesions, more accurate than X-ray. SPECT provided us more important information about the preservation of the jaw and extent of bone resection.

[Studies on warming the middle-jiao and analgesic effect of flos Caryophylli].

The effects of water extract and ether extract of Flos Caryophylli have been investigated by four experimental gastric ulcer models, choleresis in rats, two diarrhea models in mice, two pain stimulations in mice, and acute liver injury induced by CCl4 in rats. The results suggest that the action of Flos Caryophylli on dispelling the cold by warming the middle-jiao and curing abdominalgia is related to its anti-ulcer, choleresis increasing, anti-diarrhea and analgesic functions.

[The relation between results of thyrotropin-releasing hormone stimulation tests (TRH-ST), dexamethasone suppression test (DST) and urinary MHPG. SO4 excretion in depressive patients].

Thyrotropin-releasing hormone stimulation test (TRH-ST) was performed in 68 depressive patients, 25 normal subjects and 4 schizophrenics. Dexamethasone suppression test (DST) and urinary MHPG. SO4 determination was also administered in part of this depressive patients. Our data show that some of depressive patients suffered from the dysfunction of hypothalamic-pituitary-thyroid axis, and this dysfunction was not related to the dysfunction of hypothalamic-pituitary-adrenal axis and urinary MHPG. SO4 excretion. A blunted TSH response to TRH was a "STATE MARKER" for some of depressive patients.

Cooperation between bovine leukaemia virus transactivator protein and Ha-ras oncogene product in cellular transformation.

Human T-lymphotropic viruses (HTLV-I and -II) and bovine leukaemia virus (BLV) express transactivator proteins able to increase long terminal repeat (LTR) directed viral expression. These transacting factors are though to be involved in the induction of leukaemia by these viruses. Transfection of BLV transactivator p34tax together with Ha-ras immortalizes and transforms rat embryo fibroblasts, in vitro. The transformed cell induce tumours in nude mice. These data emphasize the causal role exerted by p34tax in in vivo tumorigenesis.

In vitro investigations on induction and reversal of cisplatin resistance in a rat ovarian tumor cell line.

Resistance of a rat ovarian tumor cell line (O-342/DDP) to cisplatin was induced in vitro by stepwise increase of cisplatin concentrations. Both chemosensitive parental cells (O-342) and resistant O-342/DDP cells grow in a monolayer and enter log-phase growth about 24 h after seeding (cell population doubling time in log-phase growth is about 24 h). O-342/DDP cells show a 33-fold resistance to cisplatin as compared to O-342 cells (ID50 = 33 microM in O-342/DDP vs 1 microM in O-342 cells). The intracellular total glutathione (GSH + GSSG) of O-342/DDP cells was twice as high as that of O-342 cells (3.04 vs 1.37 nmol/10(6) cells), while the intracellular GSSG was increased by 26% in O-342/DDP cells compared to O-342 cells. DNA interstrand crosslinks were found to be 8.5 times higher in O-342 cells than in O-342/DDP cells (204 vs 24 rad equiv.), following cisplatin treatment. DNA single-strand breaks were approximately doubled in the sensitive line as compared to the resistant line following exposure to cisplatin. Chromosome analysis uncovered a change in the karyotype of O-342/DDP cell as compared to O-342 cells. In the sensitive line hyperploid (3n) clones, in the resistant line near-diploid clones predominated. Both DL-buthionine-(S,R)-sulfoximine and 3-aminobenzamide were able to sensitize the resistant line towards cisplatin. Thus, the present results suggest that mechanisms for cisplatin resistance in this tumor line apparently are multi-factorial, and include a higher intracellular GSH content and an increased repair activity.

Enhancement of cisplatin (DDP) antitumor activity by 3-aminobenzamide in rat ovarian tumors sensitive and resistant to DDP in vivo.

A cisplatin (DDP)-resistant rat ovarian tumor cell line (O-342/DDP) and its parental sensitive counterpart (O-342) were used to investigate the combination effect of DDP plus 3-aminobenzamide (3-AB), an inhibitor of adenosine diphosphate ribose transferase (ADPRT). Treatment with six doses of DDP in NMRI nude mice bearing O-342/DDP produced an increase in mean survival of only 1 day over that of controls (P less than 0.05). The addition of nontoxic doses of 3-AB (5 mM/kg x 6) increased the mean survival to 6.4 days compared with that obtained with DDP treatment alone (P less than 0.001). In the sensitive ovarian tumor line (O-342), the combination effect of DDP plus 3-AB was even more impressive: simultaneous treatment of NMRI nude mice bearing O-342 with three doses of DDP plus 3-AB increased the mean survival by 2 weeks and the median survival by 3 weeks over that achieved with DDP treatment alone. Possible mechanisms involved in the potentiation of DDP activity and the possible clinical potential of this combination are discussed.

Estimates of dose to intraperitoneal micrometastases from alpha and beta emitters in radioimmunotherapy.

Intraperitoneal metastases from ovarian and other gynecologic tumors are a significant source of treatment failure. In recent years, investigators have used radiolabeled monoclonal antibodies to treat this disease with encouraging results. We have developed a dose calculational technique which generates isodose distributions from intraperitoneally administered alpha and beta particle emitters. In this study we apply the calculations to tissue biopsy samples to determine the adequacy of dose to ovarian micrometastases. Tissue samples from staging biopsies at the time of surgical debulking are scanned to identify small metastases. The patient population studied comprised those with ovarian disease who based on clinical criteria would be considered good candidates for intraperitoneal radioimmunotherapy. The regions of interest (which include the tumor and surface of the peritoneum) are digitized and tumor volumes are contoured. Dose calculations based on the modeling of intraperitoneally administered antibodies radiolabeled with various isotopes is performed and the minimum dose to tumor and normal tissue is assessed. For example, with tumor uptake of 0.1% injected dose per gram of tissue, the surface tumor dose from alpha emitters is up to 45,000 rads. The dose falls to 6000 rads at approximately 40 microns from the peritoneal surface. The surface dose from 20 mCi 90Y administered in 1500 ml saline is up to 10,000 rads, and at a 2-mm depth, approximately 2000 rads. From our calculation dose distribution from radioimmunotherapy varies as a function of physical characteristics of the isotope, absorption of activity, and amount of disease being treated.

HPLC procedure for the pharmacokinetic study of the spin-trapping agent, alpha-phenyl-N-tert-butyl nitrone (PBN).

Considerable progress has been made in the use of spin-trapping agents for the trapping of free radicals in biological systems. Radicals have been detected in both in vitro and in vivo systems using this methodology. Free radicals have not only been identified by this procedure, but also the intensity of radical generation and the duration of their production has been assessed as well. One of the most widely used spin-trapping agents in biological systems is PBN. This spin trap appears to be relatively nontoxic at the levels required for successful trapping experiments, but there is no information concerning the possible fate of PBN in such biological systems. Metabolism of PBN could alter the concentration of PBN at the site of trapping which may affect the efficiency of radical capture, especially in in vivo systems. In this study, PBN was administered intraperitoneally to rats and the concentration of the spin trap in various organs was determined by high pressure liquid chromatography as a function of time (15 min to 12 h). The concentration of PBN in plasma peaked at 15 min while the maximum in all organs tested occurred at 30 min. The time course of PBN concentrations in all tissues followed similar curves, and declined rather steeply after the 30-min maximum with a biological half-life of 134 min. However, the amount of PBN per gram of tissue was always higher in liver and kidney than in the brain, heart, and lung. PBN was detected in the urine for as long as 24 h after injection of the compound.

Transport and deamination of amino acids by a gram-positive, monensin-sensitive ruminal bacterium.

Strain F, a recently isolated ruminal bacterium, grew rapidly with glutamate or glutamine as an energy source in the presence but not the absence of Na. Monensin, a Na+/H+ antiporter, completely inhibited bacterial growth and significantly reduced ammonia production (85%), but 3,3',4',5-tetrachlorosalicylanide (a protonophore) and valinomycin had little effect on growth or ammonia production. Dicyclohexylcarbodiimide, a H(+)-ATPase, inhibitor had no effect. The kinetics of glutamate and glutamine transport were biphasic, showing unusually high rates at high substrate concentrations. On the basis of low substrate concentrations (less than 100 microM), the Km values for glutamate and glutamine were 4 and 11 microM, respectively. Strain F had separate carriers for glutamate and glutamine which could be driven by a chemical gradient of Na. An artificial delta psi was unable to drive transport even when Na was present. The glutamate carrier had a single binding site for Na with a Km of 21 mM; the glutamine carrier appeared to have more than one binding site, and the Km was 2.8 mM. Neither carrier could use Li instead of Na. Histidine and serine were also rapidly transported by Na-dependent systems, but serine alone did not allow growth even when Na was present. Because exponentially growing cells at pH 6.9 had little delta psi (-3 mV) and a slightly reversed Z delta pH (+17 mV), it appeared that the membrane bioenergetics of strain F were solely dependent on Na circulation.

[Chemical evaluation of vinegar-processing method for fresh rhizoma Corydalis].

Qualitative and quantitative analysis were carried out on the samples of Rhizoma Corydalis in both the locally vinegar-processed (fresh) and traditionally vinegar-processed forms. No difference was found in the main chemical constituents. However, both the total alkaloids and the corydalis B content of the former are higher than those of the latter.
